Expression of Aquaporin-4 and -8 Genes in Mouse Uterus during the Estrous Cycle (발정주기 동안 생쥐 자궁에서의 Aquaporin-4와 -8 유전자의 발현)

  • Aquaporins(AQPs) are a family of transmembrane water channel proteins that are widely distributed in various tissues throughout the body and play a major role in Oanscellular and Oansepithelial water movement. Uterine endometrium undergoes recurrent uterine stromal edema in response to hormonal stimuli, however, the mechanism regulating the fluid transport during the estrous cycle has not been fully understood. To investigate the possible role of AQPs in water movement in uterus during the estrous cycle, expression patterns of AQP -1, -3, -4, -5, -8, and -9 UMh in mouse uterus were analyzed by using semiquantitative reverse transcription- polymerase chain reaction(RT-nR). We employed a combination of laser capture microdissection(LCM) and RT-PCR to examine the expression patterns in specific uterine cell types luminal epithelial cells(LE) and stromal cells(S). Our results showed that the level of AQP-4 mRNA was significantly increased while the level of AQP-3 mRNA was significantly decreased during the proestous through the estrus stage. In addition LCM revealed that AQP-4 and -8 mRNAs were highly expressed in LE compared with S. Taken together, these results suggest that AQPs may have an important function in physiological changes of mouse uterus during the estrous cycle.

Silibinin Inhibits Cell Growth and Induces Apoptosis through Cell-cycle Arrest in PC-3 Prostate Cancer Cells (인간 전립선 암세포 PC-3 세포에서 Silibinin의 세포주기조절을 통한 세포사멸 유도 효과)

  • Milk thistle (silybum marianum) is a famous dietary supplement widely used in the United States and Europe. Silbinin is a major biologically active compound of milk thistle and has strong antioxidant and radical scavenger activities. Anticancer activities, as well as chemopreventive effects on various cancer cell lines, including prostate, lung, colon, skin, and bladder, have also been reported in silbinin. In the present study, we investigated the anticancer effects of silibinin and apoptosis through cell cycle arrest on prostate cancer cell PC-3. We performed cell viability by MTT assay and western blotting to confirm cell cycle check point proteins such as cyclin A/D1/E and cyclin-dependent kinase (CDK) 2/4/6. To quantify silibinin-induced apoptotic cell death of PC-3, Annexin V and PI double staining was performed by flow cytometry, by which its cell distribution was determined. As a result, silibinin inhibited the cell growth of PC-3 cells in a time- and dose-dependent manner, and its treatment resulted in cell cycle arrest at the G1 phase. Also the level of cell cycle check point proteins (cyclin, CDK) was decreased by silibinin in a dose-dependent manner. Taken together, we suggest that apoptosis of prostate cancer cell line PC-3 induced by silibinin is associated with cell cycle arrest through decrease of cell cycle check point proteins, caspase-3 activation and poly (ADP-ribose) polymerase (PARP) cleavage.

Enhanced Sensitivity and Long-Term G2/M Arrest in Adriamycin-treated DNA-PK-null Cells are Unrelated to DNA Repair Defects (DNA-PK-null 세포주의 adriamycin 처리에 의한 G2/M 세포주기 변화)

  • While the DNA-protein kinase (DNA-PK) complex, comprised of DNA-PKcs and Ku80, is primary involved in the repair of DNA double-strand breaks, it is also believed to participate in additional cellular processes. Here, treatment of embryo fibroblasts (MEFs) derived from either wild-type (Wt) or DNA-PKcs-null (DNA-$PKcs^{-/-}$) mice with various stress inducing agents revealed that adriamycin was markedly more cytotoxic for $Ku80^{-/-}MEFs$ and led to their long-term accumulation in the $G_2$/M phase. This differential response was not due to differences in DNA repair, since adrimycin-triggered DNA damage was repaired with comparable efficiency in both Wt and $Ku80^{-/-}MEFs$, but was associated with differences in the expression of important cell cycle regulatory genes. Our results support the notion that Ku80-mediated cytoprotection and $G_2$/M-progression are not only dependent on the cell's DNA repair but also may reflect Ku80's influence on additional cellular processes such as gene expression.

  • The permanent disability benefit(PDB) system whose purpose lies in helping affected workers with their self-realization through compensating them for their work-related injury or disease and facilitating their timely return to work is very important in terms of social welfare. Actually, the portions of PDB's in all WCI expenditures have been on the sharp increase. In spite of its positive aspects like this, however, an excessive amount of PDB's may have negative impacts on the affected workers' will to work or return to work, and increasing trends in the amount of PDB's payable in annuity will remain in an aging or aged society a potentially risky factor posing a threat to the soundness of WCI budgets. In this respect, the author attempted herein to identify the long-term reality of trends in the number of claimants entitled to PDB's and the amount of PDB's payable to them through making the long-term budget projections of PDB's; address problems with PDB's payable in annuity, which are or will potentially be one of the gravest burdens in securing the soundness of WCI budgets; and discuss how to optimize the amount of PDB's through studying & analyzing overseas cases and income replacement rates and what kinds of improvements are thinkable. It's recommended to reduce the absolute amount of benefits payable in annuity in a phased way considering claimants' life-cycle instead of sticking to the current system, which takes the form of lifelong pension plan.

에탄올을 첨가한 reflux법 나노 세리아 합성

  • 세리아는 고체 산화물 연료전지(SOFC, solid oxide fuel cell)의 전해질 재료와CMP( chemical mechanical polishing) 슬러리 재료, 자동차의 3원 촉매, gas sensor, UV absorbent등 여러 분야에서 사용되고 있다. 본 연구에서는 세리아의 입자의 크기와 형상을 조절하여 성능 및 물성을 향상시켜 보다 넓은 분야의 활용을 하고자 실험을 실시하였다. 세리아 합성에 사용되는 전구체인 cerium carbonate의 특성이 세리아 분말의 물리화학적 특성에 직접적인 영향을 주기 때문에 전구체의 합성 단계에서 형상과 크기를 조절하고자 하였다. 세륨염으로 cerium nitrate hexahydrate, 균일침전반응을 할 수 있는 urea를 침전제로 사용하였다. 반응 용매의 유전상수를 조절하고 반응의 과포화도 변화를 이용하기 위하여 에탄올을 첨가하여 입자의 크기 및 형상을 조절, cubic형태의 $Ce_2O(CO_3)_2{\cdot}H_2O$ 결정상을 가지는 세리아 전구체를 합성하였다. 이렇게 생성된 전구체를 $1000^{\circ}C$에서 2시간동안 하소하여 세리아를 합성하고 반응시간, 농도, 에탄올의 함량 변화에 대해 XRD, FE-SEM, particle size analyzer, micropore physisorption analyzer 분석을 통하여 입자의 결정상과 형상, 입도 분포 및 기공분포 등을 반응인자의 변화에 따라 비교 및 해석하였다.

Sleep-Wake Cycles in Man (인간의 수면-각성 주기)

  • To assess the reliability of chronobiological models of sleep/wake regulation, it is necerssary that the models predict the data which has been studied in sleep research, and they should be generalized across all ages. To date, many adult human data on such models have accumulated, yet it is evident that a comprehensive theory of the biorhythmic aspects of sleep/wake states has not established. Circadian rhythms such as the time going to bed, sleep onset, slow wave sleep pressure, periodicity of REM sleep, daytime performance, and early evening alertness are resumed everyday. Even in adult humans, sleep is inherently polyphasic. In both the disentrained and entrained states, naps when allowed tend to recur in a temporally lawful manner. The monophasic sleep pattern of most industrial societies therefore appears to be purely of social origin. The endogenous biorhythmic nature of circasemidian sleep tendency is supported by the ubiquity of the phenomenon across all ages. The NREM/REM sleep cycle within sleep with its inherent physiological, endocrine, and neurochemical fluctuations represents the best-documented ultradian sleep rhythms. Also, a daytime ultradian variation in sleepiness with a periodicity similar to nocturnal NREM/REM cycle(BRAC hypothesis) is suggested. This review article provides a brief synoptic review of the evidences for circadian, circasemidian, and ultradian sleep/wake rhythms, and then the authour will suggest the issues which expedite fuller modeling of sleep/wake system, to be further discussed.

A Dynamic Frame-skipping Scheme for Transcoder Using the Prediction Period (예측주기를 이용한 동적 프레임 삭제 트랜스코딩 기법)

  • The rapid growth of technology for network and terminal devices is able to service multimedia contents in mobile environments. But as the environment of networks and the performance of devices is varied, the same quality service is very difficult to be provided for user in internet and mobile environment. Therefore, multimedia streaming system must provide an adaptive service in considering network and device environment. Video transcoding techniques are the good solution that can provide video streaming service in various network environments adaptively. This paper proposes a dynamic frame-skipping transcoding scheme to provide adaptive streaming service in various network environments. This scheme uses the prediction period for transcoding. A proposed transcoding scheme can reduce time complexity through combination of prediction period and skipping period to control frame-skipping rate in encoder. In simulation results, the performance of proposed scheme is similar to the performance of a traditional dynamic frame-skipping scheme in PSNR value. But the performance of proposed scheme outperforms the performance of traditional scheme in time complexity.

  • Multimedia is a very popular service in the Internet. But, we cannot provide multimedia service at a uniform content, because networks and devices are various. Thus, an adaptive service is needed for multimedia transmission. Video Transcoding is the good solution that can service multimedia adaptively. This paper proposes the streaming system that is composed of encoder, transcoder, decoder The encoder enhanced time complexity and PSNR in case of transcoding using PP(Prediction Period). The decoder is almost same as the traditional media player. Transcoder reduced time complexity through combination of prediction period in encoder and skipping period to control frame rate in transcoder. In simulation results, the performances of proposed scheme outperform the system with traditional transcoder in time complexity and PSNR.

  • Real Time Locating Systems (RTLS) are used to track and identify the location of objects in real time using simple, inexpensive tags attached to or embedded in objects and readers that receive the wireless signals from these tags to determine their locations. A tag is powered an internal source such as a battery. The blink frequency of a tag affects the energy efficiency and the locating accuracy of RTLS. The mobility of a tag also affects the locating accuracy. In this paper, we introduce a RTLS tag design which improves the locating accuracy and the power efficiency. We propose an adaptive transmission-rate control algorithm using a motion sensor. By analyzing the signal pattern of the motion sensor, we can build a model to estimate the speed of the motion. Using this model, our algorithm can achieve better locating accuracy and lower power consumption than those of the conventional method. In our experiments, the number of transmission reduced as 40%, keeping similar locating accuracy.

Autonomic Period Determination for Variable Rate Limiter of Virus Throttling (바이러스 감속기의 가변 비율 제한기를 위한 자율적 주기 결정)

  • Virus throttling technique, one of many early worm detection techniques, detects Internet worm propagation by limiting connect requests within a certain ratio. The typical virus throttling controls the period of rate limiter autonomically by utilizing weighted average delay queue length to reduce connection delay time without hanving a large effect on worm detection time. In the existing virus throttling research, a minimum period of variable rate limiter is fired and a turning point which is a point that the period of rate limiter has been being decreased and starts to be increased is also fixed. However, these two performance factors have different effects on worm detection time and connection delay. In this paper, we analyze the effect of minimum period and turning point of variable rate limiter, and then propose an algorithm which determines values of performance factors by referencing current traffic pattern. Through deep experiments, it is verified that the proposed technique is more efficient in respect of reducing worm detection time and connection delay than the existing virus throttling which fixed the performance factors.
