  • 참굴 개체군 역학모델은 양식장의 환경요인들과 생물학적인 요인들이 굴 성장에 미치는 영향을 파악하기 위해 Powell et al.(1992)과 Hofmann et al.(1992)등이 미국산 참굴인 Crassostrea viginica에 적용하면서 개발되었다. 이후 Kobayashi et al.(1997)가 일본산 참굴인 Crassostrea gigas에 적용시키면서 발전되어 왔고, 우리나라에서는 현등(2001)이 가막만에서 참굴의 최적정수용량을 파악하기 위해 적용하면서 처음 시도되었다. (중략)

  • 난소기생충, Marteilioides chungmuensis는 참굴, Crassostrea gigas의 난소 내에 택적으로 기생하여, 참굴의 재생산에 영향을 미치는 것으로 알려져 있다(박 등, 1999). 특히, 1990년대에 들어 우리 나라 남해안의 굴양식장에서 이 기생충의 출현율이 급격히 증가함에 따라, 굴 종묘의 안정적 확보에 큰 차질을 빚고 있다. 따라서, 본 연구는 난소기생충(농림부, 1997)의 출현 특성과 병원성을 밝혀, 난소기 생충이 참굴의 재생산 외에 굴의 양식생태에 미치는 영향을 밝히고자 실시하였다.   • The nucleotide sequence variation of mitochondria1 DNA were investigated by eight restriction endonucleases from two oyster species, Crassostrea gigas collected from two localities of South Korea and one locality of Japan and C. nippona collected from one locality of South Korea. The total mtDNA size in the oyster, C. gigas, from the three localities was approximately 18 kb and that in C. nippona was 22 kb. The restriction fragment patterns of mtDNA in C. gigas from the three localities by BamHI, BgII, and XhoI digestions were identical to one another. The degree of mtDNA sequence divergence of C. gigas between the two localities in Korea was 2% and that between Korean and Japanese C. gigas was 5%. The amount of sequence divergence between the two species of oysters was 42%.

  • The genetic differentiation and characteristics of two oyster populations (Crassostrea gigas) in Korea were assessed based on the restriction fragment length polymorphisms (RFLP) analysis and the restriction patterns of subcloned mtDNA. The restriction fragments of twenty individuals in West Sea revealed an identical pattern, determined by 8 restriction enzymes. On the other hand, two haplotypes having variation at the HindIII site were shown in the specimens from South Sea; minor haplotypes (4 of 20) were similar to the results obtained from individuals in West Sea while major haplotypes were different from those in West Sea. It was suggested that oysters (C. gigas) of West Sea might have been introduced to South Sea. Each mitochondrial DNA from two oyster populations in Korea and from one in Japan was divided to three parts and subcloned into pUC19 to use in genetic studies effectively. Restriction map was constructed based on the cleavage pattern by multiple restriction enzymes.

  • The present work was conducted in order to survey bacterial flora inhabiting in Crassostrea gigas. The Crassostrea gigas, sea water and mud of its living environments were sampled at Goseong bay, Gyungnam and Dolsan, Yeochon-gun, Chungnam at the period of from December 1978 to February 1979. The obtained results are summarized as follows: 1. The range of bacterial counts found in Crassostrea gigas, sea water and mud was respectively 10$^4$~10$^{6}$ , 10$^{5}$ ~10$^{6}$ and 10$^3$~10$^4$. 2. Among the bacterial 382 strains isolated from the those sample sources, Vibrio genus was 43.5%, Pseudomonas 16.7%, Moraxella 11.5% and Flavobacterium-cytophaga 8.9%. 3. Among 173 strains classified as Vibrio genus, 56% of them was found in Crassostrea gigas, 54% in mud and 25% in sea water. 4. Pseudomonas type I was found only in sea water and type III/IV found in sea water, mud and Crassostrea gigas. Especially type III/IV was found much in sea water.

  • 참굴에 다환성방향족탄화수소(PAHs)의 축적성과 이 물질들이 미치는 독성을 평가하기 위해 해양환경에서 빈번히 검출되는 PAHs 중의 하나인 fluoranthene을 사용한 모델연구를 수행하였다. Fluoranthene을 0.01-1ppm의 농도로 2주간 참굴에 노출시켰을 때 참굴 조직내의 fluoranthene의 농도는 노출기간 및 노출농도에 비례하여 최고 40-70배까지 증가하였다. 소화맹낭과 폐각근의 지질 과산화물가는 노출기간 및 노출농도에 관련하여 현저히 증가하였으며 수용성단백질의 함량은 노출기간이 길어짐에 따라 서서히 감소하였다. 그러나 glycogen 함량, nucleoside/nucleotide, DNA 및 RNA 함량은 변화하지 않았다. 이 결과로 fluoranthene은 굴의 조직내로 원활하게 축적함을 확인하였다. 또한 과산화지질의 함량은 fluoranthene에의 오염에 대해 신속히 반응하는 지표의 하나로 추정되며 수용성단백질의 감소도 비록 시간적으로 다소 둔감하기는 하나 또 다른 지표가 될 수 있을 것으로 사료된다.

  • 참굴에 다환성방향족탄화수소(PAHs)의 축적성과 이 물질들이 미치는 독성을 평가하기 위해 해양환경에서 빈번히 검출되는 PAHs 중의 하나인 anthracene을 사용한 모델연구를 수행하였다. Anthracene을 0.01-1ppm의 농도로 2주간 참굴에 노출시켰을 때 참굴 조직내의 anthracene의 농도는 노출기간 및 노출강도에 비례하여 최고 25-85배까지 증가하였다. 소화맹낭과 폐각근의 지질 과산화물가는 노출기간 및 노출농도에 관련하여 현저히 증가하였으나 수용성단백질, glycogen, nucleoside/nucleotide 및 핵산의 함량은 변화하지 않았다. 이 결과로 anthracene은 굴의 조직내로 원할하게 축적됨을 확인하였으며 과산화지질의 함량은 anthracene에의 오염에 대한 예민한 생화학적 지표가 될 수 있을 것으로 사료된다.

Assessment of Immune Parameters of the Wild Pacific Oyster (Crassostrea gigas) using a Flow Cytometry and Neutral Red Retention Assay (유세포 분석기와 Neutral Red Retention Assay를 이용한 자연산 참굴(Crassostrea gigas)의 면역력 측정)

  • Hemocyte parameters of the wild Pacific oyster Crassostrea gigas inhabiting intertidal zones in small bays (Gwangyang and Jinhae Bay) on the southern coast of Korea were evaluated using flow cytometry and neutral red retention (NRR) assay. Morphological features, cell count, mortality, DNA damage, phagocytosis, and lysosomal membrane stability of hemocytes were analyzed. Three types of hemocytes were identified in the oyster hemolymph: granulocytes, hyalinocytes, and blast-like cells. Immune related functions of hemocyte including phagocytosis and lysosomal membrane stability were significantly different among the study areas (P<0.05), while cell count, mortality, and DNA damage of hemocytes were not significantly different. In Gwangyang Bay, phagocytosis of granulocytes and lysosomal membrane stability of oyster hemocytes inhabiting inside bay were significantly lower than those of oyster hemocytes in outside bay (P<0.05), indicating that oysters in inside bay of Gwangyang were relatively suppressed the immunological function in hemocytes. Contrary to Gwangyang Bay, immune parameters of oyster hemocytes in Jinhae Bay not showed the difference between sampling sites. In conclusion, flow cytometry and NRR assay using oyster hemocyte has a powerful tool to investigate the cell level in a short time due to no-preprocessing of material.

Oysters, Crassostrea gigas, as the second intermediate host of Gymnophalloides seoi (Gymnophallidae) (참굴큰입흡충(Gymnophalloides seoi)의 제2중간숙주로서 참굴의 역할)

  • Gymnophalkides seoi has drawn medical attentions since the discovery of the first human case and a highly endemic area on a southwestern coastal island of Shinan- gun, Korea. Marine bivalves especially oysters were strongly suspected as the source of infection. In this study the oysters, Crassostrea girns, naturally produced (rom the endemic area were examined whether they contain gymnophallid metacercariae. All of 50 oysters examined were infected with the metacercaviae of a gymnophallid, with the metacercarial density per oyster of 610 on average (2-4,792 in range). Later they were identified as G. seoi by obtaining adult worms from experimental mice. The metacercariae were unencysted, and firmly attached on the mantle surface of the oysters with their oral sucker. In sectioned specimens they were equipped with the ventral pit, a peculiar organ of the genus Gymophalloides, and non-muscular genital pore which was connected dorsally to the seminal vesicle. The seminal vesicle was in a great majority mono-sac. By this study, it has been confirmed that the oyster is a 2nd intermediate host of G. seoi as well as the major source of human infection with this fluke.

A Studies on the Bio-monitoring using Shell Valve Movements (SVMs) of Pacific Oyster Crassostrea gigas for Toxic Dinoflagellates, Genus Alexandrium (참굴, Crassostrea gigas의 패각운동을 이용한 유독와편모조 Alexandrium 속의 모니터링 연구)

  • We investigated the possibility of a bio-monitoring system for predicting toxic dinoflagellates (Genus Alexandrium) by the measuring shell valve movements(SVMs) of Pacific oyster, Crassostrea gigas (Mollusca: Bivalvia) using the Hall element sensor. We then described the SVMs of Pacific oyster exposed to the toxic algae under laboratory conditions. Pacific oyster used for experiment were fed Isochrysis galbana until they stabilized and kept under hunger conditions for three days to prevent the influence of food before the experiment. Pacific oyster were exposed to the toxic dinoflagellate, A. fundyense, and the potentially toxic dinoflagellate, A. affine. When Pacific oyster were exposed to A. fundyense, SVMs increased over 10 times/hr at low cell densities of 20 cells/mL. SVMs increased again at $14.1{\pm}5.7times/hr$ at 500 cells/mL, and $27.9{\pm}11.1times/hr$ at the high cell density of 5,000 cells/mL. However, in the presence of A. affine, SVMs increased at $6.7{\pm}3.9times/hr$ until 300 cells/mL, while they increased greatly to $15.3{\pm}10.8times/hr$ at 1,000 cells/mL. The SVMs of Pacific oyster indicated differences depending on species for toxic dinoflagellates. Therefore, the SVMs of Pacific oyster could be useful for A. fundyense, but would bedifficult to apply for A. affine.
