Study on the Quality Improvement of Sardine Surimi (정어리 냉동고기풀의 품질개선에 관한 연구)

  • The processing conditions and quality of sardine surimi were examined: Raw sardine meat was separated, washed in 0.2% $NaHCO_3$ and 0.15% NaCl solution, and then dewatered by centrifuge. The dewatered sardine meat was chopped, mixed with 20% emulsion curd (soybean protein : water : refined sardine oil=1:5:2.6), 4% sorbitol, 4% sucrose, 0.2% polyphosphate and 0.1% sodium erythorbate by stone mortar. The mixed sardine meat was frozen with contact freezer, packed in carton box and then stored at $-25{\pm}2^{\circ}C$. The moisture, crude protein and lipid contents of the sardine surimi product was 73.3%, 15.0% and 6.9%, respectively. Fatty acid composition of product consisted of 28.8% of saturates, 24.3% of monoenes and 47.7% of polyenes and the major fatty acids were 16:0, 20:5, 18:1, 22:6 and 16:1. The results of changes in POV, TBA value, fatty acids, texture and sensory score of products during frozen storage showed that lipid oxidation and freeze denaturation of product could be retarded, and flavor enhanced by addition 20% emulsion curd and 0.1% sodium erythorbate. In an attempt to apply sardine surimi in producing surimi-based product, it was concluded that pollack surimi could be substituted with sardine surimi up to 40% without showing any significant changes in texture and taste of surimi-based product.

  • ZITO/Ag/ZITO multilayer transparent electrodes at room temperature on glass substrates were prepared using RF/DC magnetron sputtering. Transparent conductive films with a sheet resistance of 9.4 Ω/㎡ and a transmittance of 83.2% at 550 nm were obtained for the multilayer structure comprising ZITO/Ag/ZITO (100/8/42 nm). The sheet resistance and transmittance of ZITO/Ag/ZITO multilayer films meant that they would be highly applicable for use in polymer-dispersed liquid crystal (PDLC)-based smart windows due to the ability to effectively block infrared rays (heat rays) and thereby act as an energy-saving smart glass. Effects of the thickness of the PDLC layer and the intensity of ultraviolet light (UV) on electro-optical properties, photopolymerization kinetics, and morphologies of difunctional urethane acrylate-based PDLC systems were investigated using new transparent conducting electrodes. A PDLC cell photo-cured using UV at an intensity of 2.0 mW/c㎡ with a 15 ㎛-thick PDLC layer showed outstanding off-state opacity, good on-state transmittance, and favorable driving voltage. Also, the PDLC-based smart window optimized in this study formed liquid crystal droplets with a favorable microstructure, having an average size range of 2~5 ㎛ for scattering light efficiently, which could contribute to its superior final performance.

Active Transport Characteristics of K+-Na+ Pumping System in Cell Membrane Model which Irradiated by High Energy X-ray (고에너지 엑스선을 조사한 세포막모델에서 K+-Na+ 펌프 시스템의 능동적 전달 특성)

  • The active transport characteristics of $K^+$ and $Na^+$ pumping system of cell membrane model which irradiated by high energy x-ray(linac 6MeV) was investigated. The cell membrane model used in this experiment was a $Na^+$ type sulfonated copolymerized membrane of styrene and divinylbenezene. The initial flux of the ion was increased with increase of both $H^+$ ion concentration. In this experiment range(pH 1.5-5, temperature $36.5^{\circ}C$), the initial flux of $K^+$ which was not irradiated by radiation was found to be from $2.09{\times}10^{-4}$ to $1.32{\times}10^{-3}mole/cm^2{\cdot}h$ and that of $Na^+$ from $7.09{\times}10^{-4}$ to $1.09{\times}10^{-3}mole/cm^2{\cdot}h$. the initial flux of $K^+$ which was irradiated by radiation was found to be from $21.0{\times}10^{-4}$ to $16.7{\times}10^{-3}mole/cm^2{\cdot}h$ and that of $Na^+$ from $62.0{\times}10^{-4}$ to $20.6{\times}10^{-3}mole/cm^2{\cdot}h$. The ratio $K^+$/$Na^+$ of membrane was about 1.10. And the driving force of pH of irradiated membrane was significantly increased about 9-20 times than membrane which was not irradiated. As active transport of $K^+$ and $Na^+$ of cell membrane model were abnormal, cell damages were appeared at cell.

Effect of Food Additives on Heat Sensitivity of Listeria monocytogenes H-12 and Decontamination of Kitchen Utensils (식품 첨가물이 Listeria monocytogenes H-12의 내열성에 미치는 영향 및 오염된 조리기구 제균)

  • Effect of food additives on the heat sensitivity of listeria monocytogenes H-12 inoculated into Pollack surimi was investigated and also confirmed the effectiveness of various decontamination method such as tap water washing, chlorination, ultraviolet irradiation and heat treatment haying been applied on cooking utensils. Food additives such as polyphosphate, chitosan, and potassium sorbate increased heat sensitivity of t monocytogenes H-12 and polyphosphate showed the strongest synergistic effect. The tested strain was not detected from stainless steel and plastic cutting board contaminated with $10^4{\~}10^5/cm^2$ of L monocytogenes H-12 after tap water washing for 10 seconds or 1 minute, but washing effect was not found in wooden cutting board. The chlorination of stainless steel and plastic cutting board for 10 seconds with $5{\~}50 ppm$ solution eliminated all cells of the contaminated strain, however any change of the viable cell count was not observed in the chlorination of wooden cutting board, W irradiation on stainless steel and plastic cutting board for 5 minutes with 15 W above 30 cm eliminated the contaminated strain, but the tested strain was still found after 60 seconds of irradiation on wooden cutting board. The treatment of hot water on all used cutting boards for 10 seconds at $70^{\circ}C$ resulted in complete loss of viability of the contaminated strain.

  • Chitosan was prepared from chitin which was abstracted from wasted crab shells. Then itaconic acid was graft-copolymerized onto chitosan using ceric ammonium nitrate as a reaction initiator. To investigate the optimal grafting conditions, the influences of several factors on the grafting were studied, i. e., the concentrations of CAN and itaconic acid, the reaction temperature and time. And to find out its flocculation ability. the flocculation test was carried out with a metal plating factory waste water. The state of graft-copolymer was identified through IR spectra analysis. The optimal grafting conditions and flocculation results were shown to be : concentration of ceric ammonium nitrate is $3.5{\times}10^3M$, reaction temperature is $40^{\circ}C$ and reaction time is 4hrs with 0.25M of the monomer(itaconic acid). Though flocculation tests using chitosan, grafted chitosan and cation, CODcr. metal ions removal rates were measured. The order of superiority is Itaconic acid grafted chitosan>Chitosan>Cationic polymer.

  • Enzymes are widely used in industrial applications such as detergents, food, feed production, pharmaceuticals and medical applications and major contributors to clean industrial products and processes. To screen, identify, and characterize the enzymes the zymography techniques are routinely used. The zymography technique is a simple, sensitive, and quantifiable technique that is widely used to detect functional enzymes following electrophoretic separation in sodium dodecyl sulfate (SDS)-polyacrylamide gels. The method is a versatile two-stage technique involving protein separation by electrophoresis followed by the detection of enzyme activity in polyacrylamide gels under non-reducing conditions. It is based on SDS-polyacrylamide gel (PAG) copolymerization with substrates, which are degraded by the hydrolytic enzymes restored in enzyme reaction buffer after the electrophoretic separation. Any kind of biological sample can be applied and analyzed on zymography, including culture supernatants of microbes, plants extracts, blood, tissue culture fluids, enzymes in foods extracts and metaproteome. The advantage of zymography is that it is possible to directly detect the protein with activity on the electrophoretic gel as well as confirm the activity at the nanogram level. Thus, this zymography technology can be applied in various fields. However, these advantages are rather disadvantageous and can often lead to experimental errors. In this review, the advantages, disadvantages, and problem solving of zymography technique are described.

Manufacture of Biodegradable Polymer with Wastepaper(I) - Pretreatment and Analysis of Chemical Components On Wastepaper - (폐지를 이용한 생분해성 고분자의 제조(I) - 폐지의 화학적 조성 분석 및 전처리 -)

  • Recently many scientists have tried to synthesize biodegradable polymers due to durable and non-biodegradable products of conventional synthetic plastics when these were wasted in nature. So to reuse the wastepapers for biodegradable polymer resources, ONP (old newsprint), OCC (old corrugated containerbpard) were carried out by the pretreatment of chlorinite, hypochlorite and oxygen-alkali treatment conditions. For manufacturing of biodegradable polymer with wastepaper, this study performed to investigate change of chemical components and optimal pretreatment condition. The summarized results in this study were as follows: Lignin content in ONP and OCC was was higher than in MOW and ash content was the highest in MOW. More amount of ash components were reduced by wet defiberation than by dry defiberation. Wet defiberation fiber are better than dry defiberated fiber in chemical pretreatment condition for wastepapers, and the best result was obtained in the condition of sodium chlorite at $70^{\circ}C$, because it has high delignification ratio, ${\alpha}$-cellulose contents and degree of polymerization in this treatment condition. Oxygen-alkali treatment condition is the worst method because of low yield, low degree of polymerization in this pretreatments.

Evolution of sea Urchin Strongylocentrotus intermedius Based on DNA Sequences of a Mitochondrial Gene, Cytochrome c Oxidase Subunit I (미토콘드리아 유전자, 치토그롬 옥시다제(subunit I)의 염기서열을 이용한 새치성게(Strongylocentrotus intermedius)의 진화과정 분석)

  • Sea urchin S. intermedius occurring in the Korean east coast is a cold water species that belongs to the family Strongylocentrotidae of Echinoidea. Although it is known that there are nine species in the family, species identification criteria, phylogenetic relationships, time and process of evolution of the family members have not been uncovered clearly. In the present study, I tried to find some clues to such problems for S. intermedius by means of DNA sequences. For this, cytochrome c oxidase subunit I (COI), one of the mitochondrial genes that evolve fast and follow maternal inheritance was analyzed. DNA was extracted from the female gonad of S. intermedius, a segment of COI gene amplified by polymerase chain reaction (PCR), and finally a total of 1077 base pair sequence of COI obtained by cloning and sequencing the PCR product. The sequence was compared with homologous genes of other sea urchins and echinoderm species. Phylogenetic trees of the COI gene segment revealed that S. intenedius is a sister species of S. purpuratus which lives along the east coast of the Paciflc. With reference to the fossil records of sea urchins and genetic distances in the molecular phylogenies, it is estimated that the two species were separated about 0.89 million years ago when the earth temperature fluctuated significantly. The current disjunct distribution patterns of the two species and the climate change of the earth at the time of separation suggest that speciation might have occurred by vicariance. The COI gene sequence obtained here now can be used as a molecular character which discerns S. intermedius from the other sea urchin species of Strongylocentrotidae.

  • ABA type triblock copolymer of poly(hydroxyl ethyl acrylate )-b-polystyrene-b-poly(hydroxyl ethyl acrylate), i.e. PHEA-b-PS-b-PHEA, was synthesized throughatom transfer radical polymerization (ATRP). This block copolymer was thermally crosslinked with 4,5-imidazole dicarboxylic acid (IDA) via the esterification between the -OH groups of PHEA in block copolymer and the -COOH groups of IDA. Upon doping with ${H_3}{PO_4}$ to form imidazole-${H_3}{PO_4}$ complexes, the proton conductivity of membranes continuously increased with increasing ${H_3}{PO_4}$ content. The PHEA-b-PS-b-PHEA/IDA/${H_3}{PO_4}$ polymer membrane with [HEA]:[IDA]:[${H_3}{PO_4}$]=3:4:4 exhibited a maximum proton conductivity of 0.01 S/cm at $100^{\circ}C$ under anhydrous conditions. Thermal gravimetric analysis (TGA) shows that the PHEA-b-PS-b-PHEA/IDA/${H_3}{PO_4}$ complex membranes were thermally stable up to $350^{\circ}C$, indicating their applicability in fuel cells.

Molecular cloning and characterization of β-mannanase B from Cellulosimicrobium sp. YB-43 (Cellulosimicrobium sp. YB-43의 mannanase B 유전자 클로닝과 특성 분석)

  • A mannanase gene was cloned into Escherichia coli from Cellulosimicrobium sp. YB-43, which had been found to produce two kinds of mannanase, and sequenced completely. This mannanase gene, designated manB, consisted of 1,284 nucleotides encoding a polypeptide of 427 amino acid residues. Based on the deduced amino acid sequence, the ManB was identified to be a modular enzyme including two carbohydrate binding domains besides the catalytic domain, which was highly homologous to mannanases belonging to the glycosyl hydrolase family 5. The N-terminal amino acid sequence of ManB, purified from a cell-free extract of the recombinant E. coli carrying a Cellulosimicrobium sp. YB-43 manB gene, has been determined as QGASAASDG, which was correctly corresponding to signal peptide predicted by SignalP4.1 server for Gram-negative bacteria. The purified ManB had a pH optimum for its activity at pH 6.5~7.0 and a temperature optimum at $55^{\circ}C$. The enzyme was active on locust bean gum (LBG), konjac and guar gum, while it did not exhibit activity towards carboxymethylcellulose, xylan, starch, and para-nitrophenyl-${\beta}$-mannopyranoside. The activity of enzyme was inhibited very slightly by $Mg^{2+}$, $K^+$, and $Na^+$, and significantly inhibited by $Cu^{2+}$, $Zn^{2+}$, $Mn^{2+}$, and SDS. The enzyme could hydrolyze mannooligosaccharides larger than mannobiose, which was the most predominant product resulting from the ManB hydrolysis for mannooligosaccharides and LBG.
