Antimicrobial Activity, Quantification and Bactericidal Activities of Licorice Active Ingredients (감초 성분의 항균활성, 정량 및 방부력에 관한 연구)

  • The present study was aimed at investigating the antimicrobial activities of licorice's active ingredients. Four samples of licorice ingredients (glycyrrhizin, liquiritin, liquiritigenin, and isoliquiritigenin) were evaluated for their antimicrobial activities against six skin microorganisms. The bioassay applied for determining the antimicrobial effects employed a disc diffusion assay, the minimum inhibitory concentration, and the challenge test. The ingredients showed antibacterial activities. Especially, isoliquiritigenin has significant antimicrobial activities against two Gram-positive (Bacillus subtilis, Propionobacterium acnes) and two Gramnegative (Escherichia coli, Pseudomonas aeruginosa) bacteria. These samples had much higher antimicrobial activities than synthetic preservatives. Our results reveal that liquiritigenin and isoliquiritigenin could be useful compounds for the development of antibacterial agents for the preservation of cosmetics and foods. The two flavonoids, liquiritigenin and isoliquiritigenin, sourced from Korea, China, Uzbekistan, were quantified using HPLC. The results demonstrated that Korean licorice has two flavonoids (liquiritigenin, isoliquiritigenin) in much higher quantities than was observed in the licorice obtained from the two other countries. Thus, isoliquiritigenin and Korean licorice extract represent new candidates for antimicrobial agents.

Antimicrobial Effect of Rubia akane Nakai Extract on Food-Borne Pathogens (식중독유발 세균의 증식에 미치는 천초근 추출물의 영향)

  • This study was performed to investigate the antimicrobial activity of the Rubia akane Nakai extract against food-borne pathogens. First, the Rubia akane Nakai was extracted with methanol at room temperature and the fractionation of the methanol extract was carried out by using petroleum ether, chloroform, and ethyl acetate, and methanol. The antimicrobial activity of the Rubia akane Nakai extract was determined by using a paper disc method against food-borne pathogens and food spoilage bacteria. The methanol extract of Rubia akane Nakai showed the highest antimicrobial activity against Bacillus cereus and Pseudomonas aeruginosa. Synergistic antimicrobial effect was observed when Rubia akane Nakai extract was mixed with Viscum album var. coloratum extract as compared to each extract alone. Finally, the growth inhibition curves were determined by using methanol extract of Rubia akane Nakai against Bacillus cereus and Pseudomonas aeruginosa. The methanol extract of Rubia akane Nakai had strong antimicrobial activity against Pseudomonas aeruginosa at the concentration of 4,000 ppm. At this concentration, the growth of Pseudomonas aeruginosa was retarded more than 72 hours and up to 48 hours for Bacillus cereus. From these results, it was concluded that the methanol extract of Rubia akane Nakai inhibited effectively Bacillus cereus and Pseudomonas aeruginosa.

Antimicrobial Effect of Lithospermi radix (Lithospermum erythrorhizon) Extract (자초(Lithospermum erythrorhizon) 추출물의 항균특성)

  • The optimum condition for the extraction of antimicrobial substance from Lithospermi radix was investigated. For the purpose of obtaining basic data for the development of natural preservatives and for the prevention of food poisoning accidents, its antimicrobial activity was tested against several kinds of saprophytic microbes and food poisoning bacteria. The optimum condition for extraction of antimicrobial substance was to steep Lithospermi radix into 95% ethanol for 24 hours at room temperature. Antimicrobial activity was observed at the pH range $5.0{\sim}8.0$, but its activity became stronger at acidic condition. The result of ion exchange chromatography was showed that the antimicrobial activity of anionic portion was more apparent than that of cationic portion. The antimicrobial activity against Gram positive bacteria was stronger than that of Gram negative bacteria and the growth of food poisoning bacteria such as S. aureus and V. parahaemolyticus was inhibited in the concentration of 0.1% for 48 hours. As for mold and yeast, the growth of some kinds of these organisms was inhibited in the concentration of 0.1 % for 48 hours and the growth of nearly all the fungi was inhibited in the concentration of 0.15% for 96 hours.

Antimicrobial Effect of Garlic Extract against Pathogenic Bacteria (병원성 세균에 대한 마늘추출물의 항균효과)

  • The purpose of this study is to verify the antimicrobial effect of garlic and black garlic against pathogenic bacteria. For the comparative analysis of antibacterial effects of garlic, Ampicillin $10{\mu}g$ (BBL) was used as control antibiotics. Research experiments were conducted on each of November 2013 and January 2014. Susceptibility to the antimicrobial effect was measured through Kirby-Bauer disc diffusion method and verified according to the standard proposed by the CLSI. Antimicrobial effect of fresh garlic was higher regardless of the method to extract than Ampicillin $10{\mu}g$. In contrast, the manufacturing methods of the black garlic had no effective differentiations. In antimicrobial susceptibility test, black garlic showed resistance to all of 4 strains. However, in the ethanol-extract of fermented black garlic(natural aging of 15 days.) was found the small changes of the growth-inhibition-zone against S. aureus (8 mm)and E. coli(7 mm). This study proposes a variety attempts about the extraction methods of black garlic for the possibility of food preservation.

Effects of Propolis Addition on Quality Characteristics of Oriental Medicinal Seasoning Pork (Propolis 첨가가 한방양념돼지고기의 품질특성에 미치는 영향)

  • Effects of medicinal herbs and bee propolis on pork quality were assessed using formulae A (control), B (boiling water extraction with propolis), and C (boiling water extraction without propolis). Seasoning sauce was made with medicinal herb, spices, total flavonoid, and soy sauce with/without 0.4% propolis (21.8 mg/g), mixed with pork, and stored at low temperature $(4{\pm}1^{\circ}C)$ for 20 days. In formula B peroxide value and thiobarbituric acid content increased with increasing storage period. Formula B showed highest Volatile Basic Nitrogen (VBN) and lowest microbial counts, followed by formulae C and A, receiving higher scores for taste and fragrance.

  • DMZ는 살아있는 생물다양성의 보고로 지난 60여년동안 자연적으로 재생이 일어나고 환경적인 강제 보존 영향으로 높은 생태학적인 가치가 유지되고 있으며, 최근에는 남북교류에 대한 활발한 의지로 DMZ생태자원의 남북공동활용 방안에 대한 이슈가 급부상하고 있다. 이에 본 연구진은 3년전부터 DMZ에서 자생하는 식물에 대한 조사를 진행하여 총 200여종 이상의 자생식물의 표본과 추출물들을 보유하고 있으며, 이 추출물들을 활용 in vitro 와 in vivo 평가를 통해 비임상 평가에서 유효한 효과를 나타내는 후보물질들을 다수 찾아낼 수 있었다. 그 중 조팝나무(Spiraea prunifolia var. simpliciflora)는 쌍떡잎식물 장미과에 속하는 낙엽활엽관목으로 동북아시아 지역에 널리 분포되며 우리나라에서는 중부지방에 주로 서식한다고 알려져 있다. 예로부터 해열 및 소염, 신경통완화 치료등에 이용해왔다고 알려져 있으며 그 속에는 다양한 terpenoids, flavonoid 및 phenolic 화합물이 다량 함유되어 있다고 알려져 있다. 본 연구에서는 조팝나무 추출물을 이용하여 전구지방세포에서의 지방세포분화 억제 및 관련 유전자들의 활성을 확인한 후 고지방식이로 유도된 high-fat diet mouse model을 이용하여 체지방 감소 및 내장지방감소, 간 조직내의 지방량 감소등을 확인하였으며, 혈액분석을 통해 총콜레스테롤과 고중성지방등 동맥경화와 심혈관계 질환을 유도시킬수 있는 지표들에서 억제 활성도 확인하였다. 특히 내장 지방의 경우는 Micro-CT를 통해 정밀한 분석을 진행하였고, 체지방뿐만 아니라 전체 체중감소도 나타나는 것을 확인하였다. 현재 실험을 통해 적출된 간 조직과 지방조직을 이용하여 항 비만 활성의 작용기전을 지속적으로 확인하고 있으며, 이 결과는 국제적인 연구저널에 보고되어 향후 체지방 감소 또는 항 비만 치료제로 개발되는 비임상 연구자료로 활용될 계획이다. 이미 조팝나무에 대한 연구결과는 특허로 출원이 완료되어 PCT출원까지 진행중에 있으며 개별인정형 건강기능식품 개발 기업에 기술이전이 될 예정이다. 또한 원활한 원료 수급을 위해 기초단체 소속 농업기술센터와 원료 재배 및 대량 수급에 관한 논의를 마친 상태로 접경지역 근처 농가소득 증대로도 이어지는 제품화 사례이기도 하다. 이는 접경지역에서 자생하는 원료의 활성을 과학적으로 검증하여 기업과의 연계를 통해 기초시군 단체의 농가 소득과도 연계한 우수한 제품개발 사례로 향후에도 이와 같은 연구성과가 지속적으로 도출되기를 기대해본다.

Utilization of Obosan (Dietary Herbs) I. Effects on Survival, Growth, Feed Conversion Ratio and Condition Factor in Olive Flounder, Paralichthys olivaceus (한방사료 첨가제인 어보산의 효과 I. 넙치의 생존율, 성장, 사료효율 및 비만도에 미치는 영향)

  • The effects of differen concentrations of Obosan as a feed additive dietary herb were examined on survival rate, growth, feed conversion ration and condition factor in olive flounder (paralichthys olivaceus). Effectiveness of dietary Obosan with optimized concentration for 48 weeks were also observed with regard to growth performances and yields. All groups fed diets containing 0.15, 0.3 and 0.6% of Obosan revealed significantly higher survival rate than control group (P<0.05). Growth, feed conversion ratio and condition factor of olive flounder fed diets containing Obosan were considerably improved when compared to those of controls (P<0.05). The 0.3% of dietary Obosan was proven to be the optimal concentration in all parameters tested. The dietary Obosan (0.3%) for 48 weeks showed significantly higher surval rate than control (P<0.05), and also improved yields in weight gain (19.0% improvement), specific growth rate (4.8%), feed conversion ratio(13.6%) and condition factor (10.8%), significantly (P<0.05).

Antimicrobial Effects against Food-Borne Pathogens and Antioxidant Activity of Rhododendron brachycarpum Extract (만병초 추출물의 식중독 유발균에 대한 항균효과 및 항산화활성)

  • This study was performed to investigate the antimicrobial effects against food-borne pathogens and antioxidant activity of Rhododendron brachycarpum ethanol-extract. The antimicrobial activity of the extract was determined using a paper disc-diffusion method, and the diameter of the clear zone was measured. The diameter of the clear zone in the presence of 10 mg of extract was maximal against Bacillus cereus among the three tested Gram-positive bacteria and against Escherichia coli O157:H7 among the five tested Gram-negative bacteria. Analysis of the minimum inhibitory concentration (MIC) showed that the extract exhibited a similar efficacy as that of sorbic acid, a well-known chemical preservative. The growth inhibitory effects of the extract at concentrations of 250, 500, 1,000, and 2,000 mg/L on food-borne pathogens were determined against Staphylococcus aureus, Listeria monocytogenes, Salmonella Typhimurium, and Escherichia coli O157:H7. Growth of the microorganisms was not affected by the extract at concentrations up to 250 mg/L, but it was significantly (p<0.05) inhibited by the extract at concentrations higher than 1,000 mg/L. The antioxidant effects of the extract were examined via measurement of DPPH radical scavenging activity, inhibition of reactive oxygen species (ROS) generation using fluorescent dichlorofluorescien (DCF) assay, and prevention of peroxyl radical- and hydroxyl radical-induced supercoiled DNA breakage. The $IC_{50}$ of the extract for DPPH radical scavenging activity was about half that of ${\alpha}$-tocopherol, which was used as a positive control. DCF fluorescence intensity decreased as the concentration of the extract increased, demonstrating that ROS generation was inhibited in a concentration-dependent manner. The ROS inhibitory effect of the extract was higher than that of ascorbic acid. The extract prevented supercoiled DNA strand breakage induced by peroxyl radical and hydroxyl radical. Thus, the results of the present study demonstrate that the extract exhibits antimicrobial effects against food-borne pathogens as well as potent antioxidant capacity, suggesting that R. brachycarpum could be used as a natural antibacterial agent and effective antioxidant in food.

Comparison of Anti-Oxidative Activities of Gamma-Irradiated Aralia continentalis Extracts for Long-Term Storage of Oriental Medicine (한약재 장기 보존을 위해 감마선 조사한 독활(Aralia continentalis) 추출물의 항산화 효과 비교)

  • In this study, extracts of gamma-irradiated Aralia continentalis for health and beauty food resources were examined on antioxidant activities and increasing extraction yield. In photostimulated luminescence analysis, non-irradiated (0 kGy) sample showed an intermediate result of 891.00 photon counts (PCs), whereas irradiated (5 and 10 kGy) samples showed positive results of 79,956.67 and 282,498.67 PCs, respectively. The thermoluminescence ratio of non-irradiated samples was 0.018, whereas the values of irradiated samples (5 and 10 kGy) were 0.395 and 0.464, respectively. Electron spin resonance (ESR) analysis as a physical technique for irradiation identification of Aralia continentalis showed a pair of peaks on a space of 6.0 mT in the symmetric ESR spectrums. For phenolics of gamma-irradiated Aralia continentalis, water extracts had the highest amounts of 7.61 mg/g at non-irradiated condition (0 kGy) while 50% ethanol extracts had the highest amounts of 6.86 mg/g at 10 kGy irradiation. 2,2-Diphenyl-1-picrylhydrazyl scavenging activity of water and 50% ethanol extracts from non-irradiated and irradiated Aralia continentalis were very high (94.99~100.00%). 2,2'-Azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) radical cation inhibitory activity was over 95.00% at $200{\mu}g/mL$ phenolics. Antioxidant protection factor (PF) showed a very high level of activity (1.38~2.51 PF). Thiobarbituric acid reactive substance inhibitory activity was high (70% and 95%, respectively). Thus, treatment of gamma irradiation can be used as a method of storage for long-term preservation of Oriental medicine.

Effects of Quinone Reductase Induction and Cytotoxicity of the Angelica radix Extracts (당귀 추출성분의 세포독성 효과와 Quinone Reductase 유도활성 효과)

  • Various lines of evidence suggest that dietary components protect the initiation of carcinogenesis. In this study, the ethanol extracts (AGE) and the methanol and hexane partition layers (AGEM, AGEH) of the Angelica radix were screened for their cytotoxic effects using the MTT assay on HepG2, HeLa, MCF7 and SW626 cells and for their ability to induce quinone reductase (QR) in HepG2 cells. AGEM and AGEH of the Angelica radix showed the strongest cytotoxic effects on HepG2 and HeLa cells. Cell growth was inhibited by 99.8% and 99.8% on HepG2 cells and 99.3% and 99.4% on HeLa cells, at dose of $100\;\mu\textrm{g}/ml$ of AGEM and AGEH extracts respectively. AGE and AGEH significantly induced QR activities in the HepG2 cells. The QR activities of HepG2 cells grown in the presence of AGE, AGEH, and AGEM at the concentration of $50\;\mu\textrm{g}/mL$ were 313.5, 273.3 and 133.3 nmol/min/mg protein, respectively. Therefore, based on these studies, Angelica radix may be developed into a potentially useful cancer chemopreventive agent.
