A study on degaradation stabilization of organic material through aerobic treatment before landfill of domestic waste (생활폐기물의 호기성처리를 통한 유기물 분해안정화에 관한 연구)

  • The purpose of this study is to investigate appropriate environmental factors when domestic waste is decomposed as aerobic digestion. So stabilization degree was measured after the waste is mixed as certain rates and water content was controlled by 55% and 60%. Variation of VS showed food waste in reactors of number 1, 2, 3, 4 and 5 was decomposed fully except reactor of number 6. Decomposition degree of VS in reactors of number 1, 2, 3 and 4 was not different high because Vinyl and plastic inserted played role bulking agent in reactor number 1, 2, 3 and 4. In reactors, maximum temperature indicated $57{\sim}59^{\circ}C$ and temperatures in reactors 1, 2, 3 and 4 were higher and remained longer than in reactor 5 and 6 for 2~4 days. Variation of $CO_2$ was similar to that of VS. The reduction rate of water content was low because moisture generated by oxidation fever of microorganism did not evaporated well. pH was low in the beginning of the reaction however, as time passed, it increased slightly and remained regular pattern. EC and C/N showed the same pattern as pH. Settlement and weight reduction rates were similar to the factors above. Reactor 1, 2, 3, and 4 showed higher settlement and weight reduction rate than reactor 5 and 6.

Characterization of Bacterial Cellulose Production by Gluconacetobacter sp. JH232. (Gluconacetobacter sp. JH232의 Bacterial Cellulose 생성 특성연구)

  • Previous study (J. of Chem. Technol. Biotechnol. 2004, 79, 79-84) showed that bacterial cellulose (BC) produced by a bacterial strain JH232 has potential as a source for environmentally friendly ion exchange membranes. In this study, strain JH232 was investigated for phylogenetic classified and characterized for BC production. Comparative analysis of 16S rRNA gene revealed that the strain belongs to the genus Gluconacetobacter. Maximum production of BC was observed when JH232 was cultured in CSL medium (pH 5.5) at $30^{\circ}C$ as determined by flask experiment. When batch and fed-batch cultures of JH232 were performed in the fermenter experiment to compare BC productivity of the strain, BC productivity of fed-batch culture was 1.56 times higher than that of batch culture.

Characteristic of Glasswort (Salicornia Herbacea L.) Mixture Fermentation Utilizing Aspergillus oryzae (황국균을 이용한 퉁퉁마디(Salicornia herbacea L.) 혼합물의 발효 특성)

  • In this study, we investigated the quality of glasswort (Salicornia herbacea L.) mixture fermented by Aspergillus oryzae at $30^{\circ}C$ for 7 days. Changes of pH, titratable acidity, amino-nitrogen content, reducing sugar content, activities of $\alpha$-amylase and protease and number of mold were determined in the course of the fermentation. Angiotensin converting enzyme (ACE) inhibition activities and electron donating ability (EDA) were measured after 7 days fermentation. The pH lowered from 6.19~6.22 into the level of 5.41~6.08 after fermentation for 7 days. Titratable acidity increased from 0.59~0.68 into the highest level of 0.95~1.13% after 6 days fermentation. Furthermore, the amino-nitrogen content increased from 128.0~167.2 mg% to 159.4~209.0 mg% after fermentation for 7 days. For the reducing sugar content, it decreased from 2.0~5.9% into the level of 0.4~1.1% during 7 days fermentation. The number of molds decreased from $10^7\;CFU/g$ to approximately $10^6\;CFU/g$ after 5 days fermentation. $\alpha$-Amylase activity showed from the beginning of the fermentation in all samplings, but protease activity reached the value of food code standards after 5 days of fermentation. ACE inhibition activities were slightly increased from 9.5~16.6% to 19.3~22.7% and electron donating ability were slightly increased from 55.6~57.8% to 60.9~62.7% after 7 days of fermentation.

Effect of Mineral Nutrient Control on Nutrient Uptake, Growth and Yield of Single-Node Cutting Rose Grown in a Closed Hydroponic System (순환식 수경재배시 무기이온 조절이 Single-Node Cutting 장미의 양분흡수, 생육 및 품질에 미치는 영향)

  • This study was conducted to observe the characteristics of mineral nutrient uptake of single-node cutting rose 'Versilla' and to determine optimal nutrient solution control method for soilless culture of 'Versilla' in a closed hydroponic system. Nutrient solution was managed by five different control methods: macro- and micro-element control in aeroponic system (M&M), macroelement control in aeroponic system (M), nutrient solution supplement in aeroponic system (S); electrical conductivity (EC) control in aeroponic system(EC-A); EC control in deep flow technique system(EC-D). The concentration of $NO_3$-N exceeds optimal range whereas P and Mg decreased at the later stage of plant growth with the EC control method, EC-A and EC-D. The overall mineral nutrient content increased with S. On the other hand the nutrient content at the root environment was maintained optimal with M&M and M. The nutrient solution control methods had significantly effect on the cut-flower quality. In the M&M and M, flower length, fresh weight and root activity were higher than those with the other mineral nutrients control methods. The maximal efficiency of photochemistry (Fv/Fm) was higher for M&M, M and S than that with EC-A and EC-D. Based on the above results, it is highly recommended to control nutrient solution by mineral nutrient control methods (M&M and M) in a closed hydroponic system for single-node cutting rose, 'Versillia'.

Uranyl Peroxide Compound Preparation from the Filtrate for Nuclear Fuel Powder Production Process (핵연료분말 제조공정 여액으로부터 Uranyl-peroxide 화합물의 제조)

  • Uranyl-peroxide compound was prepared by the reaction of excess hydrogen peroxide solution and trace uranium in filtrate from nuclear fuel conversion plant. The $CO_3{^{2-}}$ in filtrate was removed first by heating more than $98^{\circ}C$, because uranyl-peroxide compound could not be precipitated by $CO_3{^{2-}}$ remaining in filtrate. The optimum condition for uranyl-peroxide compound was ageing for 1 hr after controling the pH with $NH_3$ gas and adding the excess $H_2O_2$ of 10ml/lit.-filtrate. Uranium concentration in the filtrate was appeared to 3 ppm after the precipitation of uranyl-peroxide compound, and the chemical composition of this compound was analyzed to $UO_4{\cdot}2NH_4F$ with FT-IR, X-ray diffractometry, TG and chemical analysis. Also, this fine particle, about $1{\sim}2{\mu}m$, could be grown up to $4{\mu}m$ at pH 9.5 and $60^{\circ}C$. The separation efficiency of precipitate from mother liquor was increased with increase of pH and reaction temperature. Otherwise, the crystal form of this particle showed octahedral by SEM and XRD, and $U_3O_8$ powder was obtained by thermal decomposition at $650^{\circ}C$ in air atmosphere.

  • Production of 4-androstene-3,17-dione(AD) from cholesterol by microbial conversion was investigated. To facilitate the solubilization of cholesterol in the fermentation broth, ethanol was used as an organic solvent. Inhibition on cell growth by ethanol was observed to be negligible upto 2% (V/V) concentration. Microbial conversion was successfully carried out with high yield when the cholesterol was added at early logarithmic growth phase with pH control at 7.0. In order to improve the process productivity, bioconversion was conducted at various mode of cholesterol addition ; 0.1% (V/W) of cholesterol was found to be most appropriate for solubilization in ethanol and was added intermittently. When added three time(total 3 g/$\ell$), overall bioconversion yield reached upto 65% while single addition of same amount of cholesterol (3 g/$\ell$) yielded about 40% conversion.

Ultrafiltration for Quality Improvement of Wine (한외여과공정을 이용한 포도주의 품질개선)

  • Physicochemical and microbiological changes of grape wine fermented and aged at 25 and $15^{\circ}C$ for 2 and 14 weeks, respectively, were investigated. Viable bacterial cell number, $3.3{\times}10^2\;CFU/mL$ at the beginning of fermentation, increased to $2.3{\times}10^6\;CFU/mL$ after 2 weeks, then decreased to $1.9{\times}10^3\;CFU/mL$ after 14 weeks. Viable yeast cell number increased from $2.8{\times}10^2\;to\;2.2{\times}10^7\;CFU/mL$ during fermentation, then decreased to $1.6{\times}10^4\;CFU/mL$ after aging. Turbidity, pH, total sugar content, reducing sugar content, and solid content of grape wine decreased during fermentation, whereas acidity and alcohol content increased to 0.64 and 8.4%, respectively. Most physicochemical properties did not change significantly during aging. When grape wine was filtered through $0.45-{\mu}m$ nitrocellulose membrane, followed by various ultrafiltration membranes with different molecular weight cut-off values, Biomax 100K membrane with $100\;L/m^2/hr$ (LMH) of initial flux was chosen for ultrafiltration process. These membrane filtration treatments resulted in complete removal of microorganisms and decreases in turbidity, reducing sugar, and solid content. Physicochemical properties of wine did not change, and no microorganisms were found during storage at $30^{\circ}C$ for 12 weeks.

Effects of Artificial Acin Rain on Seed Germination, Growth and Needle of Several Conifers(2) (인공산성우가 몇 침엽수종의 종자발아, 생장 및 침엽에 미치는 영향(2))

  • Artificial acid rain (pH 3.0, 4.0 and 5.0) and ground was treated on the seeded pots of 4 species to examine its effects on germination and survival rate, seedling growth and contact angles on needle surface. Artificial acid rain was prepared by diluting sulfuric acid with ground water and ground water (pH 6.5) was used as control. Artificial acid rain was sprayed to seeded pots two or three times per week for growing season, one times per week for winter seasons. About 5mm of artificial acid rain was treated each time from early April, 1991 to early October, 1993. Germination and survival rate, soil acidity, seedling growth and contact angles on the needle surface were measured and compared among the treatments. The results were summarized as follows ; Artificial acid rain might have positive effects on growth-related characteristics of the seedlings in the first and second year of acid rain treatment, and the effects were differed among four species. All growth-related characteristics of the seedlings in third year, however, decreased with decrease of pH values of artificial acid rain. This was considered to the results of acidic accumulation over soil buffer capacity. Needle injury and biomass (defoliation) was correlated with the pH values of artificial acid rain, and this character might be a good criteria for early diagnosis of acid rain injury. The differences of soil acidity were significant among the treatments for all species. Contact angles between needle surface and water droplet decreased with decrease of pH values of artificial acid rain. Measuring and comparing contact angles might be very good criteria for early diagnosis of acid rain injury.

Effects of Concentration of $NO_3^--N$, K and Ca in nutrient solution on Seedling Growth of Mudeungsan Watermelon (배양액내 $NO_3^--N$, K 및 Ca의 농도가 무등산수박 유묘의 생장에 미치는 영향)

  • 무등산 수박의 유묘 생산에 있어 적정 배양액 농도를 구명하기 위하여 일본원시균형배양액을 기준으로 하여 NO$_3$-N, K 및 Ca의 농도별 첨가량에 따른 유묘생장반응을 조사하였다. 1. 배양액내 질소농도을 증가시킬수록 초장, 엽면적, 엽수, 엽장, 엽폭이 증가하였으며 생체중과 건물중도 증가하였다. 2. 배양액내 칼륨의 농도를 증가시켰을 경우 수박 유묘의 초장은 200ppm까지는 약간 증가하지만 엽면적, 엽수, 엽장, 생체중 및 건물중은 200ppm이상으로 증가시키면 감소하는 경향이었다. 3. 칼슘처리의 경우 농도의 증가가 생장의 증가를 나타내지는 않았으며 원시균형 배양액의 표준이라 할 수 있는 80ppm처리구의 생장이 양호한 것으로 나타났다. 4. 배양액(일본원시균형배양액 1/2농도)에 N의 농도를 200ppm으로 증가시킬 경우 수박유묘의 엽병내 N, K 및 Mg의 함량이 증가하였지만 P 및 Ca의 증가나 감소의 경향은 보이지 않았다. 5. K의 농도를 100ppm으로 증가시킬 경우 수박유묘의 엽병내 N, K 및 Mg의 함량이 증가하였지만 P 및 Ca의 증가나 감소의 경향은 보이지 않았다. 5. K의 농도를 150ppm으로 증가시킬 경우 수박유묘의 엽병내 N, K 및 Mg의 함량이 증가하였지만 P 및 Ca의 증가나 감소의 경향은 보이지 않았다. 그러나 200ppm이상으로 증가시킬 경우 N과 Mg의 감소가 나타났다. 6. 양액내 Ca의 농도를 증가시킬수록 엽병내 N, K Ca 및 Mg의 농도가 증가하는 반면 P의 경우 거의 변화가 없었으며,N의 경우는 200ppm 이상의 농도에서 감소하는 경향을 나타내었다. Mg의 경우는 150ppm까지는 함량의 변화가 없었지만 200ppm 이상의 처리구에서 급격한 함량증가를 나타내었다.~21.2ml가 흘러나와 배지의 공극이 일정하지 않아 시간당 통과하는 양이 일정하지 않았다고 생각되었다. $\bigcirc$ 펄라이트 : 합섬A(비스코스+레이온)급액천의 유입은 소(1$\times$60cm)에서 21.8ml, 중(2$\times$60cm) 33.5ml, 대(3$\times$60cm) 43.4ml가 통과되었고 합섬(폴리에스텔)에서는 19.0~30.7ml로서 급액천의 규격에 따라 통과되는 차이가 있었다. 배지가 규격화되어 있어 급액천의 규격별로 일정하게 유입되었으며 급액천의 재질이 유입에 영향을 미친 것으로 사료되었다. (2) 급액관과 베드상과의 높이에 따른 유출양 : 급액과 베드상과의 낙차가 클수록 유출이 증가함을 알수 있었으나 합섬C(인견)실험구에서는 낙차가 유출에 영향을 미치지 않았다. (4) 급액된 양액의 EC 및 pH조사 : 급액된 양액의 EC 및 pH에 전혀 변화가 없어 재배 적응에 문제가 없을것으로 사료되었다.이가 가장 이상적인 것으로 생각된다.세포수에 대한 내부세포괴세포(ICM/total cells)가 20~40% 범주에 드는 비율은 처리구가 대조구보다 낮은 결과를 나타냈다. 결론적으로 돼지난포란을 이용하여 체외성숙을 유기할 때 효과적인 cysteamine의 농도는 50$\mu$M이 적당하며, 초기배발달을 유기할 때의 효과적인 cysteamine의 농도는 25~50$\mu$M인 것으로 판단된다.N)A(N)/N을 제시하였다(A(N)=N에 대한 A값). 위의 실험식을 사용하여 헝가리산 Zempleni 시료(15%$S_{XRD}$)의 기본입자분포로부터 %$S_{XRD}$를 계산한

Allergy Immunity Regulation and Synergism of Bifidobacteria (Bifidobacteria의 allergy 면역 조절과 synergism)

  • Allergic diseases have increased over the past several decade worldwide including developing countries. Allergic inflammatory responses are caused by Th (T helper)2 immune responses, triggered by allergen ingestion by antigen presenting cells such as dendritic cells (DCs). Intestinal microorganisms control the metabolism and physiological functions of the host, contribute to early immune system maturation during the early life, and homeostasis and epithelial integrity during life. Bifidobacteria have strain-specific immunostimulatory properties in the Th1/Th2 balance, inhibit TSLP (thymic stromal lymphopoietin) and IgE expression, and promote Flg (Filaggrin) and FoxP3 (Treg) expression to alleviate allergies. In addition, unmethylated CpG motif ODN (oligodeoxynucleotides) is recognized by TLR (toll-like receptors)9 of B cells and plasmacytoid dendritic cells (pDCs) to induce innate and adaptive immune responses, while the butyrate produced by Clostridium butyricum activates the GPR (G-protein coupled receptors)109a signaling pathway to induce the expression of anti-inflammatory gene of pDCs, and directly stimulates the proliferation of thymically derived regulatory T (tTreg) cells through the activation of GPR43 or inhibits the activity of HADC (histone deacetylase) to differentiate naive $CD4^+$ T cells into pTreg cells through the histone H3 acetylation of Foxp3 gene intronic enhancer.
