  • The protozoan pathogen Giardia lamblia has been major cause of waterborne enteric disease. In this study, we tried to identify G. lamlbia of human infectious species and to detect viable C. lamblia in river water samples including three sites of Han River mainstream and an its creek using PCR and RT-PCR technique. The PCR/RT-PCR methods were performed by using giardin primer based on the giardin gene targeting ventral disk of Giardia. Sensitivity testing in the DNA/RNA extraction and PCR/RT-PCR amplification steps showed that it was possible to detect a single cyst of G. lamblia and viable G. lamblia. The PCR/RT-PCR methods were compared with immunofluorescence(IF) assay by analyzing 48 samples collected from the mainstream water and the creek water. The mean concentration of the total cysts were 6.3 cysts/10 L(arithmetic mean, n = 48) and the positive detection rate were 62.5%(30/48). And the mean concentration of the cysts excluding empty cysts were 4.5 cysts/10 L and the positive detection rate were 52.1%(25/48). It resulted that 24 of 48 samples included Giardia lamblia by PCR assay and 10 of 48 samples included viable G. lamblia by RT-PCR assay. It resulted that the PCR/RT-PCR technique would be available to river water samples with low concentration of Giardia cysts. And it could support the Korean protozoan standard method, which provides useful information for species and viability.

  • No currently available methods to monitor pathogenic protozoa, Cryptosporidium and Giardia in environmental water come close to acceptable sensitivity, specificity and reproducibility, and so it has to be accompanied by thorough quality control and performance evaluation to credibly predict the distribution of them. We collected surface water samples from the Han River and spiked our prepared (oo)cysts, determined Matrix Spike recoveries using USEPA Method 1623 and considered what factors influence MS recovery and validity. As a result, average 46% of spiked oocysts and 60% of spike cysts were recovered, but repetitive sampling and statistical approach seemed to be necessary to determine the environmental pollution level of two protozoa as their variation coefficients was so much as 35oio and 26%. And MS recoveries with two acid dissociations during immunomagnetic separation were improved more 10% than that with one dissociations and the use of spiked suspension enumerated by flow cytometry instead of manual preparation enhanced the validity and reliability in spiking tests. Because fluorescence characteristics of (oo)cysts stained on well slides with FITC-labeled monoclonal antibodies and DAPI was not always same, well Elides from spiked field samples were helpful to evaluate the performance of staining. We found many (oo)cyst-like objects with typical fluorescence, not (co)cysts, from the Han River water samples, and then it was concluded that nuclei staining by DAPI (4',6-diamidino-2-phenylindole) and examination by Differential Interference Contrast Microscope should be critical for valid identification.

  • The effects of temperature, coagulants and pre-chlorination on the removal of turbidity and pathogenic protozoa by coagulation process were investigated using jar test of lab scale. In room temperature ($25^{\circ}C$), protozoa were removed over 1.0log at the proper concentration range of coagulants, and up to over 2log at the optimal concentration of coagulants. Considering the 1.5log target removal for Giardiain the processes of coagulation, sedimentation, and filtration, this results implies that the target could be satisfied. However, the removal of protozoa and turbidity was reduced, and optimal PAC concentration was narrowed in low turbidity and cold temperature ($5^{\circ}C$). These results suggest that the drop of coagulation efficiency may be occurred in winter if the conditions are not optimized. Despite the effect of water temperature, the relation of turbidity and protozoa removal appeared to be good. The various kinds of coagulants did not significantly affected for removals of turbidity and protozoa when the concentrations of $Al_2O_3$ were considered. Prechlorination did not increase or decrease the removal of turbidity and protozoa in optimum condition at room temperature, pH 7, 15mg/L of PAC concentration.

  • The removal of protozoa in the coagulation process was evaluated under the different pH and turbidity using the jar test after the addition of polyaluminium chloride (PAC) as a coagulant. Two well-known protozoa of Cryptosporidium parvum and Giardia lamblia were tested at the same time with turbidity, the critical water quality parameter of the water treatment process. Both protozoa were removed about 1log (and up to 2log) at the optimum injection of PAC. The source water turbidity and pH affected the removal of protozoa and turbidity. At neutral and alkaline pH, 1.3-1.7log removal of protozoa for low turbid water with 5NTU, and 1.6-2.3log removal for high turbid water with 30NTU were achieved. However, at acidic pH, maximum 0.8-1.0log and 1.1-1.2log were removed for low and high turbid water, respectively, at the optimum PAC injection of 15mg/L. The relation of protozoa and turbidity removals were expressed as the 1st order equation (significantly positive relation) in the most of the tested conditions. In addition, the relation of protozoan removals with residual turbidity were also expressed the 1st order equation (significantly negative relation), although the significance of the equations were reduced at acidic pH. Therefore, residual turbidity could be a good index of efficient protozoan removal in the coagulation process, probably except at the low pH condition.

  • In order to evaluate the distribution level of Cryptosporidium and Giardia in drinking water resources, distribution of Cryptosporidium and Giardia was studied in intake water of main water treatment facility with treatment capacity over 50,000 ton/day in Korea. 10 L samples from each study sites were collected quarterly for 2 years between Oct. '04 and Dec. '07. Cryptosporidium and Giardia were filtered and concentrated by capsule filter and centrifugation, and analyzed by immunomagnetic separation process and fluorescent assay. Cryptosporidium oocysts and Giardia cysts were detected in 7.0% and 9.3%, respectively, of a total 776 samples from 97 study sites. And mean detection number of Cryptosporidium oocysts and Giardia cysts in total 776 samples was 0.11/10 L, 0.16/10 L, detection range of Cryptosporidium oocysts and Giardia cysts was 0~7/10 L, 0~4/10 L, respectively. In seasonal distribution, Cryptosporidium was more frequent in spring as 9.2% than other season, Giardia was more frequent in winter as 14.6% than other season, but there was not shown significant seasonal characteristics. In correlation analysis with total 776 data, Cryptosporidium had significant correlation to total coliforms at the 0.05 level, but correlation value was too low as 0.07 (r=0.07). In case of Giardia, what had significant correlation at the 0.05 level was total coliforms and fecal coliforms, but correlation value was too low as 0.26, 0.27 respectively.

  • In order to secure the reliability of pathogenic microorganisms such as Cryptosporidium and Giaridia, which are chlorophilic protozoans, membrane filtration systems have been widely used in water purification process. hese integrity tests are classified into direct and indirect methods. Based on the bubble point theory, the pressure-based test in the direct method is presented in the USEPA Guidance Manual with sensitivity to detect a minimum size of pathogenic microorganisms of $3{\mu}m$ or more. Indirect methods are widely used in that they are capable of continuous operation in on-line state, but there is a very low sensitivity of damage detection compared to the direct method, and there is a limit that can not specify the damage area, so it is necessary to improve this sensitivity. In this study, we compared the LRVDIT and UCL values according to the type of membrane defect, number of fiber breaks, and initial set pressure value through the Integrity Test by Pressure Decay Test (PDT).

  • Although the detected amounts of those disease-causing(oo)cysts were high at each point in 2001, the detected amounts of Cryptosporidium and Giardia had decreased at low levels below 10(oo)cysts until 2004. Considered the detection results of each season, the most highly detected amounts of(oo)cysts were mostly recorded in summer and late summer(from June to September), while the site which the biggest amount was detected, was the point at Goryeung. The relation of Cryptosporidium with the water quality items was low, but the relations of Cryptosporidium and Giardia with turbidity were high($R^2$=0.4914, $R^2$=0.6176 respectively). And the detected amounts of Cryptosporidium and Giardia were relatively high, 20(oo)cysts/20L when the turbidity was more than 20 NTU. It was also found that the relation between the distribution of Cryptosporidium and Giardia was high, $R^2$=0.8432 and the density of Giardia was generally higher than that of Cryptosporidium. And Cryptosporidium and Giardia were simultaneously detected at each point. The primary sources that affected the density of those(oo)cysts were the livestock waste water and sewage.
