  • The tolerance evaluation for abalone Haliotis discus hannai embryos was performed using different concentrations of cryoprotective agents (CPAs): dimethyl sulfoxide, ethylene glycol and propylene glycol added to 0.2 M sucrose, respectively. 4-cell, trochophore and veliger were exposed in each CPA with different concentration for 10, 20 and 30 minutes of immersion time. Developmental rates were increased with decreased concentration of every CPA and decreased immersion time, and differed from types of CPA. Developmental rates of veliger in all the CPAs were higher than those of 4-cell and trochophore. The developmental rates and larval activity indices in ethylene glycol were comparatively higher than those in other CPAs and the effective CPA and its concentration for the cryopreservation of the abalone embryos was suggested as 2.0 M ethylene glycol with equilibration time of 30 minutes.

  • Comparisons of growth and survival rate in different culture systems for juvenile abalone were determined in the West Coast of South Korea from June, 2000 to April, 2001. Daily growth rate reached at the maximum by $148.6{\pm}48.96\;{\mu}m/day$ with a stagnant system and at the minimum by $95.3{\pm}21.45\;{\mu}m/day$ with a sprinkler culture system. Survival rate of the juvenile abalone was significantly higher than that in sprinkler culture with a rotary culture (P<0.05). The highest survival rate was 50.0% in the rotary culture and 38.3% was observed in the stagnant culture system. The results from this study indicate that effective survival and growth rate could be predicted in the rotary culture system.

  • Triploid abalone, Haliotis discus hannai, was induced by the cold $(0^{\circ}C\;and\;3^{\circ}C)$ or the heat $(35^{\circ}C\;and\;40^{\circ}C)$ shock procedure with fertilized eggs, 12 min. or 32 min. post fertilization with the various time intervals of shock duration. Fertilization rate of each experimental group was not significantly different from that of corresponding diploid control (P>0.05). However hatching rates and normality rates of triploid larvae were significantly different from those of corresponding diploid control (P <0.05). In heat shock groups at $40^{\circ}C$, fertilization rate of eggs was extremely low $(0\~2.7\%)$ and hatched larvae were not detected in these treatment groups. Incidence of triploidy was confirmed by chromosome count and the highest rates of triploid $(84.0\%)$ revealed in cold $(3^{\circ}C)$ shock with 15 min. treatment duration 12 min. after fertilization. The number of diploid chromosome was 2n: 36, and that of tiploid was 3n=54.

  • Elevated concentrations of suspended solids in the marine enrironment caused by coastal developments have threatened to the marine ecosystem. Effects of suspended solids on the mortality and the modifications of glycogen levels of abalone, Haliotis discus hannai were studied. Abalone were exposed to suspended solids with concentrations of 0 (control), 1,000, 1,500 and 2,000 mg/ L for 96h. These suspended solids had no effect on the mortality of abalone. Significant decreases in the glycogen content of soft tissues were observed over 1,500 mg/ L concentration of suspended solids. These results suggested that abalone could tolerate a high level of suspended solids. However, it is necessary to observe further the long term effects of suspended solids on the physiological responses of abalone.

  • Radiation at very low doses frequently has a stimulating or hermetic effect on growth of organism. Effects of growth and survival rate on various dose of gamma irradiation in the farm culture of juvenile abalone (Haliotis discus hannai) were determined in Hallim (Jeju) from February 2004 to January 2005. The initial shell length of abalone juveniles in this study was average $3.45{\pm}0.4cm$. A change of growth after irradiation $(0{\sim}20Gy)$ was observed for 48 weeks. The highest growth rate was observed in 4 Gy-irradiated group and the lowest growth rate was observed in 20 Gy-irradiated group. The additional research about biochemical changes on juvenile abalone after irradiation should be accomplished. Continuous study for gamma radiation-induced hormesis on growth and metabolism of juvenile abalone will further induce the creation of value in ocean industry.

  • Three feeding experiments were conducted to investigate the effects of herbs (Obosan-C or Obosan-O) as an additive in formulated diets on growth and body composition of juvenile abalone (Haliotis discus hannai), Three replicate groups of abalone initially average weighing 206 mg, 827 mg and 432 mg were fed the each one of experimental diet containing $0\%\;or\;1\%$ Obosan for 10 (experiment 1), 7 (experiment 2) and 19 weeks (experiment 3), respectively. In addition, an imported commercial diet for abalone was also employed to compare growth performance of juvenile abalone. Weight gain of abalone fed the diet containing Obosan-O was significantly (P<0.05) higher than that of abalone fed the control diet in the experiment 3. Moisture, crude protein and ash contents of soft whole body were not influenced by dietary herbs (P>0.05), whereas crude lipid content of abalone fed the diets containing Obosan-C was significantly (P<0.05) lower than that of abalone fed the control diet in the experiment 2. The results indicate that Obosan-O as an additive in formulated diet can improve body growth of juvenile abalone.

  • A 15-week feeding trial was conducted to investigate the effect of formulated diet or macroalgae (Undaria pinnatifida) on the survival, growth and body composition of juvenile abalone (Haliotis discus hannai) under the different rearing environmental conditions (four shelter types and two water temperatures). Water temperature was maintained to $16.4^{\circ}C$ by heated and $13.7^{\circ}C$ by not heated natural sea water. The survival rate, weight gain and shell growth of abalone were affected by diet and water temperature (P<0.001). Survival rate of abalone fed the formulated diets was higher than that of abalone fed the Undaria pinnatifida at natural sea water (P<0.05), but this value was not influenced within abalone stocked at heated sea water in each shelter group (P>0.05). Weight and shell growth of abalone fed the formulated diets were significantly higher than those of abalone fed the Undaria pinnatifida at each temperature or shelter (P<0.05). Moisture, lipid and ash contents of the soft body were mainly affected by feeding diet than temperature or shelter (P<0.05). This study indicate that formulated diet used in this experiment could improve the growth of abalone independent of water temperature or shelter. and heated water could show a better growth of abalone in winter season.

  • A 20-week growth trial was conducted in flow-through aquarum system to investigate the practical dietary carbohydrate sources for juvenile abalone (Haliotis discus hannai). Four replicate grops of the abalone averaging 0.125g were fed one of eight diets containing 24.2% wheat flour (WF), 20% dextrin (DEX), 20% sucorse (SUC), 10% $^{\alpha}$-potato starch+10% $^{\beta}$-potato starch (ab-S), 15% $^{\alpha}$-potato starch (a-S15), 20% $^{\alpha}$-potato starch (a-S20), 25% $^{\alpha}$-potato starch (a-S25), or mixture (MIX) with practical ingredients such as soybean meal, corn gluten meal, cotton seed meal and heat flour. In addition, these formulated diets were compare with macroalgae such as dried sea mustard Undaria (D-SM) or dried sea tangle Laminaria(D-ST). Survival rate, weight gain, shell growth and soft body weight of abalone were not significantly affected by the different dietary carbohydrate sources (P>0.05), whereas those fed a-S15 diet were slightly low. These values of abalone fed D-ST were lowest (P<0.05), followed by those fed D-SM. Lipid contents of soft body from abalones fed a-S25, D-ST or D-SM were significantly lower than those of abalone fed other diets (P<0.05). These data indicate that abalone can equally utilize any carbohydrate sources used in this study.

  • An 18-week growth trial was conducted in flow-through aquarium system to evaluate the practical dietary protein sources for juvenile abalone (Haliotis discus hannai). Three replicate groups of the abalone averaging 0.11g were fed one of ten diets containing casein, white fish meal (WFM), meat meal (MM), feather meal (FM), blood meal (BM), soybean meal (SM), corn gluten meal (CGM), cotton seed meal (CSM), Undaria powder (UP), or wheat flour (WF) as a dietary protein source. In addition, these dietary protein sources were cmpared with algae such as raw Undaria or dried Laminaria. Weight gain of abalone fed the diets containing casein, WFM, SM, CSM, or UP was significantly higher (P<0.05) than those of abalone fed other diets, and this value of abalone fed FM, BM, CGM, or algae was lower than other groups. Shell length, shell width, body wt./shell length ratio, and body wt./shell width ratio of abalone fed casein, WFM, SM, CSM, UP, and WF were also highe (P<0.05) than those of other groups. There were no significant difference (P>0.05) in moisture and protein contents of soft body among all diets. The data obtained in this study indicate that each of the casein, WFM, SM, CSM or UP is good dietary protein source for juvenile abalone.
