Epizootiology of Perkinsus sp. Found in the Manila Clam, Ruditapes philippinarum in Komsoe Bay, Korea (곰소만에 있어 바지락포자충, Perkinsus sp.의 출현에 관하여)

  • Mass mortality of the Manila clam, Ruditapes philippinarum has been reported all along the west and south coast of Korea for the past several years. As a pathogenic agent, Perkinsus sp., an endoparasitic protozoan has been identified in this study and believed to be responsible for the mass mortalities. Prevalence and infection intensity of Perkinsus sp. was investigated from a Manila clam population inhabiting at Komsoe Bay in the west coast where mass mortality of the clam has been reported. A total of 142 Manila clam, 50 oyster, Crassostrea gigas, 10 ark shell, Scapharca broughtonii, and 5 predatory gastropada, Rapana venosa were examined for the presence and the quantity of Perkinsus sp. Ray's fluid thioglycollate medium method (FTM method) with modified Mackin's infection intensity scale and Choi's quantitative method were used in detecting and quantifying the parasite. All individuals of R. philippinarum examined in this study were infected with Perkinsus sp., indicating $100\%$ prevalence while none of the oysters and the gastropods exhibited the parasite. Six to ten individual hypnospores of Perkinsus sp. were counted from the ark shells. The number of hypnospores in the clam tissues varied from 16,667 to 4,091,667, with a mean number of 1,077,628. Average infection intensity according to Mackin's was 2.87, indicating a moderate infection. A negative correlation was observed between the number of Perkinsus sp. in the tissue and the condition index, a ratio tissue wet weight to shell cavity volume. The clam size and the infection intensity in terms of total number of parasites were positively correlated; the bigger clam, the heavier infection. Such high number of Perkinsus sp. counted in the clams could be enough to cause physiological disturbance of clams, such as retarded growth and reproduction. It is also believed that such a high infection leads mortality of the clam via continuous draining of the energy by metabolic activities and reproduction of the parasites. Correlation between the condition index and the infection intensity observed in this study supports this hypothesis.

  • A simple and selective isocratic HPLC method for the analysis of tissue polyamine contents is described and applied on the study of the changes of the hepatic polyamine contents after partial hepatectomy in male rats. The hepatic polyamines are extracted with 0.4 M perchloric acid containing 2 mM disodium EDTA, and then the extract is redissolved in 100 ul of 1 M sodium carbonate and incubated with 300 ul of FNBT-dimethylsulfoxide (1: 100) mixture. The N-2'-nitro-4'-trifluoromethylphenyl drivatives of polyamines are separated through a ERC-ODS column in an isocratic mode with an acetonitrile-water (80:20) mobile phase within 20 min. per a sample, while monitoring the effluent at 242 nm. This improved method which could detect subnanogram of each polyamines is highly specific and reproducible as evidenced by the application of it on the study of the changes of polyamine contents in the regenerating rat liver after partial hepatectomy.

Effect of Potassium on Corn(Zea mays L.) Seedling Growth under Nutrient Solution Culture (양액재배시(養液栽培時) 칼륨 농도(濃度)가 옥수수(Zea mays L.) 생육(生育)에 미치는 영향(影響))

  • A nutrient solution culture experiment was conducted in the greenhouse at Suwon Campus. Kyung-Hee University from May 23 to July 11. 1995, to determine the effects of potassium application on seedling growth of Kwanganok and Hwengsungok cultivars of corn. Potassium was treated with four different levels, which are 0.248.496 and 744ppm for treatments without potassium (-k), standard potassium concentration (K), double(2k) and triple (3k) of standard potassium concentration, respectively. Plants were harvested at weekly interval for seven weeks. at which time plant height, leaf area and dry matter yield of corn seedling were measured, only chlorophyll content was taken for five weeks from three week after planting. Seedlings growth of Kwanganok and Hwengsungok cultivars was greatly retarded from planting to harvesting time in 3k treatment. The plant height, leaf area and dry matter yield of both cultivars during the whole period were decreased in order of K>2F>-K>3K, but dry matter yield of Kwanganok cultivar was decreased in order of 2K>K>-K>3K. Chlorophyll content of Kwanganok and Hwengsungok cultivars was irregularly fluctuated during growing period. but generally highest in K treatment and lowest in 3k treatment. The crop growth rates of seedlings of both cultivars were slow until the 3rd week after planting. but very fast thereafter. In general, CGR of both cultivars was higher in K and 2k treatments than -k and 3k treatments.

  • This study was carried out to investigate the physiochemical and microbiological changes of dandelion during fermentation. Thirty and fifty percentage raw sugar groups (SFE30 and SFE50) were introduced into dandelions and fermented for 120 days at $15-20^{\circ}C$. This study was conducted to investigate the utilization of sucrose on lactic acid bacteria from dandelions and their effect on the pH, titratable acidity, microorganism and formation of organic acids in dandelions during fermentation. The number of lactic acid bacteria increased remarkably up to 15 days of fermentation and then decreased rapidly thereafter. The maximum number of lactic acid bacteria, 7.9 log CFU/mL was reached at pH 4.17 and the pH of dandelion showed a slight decrease during fermentation and decreased steadily up to 90 days to reach an optimum pH of 4.0. The titratable acidity of dandelions fermented increased during fermentation. The concentration of organic acid, amino acid and free sugar in SFE30 was higher than both SFE50 and DWE. The results of dandelions fermented were remarkably retarded in the 50% raw sugar group compared to the 30% raw sugar group.

  • Collection of organisms for the fisheries resources community were obtained with a set net during 8 months between May, 1985 and May, 1986 on the coast of Pukchon, Cheju Island, Korea. Most of organisms, representing 36 species, were less than 20cm $(93\%)$ in length, and 130g $(91\%)$ in weight. The four most abundant species were jack mackerel, Trachurus japonicus ; squid, Todarodes pacificus : damsel fish, Chromis notatus : and rabbit fish, Siganus fuscescens, which comprised about $83\% in number and about $73\%$ of the total catch. The diversity index of the number of species and information indices for individual and catch were the highest in October and the lowest in Septemer, indicating a close relationship with the change between the species number and catch at that time. A cluster analysis of 17 species was illustrated from the similarity matrix. All of the 17 species were grouped at the 0.2 similarity level. Three groups were present at 0.60 level, whereas species in other groups were sporadic in occurrence.

The Anti-Bacterial Effect of Witch Hazel(Hamamelis virginiana) on Oral Pathogens (Witch hazel(Hamamelis virginiana)의 구강병원균에 대한 항균 효과)

  • An ideal anti-bacterial medication for oral infection requires selective effect on pathogens causing dental caries and periodontal disease but not on normal flora. In addition, it should be less toxic for human and even for environment. This study was to seek such a natural anti-bacterial medication and thus anti-bacterial effect of Hamamelis virginiana was evaluated. Many recent researches on the anti-bacterial effect of natural plant extract and essential oil have reported that natural products can be used as medication for prevention and restrainment of dental caries, halitosis and periodontitis. It has been also reported that Hamamelis virginiana has anti-bacterial effect on Porphyromonas gingivalis, Fusobacterium nucleatum, Capnocytophaga gingivalis, Veilonella parvula, Eikenella corrodens, Peprostreptococcus micros, and Actinomyces odontolyticus. This study evaluated anti-bacterial effect of Hamamelis virginiana on Streptoccoccus mutans, Haemophylus actinomycetemcomitans, and Klebsiella pneumoniae to expand its anti-bacterial effect on other important oral pathogens and eventually to develop its oral care products or apply to clinical purpose. In this study, anti-bacterial tests for antibiotic disk susceptibility, minimal inhibitory concentration and minimal bactericidal concentration were performed to evaluate anti-bacterial effect of Hamamelis virginiana against Streptoccoccus mutans, Haemophylus actinomycetemcomitans, and Klebsiella pneumoniae. The results showed that Hamamelis virginiana has anti-bacterial effect on all pathogen strains tested in this study and furthermore Hamamelis virginiana possesses bactericidal effect other than bacteriostatic effect on Streptoccoccus mutans, Haemophylus actinomycetemcomitans, Klebsiella pneumoniae. This study indicates that a natural anti-bacterial medication for oral diseases can be developed using Hamamelis virginiana.

Comparison of Effect of Various Types of Soybeans on Mutagenicity and Growth of Human Cancer Cell Lines (콩 종류별 항돌연변이 및 암세포 증식 억제 효과 비교)

  • This study compared the inhibitory effects of methanol extracts from yellow and black soybeans (black soybean, Seomoktae and Seoritae) on mutagenicity using the Ames test and growth of human cancer cells (AGS human gastric adenocarcinoma, HT-29 human colon cancer, Hep 3B hepatocellular carcinoma cells). In the Ames test system using Salmonella typhimurium TA100, aflatoxin $B_1$ ($AFB_1$)-induced mutagenicity was significantly inhibited by treatments with the methanol extracts from either yellow or black soybeans in a dose dependent manner (p<0.05). The methanol extracts from various black soybeans tended to have a greater inhibitory effect compared to those from yellow soybeans. As for N-methyl-N'-nitro-N-nitrosoguamidine (MNNG)-induced mutagenicity, the methanol extracts (5 mg/assay) from black soybean, Seomoktae and Seoritae showed 51%, 61% and 53% inhibitory rates, respectively, indicating that Seomoktae, a type of black soybean, had a stronger antimutagenic activity against mutagens (both $AFB_1$ and MNNG). Methanol extracts from black soybeans showed an inhibitory rate of greater than 50% on the growth of human cancer cells (AGS, HT-29 and Hep 3B) and the inhibition was more effective in the methanol extract from Seomoktae. Our results suggested that the methanol extracts from black soybeans showed stronger inhibitory effects on mutagenicity and growth of cancer cells than those from yellow soybean. It is concluded that intake of black soybean can be recommended for improving health.

A Study on Eating Habits of Elementary School Students and the Perception on the Nutrition Education in Curriculum (초등학생의 식습관 실태 및 학교 교육과정에서의 영양교육에 대한 인식)

  • The purpose of this was to present the efficient directions of nutrition education for elementary school children. We examined the problems of eating habits. nutrition knowledge, and perception and expectations on nutrition education of elementary school children in school. This study was carried out using a self-administered questionnaire and subjects were 562 elementary school children in Gyeonggi Province. All data in this research was analyzed through SAS program, and the frequency$(\%)$ and the $mean{\pm}S.D.$ on each question were calculated. Data for each group was analyzed according to sex and the difference among the groups were treated for significance by using chi-square test and Student t-test. About $87.6\%$ of the children have a meal over three times a day, and $76.7\%$ answered that they ate at a certain time regularly everyday. Only $69.6\%$ of subjects said that they ate breakfast everyday, showing very high levels of skipping breakfast. The most common frequency of eating snacks was 'once a day' in both sexes(male $52.2\%$, female $48.6\%$). In the perception on the eating habits, $69.3\%$ of children recognized problems on dieting habits, such as unbalanced diet. irregular eating time, and overeating. The perceived and correct knowledge of nutrition were $99.5\%$ and $86.9\%$, respectively. Most of the children$(99.1\%)$ acknowledged that nutrition education need to be carried out in school. And children want to deeply deal with 'cooking', 'growth and nutrition', 'good eating habits', 'nutrition and disease', These results indicated that elementary school children tended to have undesirable eating behaviors, but the rate of children who recognize the importance and the needs of nutrition education in school was high. Therefore, their parents and teachers need to perform continuous education through a curriculum and life instruction so that students have healthy habits by considering what the problems of nutrition habits are, as the elementary students considerably recognize problems of their own dietary habits and needs of nutrition education.

Relation between ERCC1 Expression in Sputum and Survival after Cisplatin-Based Chemotherapy in Patients with Non-Small Cell Lung Cancer (비소세포 폐암환자의 객담 내 ERCC1 발현정도와 cisplatin 복합화학요법 후 치료반응)

  • Background : Excision repair cross complementing gene 1 (ERCC1) not only has a protective role against carcinogens, but plays an important role in cisplatin-resistance via the repair of cisplatin-DNA adducts. This study investigated the association between the ERCC1 expression levels in sputum and survival after cisplatin-based chemotherapy in patients with inoperable non-small cell lung cancer (NSCLC). Methods : Using the sputum collected from 67 inoperable (stage IIIa-IV) NSCLC patients treated with either taxanes (33 cases) or gemcitabine (34 cases) plus cisplatin, the relative expression levels of ERCC1 and the expression of the tumor specific antigen, MAGE, were examined by the quantitative RT-PCR and RT-PCR, respectively. The response and survival were compared with the relative level of ERCC1 or MAGE expression and the treatment modality. Results : In the sputum, ERCC1 and MAGE was detected in 74.6% and 40.2% of patients, respectively. Using the median ERCC1 level, the patients were classified as having high or low ERCC1 expression. The median overall survival (MST) was significantly longer in patients with a high ERCC1 expression level than those with a low expression level (84 weeks vs. 44 weeks respectively, P=0.017). In the taxene-based treatment group, the MST was longer than the gemcitabine group (79 weeks vs. 47 weeks, respectively, P=0.03). The levels of ERCC1 were significantly higher in patients who were MAGE-positive (P=0.003). In the MAGE-negative patients, the MST was longer in the high ERCC1 group (103 weeks vs. 43 weeks, P=0.008), but not in the MAGE-positive patients (62 weeks vs. 44 weeks, P=0.348). Conclusion : ERCC1 expression in the sputum can be a prognostic factor for survival after chemotherapy in patients with inoperable NSCLC.

Characteristics of Hydrodynamics, Heat and Mass Transfer in Three-Phase Inverse Fluidized Beds (삼상 역 유동층의 수력학, 열전달 및 물질전달 특성)

  • Three-phase inverse fluidized bed has been widely adopted with its increasing demand in the fields of bioreactor, fermentation process, wastewater treatment process, absorption and adsorption processes, where the fluidized or suspended particles are small or lower density comparing with that of continuous liquid phase, since the particles are frequently substrate, contacting medium or catalyst carrier. However, there has been little attention on the three-phase inverse fluidized beds even on the hydrodynamics. Needless to say, the information on the hydrodynamics and transport phenomena such as heat and mass transfer in the inverse fluidized beds has been essential for the operation, design and scale-up of various reactors and processes which are employing the three-phase inverse beds. In the present article, thus, the information on the three-phase inverse fluidized beds has been summarized and reorganized to suggest a pre-requisite knowledge for the field work in a sense of engineering point of view. The article is composed of three parts; hydrodynamics, heat and mass transfer characteristics of three-phase inverse fluidized beds. Effects of operating variables on the phase holdup, bubble properties and particle fluctuating frequency and dispersion were discussed in the section of hydrodynamics; effects of operating variables on the heat transfer coefficient and on the heat transfer model were discussed in the section of heat transfer characteristics ; and in the section of mass transfer characteristics, effects of operating variables on the liquid axial dispersion and volumetric liquid phase mass transfer coefficient were examined. In each section, correlations to predict the hydrodynamic characteristics such as minimum fluidization velocity, phase holdup, bubble properties and particle fluctuating frequency and dispersion and heat and mass transfer coefficients were suggested. And finally suggestions have been made for the future study for the application of three-phase inverse fluidized bed in several available fields to meet the increasing demands of this system.
