Analysis of Bacterials Community Structure in Leadchate-Contaminated Groundwater using Denaturing Gradient Gel Electrophoresis (Denaturing Gradient Gel Electrophoresis를 이용한 매립지 침출수로 오염된 지하수의 세균 군집 분석)

  • This research has been performed to clarify the relationship between hydrogeochemistry and bacterial community structure in groundwater contaminated with landfill leachate. We collected and analyzed samples from 5 sites such as leachate (KSG1-12), treated leachate (KSG1-16), two contaminated groundwaters (KSG1-07 and KSG1-08) and non-contaminated groundwater (KSG1-13). pH was 8.83, 8.04, 6.87, 6.87 and 6.53 in order; redox potential (Eh) 108, 202, 47, 200 and 154 mV; electric conductivity (EC) 3710, 894, 1223, 559 and 169.9 $\mu$S/cm; suspended solids (SS) 86.45, 13.74, 4.18, 0.24 and 11.91 mg/L. In KSG01-12, the ion concentrations were higher especially in $Cl^-$ and $HCO_3^-$ than other sites. The concentrations of Fe, Mn and $SO_4^{2-}$ were higher In KSG1-07 than in KSG1-08, and vise versa in $NO_3^{2-}$. In the comparison of DGGE fingerprint patterns, the similarity was highest between KSG1-13 and KSG1-16 (57.2%), probably due to common properties like low or none contaminant concentrations. Otherwise KSG1-08 showed lowest similarities with KSG1-13 (25.8%) and KSG1-12 (27.6%), maybe because of the degree of contamination. The most dominant bacterial species in each site were involved in $\alpha$-Proteobacteria (55.6%) in KSG1-12, $\gamma$-Proteobacteria (50.0%) in KSG1-16, $\beta$-Proteobacteria (66.7%) in KSG1-07, $\gamma$-Proteobacteria (54.5%) in KSG1-08 and $\beta$-Proteobacteria (36.4%) in KSG1-13. These results indicate that the microbial community structure might be changed according to the flow of leachate in grounderwater, implying changes in concentrations of pollutants, available electron accepters and/or other environmental conditions.

Effect of Water-Extracts from Sludge Compost on Seed Germination (퇴비의 부숙 과정 중 추출물이 종자 발아력에 미치는 영향)

  • In order to evaluate the physicochemical properties of sludge compost and to identify the effects water-extracts from sludge compost in 2, 6, 8, 12 weeks of decomposing process on seed germination and root elongation in cabbage, lettuce, soybean and barley plants was investigated. The content of total nitrogen in sludge compost increased slightly in 6 weeks decomposing process, and then decreased gradually. Organic matter content decreased continuously overall decomposing process. As decomposition was processing, pH of sludge compost decreased slightly, and EC increase within 6 weeks decomposing process, and then decreased. The content of nitrogen in water-extracts from sludge compost increased within 8 weeks decomposing process and decreased in 12 weeks decomposing process. The content of ammonium nitrogen was similar with that of total nitrogen, and the ratio of ammonium and nitrate increased within 8 weeks, and then decreased. Cation content and EC decreased the late of decomposing process and pH didn't change. The water-extracts from sludge compost during decomposing process inhibited seed germination and root elongation in cabbage (Brassica campestris), lettuce (Lactuca sativa), barley (Hordeum vulgare) and soybean (Glycine max). The inhibition of root elongation in cabbage was greater than that of relative seed germination, whereas relative seed germination in lettuce was more inhibit than root elongation. Relative seed germination and root elongation in soybean were inhibited slightly, but those of in barley was inhibited strongly. In this study, we would identify the effects of water-extracts from sludge compost on seed germination and root elongation was different to the species of seed. The inhibition of seed germination and root elongation treated with the water-extracts which extracted from sludge compost in the early stage of decomposing process was greater than that of in the late stage of decomposing process.

Composition and emission characteristics of fine particulate matters at the 1100 Site of Mt. Halla during 2011-2012 (한라산 1100고지 대기 미세먼지의 조성 및 배출 특성: 2011~2012년 측정)

  • PM10 and PM2.5 samples were collected at the 1100 site of Mt. Halla in Jeju Island during 2011~2012, and their ionic and elemental species were analyzed, in order to investigate the characteristics of emission sources as well as aerosol compositions. The mass concentrations of PM10 and PM2.5 were 22.0±13.1 µg/m3 and 11.3±6.1 µg/m3, respectively, showing 2.4~2.6 times lower than those of the capital city area of Korea. The composition ratios of major secondary pollutants (nss-SO42−, NH4+, and NO3) were the highest as 85.5 % for PM10 and 91.3 % for PM2.5, and followed by the order of marine (Na+, Cl, and Mg2+), organic acid (HCOO and CH3COO), and soil (nss-Ca2+) sources. Among the elemental species in PM10, soil-originated components (Al, Fe, and Ca) were consisted of 50.9 %, which was higher proportion than marine and anthropogenic elements. The acidification of the fine particulate matters was found to be influenced mostly by sulfuric and nitric acids, and these acids were mainly neutralized by calcium carbonate in PM10 and by ammonia in PM2.5. The clustered back trajectories showed that 47 % of total air mass inflows was from the China, and the concentrations of NO3 and nss-Ca2+ were especially high corresponding to the inflows.

Effect of Nitrate Concentration in Culture Solution on the Growth and the Uptake of Inorganic Elements of Tomato Plants(Lycopersicum esculentum Mill) (배양액(培養液)의 질산태질소농도가 토마토(Lycopersicum esculentum Mill)의 생육 및 무기원소(無機元素) 흡수에 미치는 영향)

  • This experiment was carried out to investigate the effects of nitrate concentration in culture solution on the growth and the uptake of inorganic elements in Tomato plant in the greenhouse. Tomato plants(cv. TVR-2) were grown with nitrate concentrations 8, 16, 24, 32cmol/l, based on Japan ENSI standard solution. Dry weights of lamina and petiole increased with the nitrate concentration. However, the dry weight of fruit was the highest in the treatment of nitrate concentration of 16cmol/l. The proportion of dry weights of vegitative organ to reproductive organ was the lowest in the treatments of nitrate concentrations of 16cmol/l and it increased with the nitrate concentration. The fruit yield was the highest at the treatment of nitrate concentration of 16cmol/l. With the increase of nitrate level the concentrations of N, $NO_3-N$, Ca and Na increased in lamina and petioles. The concentrations of K, P, S and Cl tended to decline in the nitrate concentration of 16 and 32cmol/l. These results indicate that optimum nitrate concentrations in a tomato grown by hydroponics change with growth stage, and the optimum concentrations for vegitative and reproductive stage were 8 and 16cmol/l, respectively. It also was proved that the nitrate concentrations in the culture solution affected antagonistically the uptake of inorganic anion in tomato : In low nitrate level $Cl^-$ uptake was affected much, while $SO_4{^{2-}}$ and $H_2PO_4{^-}$ uptake were affected in high nitrate level.

The Impact of monsoon Rainfall (Changma) on the Changes of Water Quality in the Lower Nakdong River (Mulgeum) (장마기의 강우가 낙동강 하류 (물금) 수질에 미치는 영향)

  • The impact of summer monsoon on water quality of the lower Nakdong River was evaluated during the summer (June-August) in 1997. Several limnological variables were measured in the interval of $1{\sim}3$ day using an automatic monitoring system (Hydrolab $Recorder^{TM}$) to detect water quality changes caused by rainfall on onehour basis. During the monsoon period (from late June to mid July), 5 times of major rainfall events of >50 mm were recorded in the river basin. Dynamic changes of water quality were observed during the monsoon, and the first rainfall event (June$25{\sim}27$) had a significant influence on the water quality at the lower part of the river. All Parameters were largely changed due to the first rain event, and the changed level was maintained until the end of monsoon period. Nutrient concentrations and turbidity increased and values of the other parameters were declined as a result of water dilution. This rainfall event, Changma, is a meteorological phenomenon caused by the East-Asian monsoon climate. The magnitude and frequency of the rainfall during the early monsoon play an important role in change of water quality and ecosystem characteristics of large river systems.

Hydrogeochemical Characterization of Groundwater in Jeju Island using Principal Component Analysis and Geostatistics (주성분분석과 지구통계법을 이용한 제주도 지하수의 수리지화학 특성 연구)

  • The purpose of the study is to analyze the hydrogeochemical characteristics by multivariate statistical method, to interpret the hydrogeochemical processes for the new variables calculated from principal components analysis (PCA), and to infer the groundwater flow and circulation mechanism by applying the geostatistical methods for each element and principal component. Chloride and nitrate are the most influencing components for groundwater quality, and the contents of $NO_3$ increased by the input of agricultural activities show the largest variation. The results of PCA, a multivariate statistical method, show that the first three principal components explain $73.9\%$ of the total variance. PC1 indicates the increase of dissolved ions, PC2 is related with the dissolution of carbonate minerals and nitrate contamination, and PC3 shows the effect of cation exchange process and silicate mineral dissolution. From the results of experimental semivariogram, the components of groundwater are divided into two groups: one group includes electrical conductivity (EC), Cl, Na, and $NO_3$, and the other includes $HCO_3,\;SiO_2,$ Ca, and Sr. The results for spatial distribution of groundwater components showed that EC, Cl, and Na increased with approaching the coastal line and nitrate has close relationship with the presence of agricultural land. These components are also correlated with the topographic features reflecting the groundwater recharge effect. The kriging analysis by using principal components shows that PC 1 has the different spatial distribution of Cl, Na, and EC, possibly due to the influence of pH, Ca, Sr, and $HCO_3$ for PC1. It was considered that the linear anomaly zone of PC2 in western area was caused by the dissolution of carbonate mineral. Consequently, the application of multivariate and geostatistical methods for groundwater in the study area is very useful for determining the quantitative analysis of water quality data and the characteristics of spatial distribution.

Soil properties in Panax ginseng nursury by parent rock (모암별 인삼묘포지의 토양특성에 관한 연구)

  • A research has been done for growing characteristics of Korean ginseng in Geumsan of Chungnam Province. It had been made to determine the transitional element concentrations of the rocks, divided by biotitic granite(GR) and phyllite(PH). The physical and chemical properties of their weathering soils and ginseng nursery soils were analyzed. The texture in the GR weathering and ginseng nursery soils were sandy clay, and the texture of the PH weathering and ginseng nursery soils were heavy or silty clay. The bulk densities of the GR and PH weathering soils were $1.21{\sim}1.32g/cm^3$ and $1.26{\sim}1.38g/cm^3$, respectively. Also, the bulk densities of the GR and PH ginseng nursery soils were $1.02{\sim}1.10g/cm^3$, respectively. The pH (4.80) of the GR weathering soil were lower than the pH of the PH(5.34) weathering soil. The pH in the 2 year and 4 year-ginseng nursery soil of the GR were 4.39 and 4.40. In addition, those of the PH were 5.24 and 5.34, respectively. The difference in pH of the two nursery soils could be from the pH difference between the two parent materials. The organic matter contents of the GR weathering soils(0.24%) were higher than those of the PH(1.02%) weathering soils. The organic matter of the 2 and 4 year-ginseng GR nursery soils were 0.87% and 1.52%, and of the PH nursery soils were 2.06% and 2.96%, respectively. The total nitrogen contents of the GR weathering soils were 259.43ppm and of the PH weathering soils were 657.22ppm. Those of 2 and 4 year-ginseng GR nursery soils were 588.04ppm and 657.22ppm and those of the PH nursery soils were 1037.72ppm and 1227.96ppm, respectively. The nitrate and ammonium contents of the GR weathering soils were the extremely small, and those of the PH weathering soils were 6.7ppm and 9.94ppm. Those of 2 year-ginseng GR nursery soils(223.09ppm and 26.96ppm) were higher than those of PH(19.46ppm and 8.23ppm) nursery soils. And those of 2 year-ginseng PH nursery soils(14.22ppm and 16.84ppm) were lower than those of PH(306.93ppm, 34.21ppm) nursery soils. The difference was due to fertilizer types and more deposits of nitrate after oxidation of ammonium. The phosphate contents of the GR and PH weathering soils were 14.41ppm and 38.60ppm. Those of GR 2 and 4 year-ginseng nursery soils were 46.89ppm and 102.44ppm and those of the PH nursery soils were 147.04ppm and 38.60ppm. The cation exchange capacities of the GR weathering soils were 12.34me/100g and those of the PH weathering soils were 15.40me/100g. Those of 2 and 4 year-ginseng GR nursery soils were 15.80me/100g and 7.70me/100g and those of PH nursery soils were 12.14me/100g and 12.83me/100g. All of exchangeable cation($K^+$, $Ca^{2+}$, $Mg^{2+}$, $Na^+$) contents in the nursery soils were higher than those in the weathering soils. The $SO_4{^2-}$ contents of the weathering soils in both of the GR(5.98ppm) and PH(9.94ppm) were higher than those of the GR and PH ginseng nursery soils. The $Cl^-$) contents of the GR and PH weathering soils were a very small and those of the nursery soils(2-yr GR: 39.06ppm, 4-yr GR: 273.43ppm, 2-yr PH: 66.41ppm, 4-yr PH: 406.24ppm) were high because of fertilizer inputs.

Effect of Chicken Manure Compost Application on the Growth of Vegetables and Nutrients Utilization in Upland Soil (계분퇴비 시용이 채소류 생육과 양분이용율에 미치는 영향)

  • To evaluate the effect of chicken manure compost(CMC) application on nutrient utilization and reduction of N fertilizer application for vegetables such of cabbage, reddish, and eggplant, four different application rates of CMC such as 0, 10, 30. $50Mg\;ha^{-1}$ were amended with three different rates of chemical fertilizer of 0, 15, $300kg\;ha^{-1}$. The efficiency of nutrient utilization on CMC were in the order of N, P, K. For each respective nutrient utilization by cabbage, reddish, and eggplant, N were 29%, 20%, 14%; P were 10%,<1%, and <1% ; K were 5%, 22%, 32%. The greater application of CMC, the less the efficiency, while the efficiency of P was increased with increasing application of CMC. The highest efficiency of nutrient utilization was found in $10Mg\;ha^{-1}$. The amounts of reduction of N fertilizer application in soils amended with $10Mg\;ha^{-1}$ of CMC with respect to maintain the normal production of each vegetables observed in this experiment were as follows: $25kg\;ha^{-1}$ and $15kg\;ha^{-1}$ less, and $13kg\;ha^{-1}$ more than the recommended rate for cabbage, eggplant, and reddish, respectively. For elution of available of N by the application of CMC, there was a gradual increase up to 30 day, then gradually decreased. However, there was increase of N eluted in the application of $50Mg\;ha^{-1}$. For soil chemical properties, pH, T-N, OM, and $P_2O_5$ were increased with increased application of CMC, as well as did cations such as K, Ca, and Mg. Yield and glucose of cabbage was significantly increased at the application of $30Mg\;ha^{-1}$ CMC, but content of nitrate in eggplant and raddish was highly increased with increasing application of N fertilizer and CMC. By the way, the yields of cabbage, reddish, and eggplant were slightly decreased with CMC application greater than $50Mg\;ha^{-1}$.

Changes in Chemical and Microbiological Properties of Spring Waters in Tongyeoung Area (통영시내 약수의 화학적 및 세균학적 품질변화)

  • This study was carried out to changes in chemical and microbiological properties of spring waters in Tongyeoung area. In this paper, ninety spring water samples were collected from 9 station for 11 month to evaluated chemical and bacteriological water quality. Range and mean values of constituents of the samples are as followed; water temperature 5.2~25.8$^{\circ}C$, 16.3$^{\circ}C$, pH 6.0~7.2, 6.7, total residue 33.6~210 mg/1, 90.6 mg/1, turbidity 0.35~5.48, 1.45NTU, KMnO4 consumed 0.51~4.21 mg/1, 1.39 mg/1, chloride ion 6.23~42.5, 16.7 mg/l, phosphate-phosphorus ND-0.04, 0.02 mg/1, nitrite-nitrogen ND~0.02, 0.01 mg/1, nitrate-nitrogen ND~3.56, 1.42 mg/1, ammonia-nitrogen ND~0.20, 0.14 mg/1, dissolved total nitrogen ND~3.78, 1.57 mg/1, iron 0.04~0.28, 0.13ppm, zinc 0.03~0.66, 0.20ppm, mangan ND~0.01, allumium 0.14~0.58, 0.39ppm, copper ND~0.01, 0.01, lead ND~0.01, 0.01ppm, Arsenic ND~0.01, 0.01ppm, mercury ND~0.02, chrome not detected, cadmium not detetced respectively. The viable cell counts of the spring waters ranged 5.0~760/m1(means 130/m1). Range and mean value of total coliform and focal coliform MPN's of the spring waters were 0~2,400MPN/100 ml, 73MPN/100 ml and 0~540MPN/100 ml, 21MPN/100 ml. Spring water quality was usually poor with viable cell counts exceeding 130 CFU/liter and the coliform counts in spring waters of 73 MPN/liter. Composition of coliform by IMViC reaction was 33.3% E. coli, 15.6% Citrobacter freundii, 35.6% Klebsiella aerogenes and others.

Geochemical Studies of $CO_2$-rich Waters in Chojeong area II. Isotope Study (초정지역 탄산수의 지화학적 연구 II. 동위원소)

  • The $CO_2$-rich waters in the Chojeong area are characterized by low pH (5.0~5.8), high $CO_2$pressure (about 1 atm) and high amounts of total dissolved iou (up to 989 mg/L) and chemically belong to Ca-HC $O_3$type. The oxygen. deuterium and tritium isotope data indicate that the mixing process occurred between $CO_2$-rich water and surface water and/or shallow groundwaters and also suggest that the $CO_2$-rich water has been derived from meteoric waters. According to $\delta$$^{13}$ C values (-8.6~-5.3$\textperthousand$). the $CO_2$ in the water is attributed from deep seated $CO_2$gas. The high dissolved carbon (-14.4~-6.8$\textperthousand$. $\delta$$^{13}$ C) in groundwater of the granitic terrain might be affected by $CO_2$-rich water, whereas the dissolved carbon (-17.9~-15.2$\textperthousand$. $\delta$$^{13}$ C) in groundwater of the metamorphic terrain is likely controlled by soil $CO_2$ and from the reaction with calcite in phyllite. Sulfur isotope data (+3.5~+11.3$\textperthousand$,$\delta$$^{34}$ $S_{SO4}$) also support the mixing process between $CO_2$-rich water and shallow groundwater. Strontium isotopic ratio ($^{87}$ Sr/$^{86}$ Sr) indicates that the $CO_2$-rich water (0.7138~0.7156) is not related to vein calcite (0.7184) of Buak mine or calcite (0.7281~0.7346) in phyllite. By nitrogen isotope ($\delta$$^{15}$ $N_{NO3}$) the sources of nitrogen (up to 55.0 mg/L, N $O_3$) in the $CO_2$-rich water are identified as fertilizer and animal manure. It also indicates the possibility of denitrification during the circulation of nitrogen in the Chojeong area. The possible evolution model of the $CO_2$-rich water based on the hydrochemical and environmental isotopic data was proposed in this study. The $CO_2$-rich waters from the Chojeong area were primarily derived from the reaction with granite by supply of deep seated $CO_2$. and then the $CO_2$-rich water was mixed and diluted with the local groundwater.ter.
