Processing of Powdered Smoked-Dried Anchovy Soup and Its Taste Compounds (훈건멸치 분말수프의 가공 및 정미성분)

  • The study was carried out to develop the powdered smoked-dried anchovy products as a natural flavoring substance. The processing conditions, chemical and taste compounds of products were as follows: The raw anchovy were washed, and then boiled 5 minutes in $5{\sim}6%$ NaCl and 1.0% sodium erythorbate solution. Boiled anchovy were smoked in smoking house at $40^{\circ}C$ for 4 hours as the first stage, and then increased temperature up to $80^{\circ}C$ as the second stage, and finally smoked 8 hrs at $80^{\circ}C$ to maintain the moisture content between 9 and 10 percent. The smoked-dried anchovy were pulverized and screened to be 50 mesh of particle size, and finally packed in PET/Al/CPP film bag. The moisture, crude lipid content and salinity of powdered smoke-dried anchovy were 9.4%, 9.6% and 6.9%, respectively. Fatty acid composition of product was mainly consisted of polyenes (43.4%) such as 22 : 6 and 20 : 5, followed by saturates (36.9%), monoenes (19.7%). The principal taste compounds of product were IMP, 466.5 mg/100g; free amino acids such as His, Tau, Pro, Lys, Ala and Glu, 1179.2 mg/100g; non-volatile organic acids such as lactic acid and succinic acid, 617.9 mg/100g; total creatinine, 595.9 mg/100g; small amount of betaine and TMAO. To make a instant soup, it was desirable for taste of products that powdered smoked-dried anchovy were mixed with 20% salt, 4.0% sugar, 3.0% MSG, 1.0% onion powder, 1.0% garlic powder and 1.0% black pepper.

호소의 부영양화 및 관리

  • 담수는 인간의 생존에 필수적인 요소이다. 우리는 매일 일정량의 물을 섭취하고 있으며, 일상에서 직접, 간접적으로 물과 관련되지 않고서는 생활할 수 없다. 지구 표면의 약 70%가 물로서 덮여 있다고는 하지만 인간의 생활에 직접적인 이용가치가 있는 담수(지표수)는 전체 물의 용적의 2%에 지나지 않는다. 한국의 연평균 강수량은 1,274mm로서 세계 연평균 강수량인 970mm보다 많은 편이나 인구 1인당 연간 강수량은 약 3,000m3으로서 세계 평균인 34,000m3의 11분의 1에 지나지 않는다. 수자원총량중 지하침투와 증발에 의해 손실되는 양을 제외한 하천유출량은 전체의 55%인 697억m3으로서 이것이 곧 가용수자원량이다. 그러나 하절기 강우집중도가 높은 우리나라 실정에서는 가용수자뭔량중 수자원총량의 37%가 홍수시 유출되고 실제 이용량은 수자원총량의 18%인 230억m3에 불과하여 인구증가와 함께 수자원의 가치가 더욱 증대하고 있는 실정이다. 더우기 지하수 이용량은 19억m3에 불과함에 따라 우리나라는 실제 이응수자 원량의 90% 이상을 지표수에 의존하고 있으며, 그 중 약 40%가 호소수로 구성되어 있다. 이러한 수자원 이용효율의 취약성으로 인해 제한된 수자원 용량과 생활 및 농공용수의 과수요에 따른 불균형으로 근래 많은 하천이 갈수기나 평수기시 친천화되어 가고 있어 수중생태계에 큰 위협을 주고 있다. 또한 이에 더하여 산업발달과 함께 수질오염원은 양적으로는 물론 질적으로도 증가일로에 있어 기존의 유기 및 중금속 오염물질을 포함해 수많은 신생 유기화합물질이 수계에 유출되고 있으며, 이에 더해 질소 및 인으로 대표되는 영양물질의 유입과다로 국내 대다수의 호수가 부영양화의 새로운 위협에 직 면해 있다. 우리나라에는 적은 유입유량에 큰 저수용량 및 긴 체류시간으로 대표되는 대규모 자연호는 없으며, 대부분이 매단위 강을 막아 형성된 체류시간 1년 이하의 인공호로서 그 중 안동호, 충주호, 대청호 등은 비교적 체류시간이 긴 호소형 인공호로, 팔당호나 기타 대부분의 호수는 체류시간이 짧은 하천형 인공호로 대별된다(Table 1). 이처럼 국내 호소는 국외의 자연 호와는 다른 구조적 특성을 가짐에 따라 부영양화 특성 역시 매우 상이하고, 호소형 인공호와 하천형 인공호간의 차이 역시 현저하여 일률적인 관리대책을 설정하기가 어려운 실정에 있다. 또한 각 호수의 유역특성이 상이함에 따라 호수별로 유역 오염원의 오염부하율이 현저히 달라(Table 2) 호수에 따른 특성적인 유역관리(Lake-specific management)가 요구되고 있다 정상상태(Steady state)가 아닌 국내 호소에 대해 국외 자연호의 인단순모델 (Simple phosphorus budget model)을 그대로 적용하는 것에 다소 무리한 점이 따르나, 평균적인 개념으로 OECD의 유역부하량 및 호소특성에 따른 부영양화 판정모델을 적용한다면, 국내 주요 호수는 모두 부영양 수준에 있다. 또 각 호수별로 부영양화 제어를 위해 요구되는 인의 삭감부하량은 상당량이 되어 현실적으로 관리가 이미 어려운 실정에 와 있다. 호수의 부영양화는 조류 발생으로 이어지며, 그에 따른 폐해는 각종 형태로 나타나는데 대표적으로 이취미 발생이나 유해조류에 의한 독성물질의 배출, 정수처리 과정의 THM 발생등이 주로 문제가 되고 있다. 따라서 국내 호수의 구조적 특성별 부영양화 실태를 파악하고 그에 따른 영향 및 관리방안에 관한 종합적 고찰이 차후 수질관리를 위해 요구되고 있으며, 특히 납조류로 대표되는 유해조류의 독성영향에 관한 연구는 시급한 실정에 있다.

Estimation of Kinetic Coefficient in Submerged Membrane Bioreactor for Biological Nutrient Removal (도시 하수의 생물학적 고도처리를 위한 분리막 공정의 개발 및 동역학적 계수 산정 연구)

  • This study was performed to investigate the characteristics of nutrient removal of municipal wastewater in membrane bioreactor system. Membrane bioreactor consists of four reactors such as the anaerobic, the stabilization, the anoxic and the submerged membrane aerobic reactor with two internal recycles. The hydraulic retention time (HRT), sludge retention time (SRT) and flux were 6.2 h, 34.1 days and 19.6 L/$m^2$/hr (LMH), respectively. The removal efficiency of $COD_{Cr}$, SS, TN and TP were 94.3%, 99.9%, 69.4%, and 74.6%, respectively. The estimated true biomass yield, specific denitrification rate (SDNR), specific nitrification rate (SNR), specific phosphorus release rate (SPRR) and specific phosphorus uptake rate (SPUR) were 0.653 kgVSS/kgBOD/d, 0.044 $mgNO_3$-N/mgVSS/d, 0.035 $mgNH_4$-N/mgVSS/d, 51.0 mgP/gVSS/d and 5.4 mgP/gVSS/d, respectively. The contents of nitrogen and phosphorus of biomass were 8.86% and 3.5% on an average.

Effects of Mugwort, Onion and Polygalae Radix on the Intestinal Environment of Rats (쑥, 양파 및 원지가 흰쥐의 장내환경에 미치는 영향)

  • The contents in large intestine of Sprague Dawley rats fed polygalae radix(Polygala tennuifolia), onion(Allium cepa) and mugwort(Artemisia asiatica)-supplemented diets for 14 days were analysed for changes of major intestinal microflora, activities of ${\beta}-glucosidase\;and\;{\beta}-glucuronidase$ and amounts of putrefactive products such as indole and volatile basic nitrogen. The rats having ingested $5%{\sim}10%$ mugwort water or ethanol extract-supplemented diets showed a significant increase in intestinal bifido-bacteria and a decrease in clostridia and E. coli (p<0.05). And 10% onion juice group also showed a similar beneficial microflora change. In 5% mugwort powder-supplemented group, ${\beta}-glucosidase\;and\;{\beta}-glucuronidase$ activities in the intestinal contents were lowered, but the changes were not significant. Indole contents and pH in this group were significantly low compared with that of control (p<0.05). However, the activities of ${\beta}-glucosidase$ in 5% polygalae radix water extract and 10% onion juice-supplemented group and ${\beta}-glucuronidase\;in\;5%{\sim}10%$ mugwort water and ethanol extract-supplemented group were significantly higher than those of control (p<0.05). The intestinal indole contents of rats were significantly increased by feeding diet with water extract of polygalae radix and ethanol extract of mugwort which had brought comparatively large amount of protein in intestine (p<0.05). However, polygalae radix, onion, and mugwort-supplemented group had no effect on volatile basic nitrogen.

Comparison of Physicochemical Properties between Organic and Conventional Soybean by Steaming Treatment (증자 처리한 일반콩과 유기콩의 이화학적 특성 분석)

  • The differences between organic and conventional soybean were studied based on their physicochemical properties by steaming treatment. There was no difference between the two kinds of soybean regarding crude protein, crude fat and crude ash contents. Reduction in sugar content was higher in raw organic soybean than raw conventional soybean. However, there was no difference between the two soybeans after steaming. Amino nitrogen content was higher in organic than conventional soybean. Further, there was no difference between the two soybeans after steaming. Regarding pH, acidity and salinity, there was no differences between the two soybean after steaming. Organic soybean was more bright than conventional soybean after steaming. The yeafercontent and number of bacteria in raw soybean were not different between the two soybean types. The number of fungi was higher in organic than conventional soybean. Upon steaming, the yeast number was different between two kinds of soybean. Further, the number of bacteria was higher in organic than conventional soybean. The number of fungi was not different between the two soybeans.

Quality Changes in Doenjang upon Fermentation with Two Different Bacillus subtilis Strains (Bacillus subtilis HJ18-9를 이용하여 제조한 콩알메주 된장의 발효숙성 중 특성)

  • This study investigated the quality characteristics of doenjang prepared with different Bacillus strains (Bacillus subtilis KACC 15935 and Bacillus subtilis HJ18-9). Changes in enzyme activities (protease, cellulase, and ${\alpha}-amylase$), amino-type nitrogen and ammonia-type nitrogen contents, and reducing sugar were investigated during the fermentation period. Enzymes such as protease, cellulase, and ${\alpha}-amylase$ play important roles in the composition of nutrients, as well as in the flavor and taste of doenjang. After 60 days of fermentation, protease activities in control doenjang, and doenjang fermented with B. subtilis KACC 15935, and B. subtilis HJ18-9 increased significantly up to $382.58{\pm}4.02$, $342.58{\pm}7.94$, and $392.58{\pm}1.91unit/g$, respectively (p<0.05). At the beginning of fermentation, protease activities were in the range of 156.88~182.71 unit/g. Cellulase and ${\alpha}-amylase$ activities of doenjang in HJ18-9 were higher than those in other samples. After fermentation, amino-type nitrogen in doenjang fermented with control, B. subtilis KACC 15935, and B. subtilis HJ18-9 increased significantly up to $143.25{\pm}1.62$, $141.86{\pm}2.14$, and $150.23{\pm}1.62mg%$, respectively (p<0.05). These results suggest that B. subtilis HJ18-9 is a suitable starter for the preparation of doenjang.

Changes in Soil Chemical Properties after Thinning in Quercus acuta stand (간벌 후 붉가시나무 임분의 토양 특성 변화)

  • This study was carried out to investigate the changes in soil chemical properties after thinning for a Quercus acuta stand in Wan-do Arboretum, Jeollanam-do. The stem density after thinning was determined referring to that of Quercus acutissima with 10 cm diameter at breast height and soil samples were collected 7 years after thinning. There was no considerable difference in soil pH values after thinning. However, the concentrations of soil organic matter, total nitrogen and available phosphorus significantly increased after thinning. In addition, thinning significantly increased cation exchange capacity (C.E.C) and the concentrations of exchangeable cations ($K^+$, $Na^+$, $Ca^{2+}$, and $Mg^{2+}$). From this study, it was revealed that the nutrient concentrations of this study site were generally lower than those of other forest soils. Therefore, it is need to improve the warm temperate forests through thinning and to develop forest treatment for regeneration.

Production of Poly(Hydroxybutyric-Co-Hydroxyvaleric) Acid by Pseudomonas sp. HJ (Pseudomonas sp. HJ에 의한 Poly(Hydroxybutyric-Co-Hydroxyvaleric) Acid의 생산)

  • To produce PHA(polyhydroxyalkanoic acid) from microbr, dozens of microorganism have been screened from sewage sludge. Selected a strain HJ out of 50 strains of PHA producing bacteria has a capability of accumulating large amounts of PHB/HV copolymer when grown in batch culture with a single carbon source (glucose) that was not generally considered as precursor of hydroxyvalerate monomer unit. The strain HJ was identified as the genus Pseudomonas with respect to morphological, cultural, and biochemical characteristics. The optimal temperature and pH for cell growth were $37^{\circ}C$ and 7.0. The optimal medium compositions for cell growth were glucose 1% as a carbon source, (NH4) 2SO4 0.2% as a nitrogen source, K2HPO4 0.3%, and KH2PO4 0.45%. TO investigate she optimal condition for PHA production two-step cultivation method was employed. PHA production was inducted by deficiency of NH4+, SO4-2, Mg+2. Besides carbon source, deficiency of all nutrients stimulated PHA productivity but deficiency of NH4+ stimulated the most HV monomer content. The highest PHA production was C/N molar ratio 95.2. Pseudomonas sp. HJ was also able to pyoduc PHB/HV copolymer when cultivated on alkane, alkanoate, alcohol as carbon sources. The contents of PHA and she proportions of hydroxyvalerate monomer units varied depending on the carbon sources. Especially Pseudomonas sp. HJ was able to incorporate hydroxyvalerate into PHB/HV to level as high as from 49 to 74 mol% when grown in a medium containing hexadecane and propionate. The purified PHA was identified PHB/HV copolymer by HNMR analysis.

Removal of Pb2+ Ions from Water by Surfactant-templated Mesoporous Silicates (계면활성제가 담지된 메조포러스 실리케이트에 의한 수중 납이온 제거)

  • Mesoporous silicate materials have been used as adsorbents for the removal of heavy metals from water by introducing functional groups such as thiol and amine. In this research, it was investigated whether surfactants used as templating agents in synthetic processes can act as adsorption sites for heavy metals. Three mesoporous silicates-SBA-15, MCM-41, and HMS were synthesized using, respectively, block copolymer, hexadecyltrimethylammoniumbromide (HDTMA), and dodecyamine as surfactants. X-ray diffraction and $N_2$ gas adsorption analysis confirmed that the mesoporous silicates were well prepared and FT-IR spectra resulted in the existence of the surfactants in as-synthesized mesoporous silicates and the removal of surfactants after calcination. The interactions between $Pb^{2+}$ ions and the mesoporous silicate materials with/without surfactants were observed. In adsorption kinetic experiments, it revealed that the calcined mesoporous silicates and the surfactant-loaded SBA-15 almost had no adsorption capacity for $Pb^{2+}$ ions. In contrast, the surfactant-loaded MCM-41 and HMS showed, respectively, the adsorption capacities of 26.60 and 115.16 mg/g which were acquired through the fits of adsorption kinetic data to the pseudo second order kinetic model. The adsorption capacities were comparable to those of other mesoporous adsorbents for heavy metals.

Optimization for Mycelial Growth and Inhibitory Effect on Nitric Oxide Production of Cordyceps nutans Pat. (노린재동충하초의 배양 최적화 및 NO 생성 저해 효과)

  • Cordyceps (vegetable wasp and plant worm), an entomopathogenic fungi, has been used as a herbal medicine in Asian countries since ancient times. Cordyceps nutans is common but there is little research on this species. This study investigated the optimal culture conditions of C. nutans and the inhibitory effect on nitric oxide (NO) production in RAW 264.7 cell treated culture broth. The optimal conditions for the mycelial growth were $25^{\circ}C$ and pH 7.0-8.0. Mycelial growth was highest on mushroom complete medium (MCM), V8 juice agar (V8A), and yeast malt dextrose (YMD) medium. Mycelial growth on mushroom minimal medium (MMM) did not occur, so nutrient source was essential. Dextrose and sucrose as carbon sources, and ammonium citrate as a nitrogen source were satisfactory for mycelial growth. Cytotoxicity of C. nutans culture broth was not found in RAW 264.7 cells. C. nutans culture broth suppressed NO production of lipopolysaccharide (LPS)-stimulated RAW 264.7 cell in a dose-dependent manner. Thus, our results provided the optimal conditions for cultivation of C. nutans and showed that C. nutans may have excellent physiological activities.
