Characterization of NAD(P)H-nitroreductase Purified from the TNT-degrading Bacterium, Stenotrophomonas sp. OK-5 (폭약 TNT 분해세균 Stenotrophomonas sp. OK-5에서 분리된 NAD(P)H-nitroreductase의 정제 및 특성 연구)

  • The purpose of this work was to perform the characterization of NAD(P)H-nitroreductase isolated from Stenotrophomonas sp. OK-5 capable of degrading 2,4,6-trinitrotoluene (TNT). Initially, NADP(H)-nitroreductase by a series of purification processes including ammonium sulfate precipitation, DEAE-sepharose, andQ-sepharose was prepared. From samples harvested from fraction collector, three different fractions (I, II & III)having the enzyme activity of NAD(P)H-itroreductase were detected. Specific activities of three fractions I, II,and III of NAD(P)H-nitroreductase were determined to approximately 5.06 unit/mg, 4.95 unit/mg and 4.86 unit/mg, and concentrated to 10.5, 9.8, and 8.9-fold compared to crude extract, respectively. Among these three fractions,the fraction I of NAD(P)H-nitroreductase demonstrated the highest specific activity in this experiment. Several factors affecting on the enzyme activity of NAD(P)H-nitroreductase (fractions I, II & III) were investigated.The optimum temperature of all NAD(P)H-nitroreductase (fractions I, II & III) was 30oC, and the optimal pH was approximately 7.5. Metal ions such as Ag+, Cu2+, Hg2+ inhibited approximately 80% enzyme activity of all NAD(P)H-nitroreductase, and the enzyme activities were decreased about 30-40% inhibition in the presence of Mn2+ or Ca2+. However, Fe3+ showed stimulatory effect on the enzyme activity. The molecular weights of NAD(P)H-nitroreductase (fractions I, II & III) were measured about 27 kDa on the SDS-PAGE.

  • A bacterial strain producing intracellular phytase was isolated from cultivable soil near cowsheds and identified as a fluorescent Pseudomonas sp. BUN1. The BUN1 phytase, partially purified by cation and anion exchange chromatography, exhibited its optimal activity at $40^{\circ}C$ and pH 5.5. As for substrate specificity, it was very specific for phytate and showed little activity on other phosphorylated conjugates. Its activity was greatly inhibited by metal ions such as $Cu^{2+}$, $Cd^{2+}$, and $Zn^{2+}$. Addition of corn starch to PSM (phytasesynthetic medium) [0.5% sodium phytate, 0.5% $(NH_4)_2SO_4$, 0.5% KCl, 0.01% $MgSO_4\cdot7H_2O$, 0.01% $CaCl_2\cdot2H_2O$, 0.01% NaCl, 0.001% $FeSO_4\cdot7H_2O$, 0.001% $MnSO_4\cdot4H_2O$; pH 6.5] for the phytase production significantly induced its enzyme activity in comparison with other carbon sources tested.

Ecological Characteristics of Actinomycetes from Mercury and Chrome Polluted Soil (수은, 6가크롬 오염토양으로부터 분리된 방선균군의 생태학적 특성)

  • Ecological characteristics of microbial populations inhabiting heavy metal polluted soil were investigated. The samples were collected from 293 sites around an factory and industry at Gyeoungsangbuk-do. We measured the contents of seven heavy metal elements (Cd, Cu, As, Hg, Pb, $Cr^{6+}$, CN), seven sites have been seriously contaminated by mercury and chrome. A quantitative evaluation of microbial populations in mercury and chrome contaminated soil was examined by using plate count method. Bacterial numbers in polluted soil samples ranged from $7.4X10^5\;to\;9.3X10^7\;cfu\;g^{-1}$, about $10\sim100$ fold less than the count for the unpolluted soil. Moulds were not detected in chrome polluted soil. The log values of actinomycetes of each contaminated soil samples were log ranged from 6.18 to 7.52. The ratio of actinomycetes was similar to unpolluted soil. The investigation showed actinomycetes to be the major microbial population inhabiting the mercury and chrome polluted soil. Thirty-one isolates among the total isolates were examined for antibacterial activity. These isolates were identified based on a phylogenetic analysis using 16S rRNA gene nucleotide sequences, they were categorized in three major phylogenetic groups, belong to the Streptomyces (6 strains), Saccharopolyspora (3 strains), Nocardiodes (1 strain). On the phylogenetic tree, the clade consisting of five isolates were distantly related to all of the established Streptomycetes genera, indicating the possibility as members of new species.

  • The catalytic glycolysis process is the method of chemical recycling where the polyol and carbamate compounds recovered by transesterification reaction are reused to produce new polyurethane foams. In this work, ethylene glycol, diethylene glycol, and 1,4-butanediol were used to decompose polyurethane foams and various metallic acetates were provided as catalysts. The catalytic glycolsis of polyurethane foams was taken place in the reaction temperature of $180{\sim}200^{\circ}C$. The reaction rates of catalytic glycolysis reaction were indicated by the viscosity of the reaction products at different reaction times. IR and GPC analysis showed the types and the molecular weight distributions of the products. The catalytic glycolysis was profitable for using ethyleneglycol at high temperature. The activities of the catalysts are suitable for K, Na, Tl acetate, and the products are composed of comparatively high-contained amine compounds and carbamate compounds. In the case of Sr acetate and Quinoline, the reaction rate was somewhat low. However, the content of polyol was high and the content of the side-products was low. The foams which were prepared by blending up to 20wt% of recovered polyol with virgin polyols showed better physical properties in tensile strength, hardness, tear strength, and compressive strength compared to those of polyurethane foams from virgin polyol.

  • Fatty acid salts derived from soap can be transferred into a typical derivative with p-bromophenacyl bromide using crown ether, a catalizer by the solid-liquid phase transfer reaction in nonpolar, aprotic solvents and separated by the reverse phase high performance liquid Chromatography (RP-HPLC) and determined using UV detector. The minimal limit of detection was defined at approximately 10~50ng in accordance with the chain length. The derivatization reaction in the presence of EDTA can be applied mot only to the calcium salts but also to the other various metal salts. The recoveries of fatty acid derivatizations in the absence and presence of the midium containing the yeast extract were obtained $95.4{\pm}1.2$, and $85.2{\pm}2.4%$ respectively. The analytical method would be applicable to determine the biodegradation of fatty acid salts in nature as well as in artificial condition such as shaker flask-medium method.

  • This paper deals with a waterproof and mechanical feature of concrete using an inorganic self waterproof agent. The waterproof agents having been used in our country were a membrane agent, penetration agent and an organic waterproof agent. However, these agents have a lot of problems such as losing the effect of waterproof in the environment of lots of water, the difficulty of dispersion. For the clear of problems of these water -proof agents, we used the inorganic waterproof agent. This agent was made from inorganic fluosilicate. Generally, a waterproof agent has been used only for the waterproof effect. In this paper, however through the some tests of concrete using the inorganic self waterproof agent, we recognized that the concrete using the agent is more excellent in some peculiar properties than general concrete's properties. In this paper, we performed compressive strength, permeability, pore volume test, etc. As a result, the concrete of using the agent is more excellent in economy, waterproof, compressive strength.

Effect of Inhibitor on Lipoxygenase Inactivation in Soybean Homogenates (대두 현탁액의 Lipoxygenase의 활성저해 인자들의 영향)

  • The effect of several inhibitors such as ascorbic acid on the lipoxygenase in soybeans known to catalyze reaction resulting in racid off-flavors was examined in the soybean homogenates by the oxygen electrode method. Among 8 compounds added at homogenizing process, 10 mM ascorbic acid inhibited lipoxygenase-1 and lipoxygenase-2/3 activities to 41.7 and 49.8%, respectively. Inactivation of lipoxygenase-2/3 was highly accelerated by homogenization for 15 min at room temperature, so the activity was inhibited 70.8% comparing with the homogenization of 3 min. When soybean homogenates with 10 mM ascorbic acid was stored at $25^{\circ}C$ for 72 hrs, lipoxygenase-2/3 activities lowered to 52.8% whereas L-1 activities lowered to 15.8%. Since it is reported that lipoxygenase-2 is responsible for the off-flavor of soybean products, the inhibitory effect of ascorbic acid among several inhibitors investigated might be useful in soybean processing.

Optimization of Conditions for the Preparation of W/O Emulsion Containing Eugenolchitosan (Eugenolchitosan 함유 유중수적형 유화 형성 조건 최적화)

  • Stabilities of W/O emulsions containing eugenolchitosan (EuCs) prepared from chitosan and eugenol were compared to determine the optimal conditions for the ratio of water (core phase) to corn oil (continuous phase), the concentration of EuCs, storage temperature, and the extent of homo-mixing. The optimal ratio of water to corn oil was 2:3 (w/w). The effects of EuC concentrations, and singular vs. binary system of emulsifiers on the storage stability of the emulsion were investigated with EuCs and polyoxyethylene sorbitan monolaurate. The emulsion was stable, showing more than 95% emulsion stability index (ESI) value, when the concentration of EuCs was more than 0.18% (w/v). ESI value of binary emulsifier system was almost equal to that of singular emulsifier system at the concentration of 0.18% (w/v). At this singular emulsifier system, the W/O emulsion formed by EuCs had ESI value of 100%. The optimal concentration of EuCs was determined as 0.18% (w/v). The highest stability of the emulsion was obtained from the homo-mixing at 11,000 rpm for 10 sec and the storage temperature ranging $25{\sim}65^{\circ}C$. EuCs produced from this study was mutagenecity-negative on Ames test and contained no heavy metal ions.

Effect of Vapor-Cooled Heat Stations in a Cryogenic Vessel (극저온액체 저장용기에서 열전도 차폐단의 영향)

  • An experimental study on effect of vapor-cooled heat stations in a 5.5 liter cryogenic vessel has been performed. The cryogenic vessel is made of stainless steel of thickness of 1mm and insulated by the combined insulation of vacuum, MLI(multi-layer insulation) and vapor-cooled radiation shield. Vapor-cooled heat stations are also constructed based on the 1-dimensional thermal analysis to reduce the heat inleak through a filling tube. Thermal analysis indicates that the vapor-cooled heat stations can substantially enhance the performance of vessel for cryogenic fluids with high $C_p/h_{fg}$ where $C_p$ the specific heat and $h_{fg}$ the heat of vaporization, such as $LH_2$ and LHe. The experimental results for $LN_2$ shows that the total heat inleak into inner vessel consists of 14% radiation and 86% conduction through the filling tube. Therefore, it is expected that the conduction heat in leak of the vessel for high $C_p/h_{fg}$ cryogenic fluids can be significantly reduced. powders. The amount of copper coating was 20wt%. In order to examine corrosion behavior of the electrodes, the corrosion current and the current density, in 6M KOH aqueous solution after removal of oxygen in the solution, were measured by potentiodynamic and cyclic voltamo methods. The results showed that Co in the alloy increased corrosion resistance of the electrode whereas Ni decreased the stability of the electrode during the charge-discharge cycles. The electrode used Si sealant as a binder showed a lower corrosion current density than the electrode used PTFE and the electrode used Cu-coated alloy powders showed the best corrosion resistance.

Studies on Saccharification and Citric Acid Fermentation of Alcoholic Distillery Waste(I) (주정증류 폐기물의 당화 및 구연산 발효에 관한 연구(I))

  • Alcoholic distillery waste was utilized as dual purposes to produce citric acid and to reduce the amount of waste to be treated. Enzyme and acid hydrolysis of this waste were studied to suggest effective way of present purpose. Enzymatic hydrolysis of this naked barley alcoholic distillery waste by $\alpha$-and $\beta$-amylase gave glucose as 8g/l concentration at $55^{\circ}C$ for 6 hours, which produced 1g/l citric acid and 5.33g/l mycelial. This waste material hydrolyzed with 25% HCl at $120^{\circ}C$ showed 21.5g/l glucose and produced 1.75g/l citric acid with 4.9g/1 mycelial. The glucose concentration was decreased to 3.44g/l by further 2nd acid hydrolysis because the monosugars were decomposed at prolonged hydrolysis conditions. The addition of 3g/l $NH_4NO_3$ increased the mycelial growth but reduced the amount of citric acid formed. The formation of citric acid was increased at low concentration of manganese ion.
