Genotoxicological and Acute Toxicological Safeties of Gamma Irradiated Beef (감마선조사 쇠고기의 유전독성 및 급성독성학적 안전성평가)

  • Gamma irradiation at 5 kGy was applied to beefs for evaluation of their possible genotoxicity and acute oral toxicity. The genotoxicity of 5 kGy irradiated beef was evaluated by Salmonella typhimurium reversion assay and in vivo micronucleus assay using mouse bone marrow cells. The results were negative in the bacterial reversion assay with S. typhimurium TA98, TA100, TA1535, TA1537. Clastogenic effects were not shown in vivo mouse micronucleus assay at 5 kGy dose tested. In an acute toxicity test, 5 kGy-irradiated beef was administrated orally at a dose level of 313 to 5,000 mg/kg, and then number of deaths, clinical signs, body weights, and pathological examinations were examined daily for 14 days post-administration. The results indicate that 5 kGy irradiated beef did not show any toxic effect on mice and oral $LD_{50}$ value was over 5,000 mg/kg on ICR mice.

Study on the Growth Factors for Rapidly Cultivating Mycobacterium spp. (마이코박테리움을 신속하게 배양할 수 있는 성장 인자에 관한 연구)

  • Mycobacteria grow slowly. Therefore, a solid medium should be used for eight weeks and a liquid medium for six weeks. The purpose of this study was to find the growth factors that can grow Mycobacterium rapidly and to help develop a solid medium for rapid identification. Three types of Mycobacterium growth factors were evaluated with 10 Mycobacteria by adding activated charcoal, defibrinated sheep blood, and L-ascorbic acid to $Difco^{TM}$ Mycobacteria 7H11 agar (Becton, Dickinson and Company, Sparks, MD, USA). The time to detection and the distinguishability of a colony were compared with that of the current method. In the rapidly growing Mycobacterium, the difference in detection time between the new media and conventional media confirmed that the new media was faster. M. kansasii and M. intracelluare grew faster in 7H11 C than in 7H11 medium. MTB grew faster than the other media in 7H11 C. This study confirmed that the two growth factors affect fast-growing Mycobacteria and slow-growing Mycobacteria. 7H11 C showed better distinguishability than the conventional media in all 10 Mycobacterium due to the color contrast. In particular, when the MTB was grown, the size of the colonies was larger than with other media, so visualization was easy.

The First Neonatal Case of Panton-Valentine Leukocidin-Positive Staphylococcus aureus Causing Severe Soft Tissue Infection in Korea

  • Staphylococcus aureus (SA) is a common cause of skin and soft tissue infections. Panton-Valentine leukocidin (PVL) toxin-producing strain of SA has been discovered worldwide and is known to cause serious infections. However, reports of neonatal infections caused by PVL-positive SA are rare. Here, we report a case of severe skin and soft tissue infection caused by PVL-positive SA in a 7-day-old neonate. The patient was admitted to the emergency room with a history of fever for one day, tenderness, and sensation of buttocks heating. The infant presented with fever, tachycardia, poor general health, progressive tenderness, and edema of the buttocks on the day of admission. Ultrasonography and magnetic resonance imaging revealed necrotizing fasciitis involving the skin, soft tissue, and muscles. Specimens drained from the buttock lesions confirmed the presence of PVL-positive methicillin-resistant SA (MRSA), and there was no bacteremia. She recovered after one month of intravenous antibiotics and surgical drainages. One month after discharge, she was rehospitalized for otitis externa and was infected with MRSA again. Considering the PVL-positive strain, the patient was treated with intravenous linezolid and dressing. The patient underwent decolonization therapy in a 0.5% chlorhexidine bath and recovered completely without sequelae. This case suggests that aggressive drainage and antibiotic treatment are essential for PVL-producing MRSA infections, and additional decolonization is needed to prevent recurrence and community spread.

A study on an evaluation system by factor loadings (요인적재값 가중치를 사용한 평가 시스템에 대한 연구)

  • To quantify an concept we often use Likert summated rating scale of original or standardized variables in case the variables are relatively less. When variables have different scales, standardized values tends to be used rather than the original values. This is also true in evaluating systems. For example, we may use standardized values of local tax levy, population, and etc. and use the summed value of the standardized values to access the degrees of development. In this paper, we propose using a data-driven weighted sum for a scoring system and the way how to obtain the weights. We apply the proposed method to a real data set and find that proposed method is better than the usual summated rating scale.

  • To investigate the nature of the waste materials in the Nanjido Landfill, we have conducted multivariate statistical analysis of geophysical data set comprised of magnetic, gravity, LandSat TM thermal band and surface depression measurement data. Because these data sets show different responses to the depth, we have transformed the observed total field magnetic data and gravity data to the residual reduced-to-pole(RTP) magnetic anomalies and the three dimensional density anomalies, respectively, and utilized the informations about the upper shallow part of the landfills only in the following process. For the statistical analysis at the points of depression measurement, the magnetic, density and LandSat data values at these points are determined by interpolation process. Since the multivarite statistical analysis technique utilizes a clustering algorithm for classification of data set and we have measured the dissimilarity between objects by using Euclidean distance, standardization was applied prior to distance calculation in order to eliminate any scaling effects due to different measurement unit of each data set. The hierarchial grouping technique was used to construct the dendrogram. The optimum number of statistical groups(clusters), which are classified on the basis of geophysical and geotechnical characteristics, appeared to be six on the resulting dendrogram. The result of this study suggests that the dimension and nature of the multicomponent waste landfills can be identified by application of the multivarite statistical analysis technique to integrated geophysical data sets.

Genotoxicological Safety of Hot Water Extracts of the γ-Irradiated Astragali Radix, Atractylodes Rhizoma, and Cimicifugae Rhizoma in Vitro (감마선 조사 황기, 백출 및 승마 열수 추출물의 in vitro 유전독성학적 안전성 평가)

  • As the utilization of medicinal herbs in food and bio-industry increases, safe hygienic technologies for them are demanded. To consider the possibility of application of radiation technology for this purpose, the genotoxi-cological safety of three r -irradiated medicinal herbs were studied. Astragali Radix, Atractylodes Rhizoma and Cimicifugae Rhizoma were irradiated at 10 kGy, and then were extracted with hot water. The genotoxicity of the extracts was examined in two short-term in vitro tests: (1) Salmonella reversion assay (Ames test) in strains of TA98 and TA100; (2) Micronucleus test in cultured Chinese hamster ovary (CHO) cells. The extract was treated at maximum doses of 5 mg/plate in Salmonella reversion assay, and 1 mg/mL in micronucleus test where growth of CHO cells was inhibited by 50%. In Salmonella reversion assay with or without metabolic activation, both ex-tracts of irradiated and non-irradiated herbs showed no significant differences in formation of revertant colonies compared with the negative control. And also in micronucleus test, the incidences of micronucleus in CHO cells cultured with extracts of irradiated herbs were almost same as negative control in less than 3%. These results of two in vitro tests suggest that ${\gamma}$-irradiated herbs do not show mutagenicity and cytogenetic toxicity. Further tests of in vivo genotoxicity and chronic toxicity are needed to ascertain the safety of ${\gamma}$-irradiated herbs.

Distribution and Characterization of Heterotrophic Bacteria Isolated from Deodorizing Biofilters of a Nightsoil Treatment Plant (분뇨처리장의 질소성 악취물질처리를 위한 생물탈취탑내 종속영양세균의 분포 및 특성)

  • The distributions and characteristics of bacteria on the deodorizing biofilters were studied as a part of biofiltration research for odor gas removal. The odor gas originated from a currently operating nightsoil treatment plant were mainly nitrogenous compounds of ammonia and amines. The filter media were consisted various ratio of pine bark, nightsoil cast and compost, and were sampled before and after operation of 153 days. Before operation, the level of bacteria on nitrification agar medium, nutrient agar medium and thiosulfate agar medium were around $10^7{\sim}10^8cfu/g$ media. After operation, the level decreased but still high as $10^5{\sim}10^7cfu/g$ media. The bacteria from reactors before and after operations were either rod or coccus after growth on nitrification agar media. The 45 isolated heterotrophic bacteria were further characterized and Pseudomonas, Bacillus, Flavobacterium, Alcaligenes were dominant genera. Genus Alcaligenes was particularly dominant when the isolates were incubated on nitrification agar media first and then transfered to nutrient media. The portion of Alcaligenes in the biofilter increased after operation, and was higher in the media contained nightsoil cast, which showed better treatment than the media with pine bark only.

  • E. coli LE392, transformed with CDDP-treated pBR322 DNA, was plated on ampicillin containing media. The number of colonies formed on ampicillin containing agar plate was reduced to undetectable level after treat the DNA with 13.3 ${\mu}M$ CDDP. The CDDP-treated pBR322 DNA was susceptible to sing1e strand DNA specific S1 nuclease and it's migration Pattern in agarose gel electrophoresis was changed. These results suggest that CDDP adduction to pBR322 DNA resulted in denaturation of the double helix and changes in it's conformation which ultimately leads In the inactivation of the ampicillin resistant sere.

Suppression of Bacterial Wilt with Bacillus subtilis SKU48-2 Strain (Bacillus subtilis SKU48-2에 의한 풋마름병 발병 억제)

  • Bacterial populations from the rhizosphere were obtained and the efficacy of the bacterial wilt suppression, root colonizing ability and resistance to three kinds of chemical pesticides were assayed. According to these results, SKU48-2 was selected as a potential biological agent to control the bacterial wilt caused by Ralstonia solanacearum. SKU48-2 strain at $10^8CFU/ml$ inoculum was able to suppress the bacterial wilt up to 60% in greenhouse trials. Also, the resistance of SKU48-2 to chemical pesticides make possible to use in combination with chemical pesticides for the control of bacterial wilt. Three different powder formulations of SKU48-2 were developed. The shelf-life of powder formulations was effective up to 6 months of storage. Unformulated bacterial suspension could not be stored for 2 weeks, at which time cell viability was completely lost. According to 16S rDNA sequence data, the SKU48-2 stain was identified as Bacillus subtilis.

Effects of a Ventilator-associated Pneumonia Prevention Program on Incidence Rate and Endotracheal Colonization (인공호흡기 관련 폐렴 예방 프로그램이 폐렴 발생률과 기관내 균집락화에 미치는 효과)

  • Purpose: This study examined the effects of a program designed to prevent ventilator-associated pneumonia (VAP) on VAP rate and endotracheal colonization. The program focused on aspiration prevention and oral care. Methods: A nonequivalent control group post-test only design was utilized. One hundred patients admitted to a medical intensive care unit (MICU) or coronary care unit (CCU) were assigned to either a experimental group (n=50) or a control group (n=50). The participants were selected 48 hours following an endotracheal intubation. VAP prevention program given to the experimental group includes keeping the head of the bed to $30^{\circ}{\sim}45^{\circ}$ high, maintaining continuous endotracheal cuff pressure at 25 cm $H_2O$, performing endotracheal suction before change position, and providing oral care with 0.1% chlorhexidine every four hours. The control group received usual care. Data were analyzed using t-test, $x^2$ test, Mantel-Haenszel $x^2$ and Cox proportional harzard regression model. Results: The experimental group showed a lower VAP rate than the control group although the difference was not statistically significant ($x^2=0.79$, p=.375). The experimental group showed lower colonization in tracheal secretion than the control group ($x^2=14.59$, p<.001). Conclusion: Results showed that a VAP prevention program is effective in reducing colonization of tracheal secretion. Therefore, VAP prevention programs are recommended as an ICU nursing intervention.
