Effect of Natural Plant Components on the Nitrite-scavenging (천연식물성분이 아질산염 소거에 미치는 영향)

  • The purpose of this presents is to investigate, using natural food extracts(Green tea; Camellia sinesis, Du'chung; Eucommia ulmoides Oliver, Eu sung cho; Houttuynia cordate Thunb, Sam back cho; Saurus Chinensis, Baek hwa sa seal oho; Oldenladia diffusa Roxb., Laver; Porphyra tenera, Sea mustard; Undaria pinnatifda and Sea staghorn; Condium fragile) and juices(Sweet pepper; Capsicum annuum var. angulosum, Kale; Brassia oleracea var. acephala, Cucumber; Cucumis sativus, Onion; Allium cepa, Tomato; Lycopericon esculentum, Maesil; Prunus mume, Plum; Prunus salicina and Grape; Vitis spp.), the effect of natural plant components on the nitrite-scavenging under the different levels of pH. From the above mentioned extracts and juices, the content of vitamin C was detected, containing 65.1∼77.1 mg/100g, at the highest level in the Green tea, and followed by vegetables and fruits in order. The nitrite scavenging effect of teas, medicinal plants, and seaweed extracts, in the reaction system under the condition of pH 1.2, were 57.0∼100%, 50.0∼100%, and 18.0∼99.0%, respectively.   • Medicinal plants resources are becoming important assets since their usages have been expanded to the development of functional foods for human health, natural cosmetics, and pharmaceutical industries. However, names are different from each country and their phylogenetic origins are not clear. These lead consumers to be confused. In particular, when they are morphologically similar and distributed as dried roots, it is extremely difficult to differentiate their origins even by specialists. Recently, molecular markers have been extensively applied to identify the origin of many crops. In this review, we tried to overview the current research achievements for the development of suitable 'origin identification' regarding to the differentiation of Angelica species. Furthermore, more advanced techniques including amplification genome based marker analyses are also discussed for their practical applications in the authentication of particular medicinal plant in Angelica species.

  • 눈개승마 (Aruncus dioicus)는 장미과(Rosaceae) 눈개승마속(Aruncus)에 속하는 다년생 초본식물로 울릉도에서 자생하는 잎줄기 채소이다. 독특한 향기가 특징이며, 인삼맛, 두릅맛, 고기맛이 나는 산채로 세가지 맛이 난다고 하여 예로부터 삼나물로도 불러왔다. 눈개승마에는 caffeic acid, 1-O-Caffeoyl-β-D-glucopyranose 등의 항산화 성분이 다량 함유되어 있으며, 전초는 해독, 편도선염에 효과가 있어 약용으로 쓰이고 어린순은 탄수화물과 무기물 함량이 풍부하다. 비지는 대두를 이용하여 두부를 제조하는 과정에서 발생되는 것으로서 대두로부터 수용성 단백질이 추출되었지만 나머지 약 23%의 단백질, 57%의 식이섬유와 이소플라본 등의 유용한 성분을 다량 포함하고 있다. 또한 비지의 단백질은 다른 식품에는 부족한 황 함유 아미노산과 리신의 함량이 비교적 많고, 두부에 거의 없는 식이섬유가 다량 함유된 고 식이섬유 소재이다. 그리하여, 이러한 콩비지를 고부가가치 식품 소재로 활용하기 위해 건조 미세분말 비지를 제조하고, 이를 산채와의 혼합 가공을 통한 기능증진 효과를 연구하였다. 일반적으로 산채는 쓴맛 제거와 보관성 증진을 위해 데침공정을 주로 하나 산채를 물에 데칠 경우 산채의 유용성분이 대부분 손실된다. 눈개승마의 경우 유용성분이 17.25%, 그 중 폴리페놀 성분이 약 90% 손실된다. 본 연구에서는 기존의 데침공정 대신 찜공정을 이용하여 산채의 유용성분을 보존하고 콩비지를 첨가하여 산채 유용성분의 생이용성 증진 효과를 연구하였다. 눈개승마를 콩비지와 혼합하여 고온고압 가공함으로써 산채의 쓴맛을 줄이고 보관성을 높이는 동시에 눈개승마 폴리페놀의 단량체화를 통해 눈개승마 단독가공에 비하여, 단량체 비율이 약 100~200배 증가하는 것을 확인하였다. 본 연구를 통하여 콩비지의 고부가가치 소재로의 가능성을 확인하였을 뿐만 아니라 산채의 다이어트·건강기능식품 소재로의 산업화에도 기여할 수 있을 것으로 기대된다.

Comparison on Anti-oxidant Effects of Pigment Extracts from Fabaceae 5 species (국내 자생 콩과 5종 염료 추출물의 항산화 효과 비교)

  • The Fabaceae family are being used as traditional medicine. The aim of this study was to compare the antioxidant effects as well as the cell protecting effects of extracts of 5 species (Astragalus membranaceus, Caesalpinia sappan L., Glycyrrhiza uralensis F., Pueraria lobate O., Pterocarpus santalinus L.) in Fabaceae family. The extracts from 5 species were tested by radical scavenging activity test, total phenolic contents and the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay on human liver carcinoma (HepG2) cell line. Anti-oxidant effects of the extracts (5 mg/mL) from C. sappan was 93.49% by radical scavenging activity test. In addition, A. membranaceus extracts showed a weak radical scavenging activity. Anti-oxidant effects of the extracts (5 mg/mL) from A. membranaceus was 7.83% by radical scavenging activity test. Total phenolic contents of the extracts from C. sappan and A. membranaceus were 310.93 mg GAE/g extract, 15.33 mg GAE/g extract, respectively. Cell protecting effects against H2O2 treatment were observed at 100 ㎍/mL concentration of C. sappan and P. santalinus extracts. These results suggest that C. sappan and P. santalinus might be best anti-oxidant in Fabaceae family.

Anticancer Effects of Natural Medicinal Plant Extracts on Oral Carcinoma Cells (천연 약용식물 추출물의 구강상피세포암 세포주에 대한 항암효과)

  • The anticancer effect of medicinal plants against two oral carcinoma cells, A253 and SCC-25 were investigated in this study. Methanol extracts from 63 medicinal plants, which have anticancer activities against other cancers such as stomach, hepatocellular or colon carcinomas, were prepared and screened for their anti- oral cancer activity by using MTT assay. Thirty one samples showed anti-oral cancer activity against either cell line used, however, other 32 samples had no anti-oral cancer activity. Among these samples methanol extract of Caesalpinia sappan revealed the strongest anti-oral cancer activity. The $IC_{50}$/ values of this extract against A253 and SCC-25 cells were 16 and 25 $\mu$g/m1, respectively. Fractions of n-hexane, dichloromethane, ethylacetate, n-buthanol and water were prepared from methanol extracts of Caesalpinia sappan, Anthriscus sylvestris, Rhus japonica, Curcuma arowatica, Inula helenium, Sinoarnudinaria reticulata, and Polygonum cuspidatum, respectively. Among these 35 fractions the n-hexane fraction of Inula helenium showed the strongest anti-oral cancer activity, the $IC_{50}$/ value was 1.6$\pm$0.3 $\mu\textrm{g}$/ml. Ten other fractions showed $IC_{50}$/ values lower than 10 $\mu\textrm{g}$/ml.

Characterization and Anti-Helicobactor pylori Activity of Xanthium strumarium L. Extract on Lactic acid Fermentation (도꼬마리 추출물의 유산발효 특성 및 Helicobactor pylori에 대한 항균활성)

  • This study characterized the anti-Helicobactor pylori activity of Xanthium strumarium L. extract obtained by lactic acid fermentation. The growth of the Lactobacillus strains was typically robust upon lactic acid production in monocultures containing Xanthium strumarium L. extract. Lactic acid fermentation in mixed cultures of Lactobacillus brevis KCTC 3498 and Lactobacillus casei KCTC 3109 produced higher of anti- H. pylori activity than monocultures. Concerning antioxidant activity of fermented extracts, total polyphenol contents were the highest in co-cultures of Lactobacillus brevis KCTC 3498 and Lactobacillus helveticus KCTC 3545. Electron donating ability using diphenyl picryl hydrazyl (DPPH) showed 70% scavenging in Lactobacillus casei KCTC 3109.

Antioxidant and Antiinflammation Activities of Prunus persica Tree Extracts (야생 복숭아나무 추출물의 항산화 및 항염증 효과)

  • Reactive oxygen species (ROS) are continuously produced at a high rate as a by-product of aerobic metabolism. Several lines of evidence provided that ROS appears to cause to develop aging and various diseases. High level of hyaluronic acid with decreased molecular weight has been detected in patients with inflammatory diseases including rheumatoid arthritis. In this study, we have conducted to investigate the antioxidant and hyaluronidase inhibitory activities of Prunus persica Batsch var. davidiana Maximowicz in order to screen the bioactive substances which can be developed as possible anti-inflammatory agents. Among the extracts of Prunus persica Batsch var. davidiana Maximowicz, EtOAc extract exhibited the strongest effect on antioxidant experiment such as DPPH, Ferric- Thiocyanate and Rancimat. Also, EtOAc extract showed a potent hyaluronidase inhibitory activity.

Antimicrobial Effect of Methanol Extracts from Some Medicinal Herbs and the Content of Phenolic Compounds (약용식물 추출물에 대한 항미생물 활성 검색과 폴리페놀 함량)

  • Methanol extracts were prepared from 32 medicinal herbs of the extracts were tested their microbial inhibition activities against food borne pathogens and/or food poisoning microorganisms, food-related bacteria and yeast. Methanol extracts of Cinnamomum cassia, Paeonia suffruticosa, Alnus japonica, Eugenio caryophyllata and Illicium verum exhibited antimicrobial activity for the microorganisms tested, except lactic acid bacteria and yeast. Minimum inhibitory concentrations(MIC) were about 5 mg/mL for Bacillus subtilis, Staphylococcus epidermidis and Pseudomonas aeruginosa. Cell growth of lactic acid bacteria was inhibitied, but greatly on Leuconostoc mesenteroides. The phenolic compound contents were 10.98 mg/g, 10.31 mg/g, 8.55 mg/g and 6.69 mg/g in Thea sinensis, Eugenia caryophyllata, Alnus japonica and Artenisia capillaris, respectively. Antimicrobial activity appeared to be related to phenol compound content in medicinal herbs. The methanol extracts of medicinal herbs could be suitable for the development of a food preservative.

Antimicrobial Characteristics of Edible Medicinal Herbs Extracts (식용가능한 약용식물 추출물의 항균 특성)

  • To develop a natural food preservatives from medicinal herbs (62 species), antimicrobial test was performed with hot water and ethanol extracts against Escherchia coli, Listeria monocytogens and Candida albicans by paper disk method. And its result as a first screening test, 7 kinds of medicinal herbs were selected, and more precise analysis was conducted at last 3 kinds of medicinal herbs was finally selected. Therminalia chebula Retz and Rhus javanica has a broad antimicrobial spectrums and has a low MIC values. In growth inhibition test, they showed a very strong antimicrobial activity aganist Listeria monocytogens. If there has a elaborate approaches in its characteristics and properties, some good result must followed in making a powerful natural antimicrobial preservatives.

  • In order to development of new denture cleaners using natural antimicrobial agents, we have investigated various species of medicinal plants for the antimicrobial activity against Streptococcus matans and Candida albicans. Citrus extract showed significant antifungal activities in a dose-dependent manner against Candida albicans. Candidasis is also a common problem affecting older adults using full denture. We also developed liquid type denture cleaner formulation using citrus extracts.
