Interlaboratory Comparison of Blood Lead Determination in Some Occupational Health Laboratories in Korea (일부 산업보건기관들의 혈중연 분석치 비교)

  • The reliable measurement of metal in biological media in human body is one of critical indicators for the proper evaluation of its toxic effect on human health. Recently in Korea the necessity of quality assurance of measurement in occupational health and occupational hygiene fields brought out regulatory quality control program. Lead is often used as a standard metal for the program in both fields of occupational health and hygiene. During last 20 years lead poisoning was prevalent in Korea and still is one of main heavy metal poisoning and the capability of the measurement of blood lead is one of prerequisites for institute of specialized occupational health in Korea. Furthermore blood lead is most important indicator to evaluate lead burden of human exposure to lead and the reliable and accurate analysis is most needed whenever possible. To evaluate the extent of the interlaboratory differences of blood lead measurement in several well-known institute specialized in occupational health in Korea, authors prepared 68 blood samples from two storage battery industries and all samples were divided into samples with 2 ml. One set of 68 samples were analyzed by authors's laboratory(Soonchunhyang University Institute of Industrial Medicine: SIIM) and 40 samples of other set were analyzed by C University Institute of Industrial Medicine(CIIM) and the rest 28 samples of other set were analyzed by Japanese institute(K Occupational Health Center:KOHC). Authors also prepared test bovine samples which were obtained from Japanese Federation of Occupational Health Organization (JFOHO) for quality control. Authors selected 2 other well-known occupational health laboratories and one laboratory specialized for instrumental analysis. A total of 6 laboratories joined the interlaboratory comparison of blood lead measurement and the results obtained were as follows: 1. There was no significant difference in average blood lead between SIIM and CIIM in different group of blood lead concentration, and the relative standard deviation of two laboratories was less than 3.0%. On the other hand, there was also no significant difference of average blood lead between SIIM and KOHC with relative standard deviation of 6.84% as maximum. 2. Taking less than 15% difference of mean or less than 6 ug/dl difference in below 40 ug/dl in whole blood as a criteria of agreement of measurement between two laboratories, agreement rates were 87.5%(35/40) and 78.6%(22/28) between SIIM and CIIM, SIIM and KOHC respectively. 3. The correlation of blood lead between SIIM and CIIM was 0.975 (p=0.0001) and the regression equation was SIIM = 2.19 + 0.9243 ClIM, whereas the correlation between SUM and KOHC was O.965(p=0.0001) with the equation of SIIM = 1.91 + 0.9794 KOHC. 4. Taking the reference value as a dependent variable and each of 6 laboratories's measurement value as a independent variable, the determination coefficient($R^2$) of simple regression equations of blood lead measurement for bovine test samples were very high($R^2>0.99$), and the regression coefficient(${\beta}$) was between 0.972 and 1.15 which indicated fairly good agreement of measurement results.

Mineralogical and chemical characterization of arsenic solid phases in weath-ered mine tailings and their leaching potential (풍화광미내 고상 비소의 광물학적${\cdot}$화학적 특성 및 용출 가능성 평가)

  • Arsenic contamination around Au-Ag mining areas occurs mainly from the oxidation of arsenopyrite which is frequently contained in mine tailings. In weathered tailings, oxidation of sulfide minerals typically results in the formation of abundant ferric (oxy)hydroxides or (oxy)hydroxysulfates near the tailings surface, and arsenic may be associated with these secondary precipitates. In this study, solid phases of arsenic in weathered tailings of some Au-Ag mines were investigated through the SEM/EDS and sequential extraction analyses. The stability of As solid phases and the leaching potential were assessed with the variation of pH and Eh conditions. Oxidation of sulfides in the tailings samples was indicated by depletion of S molar concentrations compared to As and heavy metals. Under XRD examinations, jarosite as an Fe-oxyhydroxysulfate was found in the tailings of Deokeum, Dongil and Dadeok, and scorodite as an As-bearing crystalline mineral was identified from Dadeok which has the highest concentration of As (4.36 wt.%). Beudantite-like phases and some Pb-arsenates were also found under SEM/EDS analysis, and most of As phases were associated with Fe-(oxy)hydroxides and (oxy)hydroxysulfates despite a few arsenopyrite from Samgwang and Gubong. Sequential extraction analysis also showed that As was present predominantly as coprecipitated with Fe hydroxides from Dongil, Dadeok and Myungbong (72∼99%), and as sulfides (58%) and Fe hydroxide-associated forms (40%) from Samgwang and Gubong. In the tailings leaching experiment, As was released with high amounts by the dissolution of As-bearing Fe(oxy)hydroxysulfates in the lowest pH (2.7) conditions of Deokeum, and by desorption under alkaline conditions of Samgwang and Gubong. Higher leaching rates of arsenite(+3) were found under acidic conditions, which pose a higher risk to water quality. Changes in pH and Eh conditions coupled with microbial processes could influence the stabilities of the As solid phases, and thus, time amendments or landfilling of weathered tailings may result in enhanced As mobilization.

Comparative Analysis of Nutrients and Hazardous Substances in Zophobas atratus Larvae (탈지 전, 후 아메리카왕거저리(Zophobas atratus) 유충의 영양성분 및 유해물질 비교분석)

  • The nutritional composition and optimal eating stage of the super mealworm, Zophobas atratus (Coleoptera : Tenebrionidae), were investigated to explore its use as a food ingredient. It was determined that 10th instar larvae were most suitable for eating in terms of nutritional value as well as economic aspects. To improve the quality of powder production, the nutritional value of 10th instar larvae before and after degreasing was analyzed. After drying the larvae powder, crude protein was the most abundant nutrient both before (52.3%) and after (60.6%) degreasing while crude fat measured 36.3% and 21.7% before and after degreasing, respectively. In terms of essential amino acids, leucine levels were highest and 1.3 times greater after degreasing (4.5%) than before (3.5%). Oleic acid, the highest unsaturated fatty acid in larvae, was 31.7% after degreasing which was 1.1 times higher than before (33.2%). Among various major minerals, potassium was most abundant and 1.4 times higher after degreasing (1267.0 mg/100 g) than before (879.3 mg/100 g). Harmful substances were 1.3 to 2.0 times lower in the degreased larvae, although mercury or pathogenic bacteria were not detected in either group. We therefore conclude that degreased Z. atratus larvae are more suitable for eating than before degreasing.

A Study of Fish Community on Up and Downstream of Hwabuk Dam Under Construction in the Upper Wie Stream. (위천 상류에 건설 중인 화북댐 상 하류 어류군집에 관한 연구)

  • Hwabuk Dam has been under construction to reduce flood damage in Nakdong River watershed and to supply stable water for middle area of Gyeongbuk Province. Therefore, fish investigation in up and downstream of the dam was conducted from 2004 to 2008 in order to determine any negative effect on fish community due to dam construction and to use as fundamental data for conserving species diversity and maintaining stream health. According to data analysis on water quality, temperature, dissolved oxygen, pH, suspended solids, and total E-coli had seasonal variation, but they did not significantly differ in sites. However, biological and chemical oxygen demand, chlorophyll-a, nitrogen, and phosphorus representing organic matter and nutrient concentration were higher in upper site and decreased to lower site so that they differed by site. Concentration of arsenic among the heavy metals was less than 0.05 mg $L^{-1}$, which is regulated for protection of human health in water quality standard, except for 0.092 mg $L^{-1}$ in June 2005. During the study period, the total number of fish caught from the 6 sites was 10,263 representing 7 families 19 species. Among them, dominant and subdominant species were Korean chub (Zacco koreanus, 62.5%) and Chinese minnow (Rhynchocypris oxycephalus, 10.6%) which inhabit mostly in mid and upper streams, Korea. Among the 19 species, Korean endemic species were 9 species (47.4%) including Korean slender gudgeon (Squalidus gracilis majimae), Korean dark sleeper (Odontobutis platycephala), and Korean shiner (Coreoleuciscus splendidus). There was several individuals of the $1^{st}$-class endangered species, Naktong nose loach (Koreocobitis nahtongensis), caught in 2005${\sim}$2007, and no introduced species of fish was found in entire sampling period. According to result of community analysis, dominance index decreased toward lower site, but diversity and richness indices increased toward lower site. The equation of length-weight relationship on the dominant species was TW=0.000003$(TL)^{3.2603}$. The parameter b in the equation was greater than 3.0 indicating good nutritional condition in the populations. Compared to populations of Korean chub in other streams, the population in Hwabuk Dam watershed had higher mean of condition factor by size indicating better growth rate. With fish fauna and multi-metric health assessment model in each sampling attempt, index of biotic integrity (IBI) was evaluated and it resulted mostly in good (26${\sim}$35) and excellent (36${\sim}$40) condition in all sites, and the mean of IBI was the highest in site 5. The results indicate that it is very important to study not only environmental impact assessment with fish composition but also stream health assessment in order to conserve healthy aquatic ecosystem.

Meiobenthic community structure in the coastal area of Hallyeohaesang National Park (한려해상국립공원 해역에 서식하는 중형저서동물의 계절별 군집 변동 특성)

  • To assess the characteristics of meiofaunal community fluctuations related to environmental factors, seasonal surveys were conducted in the subtidal zone of Hallyeohaesang National Park. The average depth of the study area was about 20 m, and the average water temperature at the bottom was low in winter(11.33℃) and high in summer(17.95℃). The sedimentary particles mainly comprised silt and clay at most stations. The abundance of meiofauna ranged from 81.7 to 1,296.5 Inds. 10 cm-2, and the average abundance was 589.3 Inds. 10 cm-2. The average abundance of meiofauna in each season was the lowest at 416.5 Inds. 10cm-2 in winter and the highest at 704.5Inds.10 cm-2 in spring. The dominant taxa were nematodes (about 92%) and harpacticoids (about 5%). In the cluster analysis of meiofaunal communities, they were divided into four significant groups. The largest group mainly contained spring and summer samples, and contained stations with a high nematode density of over 500 Inds. 10 cm-2 and harpacticoids below 50 Inds. 10 cm-2 with a high composition ratio of nematodes. In the cluster analysis, no regional division was found between the stations, and it was thought to be divided by the seasons with high abundance according to seasonal variation and the composition ratio of nematodes and harpacticoids. In the Spearman rank correlation analysis, the density of total meiofauna and the most dominant taxa, nematodes, was not significantly related to environmental factors. However, the density of harpacticoids had a significant positive correlation with water depth and a negative correlation with sediment particle size.

Preparation of Nickel Hexacyanoferrate Ion Exchanger for Electrochemical Separation of Cations (양이온의 전기화학적 분리를 위한 페리시안니켈 이온교환체의 제조에 관한 연구)

  • Although chemical sedimentation and ion exchange are usually applied to the treatment of heavy metal ions and radioactive cations, they have some serious disadvantages like a great consumption of chemicals, the disposal of valuable metals, and the secondary pollution of soil by the solid-waste. The advanced countries recently have studied the electrochemical ion exchange, combined electrochemical reduction and ion exchange, for the development of the alternative technique. This study has been performed to investigate the optimum condition for the preparation of the nickel hexacyanoferrate (NiHCNFe) which is an electrochemical ion exchanger. NiHCNFe film was deposited on the surface of nickel plate by chemical method or electrochemical method. The morphology and composition of NiHCNFe were observed by SEM and EDS, respectively. The peak current density of NiHCNFe was measured from the cyclic voltammograms of the continuous oxidation-reduction reaction in a parallel plane ion exchange electrode reactor. It was found that the chemical preparation method was better than the electrochemical method. The concentrated NiHCNFe was apparently deposited on nickel plate when dipping in the preparing solution for 118 h, especially. It also had a best durable performance as an ion exchange electrode.

  • The purpose of this study was to analyze harmful heavy metals and residual pepsticides in 30 kinds of oriental medical materials. This study was carried out on 32 samples with 30 kinds of Oriental medical materials. The GC-ECD(Varian, CP-3800) was used to analyze residual pepsticides. ICP-OES(Varian, Vista-MPX) was used to analyze lead, arsenic, cadmium. Mercury was analyzed by amalgamation method. Arsenic was detected less than 3(mg/kg) in 21 samples and was not detected 11 samples. Lead was detected less than 5(mg/kg) in 25 samples and was not detected 7 samples. Mercury was detected less than 0.2(mg/kg) in 12 samples and was not detected 20 samples. Cadmium was detected more than 0.3(mg/kg) in 3 samples was detected less than 0.3(mg/kg) in 17 samples and was not detected 12 samples. A few residual pepsticides was detected, but all residual pepsticides was safe.

Characteristics of By-product Ochre from Acid Mine Drainage (AMD) Treatment and Its Potential Use (산성광산배수 (AMD) 처리 부산물 ochre의 특성과 활용)

  • This study was carried out to find out potential use of ochre as an agent to reduce phosphorus content in water. Ochre is a by-product from treatment of acid mine drainage (AMD) which is composed mostly of $Fe_2O_3$, $Fe_2O_3{\cdot}H_2O$, $FeO{\cdot}OH$ and $Fe(OH)_3$. Three ochre samples (ochre-H, ochre-D and ochre-S) were collected from three treatment facilities in Gangwon province. Physico-chemical characteristics of three ochre samples including pH, electrical conductivity, total phosphorus, available phosphorus, particle size distribution were analyzed. Scanning electron microscopy (SEM) energy dispersive spectroscopy (EDS), X-ray diffraction (XRD) and X-ray fluorescence (XRF) analysis were also carried out. In addition, experiments for phosphorus removal from water was performed. Calcium content of ochre-H was higher than that of ochre-D and ochre-S, whereas iron content of ochre-H was lower than that of ochre-D and ochre-S. All the phosphorus in water up to maximum 191,411 mg $kg^{-1}$ per unit mass of ochre was removed with ochre-H. Ochre has immense potential as an agent to reduce phosphorus content in water.

Development and Evaluation of Bioretention Treating Stormwater Runoff from a Parking Lot (주차장 비점오염원 관리를 위한 식생체류지 개발 및 평가)

  • Urbanization increases the impervious cover, which affects the discharge of stormwater runoff and non-point source pollutants to the waterbodies. In order to improve the water quality and restore the aqua-ecosystem, the Ministry of Environment (MOE), Korea MOE introduced the Low Impact Development(LID) techniques on development projects. Therefore, research was performed to develop the bioretention technology for managing the stormwater runoff from urban areas. The test-bed was established on 2013 up to evaluate the performance of pollutant and runoff reduction. A total of 11 storm events have been monitored from November 2013 to present. Even though the SA/CA (surface area of bioretention/catchment area) is approximately 2.2%, the facility shows high pollutant and runoff reduction during storm events by increasing retention and infiltration capacities. The bioretention shows a 100% total runoff reduction at 0mm < R < 10mm rainfall range and more than 90% of runoff reduction at a rainfall range of 10mm < R < 20mm. Due to runoff volume reduction, more than 90% of nonpoint source pollutant were also removed by the bioretention.

Change In Soil Properties After Cucumber Cultivation Under Plastic Film House At Gunwi-Gun Area (군위지역 오이 시설 재배지의 토양특성 변화)

  • The soils of plastic film houses in which intensive cultivation takes place suffer from the serious problems such as severe salinity and accumulation of heavy metals etc. The objective of this research was to examine the change in soil properties with repeated cucumber cultivation under plastic film house at Gunwi-Gun area. The soils were classified mainly to Danbuk and SinJung series. Clay content is lower in the soils of plastic film house than in the field soil. Available $P_2O_5$ and exchangeable cations were accumulated at the level higher than the optimum range. The content of extractable heavy metals were higher in the soils of plastic film house than in those of the field. For optimum yield, much attention should be paied to the management of soils by the pre-soil survey.
