Effects of Bambusae Caulis in Liquamen on Blood Sugar in db/db Mice (죽력이 db/db mouse의 혈당강하에 미치는 영향)

  • This study was carried out to understand the effects of Bambusae Caulis in Liquamen on blood sugar in the db/db mice. Refined Bambusae Caulis in Liquamen C. D(BCL.C. D)manufactured by high temperature production process and Bambusae Caulis in Liquamen(H-BCL) manufactured & distributed by HANLIM PHARM.COM., LTD were used. The Bambusae Caulis in Liquamen extracted from bamboo charooal manufacturing process was filtered and refined. The effects of Bambusae Caulis in Liquamen were administered orally to mice for 6weeks and its anti-diabetic effect examined. The effects of BCL.C. D and H-BCL were observed in terms of blood sugar. creatinine. BUN and GPT in db/db mice. The results were as follows : The amount of glucose was slightly decreased (P<0.05) in the B CL.C-treated groups compared with the control. The amount of glucose was significantly decreased (P<0.01) in the BCL.D and H-BCL-treated groups compared with the control. The amount of creatinine did not show any differences among four groups. The amount of blood urea nitrogen did not show any differences in the case of BCL.C-treated groups. but observed significant decrease in the case of BCL.D and H-BCL-treated groups. The amount of GPT did not show any differences in the case of BCL.D-treated groups. but observed significant increase in the case of BCL.C and H-BCL-treated groups.

Effects of Mixed Extracts with Bambusae Caulis in Liquamen on the Blood Sugar of Diabetic mice induced with Streptozotocin (죽력배합약물이 Streptozotocin으로 유발된 당뇨 생쥐에 미치는 영향)

  • This study was carried out to investigate the optimal mixed extract with Bambusae Caulis in Liquamen in order to strengthen anti-diabetic effects on the hyperglycemia induced by streptozotocin in mice. The original Bambusae Caulis in Liquamen filtered and refined. The effects of Bambusae Caulis in Liquamen and Mixed extracts(Bamboo Juice) with Bambusae Caulis in Liquamen were administered to mice for 4weeks and its anti-diabetic effect examined. Mice used in this experiment were divided into three groups and saline(control), Bamboo Juice mixed with refined Bambusae Caulis in Liquamen(BJ+BCL.D) and distrilled water mixed with refined Bambusae Caulis in Liquamen(DW+BCL.D) were given orally for 28days respectively. And then, experemental groups were observed in terms of blood sugar, creatinine, BUN and GPT. The amount of glucose was significantly decreased in the Bambusae Caulis in Liquamen and Mixed extracts(Bamboo Juice) with Bambusae Caulis in Liquamen-treated groups compared with the control group(P<0.01). The amount of creatinine, BUN and GPT did not show any differences among Control, BJ+BCL.D and DW+BCL.D groups. In conclusion. it was found that Bambusae Caulis in Liquamen and Mixed extracts(Bamboo Juice) with Bambusae Caulis in Liquamen were nontoxic to kidney and liver and also effective on murine hyperglycemia induced with STZ. Mixed extracts(Bamboo Juice) were more effective for decreasing blood glucose than Bambusae Caulis in Liquamen D. BJ+BCL.D can be used as optimal mix material with Bambusae Caulis in Liquamen D for control Diabetes.

Effects of BCL oral administation and herbal acupuncture at BL18, BL19 on Liver function changes induced by Alcohol in the mice (죽력(竹瀝)(죽작액(竹昨液)) 경구투여(經口投與)와 간유(肝兪)·담유(膽兪) 약침(藥鍼)이 Alcohol 대사(代謝) 및 간기능(肝機能)에 미치는 영향(影響))

  • Objective : This dissertation was designed to evaluate the effect of BCL(refinded Bambusae Caulis in Liqua-men) oral administration and herbal acupuncture on alcohol metabolism and liver function. Methods : Mice were damaged by a large quantity of alcohol and received treatment of either BCL $1mg/kg$ in oral or BCL $250{\mu}g/kg$ in herbal acupuncture-BL18 BL19 bilateral. and then such parameters as GOT, GPT, catalase and superoxide dismustase(CuZn-SOD, Mn-SOD) were measured. Results : 1. Compared with control group, the activity of GOT, GPT in serum was significantly reduced and the proper degree of alcohol in serum was not significantly differ from oral administration group and herbal acupuncture group. 2. The activity of catalase in liver cell tissue and the activity of CuZn-SOD in liver cell tissue, compared with control group, was not sigificantly affected either by oral administration and herbal acupuncture group. But the activity of Mn-SOD was significantly increased in oral administration group, while it was not the case in acupuncture group. Conclusion : we consider that BCL oral administration and herbal acupuncture is highly effective in recovering alcohol metabolism and liver disfunction induced by alcohol.

  • This dissertation was designed to evaluate the effect of BCL(refinded Bambusae Caulis in Liqua-men) oral administration and herbal acupuncture on alcohol metabolism and liver function. The activity of CuZn-SOD in liver cell tissue was not significantly change in herbal acupuncture and oral administration group. The activity of Mn-SOD was significantly increased in oral administration group. while it was not the case in acupuncture group. In conclusion. we consider that BCL oral administration and herbal acupuncture is highly effetive in recovering alcohol metabolism and liver disfunction induced by alcohol.

  • This study was carried out to understand the effects of Bambusae Caulis in Liquamen manufactured by different production facilities on blood glucose of the mice induced with streptozotocin. Filtered Bambusae Caulis in Liquamen manufactured by two different facilitiesproduction process were used. The physiochemical properties of filtered Bambusae Caulis in Liquamen manufactured by two different facilities(300L×300W×400H and 700L×700W×800H electric furnace) were compared. Furthermore, the effects of filtered Bambusae Caulis in Liquamen were observed in terms of blood glucose, BUN and AL T in the mice induced with streptozotocin. The results were as follows : The pH value was the highest in L-BCL(E)B(pH 2.83) between manufactured by two different facilities. The L-BCL(E)B which observed low values of Hunter's color values show decreased concentration of methanol and phenolic compounds. The blood glucose contents was decreased in the experimental groups compared with the control. The amount of BUN and ALT did not show any differences between control and experimental group.

  • This study was carried out to understand the physiological properties and the effects of Bambusae Caulis in Liquamen manufactured by three different production process on blood glucose of the mice induced with streptozotocin. Original, filtered and refined Bambusae Caulis in Liquamen manufactured by three different production process were used. The physiochemical properties of original, filtered and refined Bambusae Caulis in Liquamen manufactured by three different production process were compared. Furthermore, the effects of original, filtered and refined Bambusae Caulis in Liquamen were observed in terms of blood glucose, BUN, ALT in the mice induced with streptozotocin. The results were as follows : The range of the pH manufactured by three different production processes was $2.28{\sim}3.17$ Concentration of methanol and phenolic compounds which are related to toxicity were lowest in filtered Bambusae Caulis in Liquamen(L-BCL [E] B) manufactured by using electric furnace at $215{\circ}C$ during 8.10/hour. The blood glucose contents was decreased in the experimental groups compared with the control. The amount of BUN and ALT did not show any differences between control and experimental group.

Anti-apoptotic and Neuroprotective Effects of Bambusae Caulis in Liquamen Manufactured by Different Production Process (생산공정 차이에 따른 죽력(竹瀝)이 apoptosis 및 신경세포 보호 효과에 미치는 영향)

  • Bambusae Caulis in Liquamen(BCL) has been commonly prescribed for stroke patients in the traditional Oriental medicine. So this study is aims to investigate the anti-apoptotic and neuroprotective effects of Bambusae Caulis in Liquamen(BCL) manufactured by different production process on the focal ischemia induced by intraluminal filament insertion in rats. The focal ischemia was induced by intraluminal filament insertion into middle cerebral artery. The animals were divided into four groups (n=15 in each group). The ischemia induced and not treated group : Control group, the ischemia induced and oral medication of the three kinds of BCL : BCL-A group, BCL-B group, BCL-C group. BCL-A was produced by heating at a low temperature$(250^{\circ} C)$ in electric kiln and filtering. BCL-B was produced by heating at a high temperature$(900^{\circ} C{\sim}1,000^{\circ}C)$ in yellow earth kiln and refining and filtering. BCL-C was produced by heating at a low temperature$(400^{\circ} C)$ yellow earth kiln and no refining and filtering. The anti-apoptotic and neuroprotective effects of the oral medication of BCL were observed by Bax, BCL-2, cytochrome c, mGluR5, cresyl violet and ChAT-stain. Our study suggests that BCl-A(was produced by heating at a low temperature in electric kiln and filtering) and BCL-C(was produced by heating at a low temperature in yellow earth kiln and no refining and filtering) show anti-apoptotic and neuroprotective effects on the focal ischemia induced by intraluminal filament insertion in rats and BCL-C is more effective than BCL-A.

  • Objectives : Ischemic brain injury is a worldwide problem that often causes irreversible brain damage. Moreover, prevention of ischemic brain injury is more important than anything else, since after-effects of stroke significantly threat the quality of life. Jukryukjichul-hwan (JRH) is an oriental medicinal formula for stroke patients in Korea. This study evaluated neuroprotective effects of methanol extract of JRH on global cerebral ischemia in rats. Changes of the pyramidal neurons, Bax and TUNEL immuno-positive neurons in CA1 hippocampus were observed using immunohistochemistry. Methods : Sprague-Dawley Rats were induced with temporal global cerebral ischemia (TGI) by occluding the bilateral common carotid artery with hypotension, The rats were divided into 3 groups. We treated one group with methanol extract of JRH after operation, another group before and after the operation. We observed Bax expressions inducing apoptosis of neurons and TUNEL-positive Pyramidal Neurons as an index of survival and apoptosis of pyramidal neurons in CA1 Hippocampus. Results : JRH treatment before and after TGI inhibited Bax expression in CA1 hippocampus. JRH treatment before and after TGI reduced the cell death of pyramidal neurons in CA1 hippocampus. JRH treatment after TGI reduced the cell death of pyramidal neurons in CA1 hippocampus. JRH treatment before and after TGI reduced TUNEL-positive cells in CA1 hippocampus. Conclusion : These results suggest that JRH has a neuroprotective effect (by anti-apoptosis) against cerebral ischemia.

The original Bambusae Caulis in Liquamen filtered and refined. Bambusae Caulis in Liquamen D(H-BCL.D) extracted at high temperature(1000℃), Bambusae Caulis in Liquamen A(L-BCL.A) and Bambusae Caulis in Liquamen B(L-BCL.B) extracted at low temperature (250~450℃) were administerd to mice for 4weeks and its anti-diabetic effect examined. Mice used in this experiment were divided into four groups(Control, H-BCL.D, L-BCL.A and L-BCL.B). Experemental groups were observed in terms of blood sugar, Creatinine, BUN and GPT. The amount of glucose was significantly decreased in the Bambusae Caulis in Liquamen-treated groups compared with the control(P < 0.05). The amount of Creatinine did not show any differences among four groups. The amount of Blood Urea Nitrogen did not show any differences in the L-BCL.A and B-treated groups, but did show slightly decrease(P<0.05) at H-BCL.D-treated group. The amount of GPT did not show any differences among four groups. In conclusion, it was found that Bambusae Caulis in Liquamen from extracted at high or low temperature were effective on murine hyperglycemia mice induced with STZ respectively.

The effects of BCL.D + Silk Worm Powder and BCL.D were observed in terms of blood sugar, creatinine, BUN, GPT and histological examination of pancreatic tissue in db/db mice. The results were as follows : The amount of glucose was slightly decreased (P < 0.05) in the B CL.D+SWP groups compared with the control. The amount of glucose was significantly decreased (P < 0.01) in the BCL.D groups compared with the control. The amount of Creatinine did not show any differences among three groups. The amount of blood urea nitrogen observed significant decrease in the case of BCL.D groups. The amount of GPT did not show any differences among three groups. The intense of insulin-immunoreactivity of β cells showed the strongest in the normal group, and more strong in the BCL.D+SWP group compared with control group.
