Effect of Dietary Protein and Calcium Levels on Calcium Metabolism of the Rat (식이단백질과 칼슘 수준이 흰쥐의 칼슘대사에 미치는 영향)

  • This study was conducted to examine effect dietary protein and calcium levels on calcium metabolism of the rat. Weaned 6-week old male rats were divided into 4 groups and were fed experimental diets for six weeks. Experimental groups were HPNC group-high protein normal calcium (protein: 400 g/kg diet, calcium: 0.5%), HPLC group-high protein low calcium (protein: 400 g/kg diet, calcium: 0.1%), NPNC group-normal protein normal calcium (protein: 200 g/diet, calcium: 0.5%), NPLC group-normal protein low calcium (protein: 200 g/diet, calcium: 0.1%). The calcium excretion in urine was higher in high protein group than in normal protein group, and it was highest in HPLC group. The activation of alkaline phophatase had a tendency to low in normal calcium group, and the concentration of parathyroid hormone (PTH) was the lowest in HPLC group. The deoxypyridinoline (DPD) concentration of urine was investigated as the highest in HPLC group and it was significantly lower in HPNC group that consumed normal calcium. The bone density of the femur was the highest in NPNC group and the lowest in NPLC group. As the results of this study, calcium excretion in urine and DPD density were the highest and the bone density was the lowest in HPLC group. It may suggest that the deficiency of calcium causes adversely effect in calcium metabolism upon consuming high protein diet. Therefore, it should be emphasized to consume enough calcium to prevent the hindrance of skeletal metabolism caused by deficiency of calcium upon consuming high protein diet.

A Study on Antioxidative Effects of Sipyimiguanjungtang and Osuyubujayijungtang, Korean Traditional Prescriptions for Soum Constitutes, in Brain and Liver of Rat (소음인(少陰人) 십이미관중탕(十二味寬中湯), 오수유부자이중탕(吳茱萸附子理中湯)이 흰쥐의 뇌(腦)와 간조직(肝組織)의 항산화(抗酸化) 기전(機轉)에 미치는 영향(影響))

  • The free radical theory of aging was introduced in 1956 by Denham Harman. This aging theory proposed that normal aging results from random deleterious damage to tissues by free radical and supplying antioxidant lead to decrease oxidative damage, inhibit aging process. In this study, we investigated antioxidantive effects of four Korean constitutional prescriptions for 'Soum' constitution - Palmulgunjatang(Y1), Sipyimiguanjungtang(Y2), Osuyubujayijungtang(Y3) and Seungyangyikkibujatang(Y4). Antioxidative activity of this prescriptions was examined by 1,1-diphenyl-2-picrylhyrdazyl radicals, superoxide anion radicals, peroxyl radical, hydroxyl radical scavenging effects and erythrocyte hemolysis inhibitory effects. Y2 and Y3 were shown to have relatively high antioxidative activity on this methods. In additions, result of the cytoprotective effects of Korean constitutional prescriptions agianst 2,2'-azobis(amidinopropane) dihydrochloride (AAPH), a free radical initiator, induced cytotoxcity in human hepatoblastoma cell line was similarly obtained. On the basis of this result, we assayed the antioxidative effects of Y2 and Y3 on experimental oxidative damage, induced in mouse by 100mg/kg AAPH. Male ICR mouse were given oral administration of 500mg/kg Y2 and Y3 for 4 weeks. Thiobarbuturic acid reactive substance (TBARS) and protein degradation level in liver, plasma and brain as index of oxidative damage were decreased and thiol compound, total antioxidant status in plasma were increased by Y2 administration. But, Y3 injected group was decreased only protein degradation level in brain. Also, glutathione, a potent water-soluble endogenous antioxidant, concentration was increased by Y2 and Y3 administration in liver and brain. However, superoxide dismutase and catalase activity as a major antioxidative enzyme in vivo were not shown change by Y2 and Y3 administration. On the basis of these result, Y2 have an antioxidative effects on both water-soluble fraction and lipid-solube fraction in cell and tissues. But, Y3 has a lower antioxidative effects on lipid-soluble fraction than Y2 in cell and tissues. These results suggest that Y2 has a antioxidative effects by protect the tissue against oxygen free radical mediated oxidative damage and Y3 has a limited antioxidaitve effects on water-soluble fraction in vivo. Therefore, we make report that Y2 is more effective prescriptions for anti-aging or therapeutics of diseases.

  • The effects of bamboo (Phyllostachys nigra var. henenis Strapf) leaf extract (BLE) on the changes of ultraviolet (UV) light B radiation-induced apoptotic sunburn cell (SBC) and epidermal ATPase-positive dendritic cell (DC) in SKH1-hr or ICR mouse were investigated. The mice were treated with UVB ($200mJ/cm^2$) and were sacrificed 24 hours later. BLE (50 mg/kg of body weight) or vehicle (saline) was given i.p. at 36 and 12 hours before irradiation, and 30 minutes after irradiation. BLE cream (0.2%) or cream base (vehicle) was also topically treated at 24 hours and 15 minutes before irradiation, and immediately after irradiation. The skin of SKH1-hr mouse prepared from the back of untreated mice exhibited about 0.3 SBC/cm length of epidermis, and 24 hours after UV irradiation, the applied areas show an increased number of SBCs. But the frequency of UVB-induced SBC formation was significantly reduced by intraperitoneal injection (59.0%) and topical application (31.8%) of BLE extract. The numbers of DC in normal ICR mouse were $628.00{\pm}51.56\;or\;663.20{\pm}62.58\;per\;mm^2$ of ear epidermis. By 1 day after UVB treatment, the number of ATPase-positive $cells/mm^2$ were decreased by 39.0% or 27.1% in i.p. or topical application group with vehicle. The frequency of UVB ($200mJ/cm^2$)-induced DC decrease was reduced by treatment of BLE as 25.7% in i.p. group and 3.2% in topical application group compared with the irradiation control group. The results presented herein that BLE administration could reduce the extent of skin damages produced by UVB.

Effects of Dietary Quartz Porphyry and Feed Stimulants, BAISM Supplementation on Growth Performance and Disease Resistance of juvenile eel Anguilla japonica (사료내 맥반석과 BAISM 복합첨가가 치어기 뱀장어 Anguilla japonica의 성장과 내병성에 미치는 영향)

  • This study investigated the synergistic effects of dietary supplementation of quartz porphyry(QP) and a laboratory developed feed stimulants, BAISM(BS) on growth performance and utilization as the additives for juvenile eel Anguilla japonica. Six isoenergetic experimental diets(18.2 kJ/g) were formulated to contain 50% crude protein, 15% lipid with or without dietary QP(Song-Gang stone, Davistone, Korea) and BS supplementation. QP and BS were provided at 0% in the control diet($Q_0B_0$) and at 0.7% QP+0% BS($Q_{0.7}B_0$), 0.7% QP+0.3% BS($Q_{0.7}B_{0.3}$), 0.7% QP+0.5% BS($Q_{0.7}B_{0.5}$), 0.7% QP+0.75% BS($Q_{0.7}B_{0.75}$) and 0.7% QP+1.0% BS($Q_{0.7}B_{1.0}$) in experimental diets on dry matter basis. After four weeks of adaptation, triplicate groups of 30 fish initially averaging $15{\pm}0.1g(mean{\pm}SD)$ were randomly distributed into each aquarium, and they were fed one of the experimental diets for 8 weeks. By the end of the feeding trial, weight gain(%), specific growth rate(%), feed efficiency(%) and protein efficiency ratio of fish fed diet $Q_{0.7}B_{0.5},\;Q_{0.7}B_{0.75}\;and\;Q_{0.7}B_{1.0}$, were significantly higher(P<0.05) than those of fish fed the other diets. But, $Q_{0.7}B_{0.5},\;Q_{0.7}B_{0.75}\;and\;Q_{0.7}B_{1.0}$ were no significant differences(P<0.05). In challenge test, fish were infected by intraperitoneal injection of 0.1 mL bacterial suspension with Edwardsiella tarda per fish after the feeding trial. As a result, fish fed QP and BS supplemented diets have a significantly higher cumulative survival rate than those of fish fed control diet(P<0.05). In conclusion, these results indicated that the optimum dietary supplementation level of QP and BS could be approximately 0.7% quartz porphyry+0.5% BAISM($Q_{0.7}B_{0.5}$) of diet based on WG, FER, SGR, PER, cumulative survival rate in juvenile eel A. japonica.

Regrowth of Bulrush (Scripus juncoides Roxb.) by Seasonal Mowing (올챙고랭이(Scirpus juncoides Roxb.)의 예취(刈取)에 따른 재생력(再生力)에 관한 연구(硏究))

  • The number of regrown shoots per plant for 2 weeks after mowing sharply increased in response to late mowing until June 6, and then the increments were slow. The number of regrown shoots for July 30 were many with early mowing until May 14, and then the numbers were rapidly decreased. The plant height regrown for 2 weeks after mowing was increased until May 14, and then decreased. The plant height regrown for July 30 was not sensitive to mowing time and the development was linearly shown. The fresh weight regrown for July 30 was only increased by late mowing. The regrowth rate of shoots for 2 weeks after mowing was much higher than untreated control and the ratio was decreased with late mowing and the ratio to untreated for control for July 30 was decreased up to 50% by late mowing. The regrowth rate of plant height for 2 weeks after mowing was rapidly decreased, but slowly decreased for July 30 mowing. The regrowth rate of fresh weight was higher until May 22, and then rapidly decreased by late mowing. The decreasing tendency for 2 weeks after mowing was more prominent them for July 30. The annual regrowth rates in shoots and plant height were slowly decreased, but the decreasing rate in fresh weight was rapidly done until May 15, and then decrement became up to 30% by late mowing.

The Effects of Green Tea on the Serum Lipid and Liver Tissue of Cholesterol Fed Rats (녹차의 음용이 지방과 콜레스테롤을 투여한 흰쥐의 혈청지질 및 간의 조직에 미치는 영향)

  • This study was performed to investigate the effects of green tea on fat metabolism of rats and prevention to cardiovascular disease in drinking green tea. Male Spague-Dawley rats were divided into seven groups consisting of the control, lard and cholestrol, aqueous green tea at the level of 1% and 3%. After 4 weeks of feeding serum lipid levels were measured for experimental rats, and analyzed the total cholesterol (TC), HDL-cholesterol (HDL-C), triglyceride (TG), phospholipid (PL). And total lipid (TL) to Folch method, lipid deposition to oil red O staining on liver tissue. The results are as follows: Total cholesterol (TC) decreased by administration of 1% aqueous green tea group and increased addition to lard and cholesterol (LC) group as compared to each groups (p<0.05). HDL-cholestrol in serum increased by administration of la aqueous green tea group (1G) and decreased to the control group, 1% aqueous green tea (L-1G) added lard group (p<0.05). Triglyceride (TG) decreased by administration of 3% aqueous green tea groups (L-3G, LC-3G) and increased by lard and cholesterol group (LC) (p<0.01). Phospholipid(PL) decreased by administration of 3% aqueous green tea added lard and cholesterol group (LC-3G) and increased by control group, lard and cholestrol group (LC) (p<0.05). Total lipid of liver decreased by administration of aqueous green tea at the level of 1% group and increased by LC group (p<0.01). The fat deposition of liver was increased in fat diet groups and decrease in the drink green tea of some but did not showed significant differences from concentration of the green tea.

Determination of Energy Requirements for Maintenance in Hanwoo Steers (거세한우의 유지에너지 요구량 결정)

  • This experiment was carried out to determine energy requirements for maintenance of Hanwoo steers. Nine Hanwoo steers weighing 376.6$\pm$12.5kg were used in this experiment and fed rice straw(44%) and concentrate (56%) at three different energy levels; 0.8 times maintenance(0.8M), 1.2 times(1.2M) and 1.6 times(1.6M), respectively. Dry matter intake was 48.5, 65.9 and 86.5g/$BW^{0.75}$ for 0.8M, 1.2M and 1.6M, respectively. Increase in energy intake with the increased DM intake did not affect digestibilities of dry matter, organic matter, crude protein, crude fiber, crude fat and nitrogen-free extract. Gross energy intake averaged 190.8, 255.8 and 340.9kcal/BW0.75 for 0.8M, 1.2M and 1.6M, respectively. Energy loss was 41% feces and 0.6${\sim}$1.5% urine of gross energy intake. Further, energy loss from methane produced during rumen fermentation was 5${\sim}$9%, while body heat loss averaged 40${\sim}$60%. Intercept of the regression equation between ME intake and retained energy indicated that energy requirement for maintenance was 124.3kcal $ME/BW^{0.75}$.

Hypoglycemic Effect of Fermented Soybean Culture Mixed with Mulberry Leaves on Neonatal Streptozotocin-Induced Diabetic Rats (뽕잎 함유 대두발효물이 신생 당뇨유도쥐에 미치는 혈당강하효과)

  • The effect of fermented soybean culture of Bacillus subtilis MORI mixed with mulberry leaves on the lowering the levels of blood glucose and cholesterol was examined using neonatal streptozotocin-induced diabetic (n-STZ) rats. B. subtilis MORI produces 1-deoxynojirimycin (DNJ), an $\alpha$-glucosidase inhibitor. The content of DNJ of soybean fermented culture mixed with mulberry was higher (4.1$\pm$0.0 mg/g dry base) than that (1.5$\pm$0.0 mg/g) of mulberry. The concentration of 50% inhibition (IC50) against rat intestinal $\alpha$-glucosidase of soybean fermented culture mixed with mulberry was $5.6{\pm}0.1{\mu}g$/mL and that of mulberry was $17.0{\pm}0.5{\mu}g$/mL. Experimental groups of diabetic rats were randomly assigned to normal control group (NC group), diabetic control group (DC group) and three diabetic groups fed with DNJ food product. One of the three diabetic groups was M group (60 mg DNJ food product/kg) (DNJ food product containing 30% mulberry) and the other two were MM-60 group (60 mg/kg) and MM-120 group (120 mg/kg) (DNJ-fortified food product containing 30% fermented soybean culture mixed with mulberry leaves). The glucose in serum was significantly decreased in the MM-60 and MM-120 groups fed with DNJ-fortified food product for 4 weeks, compared with DC group. Total cholesterol and triglyceride in serum were also lower in MM-60 and MM-120 groups than the DC group. These results support that the fermented soybean culture of B. subtilis MORI mixed with mulberry leaves improved the metabolism of blood glucose and lipid in the n-STZ rat model.

Effect of Supplementation of Acanthopanax senticosus on Growth Performance, Blood Biochemical Profiles and Expression of Pro-Inflammatory Cytokines in Broiler Chicks (육계에서 가시오갈피 급여에 따른 생산성, 혈액 생화학적 성상 및 면역 사이토카인 발현에 미치는 영향)

  • This study was performed to examine the effects of dietary Acanthopanax senticosus (AS) on growth performance, immune organ weights, blood biochemical parameters and the expression of pro-inflammatory cytokines in broiler chicks. A total of 120 4-day-old birds were given a basal diet (CON) or a basal diet supplemented with 0.5% (AS1) or 1.0% (AS2) AS powder until the birds were 35 days of age. There was no difference in body weight, total gain, feed intake or immune organ weights among the treatment groups. However, the feed conversion ratio in the AS2 group was lower (p<0.05) than that in the CON group. Serum biochemical components, including AST (aspartate aminotransferase), ALT (alanine aminotransferase), albumin and total protein, were not affected by the dietary treatments, whereas glucose and triglyceride levels increased (p<0.05) in the AS2 group compared with the CON group. The AS1 group exhibited decreased mRNA expression (p<0.05) of IFN-${\gamma}$ in white blood cells and iNOS in the liver compared with the CON group. The other pro-inflammatory cytokines were unaffected by dietary AS supplementation, although there was a trend towards decreased expression of these genes, including those encoding Il-$1{\beta}$, IL-6 and TNF-${\alpha}$. In conclusion, dietary supplementation with 0.5% AS decreased the expression of several pro-inflammatory cytokines without affecting growth performance, suggesting that this supplement might be applicable as an immunoregulatory feed additive in broiler chicks.

Effects of Repeated High Ambient Temperature on Performance in Broilers Heat-Conditioned at an Early Age (반복적인 고온환경이 사전고온 적응한 육계의 생산성에 미치는 영향)

  • Heat conditioning at an early age has been known to help chickens cope with heat stress later in life. The present study was conducted to determine the effects of heat conditioning at 5 days of age in broilers repeatedly exposed to high ambient temperature later in life. A total of 256 day-old Arbor Acre boiler chicks were housed in two identical rooms with a 23-h light/1-h dark cycle and provided with feed and water ad libitum. At 5 days of age, the birds in one room were exposed to $37^{\circ}C$ for 24 hours, while those in the other room served as controls. On day 21, half of the birds in each room were moved into the other room so that each room contained both control and heat-conditioned birds. After a 7-day adaptation period, the birds in one room were exposed to high ambient temperature ($21^{\circ}C{\rightarrow}31^{\circ}C$) for 3 days, whereas those in the other room were kept at normal temperature. The same 3-day exposure to high ambient temperature was repeated two weeks later. Hence, there were four treatment groups (CON+CON: control+control; CON+HS: control+high ambient temperature; HC+CON: heat conditioning+control; and HC+HS: heat conditioning+high ambient temperature). Repeated heat stress resulted in decreased feed intake, water intake, body weight gain, and spleen weight (p<0.05) and increased rectal temperature (p<0.05), mortality, and plasma corticosterone concentrations. The relative weight of the spleen was increased in the heat-conditioned group (p<0.05). Plasma biochemicals were also influenced by high temperature. Thus, no beneficial effects of heat conditioning at an early age were detected in broilers repeatedly exposed to high ambient temperature later in life.
