Anti-oxidative Activity of Five Plant Extracts including Apios fortune, Colubrina arborescens, Croton caudatus, Osmanthus matsumuranus and Schima noronhae (Apios fortunei, Colubrina arborescens, Croton caudatus, Osmanthus matsumuranus, 그리고 Schima noronhae를 포함하는 5종 식물 추출물의 항산화 활성)

  • This study was orchestrated with the purpose of uncovering new nutraceutical resources possessing biological activities in the plant kingdom. To fulfill our objective, we analyzed several plant extracts and selected five species possessing powerful anti-oxidative activity. The anti-oxidative effect of these five plants, Apios fortunei Maxim., Colubrina arborescens Sarg., Croton caudatus Geiseler, Osmanthus matsumuranus Hayata and Schima noronhae Reinw. ethanol extracts were then evaluated by using in vitro assay, cell model system, and Western blot analysis of target proteins. As the results, all of them possessed the potent scavenging activity against 1,1-diphenyl-2-picryl hydrazyl (DPPH), similar with that of ascorbic acid, used as a common positive control. Moreover, they strongly inhibited hydrogen peroxide ($H_2O_2$)-induced reactive oxygen species (ROS), in a dose-dependent manner, in RAW 264.7 murine macrophage cells. Furthermore, they induced the protein expression of an anti-oxidative enzyme, heme oxygenase 1 (HO-1), and its upstream transcription factor, nuclear factor-E2-related factor 2 (Nrf2). Taken together, these results indicate that these five plants possess potent anti-oxidative activity and thus appear to be useful sources as potential anti-oxidant agents. Therefore, they might be utilized as promising materials in the field of nutraceuticals.

Antioxidant activities and inhibitory effects on oxidative DNA damage of leaf from Zelkova serrata with ethyl acetate fractions and hot water extracts (느티나무 잎 에틸아세테이트 분획물 및 열수 추출물의 항산화 및 산화적 DNA 손상 억제 활성)

  • Reactive oxygen species (ROS) has been played a critical role in damage of DNA. Recently, many effort is focusing to develop the natural antioxidants for controlling ROS. Zelkova serrata, Ulmaceae, is close as plants which are planted in front of Korea villages. Although Zelkova serrata is familiar with Koreans, those of antioxidant activities and protective effects on oxidative DNA damage haven't studied. We demonstrated antioxidant activities and inhibitory effects on oxidative DNA damage of Leaf from Zelkova serrata with ethyl acetate fractions (EA) and hot water extracts (HW). Between the extracts, EA showed higher activities in 1,1-diphenyl-2-picryl-hydrazyl, 2,2'-azino-bis[3-ethylbenzthiazoline-6-sulphonic acid radical scavenging, $Fe^{2+}$ chelating and reducing power than HW. Also, those of total phenolic content are 56.63 and 51.61 mg/g respectively. In addition, ${\phi}X$-174 RF I plasmid DNA cleavage assay for inhibitory effect by oxidative DNA damage was both EA and HW has significant protective effect on oxidative DNA damage. The results suggested that leaf from Zelkova serrata with ethyl acetate fractions and hot water extracts have surpassing potential as natural resources with antioxidant and inhibitory effect on oxidative DNA damage.

  • 만감류 5종(한라봉, 천혜향, 황금향, 레드향, 카라향) 부위별 에탄올 추출물의 항산화 활성에 관하여 총 폴리페놀 함량 측정, 총 플라보노이드 함량 측정, DPPH radical 소거 활성 실험을 실시하였다. 만감류 5종(한라봉, 천혜향, 황금향, 레드향, 카라향)의 총 폴리페놀 함량의 경우, 레드향 과피 처리구(93.97mg/g)에서 가장 높았으며, 천혜향을 제외한 모든 만감류 품종 부위들 가운데, 과피 처리구에서 높은 폴리페놀 함량을 나타내었다. 만감류 5종(한라봉, 천혜향, 황금향, 레드향, 카라향)의 총 플라보노이드 함량은 한라봉 전체 처리구(23.65mg/g)에서 가장 높았으며, 모든 처리구에서 30mg/g 함량보다 낮은 결과를 나타내어 총 폴리페놀 함량 대비 낮은 함량을 나타내었다. 이러한 현상은 만감류 5종(한라봉, 천혜향, 황금향, 레드향, 카라향) 가운데 과피 두께의 비율이 영향을 미친 것으로 판단되어진다. 만감류 5종(한라봉, 천혜향, 황금향, 레드향, 카라향)의 DPPH radical 소거 활성의 경우, 한라봉 전체(81.01%)를 시작으로 한라봉 중피(73.13%), 한라봉 외피(63.94%) 처리구에서만 DPPH radical 함량이 EDA 50% 이상 값을 나타내었다. 특히 총 폴리페놀 함량(mg/g)과 총 플라보노이드 함량(mg/g) 모두에서 높은 수치를 기록했던 처리구들 가운데 한라봉 중피(79.75, 23.16), 한라봉 전체(75.51, 23.65) 처리구에서 높은 항산화 활성을 보인 반면, 레드향 과피(93.97, 20.59)의 활성은 49.26%로 DPPH radical EDA 50%에 미치지 못하였다. 이러한 결과는 과피와 과육의 에센셜 오일과 성분의 차이에서 기인된 것으로 추측된다.

  • 수입귤 6종(레몬, 자몽, 청자몽, 오렌지, 만다린, 라임) 부위별 에탄올 추출물의 항산화 활성에 관하여 총 폴리페놀 함량 측정, 총 플라보노이드 함량 측정, DPPH radical 소거 활성 실험을 실시하였다. 수입귤 6종(레몬, 자몽, 청자몽, 오렌지, 만다린, 라임)의 각 부위별 생리활성 실험에서 총 폴리페놀 함량(mg/g)은 청자몽 중피(252.14mg/g)와 청자몽 외피(224.60mg/g), 청자몽 과피(213.13mg/g), 자몽 중피(197.13mg/g) 순으로 모두 150mg/g 이상의 양호한 폴리페놀 함량을 나타내었다. 수입귤 6종(레몬, 자몽, 청자몽, 오렌지, 만다린, 라임)의 각 부위별 총 플라보노이드 함량(mg/g)은 자몽 중피(147.24mg/g)와 청자몽 중피(147.08mg/g), 청자몽 과피(127.74mg/g), 청자몽 외피(122.5mg/g) 순으로 모두 120mg/g 이상의 매우 양호한 플라보노이드 함량을 나타내었다. 수입귤 6종(레몬, 자몽, 청자몽, 오렌지, 만다린, 라임)의 각 부위별 항산화 활성(EDA, %)은 레몬 과피(83.21%)를 시작으로 라임 전체(80.23%), 레몬 과육(77.02%), 레몬 전체(69.85%), 만다린 과피(68.86%), 라임 과육(64.61%) 순으로 EDA 50% 이상 값을 나타내어 양호한 항산화 활성을 보였다.

  • Sambucus williamsii var. coreana have been used as a traditional medical food. This research was conducted to investigate the antioxidants of S. williamsii var. coreana leave and stem extracts. Total phenolic content of S. williamsii var. coreana leaves and stem water extracts were 6.6 and 2.0 mg/mL. The EDA by DPPH free radical scavenging test of S. williamsii var. coreana leaves extracts were 99.5 and 89.7% in water and ethanol extracts contained phenolic 200 ${\mu}g/mL$. The stem extracts were 92.2 and 94.3% in water and ethanol extracts contained phenolic 200 ${\mu}g/mL$. The ABTS radical decolorization activity of water and ethanol extracts from leaves were 79.8 and 99.1% at phenolic 200 ${\mu}g/mL$ and water and ethanol extracts from stem were 90.8 and 97.2% at phenolic 200 ${\mu}g/mL$. The antioxidant protection factor of water and ethanol extracts from leaves were 1.1 PF and 1.1 PF at phenolic 200 ${\mu}g/mL$ and water and ethanol extracts from stem were 1.4 PF and 1.0 PF at phenolic 200 ${\mu}g/mL$. The TBARs of water and ethanol extracts from leaves were 88.7 and 98.1% at phenolic 200 ${\mu}g/mL$ and water and ethanol extracts from stem were 93.6 and 90.6% at phenolic 200 ${\mu}g/mL$. The antioxidative activities of extracts from S. williamsii var. coreana leaves and stem were higher than BHT as positive control. These results suggests that S. williamsii var. coreana extracts have the greatest property as a natural antioxidative source.

Antioxidant Effect of Hibiscus Extract (히비스커스 추출물의 항산화 효과)

  • Due to COVID-19, modern people have come to prefer natural substances as anxiety due to harmful environments and various stimuli has increased. Therefore, in order to find out the appropriateness of hibiscus, which is recognized as a non-toxic plant in traditional medicine, as a natural cosmetic material, the antioxidant effects (polyphenol, flavonoid, DPPH, ABTS) of hibiscus extract were studied, and the following results were obtained. First, the total polyphenol of hibiscus was found to be 433 ㎍/mL when the HSE was 100%. Second, the total flavonoids showed high antioxidant capacity at 488 ㎍/mL in 100% of the HSE. Third, the DPPH radical scavenging ability was found to be 94.04% in the undiluted HES and 89.54% in the diluted HSE 20%. Fourth, the ABTS radical scavenging ability was 98.95% in 100% of the HSE stock solution and 94.84% in the diluted HSE 20%, respectively, showing a high scavenging ability of more than 90%. As a result of these studies, it is thought that the hibiscus extract can be used as an antioxidant raw material for natural cosmetics in the future.

Anticariogenic Activities of Various Plant Extracts (항충치효과를 가진 식물 소재 탐색)

  • As an effort to select powerful anti-cariogenic materials from natural resources, various plant extracts were examined for their anti-S. mutans and anti-glucosyltransferase (GTase) activities. The ethanol extracts of licorice bark, which was produced after water extraction of licorice, showed the most powerful anti-S. mutans as well as anti-GTase activities. When licorice bark was consecutively fractionated with n-hexane, chloroform, ethylacetate, and butanol, the chloroform fraction exhibited the strongest anti-S. mutans activites. This fraction was further fractionated into 4 fractions through a silica gel column, and according to HPLC analysis, anti-S. mutant activities seemed to come mostly from relatively hydrophobic materials.

Effects of Phyto-Extract Mixture on the Nicotine Decomposition (식물추출혼합물의 니코틴 분해능에 미치는 효과)

  • Cotinine, one of nicotine metabolites, has been blown to reduce 4-(methylnitro samino)-1-(3-pyridyl)-1-butanone(NNK)- induced $O^{6}$MeG DNA adducts significantly in A/J mice when administered together with NNK. In order to examine the effects of phyto-extract mixture on the conversion of cotinine from nicotine, cellular and clinical experiments were carried out. When the phyto-extract mixture was added to culture media, human liver cells (FLCFR5) produced cotinine from nicotine 2~3 times compared to the control. The phyto-extract mixture which was microinjected into Xenopus oocyte along with nicotine showed the almost similar production of cotinine compared with the results of hepatic cell culture. In clinical test employing 17 to 20 healthy men, concentrations of urinary cotinine derived from smoking after taking photo-extract mixture increased up to 2 times compared to the control group. These results indicatethat the phyto-extract mixture can increase the metabolic efficiency of nicotine to cotinine, leading to the reduced formation of $O^{6}$MeG DNA adducts.

The Physiological Activities of Bark Extract of Albizia julibrissin (자귀나무(A. julibrissin) 수피 추출물의 생리활성)

  • Three bark extracts of Albizia julibrissin were prepared using water (AW), 70% (v/v) ethanol (AE), and hot water (AHW). Organic solvent fractions were analyzed for total flavonoids and polyphenols, antioxidant activities, and inhibitory activities against xanthine oxidase. Total flavonoid and polyphenol contents of the AHW extract were 8.57 mg/g and 108.67 mg/g, respectively. The SOD-like activities of all extracts, assayed at 1.0 mg/mL, were 10.46-16.73%. The nitrite-scavenging ability of the AHW extract, assayed at pH 1.2, was 60.82%, and the $IC_{50}$ value was $770.18\;{\mu}g$/mL. The electron-donating ability of the AHW extract, at 0.3 mg/mL, was 92.30%; the $IC_{50}$ values of the AW and AHW extracts were $31.31\;{\mu}g$/mL and $36.22\;{\mu}g$/mL, respectively; thus higher than that of ascorbic acid ($39.06\;{\mu}g$/mL). Xanthine oxidase inhibition by the AHW extract, at 1.0 mg/mL, was 94.05%. These results indicate that the AHW of A. julibrissin has potential as a natural antioxidant, for addition to foods and nutraceuticals.

Peroxynitrite and Hydroxyl Radical Scavenging Activity of Medicinal Plants (약용식물의 Peroxynitrite와 Hydroxyl radical 소거 활성)

  • The radical scavenging activities of 9 medicinal plants on peroxynitrite ($ONOO^-$) and hydroxyl (${\cdot}OH$) radical were investigated using in vitro system. The water extracts of 9 medicinal plants showed the protective effect against $ONOO^-$ and ${\cdot}OH$ radical. In particular, Akebia quinata, Aster scaber, Cudrania tricuspidata, Diospyros kaki, Eriobotrya japonica, Lycium chinense, Parthenocissus tricuspidata and Polygonum aviculare exhibited $ONOO^-$-scavenging activity by about 50% at the concentration of $10{\mu}g/ml$. Although those $ONOO^-$-scavenging activities were lower than that of penicillamine (94.08${\pm}$3.04%) as a positive control, Eriobotrya japonica (89.87${\pm}$4.57%) was the most potent scavenger of $ONOO^-$ at the concentration of $10{\mu}g/ml$. Also, Diospyros kaki and Urtica angustifolia showed the strong${\cdot}$OH-scavenging activity than thiourea, positive control, at the concentration of lmg/ml. Our results indicate that 9medicinal plants may act as free radical scavengers and reduce damages caused by oxidative stress associated with $ONOO^-$ and${\cdot}$OH radical.
