Growth Inhibition and Induction of Apoptosis in Human Bladder Cancer Cells Induced by Fermented Citrus Kombucha (감귤 콤부차 발효액의 인체 방광암세포에 대한 성장억제와 Apoptosis에 미치는 영향)

  • Kombucha is a slightly sour beverage fermented by symbiotic micro-organisms, including bacteria and yeasts. In this study, we examined the biological activities of citrus Kombucha (CK) produced by addition of citrus extract to original Kombucha (K). After fermentation for 10 days, radical scavenging activity examined by ABTS and DPPH assays increased by approximately 20% compared to that of K. Moreover, content of total phenolic compounds significantly increased by 60% compared to that of K. Cell proliferation assays utilizing MTT showed that CK treatment significantly inhibited growth of bladder cancer cells, T-24 and 5637, in a dose-dependent manner with $IC_{50}$ values of 4 and 7 mg/mL, respectively. Annexin V staining showed that CK treatment led to apoptosis of cells in a dose-dependent manner. T-24 cells were more sensitive to CK treatment than 5637 cells, as 8 mg/mL of CK resulted in 97% apoptosis of T-24 cells. Western blotting showed that CK treatment led to up-regulation of apoptotic proteins, including caspases-3, -8, -9, and PARP, in bladder cells not in K-treated cells. Taken together, these results demonstrate that CK may be developed as a functional beverage.

Characteristics and Genesis of the Clay Minerals in Weathering Products from the Guweol Mountain Area, Pusan (부산시(釜山市) 구월산(九月山)의 풍화물중(風化物中) 점토광물(粘土鑛物)의 특성(特性) 및 성인(成因))

  • The various weathering products derived from andesitic rocks in the Guweol mountain area of Pusan have been studied in order to investigate the occurrence, characteristics and evolutional trend of clay minerals in the weathered rock and soil using mainly X-ray powder diffraction method. Kaolinite and halloysite are very abundant in the weathered rocks and soils derived from andesitic rocks of this area. Also, $12{\AA}$- and $14{\AA}$-minerals occur in the considerable amount. $12{\AA}$-minerals have been identified as mica/vermiculite interlayer minerals showing the $25{\AA}$ basal reflection. Their crystal sizes are relatively big showing more than 10 micron in diameter. Two types of $14{\AA}$-mineral, which are vermiculite/smectite interlayer mineral and Al-vermiculite, have been observed. Kaolin minerais consist of kaolinite, $10{\AA}$ and $7{\AA}$ halloysite. Kaolinite relatively dominates in the strongly weathered soil of the area. In contrast with kaolinite. $12{\AA}$- and $14{\AA}$-minerals such as vermiculite/smectite and mica/vermiculite interlayer minerals tend to occur in the weakly to intermediately weathered products. Based on their occurrences, it strongly suggests that they are intermediate products in the course of the weathering process from the parent materials into the kaolin mineral.

  • 생명공학의 시대로 일컬어 지고 있는 오늘날, 재생의학 분야에서는 난치성 질환 치료를 목적으로 활발한 연구가 진행되고 있다. 특히, 줄기세포를 이용한 세포 대체치료 관련 연구는 최근 국내 황우석 박사의 체세포 핵이식 배아세포주 확립에 이르기 까지 괄목할 만한 발전을 보여 주고 있다. 이와 관련해 생명보험산업에 적잖은 파장이 예상되며, 생명보험사 내부적으로 기존에 판매된 상품의 사차손 관리와 함께 급속도로 발전하는 줄기세포 연구에 직접적으로 대응하는 상품개발, 언더라이팅, 지급 심사 등 보험사 내외에서의 전방위적인 변화가 필요하다는 문제 제기가 있다. 줄기세포란 조직 분화 과정에서 볼 수 있는 세포이며 근육 뼈 뇌 피부 등 신체의 어떤 기관으로도 전환할 수 있는 만능세포로서, 간 폐 심장 등 구체적 장기를 형성하기 이전에 분화를 멈출 배아 단계의 세포를 말한다. 한편, 성체줄기세포는 조직이나 기관의 분화된 세포들 사이에서 발견되는 미분화 세포로써, 자기 스스로 증식할 수 있으며, 조직이나 기관의 특수한 기능을 가지고 있는 세포로 분화할 수 있는 능력을 가진 신체줄기세포를 말한다. 배아줄기세포와 생체줄기세포를 통한 장기이식 등 난치병 정복은 윤리적, 사회적으로 많은 논란이 예상되며, 기술적으로도 해결해야 할 문제점들이 산적해 있기 때문에 아직은 요원한 것이 사실이다. 현재 유럽 대부분의 나라와 미국에서는 인간 배아의 복제가 금지되어 있으며, 일본 정부는 연구용 배아 복제를 제한적으로 허용하고 있다. 하지만, 우리 나라의 경우 2005년 1월에 '생명윤리 및 안전에 관한 법'이 발효되었지만 정부는 관련 부작용에 대한 깊은 고찰 없이 전폭적인 지원들 약속하고 있는 실정이다. 줄기세포 연구의 발달로 인해 인류가 난치병 치료의 첫 장을 열었다고 하더라도 그 영향이 당장 보험사에 미친다고 할 수는 없다. 왜냐하면 앞으로 이러한 신기술이 실제 의료행위에 적용되기 위해서는 여러 단계의 안정화 작업과 임상시험이 필요한데 이러한 작업이 기술적으로 어렵고 그 시간도 만만치 않게 걸리기 때문이다. 또한, 보험사의 보장은 크게 사망/수술/입원/암/기타보장으로 구별할 수 있는데, 줄기세포 연구의 발달과 관련이 있는 보장이 제한되어 있어 보험사에 미치는 영향이 당장 우려할 만한 수준이라 할 수 없다. 하지만 만약 치료용 줄기세포 배양으로 인한 장기 기관의 이식이나 손상세포의 대체 등과 같은 의학신기술의 예상 외로 급격하게 발전한다면 보험사의 Risk 관리에 상당한 저해요인으로 작용할 것으로 판단된다. 특히 진단 입원 수술로 대표되는 생존보장에 대한 사차 Risk 및 사차손의 급증이나 역선택 증가는 보험사의 경영수지 악화를 유발하여 보험산업 전반에 위험으로 작용할 수도 있다. 따라서, 장기적인 안목으로 업계 공동의 대응이 필요하고, 각 사에서도 상품개발, 언더라이팅, 지급심사 간의 긴밀한 협조가 요구된다. 생명보험산업의 Risk 관리는 기존의 시장환경에 영향을 받는 비차, 이차중심에서 보험회사가 어느 정도 관리를 통해 적정규모를 유지할 수 있는 사차로 그 중심축이 이동하고 있다. 보험산업이 계속 활력을 갖고 성장하기 위해서는 체계적인 Risk나 관리가 핵심일 것이며, 보험사의 사차 Risk의 중요성이 더욱 커져 가고 있는 현실에서 거시적으로 의학신기술 발달 등 위험요인에 대해 미리 분석하고 이에 대한 대비책 마련이 필요할 것으로 판단된다.

The effects of auricular acupressure on pain and quality of life in patients with lung cancer (이압요법이 폐암 환자의 통증과 폐암 관련 삶의 질에 미치는 영향)

  • This study examined the effects of auricular acupressure on the pain and quality of life of lung cancer patients. Forty-four participants, and were divided into an experimental group (n=22) and a placebo control group (n=22). The patients in the experimental group received vaccaria seed to the four auricular acupressure points. In contrast, the patients in the placebo control groups received auricular acupressure on cancer-related points unrelated to pain. The intervention was implemented for six weeks, with five consecutive days a week. The pain intensity, pain pressure threshold, and quality of life score were used to validate the effects of the treatment. The pain intensity of the experimental group showed a significant decrease compared to the placebo control group after the intervention (Z=-2.78, p=.006). The pain pressure threshold of the experimental group showed a significant increase compared to the placebo control group after the intervention (Z=-2.69, p=.007). The quality of life among lung cancer patients showed a significant increase after the intervention compared to the placebo control group (t=3.20, p=.003). Therefore, auricular acupressure can be used as a proven nursing intervention method for lung cancer patients from cancer-related pain and the quality of life.

Analysis of Blasting Vibration at the Irregular Layered Structure Ground (불규칙한 층상구조 지반에서의 발파진동 분석)

  • By comparing test blasting data experimented in three layered-structure polymorphic grounds to a geological profile, influence of blast vibration with respect to uncontrollable ground characteristics was analyzed. Inefficient blast have been performed without sufficient verifications or confirmations because insufficiencies with regard to experiments and data of blasting engineering on the layered structures to be irregularly repeated clinker layer consisted of volcanic clastic zones. It is difficult to quantify N values of clinkers within test blasting region because they have diverse ranges, or coverages. An absolute value of attenuation coefficient N in a field, estimated by blasting vibration predictive equation (SRSD), are lesser than criteria of a design instruction, meaning that vibrations caused by blast can spread far away, and the vibrational characteristics of blasting test No.1, indicating relatively small values, inferred by the geological profile, pressures of gas by the explosion may be lost into a widely distributed clinker layers by penetrating holes resulted from blast into vicinity of clinker layers located in bottom of soft rock layers at the moment of blast. As a result, amounts of spalling rocks are decreased by almost half. Also, ranges of primary frequencies in the fields are identified as similar to those of natural frequency of typical structures.

  • Carcinomas of the stomach can spread to adjacent structure by local extension or metastasize to lymph nodes, peritoneum and distant organs. However, the incidence of metastatic bladder cancer originated at the stomach is very rare. A fifty-five year-old man admitted complaining of epigastric pain for 2 months. A large ulceroinfiltrative lesion was seen in the low body, which was confirmed poorly differentiated adenocarcinoma by histological examination. Abdominopelvic CT scan showed wall thickening at the greater curvature side of gastric body and urinary bladder. Urine cytology was negative. By transurethral resection of bladder, he was diagnosed as metastatic adenocarcinoma of the bladder. We report a case of stomach cancer with metastasis to urinry bladder.

Dormancy and Germination Characteristics of Round-Leaved Sundew (Drosera rotundifolia L.) Seeds Native to Korea (자생 끈끈이주걱(Drosera rotundifolia L.) 종자의 휴면과 발아특성)

  • This research was performed to develop mass propagation method of round-leaved sundew (Drosera rotundifolia L.) for improving horticultural usefulness. Seeds were collected around Hwaseong-si, Gyeonggi-do in October of 2011 and experimented upon while being dry-stored at 4 ± 1.0℃ (darkness). Seed length of a major and minor axis were 1.58 ± 0.060 and 0.21 ± 0.016 ㎜, respecrively, and weight of 1,000 seeds was 6.24 ± 0.172 ㎎. Seeds were thus classified as ‘dwarf seeds’. Regarding the dormancy type, since round-leaved sundew seeds were dormancy broken and germinated at 20~30℃ under the light condition after wet-chilling treatment for 12 weeks, seeds were estimated to have physiological dormancy. Germination conditions of dormancy broken seeds were found to be 20℃ and light condition (54.7%), but germination decreased at higher temperature. Percent germination (PG), germination energy (GE), mean germination time (MGT) and T50 were effectively improved by chemical treatment such as GA3 200 ㎎/L + kinetin 20 ㎎/L and wet-chilling treatment for 14 weeks. In conclusion these optimal conditions were thought to be a useful method for raising seedling uniformly.

Hydrothermal Gold Mineralization of the Sambo Deposit in the Muan Area, Korea (무안 지역, 삼보 광상의 금광화작용)

  • The Sambo gold deposit located nearby the Cretaceous Hampyeong basin is composed of gold quartz fine vein(the Jija vein) within Cretaceous rhyolite showing $N10{\sim}20W$ trends as well as $N5{\sim}10E$ trending quartz veins(the Pungja, Gwangsan and Pungjaji veins) in Precambrian gneiss. The gold vein typically displays the intermittent and irregular fine veins within rhyolite. Electrum is disseminated in wallrock along the fine cracks as well as coexists with hematite replacing pyrite. Ore-forming fluids from the mineralized vein($H_2O/-NaCl$ system, Th; $340{\sim}200^{\circ}C$, Salinity <2.7 eq. wt.% NaCl) and NE-trending veins($H_2O-NaCl/-CO_2$ system, Th; $400{\sim}190^{\circ}C$, salinity <7.9 eq. wt.% NaCl) are featured by dissimilar physicochemical conditions but their fluid evolution trends(boiling and mixing) are similar with each other. Gold veins of the Sambo deposit filled along NNW-trending tension crack are related to pull-apart basin evolution. Selective gold mineralization of the deposit reflect to dissimilarity between two ore-forming fluid sources. Consequently, gold veining of the Sambo deposit formed at shallow-crustal level and could be categorized into epithermal-type gold deposit related to tensional fractures filling triggered by Cretaceous geodynamics.

Curative Radiotherapy of Supralottic Cancer (성문상부암의 근치적 방사선치료)

  • Purpose : The purpose of this study was to evaluate the efficacy of curative radiotherapy in the management of supralottic cancer. Materials and Methods : Twenty-one Patients with squamous cell carcinoma of the supralottis were treated with radiotherapy at Gyeongsang National University Hospital between 1590 and 1994. Median follow-up period was 36 months and 95% were observed for at least 2 years. Results : Actuarial survival rate at 5 years was 39.3% for 21 patients. The 5-rear actuarial survival rate was 75.0% in Stage I, 42.9% in Stage II, 33.3% in Stage III and 28.6% in Stage IV(p=0.54), The 5-year local control rate was 52.0% for 21 patients. The 5-rear local control rate was 75.0% in Stage I, 57.1% in Stage II, 66.7% in Stage III, and 28.6% in Stage IV(p=0.33). Double primary cancer was developed in 3 patients and those were all esophageal cancers. Conclusion : In early stage(Stage I and II) supralottic cancer, curative radiotherapy would be a treatment of choice and surgery would be better to be reserved for salvage of radiotherapy failure. In advanced stage(Stage III and IV), radiotherapy alone is inadequate for curative therapy and combination with surgery should be done in operable patients. This report emphasizes the importance of esophagoscopy and esophagogram at the follow-up of patients with supralottic cancer.

A Study on the Slope Analysis of Weathered Limestone Soils during Rainfalls (강우 시 석회암 풍화토 사면의 안정해석에 관한 연구)

  • A set of soil samples were picked up from a failed slope formed by rainfall in limestone zone in Jangseong-gun, Jeonnam, Korea, to find out its physical and mechanical characteristics for this study, and variation of safety factor depending on slope inclination was defined by analysing slope stability affected by rainfall. Decomposed limestone soil in the research area is composed of quartz, orthoclase, gibbsite, geothite, etc., with specific gravity of 2.73, and this soil is included in SC by unified soil classification system. Calcium ingredient decreased remarkably during weathering at its mother rock. Coefficient of permeability is 2.56×10/sup -4/ cm/ sec, similar to its value of silty clay. Cohesion decreases remarkably from 3.0 t/ ㎡ to 0.72 t/ ㎡, and Φ value of internal friction angle tends to decrease as it turns to be saturated soil from partial saturated soil in the shear test. To analyze slope stability affected by rainfall, it is reasonable to seek seepage depth with reference to rainfall* intensity. In the slope stability analysis, when the seepage depth is the larger, its safety factor is the less, which makes the slope unstable. Comparing with minimum safety factor, 1.5 of cut slope in consideration of the seep-age line, safety factor is found to be satisfactory only when inclination of cut slope of decomposed limestone soil is more than 1:1.2 slope at least considering rainfall. It is also found that decrease of cohesion has great effect on decline of safety factor of slope while partial saturated soil turns to be saturated soil.
