Physicochemical Stabilities of Carthamins from Safflower Petals as Food Colorants (홍화 Carthamin의 식품색소로서의 안정성)

  • The physical and chemical stabilities of carthamin of red pigment from safflower petals were investigated at various conditions of pHs, temperatures, inorganic ions, sugars, light and polysaccharides. The half-life values at pH 3.0, 5.0, 9.0 and 11.0 were 5.3, 5.0, 11.0 and 45.0 h, respectively, at $25^{\circ}C$. Therefore, carthamin is unstable at acidic condition. Carthamin was red, orange and yellow at acidic, neutral and alkaline solutions, respectively. At pH 3.0, carthamin was thermally unstable and the half lives were 3.62, 9.05 and 48.2min at $90^{\circ}C$, $70^{\circ}C$ and $50^{\circ}C$, respectively. Among various inorganic ions, $Al^{3+}$ stabilized carthamin at acidic condition. At pH 5.0, carboxymethylcellulose prolonged the half-lives of carthamin at $25{\sim}90^{\circ}C$. Carthamin was very sensitive to light (20,000 lux) and the half-life was 2.32 min at pH 3.0.

Antioxidative and Antimicrobial Activities of Extracts from Different Parts of Crotalaria sessiflora L. (활나무 부위별 추출물의 유지에 대한 항산화 효과 및 항균성에 관한 연구)

  • The antioxidative and antimicrobial properties of the solvent extracts of 3 parts (leaf, stem, root) of Crotalaria sessiflora L. were investigated, in order to find out new natural food additives. The antioxidative activities of the extracts were determined by peroxide value (POV) and the conjugated diene value (CDV) of corn oil stored for 30 days at $60\pm2^{\circ}C$. Each part of the extracts were added as $0.02,\;0.05\%$ and then compared with BHT. The anti oxidative activities were as follows in decreasing order: BHT > LeafEX > StemEX > RootEX > control. The induction period showed that the part of the Crotalaria sessifloria L. group added with solvent extract showed a longer induction period compared with the control group. The part of Crotalaria sessiflora L. solvent extract were shown to have antimicrobial effects on the microorganism such as Bacillus subtilis, Staphylococcus aureus, Listeria monocytogenes, Salmonella Enteritidis, Pseudomonas flrourescens and Escherichia coli. Especially the effect on the Pseudomonas flrourescens was remarkable.

Analytical Method to Quantify Sodium Metasilicate in Shark Fins (샥스핀 가공 중 사용된 메타규산나트륨 분석)

  • According to the Korea Food Additives Code, sodium metasilicate is permitted for use as a filtering agent for edible oils and fats. Sodium metasilicate is currently used as a food additives to increase the weight of shark fins. In this study, we developed an analytical method to quantify sodium metasilicate in food. Sodium metasilicate content was estimated by measuring the moisture content, pH and silicon content of shark fins. Silicon content was analyzed using inductively coupled plasma-optical emission spectrometry (ICP-OES) following microwave-assisted digestion with $HNO_3$ (65%) and $H_2O_2$ (30%). Shark fin total silicon content was $7.17{\pm}8.92mg/kg$, while the soluble silicon content was $2.34{\pm}3.80mg/kg$. After soaking raw shark fin in an aqueous solution of sodium metasilicate, fin weight, pH and silicon content were measured. These results would be used as the basic information for shark fins safety management.

A Study on the Antioxidative and Antimicrobial Activities of the Chopi(Zanthoxylum Pipperitum DC.) Solvent Extracts (초피 용매 추출물의 항산화성 및 항균성에 관한 연구)

  • The antioxidative and antimicrobial activity were carried on the Chopi(Zanthoxylum pipperitum) extracts by six kinds of solvents in order to find out new natural food additives. Six solvents were used methanol(MeOH), n-hexan(hexane), chloroform$(CHCl_3)$, ethylacetate(EtOAc), buthanol(BuOH), and water(water) and methanol extract(MEex), hexan extract(HEex), chloroform extract(CHex), ethylacetate extract(EAex), and buthanol extract(BUex), water extract(WAex). The antioxdative activities of them extracts were determined by peroxide value(POV), conjugated diene value(CDV) of corn germ oil storaged for 30 days at $60{\pm}2^{\circ}C$. These extracts were added as 0.02, 0.1, 0.2% of each extracts and compared with ${\alpha}-tocopherol$, and BHT. The antioxidative activities of 0.02% extract were as follows in decreasing order BUex > WAex, BHT, MEex > HEex, EAex, CHex > TOC, and control. While, BUex and CHex among these extracts were shown to be had antimicrobial effects on the microorganism such as Escheria coli, Salmonella typhimurium, Psedomonas aeruginosa, Staphylococcus aureus, and Listeria monocytogenes. Finally to find out the preference of chopi we had made Yugwanzageon (a kind of shallow fried meat ball)by adding chopi powder. The result were similar to control(not added chopi powder) in case of 0.1% chopi adding Yugwanzageon.(p<0.05) Therefore it was thought to be possible that chopi powder was used Yugwanzageon preperation.

Effect of Chemical Fertilizers and Organic Materials on Soil Actinomycetes Flora (화학비료(化學肥料) 및 유기물(有機物) 시용시(施用時) 방선균(放線菌) Flora의 구성변화(構成變化))

  • This investigation was undertaken to clarify the effects of application of chemical fertilizers (Compound fertilizer and commercial compost) and organic matters (straw and clover) on changes of the composition of soil actinomycetes flora. Actinomycetes were isolated from chemical fertilizer and organic materials treated soils, and then grown on HV agar plate. The isolated strains were classified by Bergey's manual based on the morphological characteristics and color of substrate mycelium of actinomycetes. The number of actinomycetes increased 4 times in control, 36 times in clover, 20 times in straw, 5 times in chemical fertilizer and 4 times in commercial compost treated soil after 14th day of incubation. This result suggests that the application of straw or clover is more effective to increase the number of actinomycetes than compound fertilizer or commercial compost. It also showed that the application of chemical fertilizer or organic materials considerably changed the composition of soil actinomycetes flora. The proportion of streptomyces strains to the isolated total actinomycetes was 62% in control, 60% in clover, 68% in straw, 67% in chemical fertilizer and 64% in commercial compost treated soils at the 14th day.

Studies on Hydrothermal Extracts from Fish Head 2. functional Properties of the Extracts (어체두부열수추출물에 관한 연구 2. 추출물의 기능특성)

  • The hydrothermal extracts were produced from fish head and their functional properties were investigated. The extracts exhibited an antioxidative effect and synergistic effect with $\alpha$-tocopherol by measuring peroxide values. The $5\%$ solution showed more antioxidative effect than $1\%$ solution. The extracts showed cryopropective effect, but the effect was less than sucrose. The solubility of the extracts was over $70\%$ in the all range of pH, and its lowest solubility was shown in pH 3.0. The extracts exhibited an emulsifying activity less than sugar ester (EAI=34.72) and little higher than or similiar to egg white (EAI= 14.69). The extracts showed a relatively high foam activity. The osmolality of the hydrothermal extracts at $1.0\%$ concentration were 23$\~$31 mOsmole which were much lower than 317 mOsmole at $1\%$ NaCl.

Development of Natural Alkaloid-feed Additive for Enhancing Yield Grade of Hanwoo Steers (거세한우 육량향상을 위한 알칼로이드계열 천연물 이용 사료첨가제 개발)

  • This study was indicated that both potato leaves and stems contained certain amount of alkaloids compounds which digested in the rumen of Hanwoo steers. Three candidate plant byproducts such as potato, tomato, and eggplant were collected and dried for making feed additives. Among the candidate byproducts, potato extract was contained 2.05±0.27mg/g of α-chaconine and 0.60±0.08mg/g of α-solanine. These glycoalkaloids was potentially activated as functional compounds to skeletal muscle in beef cattle. However, tomato and eggplant byproducts were not detected any alkaloids compounds. After potato byproducts collected and dried for 24 hours, dry matter of potato byproduct were used as feeding study for evaluating active dose titrations. For evaluating dietary rate of rumen microbes, potato dry matter were treated with in situ hybridization for 72 hours using annulated Hanwoo steers. The glycoalkaloids contained potato dry matter were digested until 12 hours after treatment. Blood concentration of glycoalkaloids were not detected after 24 hours treatment of potato dry matter. These data indicated that potato byproduct were digested with rumen microbes for 24 hours after treatments.

Physicochemical and Textural Properties, and Antimicrobial Effects of Low-fat Comminuted Sausages Manufactured with Grapefruit Seed Extract (자몽 종자 추출물을 첨가한 저지방 세절 소시지의 이화학적, 조직적 특성 및 향균 효과)

  • This study was conducted to evaluate physicochemical and textural properties, and antimicrobial effects of low-fat comminuted sausages manufactured with sodium lactate $(3.3\%,\;SL)$ and various levels $(0.1\~0.3\%)$ of grapefruit seed extract (GSE, DF-100) during refrigerated storage for 10 weeks. Low-fat comminuted sausages (LFCS) has pH ranges of $6.09\~6.26,\;74\~76\%$ moisture, $<3\%\;fat,\;16\~17\%$ protein. The addition of SL $(3.3\%)$ and GSE with various levels $(0.1\~0.3\%)$ didn't impair water holding capacity (WHC), vacuum purge (VP) and Hunter color values (L, a, b). LFCS containing SL $(3.3\%)$ increased hardness and chewiness, whereas most TPA values were not affected by the addition of various levels $(0.1\~0.3\%)$ of GSE. LFCS containing $0.2\%\;or\;0.3\%$ GSE retarded the microbial growth of Listeria monocytogenes(LM). The addition of $0.3\%$ GSE in LFCS showed similar antimicrobial effect to $3.3\%$ SL, which kept $10^3 CFU/g$ until 10 weeks of refrigerated storage. Yellowness, VP and cohesiveness tended to be increased with increased storage time. These results indicated that the addition of $0.3\%$ GSE as a replacer for synthetic particularly paI1icuiarly inhibited the microbial growth of LM, resulting in antimicrobial effect similar to those of $3.3\%$ SL treatment without quality defects.

Studies on the Repeated Toxicity Test of Food Red No.2 for 4 Weeks Oral Administration in SD Rat (SD랫드에서 식용색소 적색2호의 4주간 경구투여에 따른 반복독성시험에 관한 연구)

  • This study was carried out to investigate the toxicity of food Red No.2 in the Sprague-Dawley (SD) female rat for 4 weeks. SD rats were orally administered for 28 days, with dosage of 500, 1,000, 2,000 mg/kg/day. Animals treated with food Red No.2 did not cause any death and show any clinical signs. They did not show any significant changes of body weight, feed uptake and water consumption. There were not significantly different from the control group in urinalysis, hematological, serum biochemical value and histopathological examination. In conclusion, 4 weeks of the repetitive oral medication of food Red No.2 has resulted no alteration of toxicity according to the test materials in the group of female rats with injection of 2,000 mg/kg. Therefore, food Red No.2 was not indicated to have any toxic effect in the SD rats, when it was orally administered below the dosage 2,000 mg/kg/day for 4 weeks.

Comparison of Antioxidant Activities of Radish Bud (Raphanus sativus L.) According to Extraction Solvents and Sprouting Period (추출용매 및 발아시기에 따른 무순 추출물의 항산화 활성 비교)

  • The purpose of this study is to investigate the total polyphenol and antioxidant activities of radish buds (Raphanus sativus L.) based on sprouting periods and extraction solvents in order to present basic data that are needed for using the radish buds as functional food material. The antioxidant activities were assessed by using various antioxidant models (DPPH, TBARS, Rancimat method, POV). The total polyphenol contents according to the extraction solvents were 84.11 and 296.51 mg/g, and the ethanol extract on day 4 of showed the highest value as 296.51 mg/g. As for DPPH radical-scavenging activity, on the day 4 of sprouting, water extracts indicated the highest scavenging activity by 86.67%, and the acetone extracts indicated a rather low scavenging activity as 77.23%. As for TBARS measurement of the radish bud extracts on day 4 of sprouting the extract of 70% ethanol was highest (71.48%). On day 8 of sprouting the TBARS value was increased and the methanol extract was highest (78.99%). As for the oxidative induction period on day 4 of sprouting in Rancimat measurement, the methanol extract was highest (6.07 hours) on day 4 and the antioxidant index was 1.16. On day 12 of sprouting, the general oxidative induction period tended to be reduced to 5.25 to 5.91 hours. In the peroxide value measurement on day 4 of sprouting and beginning of the storage, the extracts showed no difference between 3.02 meq/kg oil and 4.12 meq/kg oil, and on the day 60 of storage, the water extract (43.83 meq/kg oil) and the methanol extract (45.42 meq/kg oil) were lowest with higher antioxidant effect. In conclusion, the radish bud extract with higher total polyphenol contents and antioxidant activities may serve as functional material for food additives, such as natural antioxidants and food preserving agents.
