Improve Effects of Saengshik on Patient with Fatty Liver and Hyperlipidemia in Murine (생식 섭취가 지방간 개선 및 지질 대사에 미치는 영향)

  • To examine the effect of Saengshik on parameters related to hepatoprotective, anthropometric, blood pressure, serum lipid and blood related indices, nonalcoholic fatty liver subjects were treated with two meal portion of Saengshik in the replacement of meals for a period of three months. Weight, Body Mass Index (BMI) and systolic blood pressure were significantly decreased after the treatment. Chronically elevated serum aspartate aminotransferase (AST), gamma-Glutamyl transferase (r-GTP) and Alkaline Phosphatase (ALP) levels showed reduction to the near normal range. Serum total triglyceride level were reduced following the treatment. Whereas, there were no changes of serum total cholesterol with Saengshik consumption. Also, additional study was conducted to investigate the effect of Saengshik supplementation to high cholesterol and fat diet on lipid metabolism in rats. Male Spraque-Dawley rats were administrated hyperlipidemiainducing diet containing 1% cholesterol and 10% lard to induce hyperlipidemia for 4 weeks and were fed on diet containing Saengshik (30%, w/w) for 7 weeks. The feeding of diet containing 30% Shaengshik significantly decreased total cholesterol (TC) contents and total triglyceride. These results demonstrate Saengshik may be beneficial for fatty liver patients in improving their lipid metabolism.

  • In the present work, the interaction of fatty acid with vesicle membrane of phospholipids was investigated using 3 different kinds of fatty acids such as stearic acid (SA), oleic acid (OA) and linoleic acid (LA). Basically, the same trend has been found in 3 fatty acid systems. The addition of fatty acid produced a close packing of liposome due to the penetration of fatty acid molecules into liposome vesicles, which resulted in a decrease in size and an increase in zeta potential of liposome. However, excessive addition of fatty acid produced a transition from liposomes to aggregates of lipid particles having polymorphic structure. The membrane fluidity, characterized by measuring membrane deformability and fluorescence anisotropy ratio of liposomes, was in good agreement with measurement results of transmission electron microscopy (TEM) and particle size. The minimum size and closest packing of liposome with SA, OA and LA were found when the molar ratios of fatty acid to lecithin were 0.70, 0.50, and 0.25 respectively.

  • When cultured soybean immature seed on 15th days after flowering, suitable temperature in formation of callus were $24{\sim}27^{\circ}C$, and embryogenic callus(EC) were generated at medium containing NAA with growth regulators, and then, results were subcultured the EC, a plenty of shoots and roots were formed at medium supplemented BA 2mg/1 and IAA 2mg/1, respectively, however when used at medium the same time supplemented BA 2mg/1 and IAA 2mg/1, formation of cullus was energetic, and a symptom of organization was not showed , Total lipid contents include in each cultures were increased at low temperature of cultural conditions as much as possible, but glycolipid, phospholipid, free sterol contents were a little increased at $24{\sim}27^{\circ}C$, and free sterol content was increased at a case of embryogenic structure were generated. In fatty acid compositions in each cultures, the contents of unsaturated fatty acid were plenty in EC, and unsaturation rate was 0.837. Besides, in sterol compositions, cholesterol content was remarkably high in EC than that of other cultures.

The Effects of Dietary Supplementation of Vitamin C or E on the Expressions of Endoplasmic Reticulum Stress, Lipid and Glucose Metabolism Associated Genes in Broiler Chickens (비타민 C 및 E의 첨가 급여가 육계의 소포체 스트레스와 지방 및 포도당 대사 연관 유전자의 발현에 미치는 영향)

  • This study was conducted to evaluate the effects of dietary supplementation of vitamin C or E on the expressions of endoplasmic reticulum (ER) stress, lipid and glucose metabolism associated genes in broiler chickens. A total of 216 one-day-old male broilers was randomly alloted to 4 treatments with 6 replicate pens per treatment and 9 broilers per pen for 35 days. The dietary treatments were control, vitamin C (control diet + ascorbic acid 200 mg/kg diet), vitamin E (control diet + ${\alpha}$-tocopherol 100 mg/kg diet), vitamin C + E (control diet + vitamin C 200 mg/kg + vitamin E 100 mg/kg), respectively. To evaluate gene expressions by quantitative real-time polymerase chain reaction, total RNA was extracted from the liver of the chicken at 35 days of age. Dietary supplementation of vitamins was significantly down-regulated the expression of stress marker genes including HSP70, HSP90, and HMGCR, as compared to the control (p<0.05). The expressions of ER stress associated genes also inhibited by supplementation of vitamins as well (p<0.05). Vitamin C supplementation suppressed the expression of lipid associated genes such as FASN, FATP1 and ACSL1. Vitamin supplementation did not affect the glucose transporters, GLUT2 and GLUT8, in the liver. The results of the present study indicated that dietary supplementation of vitamin C or E could be beneficial for the alleviating physiological stress in broiler chickens.

  • Seasonal changes in biochemical composition of muscle, gonad-viceral, mass and whole body of the cultured ark shell, Scapharca broughtonii in the Gamag bay of Yeosu city were studied from December 2008 to November 2009 in relation to environmental condition and reproductive cycles. Average monthly water temperature in the winter was in the range of $7-12^{\circ}C$ and $20-25^{\circ}C$ in the summer, while the salinity fluctuated in the range of 30.1%-33.8‰ on the average. Seasonal fluctuation of the concentration of nutrient salt was the highest in September ($13.04{\mu}g/L$) with average annual concentration of $4.6{\mu}g/L$. The main spawning season of the ark shell was during the months of July and August, and the gonads were in inactive stage during the winter. The gonad-visceral mass contained lower amounts of proteins than the other body parts. The most marked changes in body composition were lipids and carbohydrates within the gonad-visceral mass, and protein for each of the organs was relatively consistent throughout the year. All the parts in the visceral sac displayed the highest changes during the gametogenic cycle while the contents of moisture and lipid within the visceral act displayed somewhat inverse relations with each other. Moisture content was the lowest during the inactive stage during which the lipid content is the highest. The lipid content was the lowest immediately following spawning with increase in the moisture content as the lipid is being consumed. Protein mass within the visceral sac was low in comparison to the muscle mass. It is deemed that carbohydrates, lipids and proteins in the visceral sac play the major role as the source of energy during the development process of the gonads, and used for maintenance of base metabolism when available food is scarce.

  • A persistent major haemolyruph protein (MHP) was confirmed, and its ontogeny and physicochemical charadedstics were investigated in Helicoverpa assulta. The MHP existed continually during larval-pupal-adult development, and its ontogeny was similar to that of total haemolymph protein concentration during development. Its content increased with larval growth, and kept to high level during pupal-adult development except for temporary decrease at the early pupal and adult stages. The MHP was purified by ammonium sulfate precipitation, gel filtration and ion exchange chromatography. The purified MHP was determined to be hexamer glycolipoprotein (pI 5.9, M.W. 414kDa) consisted of single type subunit (69kDa). Amino acid analysis suggested that the MHP contained a relatively high content of aromatic amino acids (18.27 mole % of tryptophan, 7.47 mole % of tyrosine and 6.51 mole % of phenylalanine) compared to storage proteins from other insects. Immunodiffusion test and electrophoretic analysis of the organ proteins (gut, fat body, and Malphigian tubule) suggested that the major haemolymph protein was present in the fat body.

Characterization and Distribution of Transferrin from the Last Larval Haemolymph of Papilio xuthus (호랑나비 유충 혈림프 Transferrin의 특성과 분포)

  • Transferrin is a molecule carrying iron to store and maintain for iron homeostasis of living organisms. In this study, we have purified transferrin, as an iron-binding protein, from the last larval haemolymph of Papilio xuthus by KBr density gradient ultracentrifugation and gel filtration (superose 6 HR) using fast protein liquid chromatography (FPLC) and transferrin containing iron was identified by Ferene S staining. The purified haemolymph transferrin was shown to have molecular mass of 78 and 80 kDa and amino acid composition of transferrin was rich in aspartic acid, valine, leucine and glutamic acid. With immuno-diffusion assay, we confirmed the existence of the transferrin in the haemo-lymph and fat body by detection of visible and clear positive reaction. From the quantitative comparison by rocket immuno-electrophoresis process, the amount of transferrin were increased in the haemolymph of 3 days after pupation and the whole 5 days after pupation. Here, with biochemical and immunohistochemical analysis, we speculate the relationship of transferrin between the physical characteristics and distribution during metamorphosis of P. xuthus.

Components and Antimicrobial Activity of Veiled Lady Mushroom, Dictyophora echinovolvata (흰돌기망태버섯(가칭; Dictyophora echinovolvata)의 일반성분 및 항미생물활성)

  • A strain of Dictyophora echinovolvata ASI 32002 showing good fruiting body formation was selected. Analyses of chemical and nutritional components as well as antimicrobial activity of different parts of the mushroom such as mycelium, egg, and fruiting body were carried out. There were differences in the chemical compositions and the quantities depending on developmental stages of veiled lady mushroom, D. echinovolvata ASI 32002. Nitrogen, phosphate, magnesium, and calcium in inorganic chemicals were abundant in mycelium, and potassium and mineral elements were abundant in the egg and fruiting body. Mannitol and trehalose were abundant in free sugar contents. Glutamic acid and arginine in mycelium and aspartic acid and glutamic acid in egg and fruiting body were abundant in free amino acid contents. Linoleic acid, an polyunsaturated fatty acid, was abundant in all parts of the Dictyophora species, but compositions and quantities of other fatty acids varied depending on the different parts of the mushroom. It was detected that malic acid, lactic acid and acetic acid in mycelium, formic acid, acetic acid and fumaric acid in egg, and malic acid, citric acid, lactic acid, fumaric acid in fruiting body were abundant. The methanol extracts of D. echinovolvata ASI 32002 mycelium showed antifungal activity with minimal inhibition concentration (MIC) of $62{\sim}125\;{\mu}g/ml$ that was similar levels of cyclohexamide against Aspergillus awamori, Hypocrea nigricance and Trichoderma virens. The MIC of extracts from mycelium and fruiting body against Candida albicans was $250\;{\mu}g/ml$, similar to that of tetracycline.   • Mulberry leaf sprayed on at the various concentrations of cadmium (Cd) was fed on the just molted fifth instar through the 6th day of the fifth instar and content of cadmium and zinc in the feces, organs and tissues involved were analyzed by spectrophotometer. Along with the effect of those heavy metals on development of the relevant organs and tissues sexwise. 1. The residual content of cadmium on mulberry leaf was about 50% of the total cadmium and the weight of feces disposed from the cadmium fed larvae was less than the control. There was distinctive differences in female-wise disposal of feces between cadmium treatment and control ; cadmium treatment female larvae disposed less feces than female of control. 2. Fourteen percent of the total dry weight of the anterior plus the middle silkgland and of the posterior silkgland in cadmium 100ppm treatment was decreased in female but only 9% of the anterior plus middle silkgland in male was made, showing no decrease the weight of the posterior silkgland in male. 3. Decrease of the dry weight of the silkgland, fat body and hindgut was made in cadmium 100ppm treatment and contrarily the weight of midgut and malphihian tube were increased. The total larval weight of the female and male were decreasd in cadmium 100ppm treatment and the decreasing ratio was higher in famale. 4. There wasa no detection of cadmium in all of the organs and tissues of the control and in those of the cadmium treatment the high residual content was in order of the midgut, hindgut, malphighian tube and head. A slight accumulation of cadmium was shown in fat body and silkgland. 5. Cadmium accumulation in the midgut was higher than in female but in the fat body, malphighian tube, trachea and integument was contrarily high in the female than male. 6. The total zinc content of the larvae was below 60ppm in the control and 80ppm in cadmium 100ppm treantment. The increase of zinc content in cadmium treatment was made on silkgland, malphigian tube and midgut but contrarily the decrease of zinc content in fat body was resulted. 7. In cadmium treatment, much more cadmium accumulation took place in the female and zinc accumulation was made much more in the male.

Effects of Varying the Concentration of Energy Yielding Nutrients on Nitrogen Balance and Body Composition of the Growing Rats (열량 영앙소의 수준이 흰쥐의 단백질 평형과 체조성에 미치는 영향)

  • In order to investigate the utilization efficiency of dietary protein in the rat body and effects of dietary fat on its protein metabolism, 51 Sprague-Dawly rats of 6 weeks old weighing approximate)y 106g were subjected to feeding trials for 4 weeks and then subsequently to metabolic trials for 3 weeks using six different diets composed of three different levels of protein (10%, 20% and 30%) with low (20%) and high (40%) fat content for each protein level, total energy being fixed at 4000 Kcal/kg by addition of an appropriate amount of carbohydrate, and the following results were obtained. 1) The body weight gain increased in the low fat diets with increasing protein level whereas it decreased in the high fat diets. Food efficiency also increased in the low fat diets with increasing protein level, but no do deffinitive trend was observed in the high fat diets. The protein efficiency was generally higher at low protein level and tended to decrease as the protein level increased, regardless of the amount of fat intake. 2) As the protein level was increasing, digestibility of dry matters and carbohydrate were decreasing whereas that of protein was slightly increasing. On the other hand, digestibility of fat was always very high regardless of the amounts of dietary protein and fat. 3) The gross energy intake was affected both by protein and fat contents in the diets: The energy metabolism efficiency was decreasing with increasing protein level and at the same protein level the energy utilization was considerably higher in the low fat diets than in the high fat. 4) From the above-mentioned experimental results it may be concluded that the best formula of diet for growing rats is probably composed of 20% protein, 20% fat and 60% carbohydrate.
