Testicular Toxicity of 2-Bromopropane in Male Sprague-Dawley Rats (2-Bromopropane의 수컷 랫트를 이용한 고환 독성시험)

  • 2-Bromopropane, important industrial chemical, specially in electronic industry at Yangsan in Korea has been reported to cause amenorrhea for female and azoospermia, oligozoospermia or reduced sperm motility for male. 2-BP was investigated through 21 days of repeated dose in male Sprague-Dawley rats. The dose levels per body weight were 0 (control), 250,500 and 1,000 mg/kg. 2-BP dissolved in vehicle olive oil was injected into the intraperitoneum 6 times per week for 3 weeks, but 1,000 mg/kg dose group was 2 weeks because of serious illness. Male rats showed significant decreases in body weight and right and left testis showed typical weight losses depending on the 2-BP. The number of white blood cell and red blood cell , percentage of monocytes, and hemoglobin decreased significantly in high dose (P< 0.05). Red cell volume distribution width increased significantly in the high dose (P< 0.05). Histopathological findings of testes showed a decrease of spermatogenic cells, exfoliation of spermatid and spermatocyte, vacuolization of Sertoli cells and hyperplasia of Leydig cells. Protein band density between 113,000 dalton ($\beta$-galactosidase) and 53,900 dalton (ovalbumin) has decreased in 250 mg/kg dose group, but it has gradually increased to the higher density in 1,000 mg/kg dose group than in control group.

Postnatal cytomegalovirus infection in an extremely premature infant transmitted via breast milk: A case report (산모의 모유를 통하여 감염된 극소 저체중 출생아에서의 거대세포바이러스 감염)

  • Cytomegalovirus (CMV) is one of the most commonly encountered viral pathogens in newborn infants and is found in 0.3-2.4% of all live births. It has been demonstrated that 40-96% of seropositive mothers shed the virus via their breast milk. Breast milk containing CMV can cause almost one-third of CMV infections occurring in infants. A case of postnatal CMV infection in an extremely premature infant (gestational age $24^{+5}$ weeks, birth weight 750 g) transmitted via breast milk is presented. For neonatal intensive care unit (NICU) management of severe thrombocytopenia, anemia, and sepsis syndrome, the infant received repeated transfusions of platelets; intravenous (IV) immunoglobulins; and gamma- irradiated, filtrated packed red cells and was fed her mother's breast milk since the second week of life. CMV infection was diagnosed with positive CMV immunoglobulin M (IgM) and positive urine CMV culture at the second month of life. Considering the negative CMV IgM and urine CMV culture at birth, postnatally-acquired CMV infection was suspected and confirmed with completely identical nucleotide sequence alignments of the infantile blood isolate and the maternal breast milk isolate. To our knowledge, this is the first case of proven postnatal CMV infection transmitted via breast milk in an extremely premature infant in Korea.

Effect of Maternal Undernutrition on the Growth and Composition of Young Rat Brain (분만전후의 어미쥐의 영양부족이 새끼쥐의 뇌성장발육과 조성에 미치는 영향)

  • A quantitative restriction of maternal diet without changes in quality of diet was given to the Sprague Dawley rats during the third week of gestation and lactation. Half the normal average daily intake of control group was given to deficient groups in this period. Female rats of control group were fed a commercial diet ad libitum throughout the experimental period. Dietary restriction started from birth to weaning in deficient I group and from the 15th day of gestation to weaning in deficient II group. Body and brain weight of offsprings of deficient groups were significantly lower than control group, but the ratios of brain weight to body weight in deficient groups were higher than the control group. Significant difference between deficient groups (I and II) was noticed at weaning. Brain DNA, RNA and total protein of offsprings of deficient groups were significantly lower than control group, but RNA/DNA, brain weight/DNA, and total protein/DNA show that cell number were more affected than the cell size by the maternal dietary restriction during the third week of gestation and lactation. Between the deficient groups, there was a significant difference in brain DNA and RNA, but no significant difference in total brain protein. Effects of Chronic Sleep Fragmentation and Diet Restriction on Appetite-Regulating Hormones and Cardiometabolic Indicators (만성 분절수면과 식이제한이 식욕조절 호르몬 및 심혈관 위험지표에 미치는 영향)

  • Data are limited on biological mechanisms underlying the associations of sleep insufficiency with obesity and dyslipidemia. To explore these mechanisms, we investigated appetite-regulating hormones, stress-related hormones, and cardiometabolic indicators in association with sleep fragmentation, which is a type of sleep disorder. In an experimental study, we randomly allocated 40 Wistar male rats aged 7 weeks into four groups; rats with ad libitum sleep and ad libitum intake (Control), those exposed to sleep fragmentation (SF), those with diet restriction (DR), and those exposed to sleep fragmentation and diet restriction (SF+DR). Amongst them, 13-day chronic sleep fragmentation was applied to the SF and SF+DR groups while 50% reduction in chow intake was applied to the DR and SF+DR groups for 13 days. After these experiments, blood lipid and lipoprotein profiles, leptin, ghrelin, adiponectin, cortisol, epinephrine, and norepinephrine levels were compared among the four groups. In the results, the SF group showed the highest levels of serum ghrelin (P<0.001) and the lowest levels of serum adiponectin (P<0.01). All experimental groups showed higher levels of low density lipoprotein-cholesterol (LDL-C) than the Control (P<0.001). LDL-C levels and the ratio of LDL-C and high density lipoprotein-cholesterol were positively correlated with ghrelin levels (P<0.05) in the SF group, but not in the DR and SF+DR groups. In the SF group, the highest levels of serum free fatty acids were also observed and correlated with lower levels of serum adiponectin, which reflects insulin resistance (P<0.05). Based on these findings, we suggest that chronic sleep fragmentation may induce disturbances in lipid metabolism and appetite-regulating hormones independent of food intake, and these metabolic disturbances may be worse due to insulin resistance related to overeating, which is indicated by elevated ghrelin levels in sleep fragmentation. For persons with sleep insufficiency, anti-atherogenic dietary interventions may be recommended to prevent cardiovascular disease.

Anti-Obesity Effects of Lentinus edodes on Obese Mice Induced by High Fat Diet (표고버섯이 고지방식이로 유도한 비만 흰쥐에 미치는 항비만 효과)

  • The aim of this study was to investigate the anti-obesity effects of Lentinus edodes water extract powder (LEP) in mice fed a high fat diet (HF, 45% kcal fat). Mice were administrated a HF diet supplemented with 1%, 3%, or 5% LEP for 12 weeks. Consumption of HF diet caused increases in body weight, serum lipid profiles, and adipose tissue weights. Serum TC and TG levels in the LEP-supplemented groups were lower than those in the NC group. Supplementation with 5% LEP significantly suppressed body weight gain and reduced the weight of subcutaneous adipose tissue compared to the HF group. HF diet ingestion resulted in higher lipid content and increased lipid peroxidation in the liver. However, LEP supplementation inhibited accumulation of hepatic lipids induced by HF diet, considerably decreased MDA levels, and elevated total antioxidant activity in the livers of mice in the 5% LEP group. Histopathological analysis indicated that the livers of mice fed HF diet developed hepatic steatosis, whereas LEP-treated groups showed small fat droplets. These results suggest that long-term supplementation with LEP may also have an ameliorating effect on HF-induced obesity.

Effects of Aspirin on the Pathogenesis of Acute Lung Injury in Rats Subjected to Hemorrhage (출혈성 쇼크로 인한 급성 폐손상의 발병기전과 아스피린의 효과)

  • Background : For unknown reasons, the serum ferritin concentrations are higher in patients with acute lung injury. A pretreatment with aspirin reduces the acute lung injury in rats subjected severe hemorrhage, and increases the rate of ferritin synthesis in vitro. This study investigated the effect of aspirin on the serum ferritin changes in rats subjected to severe hemorrhage. Methods : Hemorrhagic shock was induced by withdrawing blood (20 ml/kg of B.W.) through the femoral artery for 5 min. The rats were pretreated with aspirin (10 mg/kg, i.v.) 30 min before hemorrhage. Results : The protein content and leukocyte count in the bronchoalveolar lavage fluid, lung tissue myeloperoxidase activities were significantly higher after hemorrhage. The aspirin pretreatment prevented these changes. The serum and lavage fluid ferritin concentrations were elevated higher after hemorrhage. These were also attenuated by the aspirin pretreatment. Conclusion : The changes in the serum and lung lavage ferritin level might be closely related to the severity of hemorrhage-induced acute lung injury. Therefore, the serum and lavage ferritin concentrations can be a useful biomarker for patients with precipitating conditions.

Effect of Starvation on Substrate Utilization of Isolated Rat Atria (적출심장의 대사기질 이용에 대한 내인성 기질의 영향에 관한 연구)

  • The abilities of metabolic substrates, glucose, pyruvate, and acetate to produce a maximal increase in the force of contraction of substrate-depleted atria from fed rats were compared to those from starved rats, in order to observe the effect of starvation on substrate utilization of the myocardium. Starvation results in a marked loss of body weight in rats. In contrast to the starved rats, the body weight of fed rats increased with time. When placed in substrate-free medium, atria from fed rats showed marked decline in contractile force. In contrast to the atria from fed rats, the substrate-depleted atria from starved rats showed much less decline of the force of contraction. In the substrate-free medium, abilities of glucose, pyruvate, and acetate to produce a maximal increase in the force of contraction of atria from fed rats were much greater than those from starved rats. The data from these studies indicate that in the substrate-free medium atria from starved rats utilize much less exogenous substrates than those from fed rats. These results suggest that starvation has no deleterious effect on contractile activity of the myocardium, and the starvation increase the storage of readily metabolizable endogenous substrstes useful for the functional activity of the isolated heart.

Aspirin Reduces Acute Lung Injury in Rats Subjected to Severe Hemorrhage (Aspirin이 출혈성 쇼크로 인한 급성 폐손상에 미치는 효과)

  • Background : Hemorrhagic shock and trauma are two of the most common causes of acute lung injury. The activation of cyclooxygenase is one of the important causes of acute lung injury. This study investigated the effect of aspirin, a well-known cyclooxygenase inhibitor, on severe hemorrhage-induced acute lung injury in rats. Methods : The hemorrhagic shock was induced by withdrawing blood; 20ml/kg of B.W., through the femoral artery in 5 min. The mean arterial pressure was recorded through the femoral artery on a polygraph. Results : In the present investigation, the lung tissue myeloperoxidase activity, protein contents and leukocyte counts, in bronchoalveolar lavage fluid, increased significantly 2 and 24 h after the hemorrhage induction. Although the decreased mean arterial pressure spontaneously recovered, acute lung injury occurred after severe hemorrhage. These changes were effectively prevented by a single intravenous injection of aspirin (10 mg/kg of B.W.) 30 min before the hemorrhage. Conclusion : These results suggest that severe hemorrhage-induced acute lung injury is mediated, in part, by the activation of cyclooxygenase. Furthermore, pretreatment of aspirin in acute lung injury-prone patients, or prophylactic treatment of aspirin to the patients with precipitating conditions, could be helpful in the prevention of acute lung injury.

Standard Index Development for Comparing the Glomerular Number (사구체 수의 증감을 비교할 수 잇는 기준 개발)

  • Background: The number of glomerulus has been considered one of the etiologic factors especially for focal segmental glomerulosclerosis. However, glomeruli are too many to calculate them correctly. Although the fractionator method has became convinced, in which they used selected sections, not whole kidney sections, with same intervals, it is also very hard to get good results. Because it is still very time-consuming and laborous work which leads to make big observers' biases. Methods and Materials: We designed the index for glomerular number to estimate the tendency of increase or decrease of the number of it in different kidneys and which was evaluated by other conventional methods including fractionator method. Index was based upon the theory by Nyengaard; "the number of glomerulus correlates with the weight of kidney, which is positively correlated with body weight". Calculating formula is the number of glomeruli/surface areas of cortices, which contain calculated glomeruli multiplies by kidney weight/body weight. Results: We applied this index to kidneys of FGS/Kist mouse and those of RFM/Nga mouse. The former is spontaneous glomerulosclerosis model with heavy protein uria and renal failure and the latter is the mother side of FGS/Kist mouse but has no glomerular disease or protein uria. The number of glomerulus of FGS/Kist mouse was decreased by 30% to those of RFM/Nga mouse. Conclusion: This index was useful and reliable for estimating the relative glomerular number between two groups.

Study on Eating Behavior of Holstein Steers by Growth Stages (Bos taurus) (Holstein 거세 비육우 성장단계별 채식 행동에 관한 연구)

  • This study was carried out to investigate the effects of a different growing stage on the eating and ruminating behavior of Holstein steer (Bos taurus). A total of nine Holstein steers were divided into three growing stage groups (T1: growth stage, 8 months old, T2: Early fattening stage, 15 months old, T3: late fattening stage, 24 months old) with 3 replicates. In the feeding method, concentrate was offered at 1.62% of the body weight per treatment, respectively. And the forage was fed ad libitum for all treatment. The results of behaviors are summarized as follows: The rate of dry matter intake compared to body weight (DMI/BW, %) was high in the order of T1(2.21%) > T2(2.09%) > T3(1.39%). Eating time, ruminating time and resting time were significantly higher in T1, T2 and T3, respectively(p<0.05, 0.01). Chewing time was significantly lower in T3 than in T1 and T2(p<0.01). The number of bolus and the number of total chews were significantly higher in T2 than those of the other treatments(p<0.01). However ruminating time per bolus and the number of bolus per minute were not significantly different among the treatments. The numbers of drinking and defecating were not significantly different among the treatments. The number of urinating was significantly higher T1 than those of the other treatments(p<0.01). Time of drinking, defecating and urinating was not significantly difference among the treatments.
