Development of Mathematical Model for the Hydrolysis Fish Oil (물고기 기름의 가수분해에 대한 수학적 모형개발)

  • The functional relationship between the number of mole of an i-fatty acid (Si) included in fish oil and the hydrolysis time(t) was expressed as a mathematical model, $S_i=-{\alpha_i}1n(t)+\beta_i$. The average errors of calculated values on the basis of the measured values were distributed in the range of less than $5\%$ for all the 15 fatty aids composing of fish oil. The equation of hydrolysis rate of each fatty acid was deduced as $v_i={\gamma_i}exp(\frac{S_i}{\alpha_i})$ from the above-mentioned $S_i=-{\alpha_i}ln(t)+{\beta_i}$. Therefore the hydrolysis yields of fatty acids were analyzed using the equation of $S_i\;Vs.\;t.$. The 15 fatty acids were categorized into 4groups from the view point of hydrolysis yield. The hydrolysis yields of the first group, including C14:0, C16:0, C16:1, C18:0, C18:1 (n-7) and 1l8:1 (n-9), were higher than $70\%$ at 48 hr of hydrolysis. Those of the second group, C20:1, C22:1, C18:3, C20:4 and C20:5, were distributed from $40\%,\;to\;60\%$, and third group were around $30\%$. The final group containing only C22:6 was very hard to be hydrolyzed and the yield was less than $20\%$ at the same time.

  • In this work, modified epoxy resins which were well melted in mild solvent were synthesized and solubility assessment was carried out for synthesized epoxy resins. Bisphenol-A type, phenol novolac type and ortho-cresol novolac type epoxy resins were used and fatty acid, dodecyl phenol (DP) and toluene diisocyanate (TDI) were added for synthesis of modified epoxy resins containing fatty acid (MEFA). Composition was epoxy resin/fatty acid = 1.0/0.5 and fatty acid/DP = 0.25/0.25 by equivalent weight and twelve MEFAs were synthesized according to epoxy resins. Viscosity and solubility were measured for twelve MEFAs. As a result, solubility of MEFA was excellent for mild solvent according to increasement of contents of benzene ring, glycidyl group and carbon number of alkyl group. And physical properties were measured for each coating of paints after preparing transparent paints of MEFA to melt well in mild solvent among twelve MEFAs. As a result, they showed an optimal performance on conditions of equivalent ratio of bisphenol-A type epoxy resin/fatty acid/DP/TDI; 1.0/0.25/0.25/0.5 and equivalent ratio of phenol novolac type epoxy resin/fatty acid/DP; 1.0/0.25/0.25 for drying time, adhesion, hardness, impact resistance and alkali resistance.

Varietal Difference of Oil Content and Omega Fatty Acid omposi tion in Korea Local Perilla (들깨 지방수집종의 기름함량 및 오메가 지방산 조성 차이)

  • This study was conducted to obtain basic information for breeding materials on the oil quality improvement of perilla seeds. Oil contents and omega(${\omega}$)fatty acid compositions of seeds were investigated to 317 varieties of domestic perillas. Oil contents of 317 perilla varieties ranged from 35.6 to 55.3 with 44.5% of varietal mean. The major omega fatty acids contained in the oil were oleic acid(${\omega}$-9) 15.2%, linoleic acid(${\omega}$-36) 13.9% and linolenic acid(${\omega}$-3) 63.1% in the mean value. Varietal variation of $\omega$-9, 6 and 3 fatty acids ranged of 9.5~21.4%, 9.1~20.4% and 50.6~70.5% respectively. Unsaturated fatty acid were averaged 92.2% of seed oil in fatty acid composition. The ratios of ${\omega}$-6 to ${\omega}$-3 ranged of 0.13~0.34%(0.22% in mean value). The highest linolenic acid variety was Yecheonjong being 70.5%. The lowest variety in rations of ${\omega}$-6 to ${\omega}$-3 was Goseongjong being 0.13%. Oil content showed positive correlation with stearic acid and linolenic acid, while the negative correlation with oil content and linoleic acid. On the other hand, A significant negative correlation were showed between lionolenic acid and the ratios ${\omega}$-6 / ${\omega}$-3 fatty acid, saturated fatty acid. Saturated fatty acid was highly correlated with unsaturated fatty acid negatively being r=-0.723$^{**}$

  • The fatty acid compositions of cow's and human milk fats of Korea were compared by Gas-Liquid Chromatography(GLC) and general chemical compositions of their milks were also analyzed. Some similarities between human and cow's milk fatty acids were found. Human milk contained little butyric, caproic and caprylic acid were rich in linoleic acid. Cow's milk contained short chain fatty acids. Methylesters of the fatty acids were prepared by methanolysis.

  • Composition of fatty acids and amino acids in water extracted material of cooks comb plant root was studied by HPLC and GC-MS. There are 14 kinds of fatty acids in water extracted material of cockscomb plant root, especially, hexadecadienoic acid$(C_{16:2})$ was contained. Tricoasnoic acid, lignoceric acid, cerotic acid and 1,1'-[3-(2-cuclopentylethyl:dene)-1,5-pentanediyl] bis-cyclopentane were identified by HPLC and GC-MS. There are 16 kinds of acids containing ${\alpha}-aminobutyric$ acid in water extracted material of cockscomb plant root.

  • Lipid extraction preceding fatty acid methyl esters preparation for gas-liquid chromatography is time-consuming and cumbersome. We performed one-step extraction/methylation method with a mixture of methanol-heptane-benzene-DMP-H$_2$SO$_4$ without prior fat extraction. The simultaneous digestion and lipid transmethylation takes place at 8$0^{\circ}C$ in a single phase. After cooling till room temperature, two phases are formed. The upper one of the phases contains the fatty acid methyl esters ready for GLC. The fatty acid composition of major industrial crops obtained by the one step extraction/methylation method (method 1 and 2) was almost identical with the fatty acid composition of the pure fats extracted with hexane by the Soxtec instrument (method 3). Due to its simplicity, speed, and reduced organic solvent the one-step extraction/methylation method (method 1 and 2) should be useful to determine overall fatty acid composition, especially in situations where many samples have to be analyzed.

  • 식이 중 지방함량이 성장기 뇌의 미토콘드리아 지방산 조성에 미치는 영향을 살펴보기 위해 옥수수기름을 각각 10, 2, 0.5% 포함한 3종류의 식이를 임신한 Sprague Dawley종의 흰쥐에게 임신말기부터 제공했다. 수유기간 중의 영향을 살펴보기 위해 대조군(10%옥수수기름 섭취)의 일부에 분만 후 0.5% 결핍식이를 섭취시켰고, 또 이유후부터 0.5% 결핍식이군의 일부에 10% 정상식이를 주어 회복효과를 관찰하고자 하였다. 전 실험기간 동안 체중과 뇌 무게, 단백질 함량을 측정하였고 뇌에서의 미토콘드리아를 분리하여 지방산 조성을 관찰하였다. 출생후 수유기간 동안에는 대조군에 비해 지방결핍식이군에서 체중과 뇌의 무게가 다소 높았으나 이유 후 부터 점점 감소하여 유의적인 차이를 볼 수 없었고 특히 0.5% 식이군의 체중은 생후 6주와 7주에 유의적인 감소를 보였다. 그러나 체중에 대한 뇌무게와 뇌의 단백질 함량에 있어에는 대조군과 결핍군 사이에 유의적인 차이를 볼 수 없었다. 따라서 지방결핍이 뇌 성장에 미치는 영향은 다른 영양소에 비해 크지 않음을 알 수 있었다. 뇌 미토콘드리아의 지방산 조성은 나이와 식이에 따라 변하였다. 나이를 먹음에 따라 단쇄 포화 지방산은 감소하고 다 불포화 지방산의 함량은 증가하였다. 또한 결핍식이군과 대조군 사이에 뇌 미토콘드리아 지방산조성을 살펴 본 결과 지방 결핍의 좋은 지표가 되고 불포화도, triene 과 tetraene의 비, 그리고 ${\omega}-6$ 계열과 ${\omega}-9$ 계열의 비에 있어서 큰 차이를 볼 수 있었다. 이들로 부터 성장초기에 지방함량을 조절한 식이에 의해 뇌 미토콘드리아 지질 조성이 크게 변하고 이에따라 미토콘드리아의 기능도 유의적인 영향을 받고 있음을 알 수 있었다.

Taxonomic Study of the Genus Candida by Cellular Fatty Acid Analysis (세포내 지방산 조성 분석에 따른 Candida속의 분류)

  • Candida속 효모 27종이 표준 균주를 대상으로 세포내 지방산 조성을 기체 크로마토그래피로 분석하여, 함유 지방산의 종류와 함량비를 근거로 27종이 균주를 3개의 분류군으로 나누었다. 이들 지방산 분석에 의한 분류군을 Candida속의 다른 분류학적 결과들인 coenzyme Q 분자종의 종류, DNA 염기함량 분석, ITS 부위의 제한효소 절단 다형질의 양상, 리보좀 소단위 RNA 유전자의 염기배열 보고와 비교 분석하였다. 그 결과 같은 분류군에 위치하는 균주들 사이의 연관 관계가 거의 일치하였다. 다라서 세포내 지방산 분석은 Candida속 규준들을 유전적 동질성을 갖는 균주들로 분류하는 수단으로 사용될 수 있으며, 유전적으로 매우 다양한 균주들의 집단으로 알려진 Candida속에 관한 분류적 고찰에 일차적으로 사용될 수 있는 유용한 수단임을 알 수 있었다.

Methodological Research on the Instruments of Fatty Acids Determination (지방산의 기기 측정 방법에 관한 연구)

  • In this study, several standard fatty acids were analyzed by three analysis instruments. And also, for the two kinds of soybean oils, fatty acids compositions were determined by three instruments. The results were obtained as follows: 1. In the case of Gas Chromatography (GC), standard fatty acids (Myristic, Stearic, Linoleic, Linolenic, Arachidonic acid) were determined with high reproducibility, but oleic acid/elaidic acid were not seperated. By Capillary Gas Chromatography (CGC), most of standard fatty acids were determined with very high reproducibility than saturated fatty acids, and palmitic acid/oleic acid were not seperated. 2. In the analytical ability of cis-trans fatty acids isomer (oleic acid/elaidic acid), CGC was shown better analytical ability of geometrical isomer than HPLC. Oleic acid/elaidic acid were not seperated by packed column (15% DEGS). The rquire time for standard fatty acids analysis was as follows; GC, 7.21 min., CGC, 9.84 min., HPLC, 24.48 min. 3. The major compositions of fatty acids of each soybean oil (CSOY; refined, DSOY; unrefined) by GC and CGC were linoleic acid, oleic acid, palmitic acid, linolenic acid and stearic acid. But in the case of HPLC, palmitic acid/oleic acid were not seperated. Analytical ability of three instruments on fatty acids composition in each soybean oil was same trend as in the standard fatty acids mixture.

The Effect of Acyl Chain Structure on the Hydrolysis of Fatty Acids from Fish Oil by Lipase-OF 360,000 (아실체인 구조가 효소 Lipase-OF 360,000에 의한 지방산의 가수분해 특성에 미치는 영향)

  • The hydrolysis characteristics of each fatty acid composing the fish oil by means of the lipase from Candida cylindracea was investigated. The saturated fatty acids, C14:0, C16:0 and C18:0, and the unsaturated fatty acids with one double bond, C16:1, C18:1(n-7), C18:(n-9), C20:1 and C22:1 were more easily hydrolyzed than the $\omega$-3 polyunsaturated fatty acids. when the number of carbon of the $\omega$-3 fatty acids was same but that of double bond was different, the hydrolysis of the $\omega$-3 fatty acids having lower number of double bond was more rapidly carried out. When the degree of polyunsaturation was same but the number of carbon was different, the lipase acted more rapidly upon the $\omega$-3 fatty acids with lower number of carbon. Docosahexaenoic acid(DHA) was most highly concentrated in the glyceride mixture after 120hr hydrolysis among the various $\omega$-3 polyunsaturated fatty acids. The weight percentage of DHA to the $\omega$-3 fatty acids in the fish oil was 31.87% but that in the glyceride mixture after 120hr hydrolysis was 51.89%.
