The fibrinolytic activities of soluble proteins extracted from seeds of Coix lacryma-jobi L., Carthamus tinctorius L. and Malva venicillata L. were studied. Fibrinolytic activity of extract from C. lacryma-jobi L. showed 1.3 times higher than plasmin used as positive control. The fibrinolytic enzyme was confirmed and extracted directly from seed of C. lacryma-jobi L. by a fibrin zymography. The protein was composed of a single polypeptide and its apparent molecular weight was found to be 7.8 kDa, as judged by SDS-polyacrylamide gel electrophoresis. The effect of temperature for the proteolytic enzyme activity were stabilized above $50^{\circ}C$ and then dramatically decreased. Also, the enzyme activity was clearly inhibited by APMSF, PMSF and TPCK, suggesting that it is a member of the chymotrypsin-like serine pretense. In addition, effects of gamma-irradiated on seed of each plants were revealed that 8 Gy and 64 Gy were higher than others. This result shown that gamma-irradiation of seeds were capable to increase the fibrinolytic activity. All these results suggest the pretense is a fibrinolytic enzyme belong to a family of chymotrypsin-like serine pretense.
In soil and sediment environment, microorganisms play major roles in biochemical cycles of ecological significant elements. Because of its ecological significance, microbial diversity and community structure information are useful as indexes for assessing the quality of subsurface ecological environment and bioremediation. To achieve more accurate assessment, it is requested to gain sufficient yield and purity of DNA extracted from various soil and sediment samples. Although there have been a large number of basic researches regarding soil and sediment DNA extraction methods, little guideline information is given in literature when choosing optimal DNA extraction methods for various purposes such as environmental ecology impact assessment and bioremediation capability evaluation. In this study, we performed a thorough literature review to compare the characteristics of the current DNA extraction methods from soil and sediment samples, and discussed about considerations when selecting and applying DNA extraction methods for environmental impact assessment and bioremediation capability evaluation. This review suggested that one approach is not enough to gain the suitable quantity and yield of DNA for assessing microbial diversity, community structure and population dynamics, and that a careful attention has to be paid for selecting an optimal method for individual environmental purpose.

암을 제외한 대부분의 성인병은 혈액순환과 관련된 혈관계 질환들로 이들에 의한 사망 비율의 합은 암의 비율과 비슷하게 나타나 혈관계 질환의 심각성을 나타내고 있다. 혈전에 의한 혈관계 질환은 혈전용해제와 트롬빈저해제를 이용하여 치료하고 예방할 수 있으며, a-glucosidase 저해제는 당뇨병 예방과 치료에 이용할 수 있다. 예로부터 민간요법에서 다양하게 이용되어 왔던 반하를 성인병 치료와 예방에 관련된 기능성식품의 자료로 사용하기 위해 반하 열수추출물을 유기 용매로 분획화하고 분획물의 혈전용해활성, 트롬빈저해활성, 혈당강하효과를 측정하였다. 혈전을 직접 용해하는 활성을 측정하기위해 100 mg/ml의 농도로 준비한 시료액을 fibrin plate 방법을 이용하여 혈전용해활성을 측정 결과 hexane 층, chloroform 층, ethyl acetate 층, butanol 층에서는 활성을 나타내지 않았지만 물 층에서 만 0.8 plasmin unit의 높은 혈전용해활성을 나타냈다. 혈전의 형성을 억제하기위해 혈전형성의 필수 효소인 트롬빈의 활성을 저해하는 트롬빈저해활성을 측정하기위해 10 배 희석한 (10 mg/ml) 시료 용액을 이용하여 트롬빈저해활성을 측정결과 hexane 층에서 75.3%의 높은 트롬빈저해활성을 나타냈으며, ethyl acetate 층과 chloroform에서도 각각 43.3%와 39.7%의 활성을 나타냈다. 또한 탄수화물의 소화를 지연시킴으로서 소장에서 포도당의 흡수를 억제하여 혈관내 당의 농도를 조절하는 혈당강하제의 개발을 위해 준비된 조효소액(10 mg/ml)을 이용하여 a-glucosidase 저해활성을 측정한 결과 ethyl acetate 층과 hexane 층에서 각각 24.9%와 23.4%의 저해활성을 확인하였다. 따라서 반하의 hexane 층과 ethyl acetate 층과 물층은 혈전관련 혈관계 질환과 당뇨병 관련 기능성 식품의 개발 소재로 이용할 수 있을 것으로 기대된다. 특히 최근 질병 치료에 효과는 크지만 부작용이 동반되는 화학약품들 대신 활성은 조금 뒤지지만 부작용이 없는 생약을 이용하려는 경향이 커지고 있다. 따라서 오랫동안 민간용법에서 사용되고 있는 반하는 안정성이 확인된 장점을 갖고 있어, 식품재료나 기능성 음료에 이용할 경우 장기간 섭취가 가능해 혈관계 질환의 치료와 예방에 큰 효과를 가져 올 수 있을 것으로 기대된다.
${\beta}-carotene$ was extracted from freeze-dried carrot by supercritical carbon dioxide $(SC-CO_2)$ and mixtures of $CO_2$ doped with ethanol or methanol as a cosolvent at temperatures of 40 to $60^{\circ}C$ and pressures of 138 to 276 bar. Solubility of ${\beta}-carotene$ in $SC-CO_2$ increased with the increase of extraction pressure and the decrease of extraction temperature. The highest solubility observed was $4.90\;{\mu}g/g\;CO_2\;for\;{\alpha}-carotene\;and\;0.604\;{\mu}g/g\;CO_2$ for ${\alpha}-carotene\;at\;40^{\circ}C$ and 276 bar. Addition of ethanol increased the solubility being the largest increase of 82% using a mixture of $CO_2$ and 17.4% ethanol. $SC-CO_2$ extraction can be used to selectively obtain natural carotenoids, free of solvent residuals, which can be used as food additives.
Marker pens belong to school things that are controlled by the regulation system called safety confirmation under special act on the safety of products for children with the formaldehyde criteria of 20 mg/kg. With nine marker pens available commercially, formaldehyde in marker pen ink was analyzed by present test standard where marking on a fabric swatch with a pen and extracting the swatch in water and derivatization with Nash reagent followed by UV/Vis spectrophotometeric measurement (Nash-UV/Vis method), giving not detected results or a false positive result in case of a colored water extract. However, the contents of formaldehyde in ink of nine marker pens were determinded to range between 3.2 ~ 93.2 mg/kg with three results above the safety criteria of 20 mg/kg by HPLC/DAD measurements on DNPH derivatives of formaldehyde (DNPH-HPLC/DAD method) in ink dissolved directly in water using an ultrasonic bath. Therefore, the DNPH-HPLC/DAD method with the extraction of ultrasonic dissolving ink in water is proposed as a proper method for analyzing formaldehyde in ink. The proposed method has advantages of lower detection limit and accuracy with colored extracts as well as a simple and fast extraction. The accuracy and precision of this method was estimated to be 90.1 ~ 105.4 % and 0.6 ~ 3.3 %, respectively by spiking tests in the ranges of 20 mg/kg and 40 mg/kg using matrixes such as highlighter pen ink, board marker ink, chalk marker pen ink and painter marker ink.
The comparative analysis of glycerin in cosmetic samples was carried out by LC/MS and $^1H$ NMR spectrometry. For the LC/MS analysis, aqueous solution was controlled in strong basic condition with sodium hydroxide, and benzoyl chloride was added to the solution for the derivatization of glycerin. The derivative was extracted using pentane and analyzed by the LC/MS. For the $^1H$ NMR analysis, sample was directly dissolved in $D_2O$ solvent without pretreatment. The quantitative analysis of glycerin was done by $^1H$ NMR ERETIC method. The analysis results of LC/MS and $^1H$ NMR showed that the calibration curves were a good linearity with $r^2=0.9991$ in the range of 0.1 to $10{\mu}g/mL$ and $r^2=1$ in the range of 25 to $500{\mu}g/mL$, respectively.
Quantitative analytical condition for nickel in ceramic, metal and plastic materials using complexation and solvent extraction followed by inductively coupled plasma-atomic emission spectrometry (ICP-AES) and atomic absorption spectrophotometry (AAS) was studied. Ceramic, metal and plastic samples were dissolved by acid digestion. Nickel was determined by ICP-AES and AAS after extraction of Ni $(DMG)_2$ in $CHCl_3$. Recovery efficiency of nickel was satisfactory, and most of matrix elements causing interference could be effectively eliminated by the separation. Nickel in the certified reference materials (BAM-376 and PACS-2) were quantitatively determined without influence of sample matrix.

This study was performed on the effects of extraction temperature, pressure, time on apparent solubility and extraction yield of perilla oil and tocopherol, and color and fatty acid composition of the residue in supercritical carbon dioxide $(SC-CO_2)$ extraction. Apparent solubility of perilla oil and tocopherol increased with the increase of $CO_2$ density and was found to strongly depend on extraction pressure rather than extraction temperature. The extract yield of tocopherol in $SC-CO_2$ extraction increased with an increase of temperature and decreased with an increase of pressure and extraction time. The perilla oil apparent solubility of dried perilla powder for $60\sim180$ min at $40^{\circ}C/276$ bar increased with an extraction time, on the other hands, tocopherol apparent solubility decreased. As the increase of $CO_2$ density, less redness and yellowness increased. Fatty acid composition of perilla oil showed that perilla oil extracted by $SC-CO_2$ had better unsaturated fatty acid and decreased in saturated fatty acid. $SC-CO_2$ extraction offers a safe natural method for gaining perilla oil from dried perilla seeds powder.

The CKD extract is wastewater from which KCl in CKD has been removed to reuse CKD as a cement raw material, and tried to reuse no extracts due to problems such as wastewater treatment facility expansion. As a result of removing KCl by the ion exchange method, the pH of the extract after ion exchange decreased from 12.7 to less than pH 2, and it was confirmed that H+ of the cation exchange resin was dissolved in the extract through ion exchange. In addition, the selectivity of the ion exchange was removed in the order of Ca2+, K+, it was determined that the increase in the contact time to remove the K+ ions. The batch system had a contact time of 6 times or more, compared to the continuous system, and showed 4 times of K+ removal efficiency and 7 times of Cl- removal efficiency. It was showed by analyzing the pH of the extract that more H+ of the cation exchange resin was extracted than OH- of anion exchange resin as the pH of the extract was changed.

This study was intended to observe possibility of which radiation technique can be used for oligopeptide production from pigskin collagen to reduce environmental pollution in processing and simplify the processing steps. Raw pigskin was ground using chopper, and then defatted in acetone cooled at $-20^{\circ}C$ freezer. Defatted dried pigskin was irradiated at 20, 40, 60, 100, 150, 200, 250, and 300 kGy using Co-60 gamma rays irradiator. With irradiation doses, the amount of soluble proteins increased, and the viscosity and turbidity of soluble proteins decreased, which could be clue of that irradiation degrade high molecular proteins directly. pH of soluble proteins from defatted pigskin increased in the sample above 150 kGy, and low molecular weight components (below 24 kDa) in SDS-PAGE increased. From gel permeation chromatography of the hydrolysates of pigskin irradiated at 300 kGy showed the major peak of 9,000, 8,200, 860, and 170 Da.
