Effect of Dietary Lipid on Fatty acid Pattern in Developing Brain Mitochondria in Rats (식이 중 지방 함량이 성장기 쥐의 뇌 미토콘드리아 지방산 조성에 미치는 영향)

  • 식이 중 지방함량이 성장기 뇌의 미토콘드리아 지방산 조성에 미치는 영향을 살펴보기 위해 옥수수기름을 각각 10, 2, 0.5% 포함한 3종류의 식이를 임신한 Sprague Dawley종의 흰쥐에게 임신말기부터 제공했다. 수유기간 중의 영향을 살펴보기 위해 대조군(10%옥수수기름 섭취)의 일부에 분만 후 0.5% 결핍식이를 섭취시켰고, 또 이유후부터 0.5% 결핍식이군의 일부에 10% 정상식이를 주어 회복효과를 관찰하고자 하였다. 전 실험기간 동안 체중과 뇌 무게, 단백질 함량을 측정하였고 뇌에서의 미토콘드리아를 분리하여 지방산 조성을 관찰하였다. 출생후 수유기간 동안에는 대조군에 비해 지방결핍식이군에서 체중과 뇌의 무게가 다소 높았으나 이유 후 부터 점점 감소하여 유의적인 차이를 볼 수 없었고 특히 0.5% 식이군의 체중은 생후 6주와 7주에 유의적인 감소를 보였다. 그러나 체중에 대한 뇌무게와 뇌의 단백질 함량에 있어에는 대조군과 결핍군 사이에 유의적인 차이를 볼 수 없었다. 따라서 지방결핍이 뇌 성장에 미치는 영향은 다른 영양소에 비해 크지 않음을 알 수 있었다. 뇌 미토콘드리아의 지방산 조성은 나이와 식이에 따라 변하였다. 나이를 먹음에 따라 단쇄 포화 지방산은 감소하고 다 불포화 지방산의 함량은 증가하였다. 또한 결핍식이군과 대조군 사이에 뇌 미토콘드리아 지방산조성을 살펴 본 결과 지방 결핍의 좋은 지표가 되고 불포화도, triene 과 tetraene의 비, 그리고 ${\omega}-6$ 계열과 ${\omega}-9$ 계열의 비에 있어서 큰 차이를 볼 수 있었다. 이들로 부터 성장초기에 지방함량을 조절한 식이에 의해 뇌 미토콘드리아 지질 조성이 크게 변하고 이에따라 미토콘드리아의 기능도 유의적인 영향을 받고 있음을 알 수 있었다.

Effect of Dietary Intake of Salicornia herbacea L. Hot Water Extract on Anti-obesity in Diet-induced Obese Rats (함초 열수추출물의 섭취가 비만유도 흰쥐의 항비만 효과에 미치는 영향)

  • Salicornia herbacea L. is an annual herb that grows in salt marshes and salt fields along the seashores. It is also commonly used as a folk remedy in Korea. The objective of this study was to investigate the beneficial effects of Salicorrnia herbacea L. (SH) hot water extract on obesity. Five-week old male Sprague-Dawley rats (n=36) were divided into two groups and provided either a normal fat diet (11.5% fat from kcal) or a high fat diet (40.5% fat from kcal) for 6 weeks to induce obesity. Then, rats were blocked into six groups of six mice each and provided either a diet containing SH (0.5% of diet; g/kg) or a normal diet for another 6 weeks. Final body weights were significantly reduced when rats were fed SH among the high fat diet groups (HNS and HHS). Serum triglyceride concentrations significantly decreased in every group provided SH. HDL-cholesterol concentrations significantly increased in SH-fed groups among the high fat diet groups. Further, atherogenic index significantly decreased when rats were fed SH diet (HHS). There were no differences in LDL-cholesterol between the high fat diet groups, and glucose concentrations decreased when rats were fed SH diet (HNS). These results indicate that dietary intake of Salicornia herbacea L. hot water extract might have beneficial effects on obesity by reducing body weight, fat weights, and improving blood lipid profile.

Anti-obesity Effect of Chopped Sauce with Medicinal Plant Extracts (약용작물 추출물을 첨가한 다진양념의 비만 억제 효과)

  • 본 연구는 항비만 기능성을 가지면서 맛과 향이 증진된 분말형 다진양념(다데기)을 개발하기 위해 첨가할 수 있는 약용작물들의 선정 및 그 기능성을 평가하기 위해 실시되었다. 오미자, 진피, 감초, 녹차엽 등을 첨가군으로 선정하여 각각의 시료들을 8시간, $80^{\circ}C$ 증류수에서 추출 후 감압농축하여 추출물을 제조한 뒤 마우스 유래 adipocyte (3T3-L1)에 처리하여 세포내 지방 축적 억제 정도를 실험하였다. 시료 처리 배양 5일 후 오미자, 진피, 감초, 녹차엽 추출물을 처리한 모든 군에서 대조군과 비교시 20% 이상 세포내 중성지방 농도가 감소하였으며 특히 오미자 군에서는 30% 이상 감소를 보였다. C57BL/6 생쥐를 이용하여 고지방식이로 비만을 유도하면서 분말형 다진양념, 오미자, 진피, 감초, 녹차엽 추출물을 경구투여한 뒤 체중증가를 조사한 결과 8주 후 고지방식이와 다데기만을 경구투여한 군은 고지방식이군과 비교하여 체중감소경향만을 보였으나 분말형 다진양념과 추출물을 함께 경구투여한군에서는 유의적으로 체중이 감소하는 결과를 보였다. 이들 추출물을 혼합하여 제조한 분말형 다진양념은 관능검사에서도 고춧가루 특유의 풋내를 감소시켜 향미가 증진되는 것으로 평가되어 오미자, 진피, 감초, 녹차엽을 첨가한 다양한 항비만 기능성 양념 개발이 기대된다.

Effect of Improve Obesity with Sprout Raw Grains and Vegetables (발아 생식을 이용한 비만개선 효과에 관한 연구)

  • This study was carried out to investigate the effect of weight loss and the change of serum lipids composition for normal and obese women after sprout raw grains and vegetables diet intake. Diet were given to the subjects which were composed of 11 normal women and 7 obese women for 4 weeks. After diet intake, 4.6% in normal group and 3.5% in obese group were shown weight loss. And the both groups of normal and obese women showed a significant reduction in the thickness of subcutaneous fat and the body girth. Particularly, the waist measure was shown to be remarkably reduced in the normal group (p< 0.0001) and to be reduced in the obese group (p<0.017) significantly. After diet intake. the levels of total cholesterol and triglycerides were reduced in both groups significantly. And in the obese women, HDL-cholesterol level increased 5.7%. As those results, it could be seen that weight and the thickness of subcutaneous fat were reduced and the component of blood serum were improved after diet intake for 4 weeks. Specially, abdomen, waist measures and hip girth were reduced remarkably, so it is considered that the diet is more effective in the diet therapy of obese women with upper obese status.

Weight Reduction and Lipid Lowering Effects of Korean Traditional Soybean Fermented Products (전통 장류의 체중감소 및 지질저하 효과)

  • Effects of Korean traditional soybean fermented foods on weight reduction and lipid lowering activities were studied using Spraque-Dawley (SD) rats fed a high fat diet. The rats were raised for four weeks after adaptation period on either a normal diet (ND, based on the AIN-93 diet), high fat diet (HFD, supplemented with 12% lard oil in the ND), or diets containing 10% of freeze dried Doenjang, Chungkukjang, Kochujang, or Samjang to HFD. The final weight, food efficiency ratio (FER) and the weight of adipose tissue were decreased significantly by the consumption of Doenjang and Samjang, compared to HFD (p<0.05). The contents of cholesterol and triglyceride increased by HFD were reduced by the fermented foods in liver and perirenal fat tissues of the rats, especially Doenjang and Samjang diets showed high lipid lowering activity. And TG and cholesterol contents in the plasma serum were simillar trend to liver and perirenal fat tissue. These results suggested that Korean traditional soybean fermented foods, especially Doenjang showed the highest weight reduction and lipid lowering activities of the rats fed high fat diet.

Hypernatremia and Intraventricular Hemorrhage in Very Low Birth Weight Infants(<1,250 g) (극소 저체중 출생아(<1,250 g)에서 고나트륨혈증 발생 및 뇌출혈과의 관계)

  • Purpose: Hypernatremia most frequently occurs in the immature newborn and be severe in association with intraventricular hemorrhage (IVH). This study examined the frequency, onset and risk factors of hypernatremia, and the relationship between hypernatremia and IVH in very low birth weight (VLBW; <1,250 g) infants. Methods: We retrospectively reviewed the medical records of 55 VLBW infants admitted between January 2006 and December 2009 to the neonatal intensive care unit of Wonkwang University Hospital and who survived over 7 days. Serum sodium concentration, sodium intake, fluid and weight loss, as suggested risk factors of hypernatremia, and the incidence of IVH were evaluated. The infants were divided into a hypernatremia group (${\geq}$150 mEq/L) and nonhypernatremia group, and were compared. Results: Incidence of hypernatremia in the VLBW infants was 52.7%, and mean starting time of hypernatremia was 2.8${\pm}$1.3 days. There were no differences in the sodium and fluid intake between the two groups. Weight loss at day 3 after birth was significantly higher in the hypernatremia compared to the nonhypernatremia group (P<0.05); thereafter weight loss was non-significantly higher. The incidence of IVH in VLBW infants was 38.2%, and the difference between the two groups was not significant. Conclusion: Hypernatremia occurs commonly in VLBW infants and is most commonly caused by weight loss in the early days after birth. Incidence of IVH is not likely influenced by hypernatremia with marginally elevated sodium concentration.

Effect of Weight Control Program on Obesity Degree and Blood Lipid Levels among Middle-aged Obese Women (중년여성대상 체중조절 프로그램이 체구성 및 혈액지질 개선에 미치는 영향)

  • This study was performed to investigate the effect of 12-week weight control program including nutrition education, diet therapy and regular exercise on body weight, obesity degree, BMI and blood lipid levels among middle aged obese women. The 31 middle aged obese women participated in the weight control program for 12-week. The body weight, body composition, blood lipid levels and food habit were measured before and after the weight control program. Changes in body weight after 12 week weight control program showed the slight reduction from $67.4{\pm}10.8kg\;to\;65.1{\pm}9.2kg$. Obesity degree and BMI were decreased from $128.4{\pm}19.1%,\;27.8{\pm}3.9\;to\;123.4{\pm}15.7%,\;26.8{\pm}3.0$, respectively. Waist-hip ratio was also decreased from $0.98{\pm}10.16\;to\;0.94{\pm}0.04$. LBM was also slightly reduced from $39.9{\pm}5.6kg\;to\;37.0{\pm}8.5kg$ with the reduction of body fat. The levels of systolic blood pressure, total cholesterol and triglyceride were slightly decreased, HDL-chol level was significantly increased from $43.1{\pm}14.1mg/dL\;to\;50.4{\pm}16.5mg/dL$. Waist-hip ratio was also slightly decreased after 12 weeks. Food habit score was significantly increased after nutriton education and personal counseling about their eating behaviors and nutritional status.

Clinical Observation on Effects and Adverse Effects of Choweseuncheng-tang on Obesity Patients (비만 환자에 대한 조위승청탕의 효과 및 부작용에 관한 임상적 고찰)

  • Objective : This study was designed to investigate the effects and adverse effects of Choweseuncheng-tang on obesity patients Methods : Choweseuncheng-tang was given to 20 obesity patients who have no abnormality of GOT, GPT and past medical history. Additionally, electroacupuncture was performed on Fuchao(腑巢) with 60Hz. We compared body weight, body fat and laboratoy test(GOT, GPT, BUN, Creatinine) between before and after treatment. Blood pressure, heart rate and clinical symptoms were checked to investigate the side effects. Decrease of body weight was checked in accordance with Sasang constitutional type, appetite type, and BMI. Results : 6 patients(BMI 23-24.9), 10 patiets(BMI 25-29.9), and 4 patients(BMI over 30) were showed decrease of body weight. We found body fat was decerased $3.16{\pm}1.25%$. BUN, Creatinine, GOT and GPT were increased within normal range. There were adverse event of autonomic hyperactivity(11 patients), gastrointestinal symptoms(7 patients) and others(3 patients). Conclusions : According to above results, Choweseuncheng-tang could be used effectively, but should be taken cautiously on obese patients.

The Toxicological Parameter Assessment in Experimental Animals for Various Dosages of Polyacetylene Compounds (Polyacetylene 화합물들의 투여량에 따른 독성)

  • The change in bodyweight, organ weight and the amount of water intake were investigated to assess the effect on a range of toxicological parameters of varying dosages of polyacetylene compounds. Concerning inhibition of solid tumor growth induced by Sarcoma-180 cells in the ICR mouse strain, relytionships between the dosage of polyacetylene compoilnds used and several toxicological parameters were observed. When 40μmoles/kg b.w. of panaxydol tvas administered for 3 consecutive days, the suppression of body weight growth in Wistar rats was about 17% that of the control group, however, the body weight gain was normalized 4 days after stopping arlnlinistration of the polyacetylene compounds. When the dosage of polyacetylenes was reduced to 20μmoles/kg b.w., the change in body weight gain was alleviated to within l0% of the control and the suppremission of the body weight gain increased in order for Panaxydol, Panaxynol, and Panaxytriol. The organ weight did not show any significant change between the control and the polyacetylene administered groups. Electron micrographs of the liver tissues of ICR mice injected with 40Umoles/kg b.w. of panaxydol gave no further significant information about toxicological changes.
