This study was undertaken to find the effect of radiation therapy on the healing ability of surgical wounds and on this basis, to find the proper time interval between the radiation therapy and surgery. Two hundred and fifty-two mice were used and a single dose of 2000 cGy was given in each instances to the hind limb of mice. Incisional wounds were produced after varying intervals in the previously irradiated areas and then they were followed up at regular intervals by the measurement of tensile strength. The wounds which received surgery immediately, 1 or 2 weeks after irradiation revealed marked delay and the wounds which received surgery 12, 16 or 20 weeks after irradiation demonstrated slight delay in wound healing in terms of tensile strength measurement. But the wounds which received irradiation 4 or 8 weeks before surgery did not differ much in the wound healing process from that of the control group. Histopathologic studies of the wounds demonstrated epithelization in most instances as quickly as in the control wounds. The appearance of fibroblasts and collagen fibers has delayed momently and appeared to have close correlation with the tensile strength healing curves.
저수준레이저를 이용하여 창상이나 병소의 치유과정에 대한 효과를 조사하기 위하여 많은 연구가 시행되었다. 연구에 의하면 갈륨비소 레이저광이 생체자극효과를 가진다고 하며, 저수준레이저를 조사하면 단백질과 핵산 (DNA) 합성을 자극하여 치은섬유아세포의 증식을 촉진한다고 보고하였다. 외상병소나 근육병소의 치료에 사용된 레이저치료법에 관한 관심이 점증함에 따라 저수준레이저요법 (LLLI)의 치유효과를 설명하기 위하여 분자생물학적 수준의 연구를 시행하기에 이르렀다. 보고에 의하면 Mutans Streptococcide 는 LLLI를 사용시 증식이 촉진되며, 다른 세균에서도 유사한 증식효과가 나타날 것이라고 주장하였다. 그러므로 LLLI가 피부감염을 야기하는 가장 흔한 원인인 Staphylococcus aureus 도 마찬가지로 증식이 촉진되는 지를 조사해볼 필요가 있으며, 또한 감염과 같이 특정 병적 상태에서의 저수준레이저광의 효과는 아직까지 명확하게 밝혀져 있지 않았다. 그러므로 본 연구의 목적은 첫째, Staphyloc occus aureus 의 증식에 대한 저수준레이저광의 효과를 조사하는 실험이며, 둘째 Staphylococcus aureus 로 가염된 피부창상에 대한 저수준레이저광의 효과를 판정하는데 있다. 34개의 Staphylococcus aureus 배양표본을 사용하여 48시간의 세포주기동안 조사기간과 조사시간, 그리고 레이저 펄스(laser pulse)형에 따라 3가지 실험을 시행하여 증식에 가장 효과적인 상태와 가장 비효과적인 상태의 갈륨비소 반도체 레이저펄스를 결정하였다. 이후 지름 약 6 mm의 개방창상을 44마리 백서의 양측 대퇴부에 형성하여 모든 창상에 S. aureus를 감염시켰다. 모든 표본은 펄스형과 조사방법 (중앙조사법과 주변조사법)에 따르는 실험을 하기 위하여 4가지로 분류하였다. 각 백서의 양측 창상중 하나는 1,3,5,7일 마다 각 실험의 방법에 따라 레이저를 조사하고 실험동물의 다른 창상은 대조군으로서 사용하였다. 모든 창상의 면적은 실험 1,3,5,7 일째에 일정한 거리에서 사진촬영하여 면적계를 이용, 측정한 후 통계적인 의의를 조사하였다. 본 연구의 결과는 저수준레이저는 특정 조건하에서 S. aureus의 증식을 촉진하였다. 그러나 S. aureus에 감염된 창상을 저수준레이저로 조사시 치유가 촉진되었다. 중앙 조사법고 주변조사법에 의한 창상치유효과는 통계적인 의의가 보이지 않았다. 따라서 결론적으로 S. aureus 에 감염된 창상에 직접 또는 간접적이든 pulse의 종류에 관계없이 조사하는 경우 치유효과가 나타나는 것은 정사주위 조직의 LLLI 자극효과가 염증의 확산을 억제한다고 말할수 있다.
This study was performed to compare the effects of hydrocolloid(Duoderm$\circledR$, HC in this study) and hydrogel (Nu-Gel$\circledR$, HG in this study) occlusive dressing materials on degree of exudate, wound contraction, epithelialization, and healing of full-thickness skin wound in dogs. Three wounds measuring 2${\times}$2 cm in size were created bilaterally(6 wounds/dog) on the dorsolateral aspect of the trunk of 12 dogs. In each dog, the wounds were treated with HC, HG, and normal saline, respectively. For a 4 week period, the wounds were evaluated gross aspects and histopathological aspects. There were no statistically significant differences between treatment groups in percentage of wound contraction, percentage of epithelialization, and percentage of wound total healing during the first week. Significant differences were first detected on day 14. On day l4(P < 0.01) and 21 (P < 0.05), mean percentage of epithelialization of HG-treated wound was significantly greater than those in HC- and normal saline-treated wound. Mean percentage of wound contraction of HG-treated wound was significantly greater than that in HC- and control wounds on day 21(P< 0.05). On day 21, mean percentage of wound healing of HG-treated wound was significantly greater than that in HC- and control wounds(P < 0.02). On day 1, 4, and 7 after wound creation, although severe infiltration of PMN (polymorphonuclear leukocyte) cells in HC- and control wounds were observed in the subcutis and moderate infiltration of PMN cells in HG-treated wound were observed in the subcutis, we did not detect significant differences. On day 14 after wounding creation, in the wounds treated with HG dressing, epithelial cells were found over the surface, and edema further decreased in the tissue under the wounds, and the granulation tissue was replaced with collagen fibers. On day 21 after wound creation, in HG-treated wound compared with other experimental material-treated wounds, regenerated epidermis covered most of the wound surface, and the granulation tissue was more replaced with collagen fibers than that on day 14. Overall results indicated that the use of hydrogel dressing materials(Nu-Gel$\circledR$) as hydrocolloid dressing (Duoderm$\circledR$) materials and normal saline treatment on full-thickness skin wounds in dogs increased the rate of healing at repair stage.
저출력레이저는 인체조직에 biostimulation effects를 가지므로 구강주위에 발생하는 여려가지 질병에 대한 저출력레이저광의 효과에 관하여 많은 연구가 시도되고 있으며 또한 치료에 응용되고 있다. 감염창상에 저출력레이저 조사시 조직치유의 기전이 세균 성장에 의한 조직손상보다 주위 정상조직의 biostimulation effects가 우세하기 때문이라는 가설을 확인하고저 본 연구를 시행하였다. 백서 7마리를 레이저 조사군과 대조군으로 나누어 감염창상의 면적차이를 비교하여 다음고 같은 결론을 얻었다. 1. 저출력레이저 조사군에서 창상수축율이 현저히 높았다. 2. 부종의 빈도는 저출력레이저 조사시 뚜렷하게 감소하였다. 위의 사실로 보아 저출력레이저 조사시 감염창상의 치유촉진은 주위 정상조직의 biostimulation effect가 세균증식에 의한 조직의 손상보다 우세하기 때문이라는 가설을 확인할 수 있었다.
To investigate the modulation of nerve growth factor (NGF) during corneal epithelial wound healing and the effect of topical NGF on corneal epithelial wound healing in dogs. An axial epithelial defect was created in the right eye using 6mm axial corneal mechanical debridement while the left served as an unwounded control. The tears were collected from both eyes during 1 week and the corneal epithelium was processed for the measurement of NGF at day 0 and 7. The NGF content of tears and corneal epithelium was determined by enzyme-linked immunosorbent assay. In another experiment, the animals were divided into 3 groups. The right eyes in each group were treated every six hours with 200 ug/ml of recombinant human (rh) NGF, murine NGF, or 600 ug/ml of anti-NGF blocking antibody. The left eye of each animal was treated with bovine serum albumin (BSA) to serve as controls. Wound healing was analyzed using NIH image software. Tear NGF was markedly increased in the wounded eyes, relative to tears from control eyes during the early healing period. The NGF content of the corneal epithelium was elevated in the wounded eye (p=0.024). Time to wound closure and rate of epithelial migration were not significantly different between the NGF treated or the NGF antibody treated, and the control BSA treated eyes. Corneal epithelial wounding increased NGF content only on the wounded side during the early healing period. Neither topical recombinant human or murine NGF affected corneal epithelial wound healing in the normal dog.

In this study, the anti-inflammatory effects of Lespedeza cuneata extract on macrophages and wound-healing in wound-induced animal experiments were investigated. In an anti-inflammatory test, 0.1 mg/mL of Lespedeza cuneata extract did not affect growth of RAW 264.7 cells, and Lespedeza cuneata extract suppressed nitric oxide (NO) generation from inflammation-induced macrophages in a concentration-dependent manner. Wounds on the skin of rats were treated with vehicle containing Lespedeza cuneata extract (SSP), vehicle (SCO), and commercial ointment (CCO). The wound and scar sizes in the SSP group were significantly reduced in comparison to the SCO and CCO groups (P<0.05). The epidermis and dermis of the SSP group also recovered faster than the SCO group based on Masson's trichrome staining. The gene expression levels of vascular endothelial growth factor (VEGF) decreased and transforming growth factor-beta 1 (TGF-${\beta}1$) increased in wound tissue from the SSP group compared to that from the SCO group. These results show that Lespedeza cuneata extract accelerates wound-healing through anti-inflammatory activity and induction of collagen regeneration as well as reduces the scar area surrounding wounds. Accordingly, Lespedeza cuneata extract could be useful as a cosmeceutical in the cosmetic industry.

The purpose of this study was to develop an aloin-loaded wound dressing with an enhanced healing effect. The cross-linked hydrogel films were prepared with poly(vinyl alcohol) (PVA) and carboxyl methyl cellulose (CMC) using a freeze-thawing (F-T) method. Their gel properties, release of drug, in vivo wound healing effect and histopathology were then evaluated. In the wound healing test, this aloin loaded PVA/CMC hydrogel showed faster healing of the wound made in rat dorsum than the aloin carbomer 934 gel or the control (carbomer 934 gel) due to phytochemical activity of aloin and moisture of CMC. In conclusion, the aloin-loaded wound dressing composed of 5% PVA, 5% CMC and 0.125% aloin is a potential wound dressing with enhanced wound healing effect.
This experiment was applied to examine the effects of ${\beta}$-glucan originated from Aureobasidium on wound healing infected by 3 strains of bacteria in the normal nude mouse. From the results it was suggested that ${\beta}$-glucan has properties, which may be beneficial in the treatment of wound healing though it does not showed any antibacterial activities against wound infections by three strains such as S. aureus, S. pyogenes and P. aeruginosa. Therefore, it is considered that ${\beta}$-glucan will be promised as a new wound management.
