Monitoring of Commercial Red Pepper Powders for Their Irradiation Status (물리적 확인시험법을 이용한 시판 유통 중인 고춧가루의 방사선 조사여부 판별 모니터링)

  • Ten commercially available red pepper powders were investigated using photostimulated-luminescence (PSL), thermoluminescence (TL) and electron spin resonance (ESR) analyses to confirm their irradiation status. The application of PSL, TL, and ESR analyses was also confirmed by in-house irradiation. In PSL-based screening, all samples gave negative photon counts (<700 PCs). The PSL calibration dose (1 kGy) showed a low sensitivity of 4 samples, while the others provided reliable screening results. TL glow curves demonstrated maximum peaks after $250^{\circ}C$ for the 6 samples; however 4 samples gave complex TL glow curves with maximum peaks in the range of $185-260^{\circ}C$ (radiation-specific), which could be the effect of an irradiated component in low concentration as the TL ratios of all samples were <0.1. Radiation-specific ESR features were absent in the all commercial samples. Variable irradiation detection properties were found; where the TL analysis showed the possible presence of an irradiated component in 4 samples requiring further monitoring and investigation.

Species Identification and Monitoring of Labeling Compliance for Commercial Pufferfish Products Sold in Korean On-line Markets (국내 온라인 유통 복어 제품의 종판별 및 표시사항 모니터링 연구)

  • In this study, based on an analysis of two DNA barcode markers (cytochrome c oxidase subunit I and cytochrome b genes), we performed species identification and monitored labeling compliance for 50 commercial pufferfish products sold in on-line markets in Korea. Using these barcode sequences as a query for species identification and phylogenetic analysis, we screened the GenBank database. A total of seven pufferfish species (Takifugu chinensis, T. pseudommus, T. xanthopterus, T. alboplumbeus, T. porphyreus, T. vermicularis, and Lagocephalus cheesemanii) were identified and we detected 35 products (70%) that were non-compliant with the corresponding label information. Moreover, the labels on 12 commercial products contained only the general common name (i.e., pufferfish), although not the scientific or Korean names for the 21 edible pufferfish species. Furthermore, the proportion of mislabeled highly processed products (n = 9, 81.8%) was higher than that of simply processed products (n = 26, 66.7%). With respect to the country of origin, the percentage of mislabeled Chinese products (n = 8, 80%) was higher than that of Korean products (n = 26, 66.7%). In addition, the market and dialect names of different pufferfish species were labeled only as Jolbok or Milbok, whereas two non-edible pufferfish species (T. vermicularis and T. pseudommus) were used in six commercial pufferfish products described as JolboK and Gumbok on their labels, which could be attributable to the complex classification system used for pufferfish. These monitoring results highlight the necessity to develop genetic methods that can be used to identify the 21 edible pufferfish species, as well as the need for regulatory monitoring of commercial pufferfish products.

  • Most classification accuracy measures for optimal threshold are divided into two types: one is expressed with cumulative distribution functions and probability density functions, the other is based on ROC curve and AUC. Unal (2017) proposed the index of union (IU) as an accuracy measure that considers two types to get them. In this study, ten kinds of accuracy measures (including IU) are divided into six categories, and the advantages of the IU are studied by comparing the measures belonging to each category. The optimal thresholds of these measures are obtained by setting various normal mixture distributions; subsequently, the first and second type of errors as well as the error sums corresponding to each threshold are calculated. The properties and characteristics of the IU statistic are explored by comparing the discriminative power of other accuracy measures based on error values.The values of the first type error and error sum of IU statistic converge to those of the best accuracy measures of the second category as the mean difference between the two distributions increases. Therefore, IU could be an accuracy measure to evaluate the discriminant power of a model.

Development and validation of a PCR method to discriminate between Branchiostegus japonicus and Branchiostegus albus (옥돔과 옥두어 판별을 위한 PCR 검사법 개발과 검증)

  • We developed and validated species-specific primers for Branchiostegus japonicus and Branchiostegus albus to prevent the sale of B. albus as B. japonicus. Primers for B. japonicus and B. albus were designed against the cytochrome b gene. Multiplex PCR showed a 288 bp amplicon for B. japonicus, a 159 bp amplicon for B. albus, and a 502 bp amplicon for the internal control. The PCR product bands for B. japonicus, B. albus, and the internal control were present at 1 ng each. The specificity and sensitivity of the primers developed in this study were validated by testing 38 B. japonicus strains and 13 B. albus strains. Using this monitoring method, fake fish did not appear due to the agreement between the experimental results and the species. Therefore, the developed multiplex PCR method was suitable for differentiating B. japonicus and B. albus.

Discrimination of geographical origin for soybeans using ED-XRF (ED-XRF (Energy Dispersive X-ray Fluorescence spectrometer)를 이용한 콩 원산지 판별)

  • In this study we developed a method for determining the geographic origin of soybeans by combining energy dispersive X-ray fluorescence spectrometry with statistical analysis. In 2018, 197 soybean samples (100 Korean domestic samples and 97 foreign samples) were collected for the construction of a geographic origin model. The mineral concentrations of 26 elements were measured and determined via the fundamental parameters approach. One-way analysis of variance, t-test, and canonical discriminant analysis were employed to reveal five elements (P, Ni, Br, Zn, and Mn) that could be used for the determination of geographic origins. The sensitivity, specificity, and efficiency for the above method were 91.0, 95.9, and 93.4%, respectively. Validation results from 60 samples collected in 2019 showed a predictive rate of 93.3% for Korean domestic soybeans and 100.0% for foreign soybeans. In conclusion, the combination of energy dispersive X-ray fluorescence spectrometry and chemometrics could be used to effectively determine the geographic origin of soybeans.

Identification of Hanwoo Meat by DNA Analysis (DNA 분석법에 의한 한우고기 판별)

  • This study was deal with the development of breed-specific DNA marker which is able to identify Hanwoo and European cattle breeds(Non-Hanwoo) meat. Genetic differentiation between Korean cattle(Hanwoo) and European cattle breeds was examined by Random Amplified Polymorphic DNA(RAPD) analysis. The RAPD patterns were identical among Non-Hanwoo, such as Holstein, Hereford, Aberdeen Angus, Brown Swiss, Limousin or Simmental, but the above pattern was different from that of Hanwoo. All bands detected in the Hanwoo samples were observed in Non-Hanwoo cattle samples, but one of the common bands found in samples was not detected in the Hanwoo samples. The band(1.4kb) may be useful as a marker for identifying a meat of Hanwoo from imported cattle meat. Actually, the detection of the DNA marker was tested by DNA analysis with 929 samples which were prepared from bloods of 673 Hanwoo cattles and 141 Holstein cattles, from 115 imported cattle meats. The DNA marker was absent in 644 of 673 Hanwoo cattles (96%) but present in 245 of 256 Non-Hanwoo cattles (95%). These results show that the DNA marker is effective to characterize Hanwoo and Non-Hanwoo meat by its detection. This DNA marker, however, was not useful in detecting unwanted crossbreeding between two cattle breeds, because the band pattern in hybrid cattle shows one of two band patterns in Hanwoo and Non-Hanwoo.

Trends in the development of discriminating between Angelica L. species using advanced DNA barcoding techniques (진보된 DNA barcoding 기술을 이용한 당귀(Angelica)속 식물의 기원 판별 기술에 관한 연구 동향)

  • We reviewed current research trends for discriminating between species of the Angelica genus, a group of important medicinal plants registered in South Korea, China, and Japan. Since the registered species for medicinal purposes differ by country, they are often adulterated as well as mixed in commercial markets. Several DNA technologies have been applied to distinguish between species. However, one of the restrictions is insufficient single-nucleotide polymorphisms (SNPs) within the target DNA fragments; in particular, among closely-related species. Recently, amplification refractory mutation system (ARMS)-PCR and highresolution melting (HRM) curve analysis techniques have been developed to solve such a problem. We applied both technologies, and found they were able to discriminate several lines of Angelica genus, including A. gigas Nakai, A. gigas Jiri, A. sinensis, A. acutiloba Kitag, and Levisticum officinale. Furthermore, although the ITS region differs only by one SNP between A. gigas Nakai and A. gigas Jiri, both HRM and ARMS-PCR techniques were powerful enough to discriminate between them. Since both A. gigas Nakai and A. gigas Jiri are native species to South Korea and are very closely related, they are difficult to discriminate by their morphological characteristics. For practical applications of these technologies, further research is necessary with various materials, such as dried or processed materials (jam, jelly, juice, medicinal decoctions, etc.) in commercial markets.

A PCR Method to Distinguish Matsumuraeses phaseoli from M. falcana Based on the Difference of Nucleotide Sequence in the Mitochondrial Cytochrome c Oxidase Subunit I (미토콘드리아 COI 영역의 뉴클레오티드 서열 차이를 이용한 팥나방과 어리팥나방의 PCR 판별법)

  • The two closely related major leguminous crop pests in Korea, Matsumuraeses phaseoli and M. falcana (Lepidoptera: Tortricidae) have very similar morphological characters, which occasionally give rise to a failure in distinguishing between the two. In this study, we report an easy PCR-SSP method to distinguish between them, with a sequence specific primer set (P-SF2, F-SF3, and C-SR3) based on single nucleotide mismatch in 3' terminal base of a primer, which is found in the mitochondrial cytochrome c oxidase subunit I DNA (mtCOI). Through application of this method, each species may be clearly identified in terms of its PCR band size and pattern, only one band (245 bp) for M. falcana and one (409 bp) or two bands (409 bp & 245 bp) for M. phaseoli.

  • 본 논문은 FM 라디오 방송의 오디오 신호를 블록 단위로 음성 음악을 판별하는 시스템을 제안하는 논문이다. 본 논문에서는 음성 음악 판별 시스템을 구축하기 위해 다양한 특정 파라미터와 분류 알고리즘을 제안 한다. 특정 파라미터는 신호처리 분야(Centroid, Rolloff, Flux, ZCR, Low Energy), 음성 인식 분야(LPC, MFCC), 음악 분석 분야(MPitch, Beat)에서 각각 사용되는 파라미터를 사용하였으며 분류 알고리즘으로는 패턴인식 분야(GMM, KNN, BP)와 퍼지 신경망(ANFIS)을 사용하였고, 거리 구현은 Mahalanobis 거리를 사용하였다.

Classification of Subjective Sensation by Surface Fibers Measured by Image Analysis Technique (이미지 분석기법으로 측정한 표면섬유에 의한 주관적 감각 판별)

  • 본 연구에서는 이미지 분석기법으로부터 측정된 표면섬유과 가와바타 측정법에 의해 측정된 직물의 표면특성과 주관적 거칠기, 따뜻함간의 관계를 고찰하였다. 시료로는 춘추용 수트직물로 사용되는 평직과 능직의 소모직물 32종을 사용하였다. 표면섬유의 분석을 위해서 이미지 분석장치로부터 촬영된 직물 표면 이미지로부터 단위길이의 직물안에 들어가는 표면섬유의 총길이(Fiber Aggregate Length)가 측정되었다. 직물의 주관적 평가를 위해 일관성 테스트와 평가능력 향상 훈련을 마친 20명 패널을 대상으로 기준직물을 제시한 9 의미미분척도를 사용하여 직물의 거칠기와 따뜻함에 대해 평가하였다. 직물의 표면섬유와 주관적인 거칠기, 따뜻함간의 상관성이 분석되었고, 직물의 표면 특성, 표면섬유로부터 직물의 감각을 판별하는 판별식을 도출하였다.
