  • 미꾸리속의 미꾸리(Misgumus anguillicaudatus)와 미꾸라지(M. mizolepis)의 종내 및 종간 유전적 변이와 유전적 차이를 알아보고자 2종 15개집단과 대만산 Paramisgumus김늠bwanus 1개 집단을 대상으로 전기영동법의 의한 동위효소분석을 실시한 결과 각. ongui각caudotus 9개 집단의 평균 유전적 변이정도는 P=34.20%, Ho=0.099, He=0 114였고. M. mizolepgs 6개 집단은 평균 P=35.55%, Ho=0 141, He=0.148이었으며, p연abwonus는 P=45.9%, Ho=0.132. 연e=0.119로서 일반적인 담수어류에 비해 높은 변이를 나타내었다 종간 평균 유전적 근연치를 비교한 결과 M. aneuflISca udatuo와 M. mfzole부랍 사이는 5=0.467이었고. M. angu연흘audatus와 P. dabrvanus 사이는 5=0.475로 비교적 근연관계가 낮았으나 M. mizolep결와 P. dabwanuo 사이는 5=0.834로 매우 가까운 유연관계를 나타냈다 유전적 차이치를 토대로 M. ongu비상라udctus와 M. mfzolep결의 종분화 연대를 산출한 결과 이들은 약 350만년전에 분화된 것으로 추정되었다.

Genetic Variation of Abies holophylla Populations in South Korea Based on ISSR Markers (ISSR 분석에 의한 전나무 집단의 유전변이)

  • Genetic diversity and genetic differentiation in six natural populations of Abies holophylla Max were investigated using ISSR marker system. From 6 ISSR primers, the average percentage of polymorphic loci was 85.6%, and the average expected heterozygosity ($H_e$) was 0.288. From the result of AMOVA, 94.4% of total genetic variation came from the differences among individuals within populations, and 5.6% was caused by those of among-populations. On the basis of Bayesian inference, genetic differentiation (${\theta}^{II}$ and $G_{ST}$) and inbreeding coefficient for all populations were 0.045, 0.038, and 0.509, respectively. The correlation between genetic distance and geographical distance was highly significant at the Mental's test (r = 0.74, P < 0.05). Six populations divided into two groups according to the results of UPGMA and PCA. One group included Namwon, Cheongdo and Mungyeong population. The other was Inje, Hongcheon and Pyeongchang population. Also, in Bayesian clustering analysis, 6 populations were divided into two clusters. But Cheongdo population was assigned into the other cluster unlike those of UPGMA or PCA. Taking the regions based on the results of the cluster analysis into consideration of AMOVA, 3.9% of genetic variation came from the regional difference. The dendrogram from UPGMA could provide the most genetically reasonable explanation for the distribution of Abies holophylla populations in South Korea.

RAPD Variation and Phenetic Relationships for Six Populations of Equisetum pratense in Korea (한국 내 물쇠뜨기 6개 집단의 RAPD 변이와 표현형 관계)

  • The phenetic relationships among six natural populations of Equisetum pratense in Korea were investigated at the population level by constructing a tree based on Random Amplified Polymorphic DNA (RAPD) markers. RAPD analysis was also conducted to estimate genetic diversity and the population structure of E. pratense. A mean of 26.7% at the six population levels indicated polymorphism. E. pratense was found to have fewer alleles per locus (1.267) and fewer effective alleles per locus (1.176). Genetic diversity (0.102) in E. pratense is lower than the average for species with similar life history traits. Total genetic diversity values (HT) varied between 0.112 (OPD-07) and 0.445 (OPD-16), for an average overall polymorphic locus of 0.141. Inter-locus variation in the within-population genetic diversity ($H_S$) was low (0.102). Asexual reproduction, small population size, and the colonization process are proposed as possible factors contributing to the observed low genetic diversity in E. pratense. On a per-locus basis, the proportion of total genetic variation due to differences among populations ($G_{ST}$) ranged from 0.129 for OPD-07 to 0.455 for OPD-09, with a mean of 0.277. This indicated that about 27.7% of the total variation was among populations. Thus, genetic variation (72.3%) resided within populations. This study contributes new information for research on the taxonomy and population genetics of E. pratense.

Geographical Variation of the Oriental Fruit Fly, Bactrocera dorsalis, Occurring in Taiwan (오리엔탈과실파리 유전변이 - 대만 지역 집단변이)

  • This study analyzed genetic variation of the Oriental fruit fly (OFF), Bactrocera dorsalis, which is designated to be a quarantine insect pest in Korea. OFF samples endemic to Taiwan were collected at three different locations (Taipei, Taichung, and Kaohsiung) for three days from July 30 to August 1 in 2018 and assessed in their age and mitochondrial DNA sequence variations. In these places, 1,085 OFF males were collected using methyl eugenol lure while 30 males of Zeugodacus cucurbitae and one male of Bactrocera tau were collected using Cuelure. A protein diet lure attracted 6 flies including one OFF and 5 flies of Z. cucurbitae. Male heads of OFF contained pterin, which increased in contents with age from 32 to $59{\mu}g/head$. There was a local variation in pterin amounts in OFF heads, in which Kaohsiung population had lower amounts of pterin than Taipei and Taichung populations. Genetic distance among these three populations were measured by random amplified polymorphic DNA and showed that Taipei population was separated from Taichung/Kaohsiung cluster. Genetic variation was also analyzed in sequence variations in cytochrome oxidase I (CO-I) and NADH dehydrogenase I (ND-I). There was 7.8% variation in CO-I sequence (360 residues) and 6.6% variation in ND-I sequence (213 residues). These polymorphic sites are proposed to be used to develop SNP (single nucleotide polymorphism) markers characteristic to Taiwan OFF populations.

  • These studies were carried out to examine the anatomical, karyological characteristics, the variation of isozymes in needle and the frost damage of tissue for the purpose of investigating the relationships among the three natural Torreya nucifera populations of Cheju-do, Namhae, and Mt. Naejang in Korea. The results obtained can be summed up as follows : 1. The numbers of endodermal cells and inner layer cells of mucilage canal of needle conductive tissue were different in each population. The number of those of Cheju-do population was the largest. 2. In somatic chromosome, Namhae population showed one more secondary constriction. The values of $b^{arm}/a^{arm}$ were the same in all the three populations, but Cheju-do population particularly showed a different minimum value. And the karyotype formulas of each population showed difference. 3. During the meiosis, each population showed no significant difference in the irregular phenomena of chromosome. 4. In isoperoxidase and esterase variations of needle, each population showed its particular number and variation of bands. Cheju-do population showed the largest number and greatest variation of bands. 5. Under the same freeging conditions, the frost damage of Mt. Naejang population was comparatively slight, and Cheju-do population suffered from a greater frost damage than the others.

Genetic variation in populations of the Korean endemic Eranthis byunsanensis (Ranunculaceae) (한국 특산식물 변산바람꽃(Eranthis byunsanensis)의 유전적 변이)

  • The genetic variation in populations of Eranthis byunsanensis, an endemic and rare species of Korea, was studied using starch gel electrophoresis. All five known populations were sampled for allozyme electrophoresis of nine enzymes coded by 10 loci. The overall genetic variation of E. byunsanensis population was shown to be considerably high within the populations (A = 2.4, P = 90.0, $H_E$ = 0.311). A positive $F_{IS}$ value of E. byunsanensis indicated an overall deficiency of heterozygotes, and a low $F_{ST}$ value (0.131) showed little differentiation among populations. The high genetic variation, less genetic differentiation among populations, and a significant amount of heterozygote deficiency propose the hypothesis that they have an experience of recent isolation and fragmentation of their habitat. Thus, the rate of gene flow has been drastically reduced, and the rate of inbreeding in E. byunsanensis populations has increased. Current habitats in Mai-san and Naro-do are vulnerable due to their small population size and the levels of anthropogenic activity in the region constantly threatening survival of this species. Because of the high genetic variation and low levels of differentiation among populations in E. byunsanensis, it is not issue which populations have a priority for protection, but we may concern the plan to maintain population continuously and diminish the rate of inbreeding.

The Leaf Morphological Variation of Ten Regions of Natural Populations of Machilus thunbergii in Korea (후박나무 10개 천연집단의 엽형질 변이)

  • This study was conducted to examine genetic variation on leaf characteristics of Machilus thunbergii populations. Ten populations were subjected to multivariate analysis for 9 characteristics of leaf morphology. Average length of leaf blade, leaf width, petiole length, vein number were 9.8cm, 4.0cm, 1.8cm, 8.4 respectively, while angle of leaf base and leaf apex were $67.9^{\circ}$ and $78^{\circ}$ respectively. The coefficient of variation (C.V.) on leaf characteristics was 20% which indicate similar features among the populations. Nested analysis showed statistically signigicant differences among populations as well as among individuals within populations. Genetic relationship between populations using complete linkage method showed four groups to Euclidean distance 1.2 and did not show a tendency to cluster into the same group. There were three principal components that had a meaningful eigenvalue over 1.0 among the 9 components. The explanatory power of the top three main components on the total variation was 92.8%. The first principal component (PC) was explained about 40.3% which is mainly correlated with maximum leaf width and the second PC was explained about 28.7% which is correlated with leaf blade length. The third PC was explained about 23.8% which is correlated with petiole length ($X_3$). These characters were important factors for analysis of the relationship among natural populations of M. thunbergii.

Genetic Variation of Larval Stripe Patterns of Spodoptera exigua(Hubner) (파밤나방(Spodoptera exigua (H bner)) 유충 줄무늬 형질의 유전변이)

  • Larval morphological characters such as body color and stripe pattern were analyzed to get morphological genetic markers of beet armyworm, Spodoptera exigua (Hiibner). Body color was varied from light green to dark brown with diets. Stripe characters were classified by the presence of dorsal and lateral lines: three stripes with both lines, one stripe with dorsal line only, and zero stripe. Proportions of each stripe character increased with successive selections for its own character. Three stripe was dominant to one stripe when they were crossed. The estimated heritability in narrow sense (h2) of the stripe pattern was 0.50k0.42. About two fold more females than males were produced in zero stripe line. Stripe pattern was not significantly changed by different diets except in welsh onion which had lower proportion of three stripe individuals than that of the expected. Larval stripe pattern was also correlated with larval and pupal developmental rates and cold hardiness but not with insecticide susceptibility.

Genetic Variation and Structure of the Relict Populations of Korean Arborvitae (Thuja koraiensis Nakai) in South Korea, Employing I-SSR Markers (I-SSR 표지자에 의한 눈측백나무 남한 잔존집단의 유전변이와 구조)

  • We investigated the genetic variation and structure in Korean Arborvitae (Thuja koraiensis Nak.), by 29 examining I-SSR polymorphic loci in 84 individuals distributed among four natural populations in Korea. The level of population genetic diversity ($A_e$=1.44, P=72.42, $H_e$=0.258, S.I.=0.385) was similar to or slightly higher than that of plants with similar ecological traits and life history (Cupressaceae). Most genetic diversity was allocated among individuals within populations (${\Phi}_{ST}$=0.13). The UPGMA dendrogram based on genetic distance failed in showing decisive geographic relationship. The Mt. Bangtae population had the lowest level of genetic diversity and was the most distinctive from the other populations. Mt. Jang population which is possessed of the highest level of genetic variation and Mt. Bangtae population which is consisted of heterogeneous was considered to be a prime candidate for the conservation studies.

Geographic Variation of Lamina Circularis in Cobitis longicorpus (Pisces: Cobitidae) (왕종개 Colitis longicowus (Pisces: Cobitida티 골질반의 지리적 변이에 관하여)

  • 한국 특산종 Cobitis longicorpus (왕종개)의 주요 분류형질인 골절반의 지리적 변이를 조사하기 위하여 남한의 12개집단 205개체를 이용, 골질반의 type분석과 계수학분석을 실시하였다. Type분석 결과, 동해유입하천집단(영덕의 송천, 축산천, 경주 형산강, 울주 회야강)의 골질반은 등근 삼각형, 삼각형, 인각형으로 남해 유입하천 및 도서집단의 타원형, 반타원형, 부채꼴과 구별되었다. 골질반의 계수화분석 결과에서도 동해유입하천의 골질반 폭은 비교적 좁아 남해유입하천 및 도서지방과의 평균 CD값이 1.64로 새종 수준 이상의 차이를 보였다. 아울러 조사, 비교된 제 1 체측반문도 동해유입하천에서는 C. longicorpus의 특징이 전혀 나타나지 않아, 원기재질단인 섬진강집단을 포함한 남해유입하천 및 도서집단과 구별된 새종수준의 종분화가 이루어진 것으로 사료되었다.
