Effects of mixing and reciprocal inoculation with compatible and incompatible races of Pyricularia oryzae on the enlargement of disease lesions of rice blast (친화성 및 비친화성균계의 혼합 및 전후접종이 벼 도열병의 병반확대에 미치는 영향)

  • Effect on blast lesion development was investigated in rice cultivar Milyang 23 and Jinheung using two races of Pyricularia oryzae compatible or incompatible to each of two rice cultivars. Discase lesions were measured in rice when inoculated with inoculum mixed with compatible and incompatible races, inoculated with a compatible race following with an incompatible race and vice versa. Lesions were smaller when rice was incoulated with an incompatible race and then inoculated with a compatible race than those inoculated in the reverse way both at the inoculated site and at 3cm-upper part from inoculated spot. The results suggested that induced resistance might play a role in suppression of lesion development although such an effect appeared to be influenced by heavy application of nitrogen fertilizer.

Height Suppression of Cucumber and Tomato Plug Seedlings Using of Brushing Stimulus (브러싱 자극을 이용한 오이와 토마토 공정묘의 초장 억제)

  • This study aimed to evaluate the effect of height suppression of cucumber and tomato plug seedlings as affected by mechanical stimulus using brushing as environment-friendly method. Cucumber (Cucumis sativus L. 'Joeunbaekdadagi') and tomato (Solanum lycopersicum L. 'Mini Chal') seeds were sown in 40-cell plug trays ($54{\times}27.5{\times}5cm$) filled with growing medium on Oct. 9, 2017. The cultivation environment in a venlo-type glasshouse was maintained as cultivation temperature range of $15-25^{\circ}C$ and the relative humidity of $50{\pm}10%$. Nontreatment and diniconazole ($7.5mg{\cdot}L^{-1}$) application at 15 days after sowing were used as the control. In addition, brushing treatments in cucumber and tomato were applied interval of 2, 4 or 6 hrs for 15 and 20 days, respectively. Plant height, hypocotyl length, and internode length were inhibited for cucumber and tomato in the diniconazole treatment than in the control. The leaf size was reduced, both cucumber and tomato, while the SPAD increased under the diniconazole treatment. However, stem diameter of cucumber was the thickest in the 2 hrs brushing interval treatment. Fresh weights of shoot and root were the significantly lowest in the diniconazole treatment. Application of brushing improved seedlings quality by promoting dry weights of shoot and root, and compactness of tomato seedlings. The chlorophyll fluorescence of tomato seedlings drastically decreased with 2 hrs treatment, indicating that mechanical stress by brushing treatment. The relative growth rate of tomato seedlings was significantly lower in the diniconazole treatment, but cucumber seedlings were not significantly different in all treatments. As a results, height suppression of cucumber and tomato seedlings was best achievement in the diniconazole treatment by the chemical as growth regulator. In an environment-friendly point of view, however, it is considered that 2 hrs brushing interval treatment can be the applicability for replacing the chemical methods in plug seedling growth of cucumber and tomato.

The effect of environmental condition to the mycelial browning of Lentinula edodes (Berkeley) Sing. during sawdust bag cultivation (환경조건이 표고톱밥배지의 갈변에 미치는 영향)

  • Recently sawdust cultivation of Shiitake mushroom (Lentinula edodes ) is getting increased because log cultivation is getting difficult to get oak logs. It is important to make mycelia browning on the substrate surface in sawdust cultivation. This browned surface plays an important role like as artificial bark of the oak log, which protects the other pests and suppresses water evaporation in the substrate. The period for mycelia browning is so long that the sawdust cultivation of Shiitake mushroom can not spread well into the mushroom farms. In this article we would like to discuss about the effect of environmental condition to the mycelial browning during sawdust bag cultivation for the To reduce the period required for browning of substrates, sawdust substrates was illuminated light with difference intensity. One hundred Lux light illumination was needed for producing normal yield of fruit body but fruit body yield was low and abnormally shaped fruit body was produced when cultured under the dark condition of incubation. Illumination over 200lux is necessary for the successful browning of substrates during incubation. Optimum incubation temperature for browning of substrates and fruiting was $25^{\circ}C$. The treatment of cotton plug with different size to identify the effect of aeration on the browning of substrates and fruiting showed rapid mycelial growth and reduced the periods for browning as the size of cotton plug was bigger. However, yield of fruit body was the highest at 16mm diameter cotton plug as compared to 20mm of that. $CO_2$ content in vessel of substrates was low as the size of cotton plug was bigger during incubation. $CO_2$ content during incubation of substrate was highest in periods between 8 week and 14 week after inoculation of shiitake when substrate was changed color into brown. $C_2H_4$ content in vessel with substrates was highest at 8mm diameter cotton plug and it was increased by order of 12, 16, 20, 0, 4 mm diameter cotton plug during substrate incubation. Sawdust substrate was soaked in cold water for different time to identify soaking effect of sawdust substrate on fruit body yield and activities of enzymes in these substrates were investigated. The fruit body yield was increased up to 40% by soaking substrates in comparison with unsoaked substrates. The soaked substrates showed 165, 175g/1,000ml at treatment of 4 and 15 hours, respectively. Cellulose activities in soaked substrates were not changed with soaking time, but activities of laccase, lignin degradation enzyme, were drastically increased up to 4 times in comparison with unsoaked substrates.

Clinical Investigation of Cervical Tuberculous Lymphadenitis (결핵성 경부 임파선염의 임상적 고찰)

  • Background : Cervical tuberculous lymphadenitis(CTL) is one of the most common extra-pulmonary tuberculosis. Recently overall incidence of pulmonary tuberculosis has decreased, but the incidence of tuberculous lymphadenitis has not decreased. Its duration of treatment is still controversial and the pathogenesis, prognosis and relationship with other site tuberculous are poorly published. So we did a retrograde study of 120 cases of confirmed CTL about its clinical manifestations. Method : All patients were applied fine needle aspiration(FNA) of palpable enlarged cervical lymph nodes and 114 patients were examined for AFB smear and 34 patients for TB culture with aspirated fluid. 57 patients were examined Mantoux test(5TU with PPD-S). With above methods, a total of 120 patients was diagnosed as having CTL. Results : 1) CTL is most prevalent in young women between the age of 20~30 years and the incidence of CTL in female is 2.5 times higher than that of male. It is located most commonly in the posterior cervical area. The most common presenting symptom is painless palpable enlarged cervical lymph nodes. 2) With FNA of enlarged cervical lymph nodes, the percentage of histopathological positivity is 82.3%. The percentage of AFB smear positivity is 38.6%, and that of TB culture positivity is 17.6%(p<0.001). 3) Pulmonary tuberculosis is noted in 79 cases(65.8%). And 42 cases(53.5%) of them had minimal pulmonary tuberculosis. In 14 cases(11.7%), other extrapulmonary tuberculosis coexsisted and pleural tuberculosis was most Common in the order. 4) CTL was treated with anti-tuberculous medication(first line drug) and median treatment duration was 18.5months. During treatment, the size of involved lymph nodes decreased gradually in 62 cases(75.8%), newly developed lymph nodes were found in 25 cases(30.4%), fluctuation formation in 22 cases(26.8%) and fistula formation in 14 cases(17.0%). Conclusion : CTL is prevalent in women between the age of 20~40 years and it involves posterior cervical area most commonly. CTL is treated with long-term anti-tuberculous chemotherapy. We think it is one manifestation of systemic disease and frequently coexisting with pulmonary tuberculosis. Despite anti-tuberculous chemotherapy, the size of involved lymph nodes was increased, new lymph nodes were developed or fluctuation and fistula formed in involved lymph nodes. After sufficient medication, when the patient felt pressure discomfort from enlarged lymph node or fistula was formed, we recommended total excision of involved lymph nodes.

  • The pH of beverages is known to be low and have, therefore, been implicated in the increasing incidence of erosion. Erosion is believed to be the predominant cause of teeth wear in children and young adults, although there will always be a contribution from attrition and abrasion. The aim of the present study was to evaluate the effect of yogurt on the progression of erosive demineralization in human enamel using demineralization model in vitro. In 4 yogurts, available on the market, pH, buffering capacity and the concentrations of calcium, phosphate and fluoride were determined. The buffering effect was determined by titration with NaOH. 50 milliliters of each drink was then titrated with 1M sodium hydroxide, added in 0.5 milliliters increments, until the pH reached about 7. Human deciduous enamel(n=40) samples were divided into four groups and exposed to 80ml of the yogurt for 30,60, 90 and 120min. Enamel surface microhardness(VHN) was examined before and after each exposure. 1. The average PH of fermented milk was 3.77 and this pH value was acidic enough to cause tooth erosion. 2. All of the fermented milks were found to be erosive(p<0.05) 3. The teeth exposed to the fermented milk all showed erosion like lesions and microhardness measurements showed that enamel surface hardness decreased proportionately with increased time of immersion in all tooth specimen groups. 4. After immersion for 30 and 60 minutes, reduction rate of microhardness values was not significantly different between the groups(p>0.05). However, after 90 and 120 minutes, reduction rate of each group was significantly different(p<0.05).

Effects of Dietary Quartz Porphyry and Feed Stimulants, BAISM Supplementation on Growth Performance and Disease Resistance of juvenile eel Anguilla japonica (사료내 맥반석과 BAISM 복합첨가가 치어기 뱀장어 Anguilla japonica의 성장과 내병성에 미치는 영향)

  • This study investigated the synergistic effects of dietary supplementation of quartz porphyry(QP) and a laboratory developed feed stimulants, BAISM(BS) on growth performance and utilization as the additives for juvenile eel Anguilla japonica. Six isoenergetic experimental diets(18.2 kJ/g) were formulated to contain 50% crude protein, 15% lipid with or without dietary QP(Song-Gang stone, Davistone, Korea) and BS supplementation. QP and BS were provided at 0% in the control diet($Q_0B_0$) and at 0.7% QP+0% BS($Q_{0.7}B_0$), 0.7% QP+0.3% BS($Q_{0.7}B_{0.3}$), 0.7% QP+0.5% BS($Q_{0.7}B_{0.5}$), 0.7% QP+0.75% BS($Q_{0.7}B_{0.75}$) and 0.7% QP+1.0% BS($Q_{0.7}B_{1.0}$) in experimental diets on dry matter basis. After four weeks of adaptation, triplicate groups of 30 fish initially averaging $15{\pm}0.1g(mean{\pm}SD)$ were randomly distributed into each aquarium, and they were fed one of the experimental diets for 8 weeks. By the end of the feeding trial, weight gain(%), specific growth rate(%), feed efficiency(%) and protein efficiency ratio of fish fed diet $Q_{0.7}B_{0.5},\;Q_{0.7}B_{0.75}\;and\;Q_{0.7}B_{1.0}$, were significantly higher(P<0.05) than those of fish fed the other diets. But, $Q_{0.7}B_{0.5},\;Q_{0.7}B_{0.75}\;and\;Q_{0.7}B_{1.0}$ were no significant differences(P<0.05). In challenge test, fish were infected by intraperitoneal injection of 0.1 mL bacterial suspension with Edwardsiella tarda per fish after the feeding trial. As a result, fish fed QP and BS supplemented diets have a significantly higher cumulative survival rate than those of fish fed control diet(P<0.05). In conclusion, these results indicated that the optimum dietary supplementation level of QP and BS could be approximately 0.7% quartz porphyry+0.5% BAISM($Q_{0.7}B_{0.5}$) of diet based on WG, FER, SGR, PER, cumulative survival rate in juvenile eel A. japonica.

  • To detect proper lytic assay conditions of the crude zumolyase from Arthrobacter luteus, effets of the various factors involved in the lytic system of Sacchromyces sake cultured with shaking in the malt extracts medium were investigated. The results are summarized as follows : 1. The susceptibilities of viable cells of S. sake from logarithmic growth phase to the lytic enzmye were much greater than those of the cells in lag and stationary phases. The cells cultured for 18 hr were the most susceptible to the enzyme. 2. Lytic activity of the enzyme toward the viable cells of S. sake was very low. It was, however, enhanced 4 folds of more by the pretreatment of the cells with 0.05 M sodium sulfite. 3. Lytic activity of the enzyme toward commercial baker's yeast cells was negligible, and the effect of sodium sulfite on the lysis of the cells also was nothing but a little. 4. The lyophilized cells of the baker's yeast showed more susceptibility to the lytic enzyme than viable cells of the yeast. No definite effect of sodium sulfite on the lysis of the lyophilized cells, however, was observed either baker's yeast of S. sake. 5. It appeared that the relationship between the reaction rate and the enzyme concentration on the lysis of the yeast cell walls followed enzyme kinetic theory, but one between the reaction rate and concentration of the yeast cells as substrates showed different pattern from that in enzyme kinetic theory. 6. After the preparation of crude zymolyase was kept at $7^[\circ}C$ for 10 days in the 0.05 M phosphate buffer, pH 7.5, the remainning lytic activity was about 80 %.

The Effect of Ishige okamurae Extracts on Antioxidant Activity and Serum Lipid Content in Ovariectomized Rats (패 추출물의 항산화 활성 및 난소절제 흰쥐의 혈중 지질 함량 변화에 미치는 영향)

  • This study investigated antioxidant activity and the lipid content of serum for the possible outcome of improving the activity of Ishige okamurae extracts in ovariectomized rats. The antioxidant effects of an Ishige okamurae water extract and an Ishige okamurae ethanol extract were measured by evaluating DPPH free radical scavenging activity and SOD-like activity. Fifty, seven-week old female Sprague Dawley rats were randomly assigned to five groups as follows: sham-operated rats (SHAM), ovariectomized rats (OVX-CON), ovariectomized rats that were treated with 17-beta-estradiol (200 ${\mu}g/kg/day$), and ovariectomized rats that were treated with Ishige okamurae extracts (50 mg/kg/day and 200 mg/kg/day, respectively). The diets were fed to the rats for seven weeks after ovariectomy. The antioxidant activities of the water and ethanol extracts of Ishige okamurae increased in a dose-dependent manner, and the ethanol extract was found to be higher than the water extract. Therefore, we examined the effect of an Ishige okamurae ethanol extract on total serum cholesterol, triglycerides, HDL-cholesterol, and LDL-cholesterol levels, and anti-platelet aggregation. The total-cholesterol and triglyceride content of the serum increased in the OVX-CON group compared to the SHAM group, but supplementation with the Ishige okamurae ethanol extract caused these factors to decrease. Notably, the serum LDL-cholesterol concentration in the supplemented 200 mg/kg/day Ishige okamurae ethanol extract group was significantly more reduced than it was in the OVX-CON group. In addition, the platelet aggregation ability was lower in the groups treated with Ishige okamurae than it was in the OVX-CON group. According to these results, the effects of Ishige okamurae extract on serum lipid content in ovariectomized rats were illuminated.

Comparison of Anticancer and Immuno-Modulatory Activities in the different parts of the Acer mono Max. and Acer okamotoanum (고로쇠와 우산고로쇠 나무의 부위별 항암 및 면역조절능 비교)

  • This study was performed to investigate anticancer activities and immuno modulatory activities in the several parts of the A. mono and A. okamotoanum. The cytotoxicity of 1 $mg/m{\ell}$ of the water extracts on normal human lung cell(HEL299) was < 19.5%. The anticancer activity of all extracts were increased in over 55% against AGS (stomach adenocarcinoma), A549 (lung adenocarcinoma), Hep3B (liver adenocarcinoma, and MCF-7 (breast adenocarcinoma) cells. The growth of human immune B and T cells was improved of A. mono and A. okamotoanum in adding 1.0 $mg/m{\ell}$ concentration. The secretion of the IL-6 and $TNF-{\alpha}$ of human immune B and T cells was increased with all extracts of A. mono and A. okamotoanum. All extracts of. A. mono and A. okamotoanum increased NK cell growth. The results showed that the barks and woods extracts of A. mono and A. okamotoanum had useful biological activities. In addition, bark of A. okamotoanuim showed the highest anticancer and immune activities.

Enhanced Production of Oleanolic Acid by the Elicitation in Oldenlandia diffusa Suspension Cell Cultures (백화사설초의 현탁세포배양에서 Elicitation에 의한 Oleanolic acid 생산성 증대)

  • Oldenlandia diffusa is a Chinese medicinal herb with antitumor activity capable of suppressing the growth of some cancer cell lines. Oleanolic acid and ursolic acid are triterpenoid compounds that exist in Oldenlandia diffusa. Recently, these have been noted for anti-inflammatory, anti-cancer, and hepato-protective effects. Application of both plant growth regulators, 2,4-D and kinetin, was found to be essential for the initiation of callus and suspension cells. Leaf blades of Oldenlandia diffusa was transformed into callus on Schenk and Hildebrandt medium supplemented with 0.5 mg/L 2,4-D and 0.1 mg/L kinetin, while optimum initiation condition for suspension cells of Oldenlandia diffusa was determined to be 0.75 mg/L 2,4-D and 0.1 mg/L kinetin. Chromatographic separation of oleanolic acid from its derivatives was achieved using Rexchrom S5-100-ODS column. Analytical conditions for oleanolic acid were determined as follows: flow rate at 1.0 mL/min, UV length at 200 nm and mobile phase of $80\%$ acetonitrile and $20\%$ water. Production of secondary metabolites was found to be increased by the treatment with elicitors or signal transducers. The maximum production of oleanolic acid was 99.6 mg/L in cultures with 0.5 mM salicylic acid. It is 1.74 times higher than that of control.
