Sexual Maturation and Feeding Habit of Korean Mandarin Fish, Siniperca scherzeri (Perciformes, Centropomidae) in the Seomjin River and Imjin River, Korea (섬진강과 임진강에 서식하는 쏘가리 Siniperca scherzeri (Perciformes, Centropomidae) 집단의 성 성숙 및 섭식특징)

  • This study was conducted to investigate the reproductive ecology of the Korean Mandarin Fish, Siniperca scherzeri in Seomjin River and Imjin River from April to October 2008. After have investigating the relationship between standard length (SL) and Gonadosomatic index (GSI) of Siniperca scherzeri, we can conclude that the body length of maturation was over 200mm SL in case of females and over 180mm SL in males. The GSI of the females that were from Seomjin river was the highest in April (7.43%), and gradually decreased after May (5.55%) and June (3.58%). The GSI was decreased sharply in August and indicated the lowest value in September (0.44%). The GSI of the females that were from Imjin river showed similar tendency of females' GSI from Seomjin river. The GSI of males from Imjin river was totally higher than that of females, it was 5.96% in April, and 8.03% in May. It was lowered in early June, and late June, it indicated 2.12%. In September, it was the lowest value (0.20%), and increased after October (0.54%). The result of histological study of ovary presented similar result of the study of GSI in Seomjin river and Imjin river. The ratio that fishes constitute in stomach contents of Siniperca scherzeri was 92% in Seomjin river and 90% in Imjin river. Digestion of food organisms and the ratio of unidentifiable fishes in stomach contents was fairly high, and it was 50% in Seomjin river and 72.2% in Imjin river. In the period of study, eating ratio was very low 38.3% (36 ind.) in Seomjin river and 21.8 (53 ind.) in Imjin river.

  • To investigate the effects of paper sludge on seasonal variations of volatile lower fatty acids in paddy soil, paper sludge was applied to pots at the rate of either 300, 600, 900 or 1,200 kg/l0a which was either preadjusted at a C/N ratio of 30 : 1 or not adjusted. The decomposition rate of paper sludge, the evolution of $CO_2$, and the fractions of volatile lower fatty acids in the soil were determined. The results are summarized as follows: 1. Paper sludge was decomposed to $35{\sim}44%$, and its C/N ratio was $55{\sim}82$, respectively, at 120 days after treatment. 2. The evolution of $CO_2$, in the soil was proportional to the amount of paper sludge added. Significant positive correlations were observed the $CO_2$ evolution was compared with the decomposition rate of paper sludge, and volatile fatty acid contents in soil. 3. Acetic, propionic, butyric, i-butyric, valeric and i-valeric acids were identified in all the soils investigated. The content of the total volatile fatty acids in the soil increased with as the application of paper sludge increased. The formation of the acids was the highest at 25 days after treatment, and thereafter the contents of the acids decreased as time elapsed. 4. The volatile fatty acids in the soil inhibited the growth of paddy rice in early stages. The contents of acetic, propionic and i-valeric acids in the soil negatively, correlated with the uptake of N, $P_2O_5$, $K_2O$, CaO, MgO and $SiO_2$ in the paddy rice at 25 days after transplantation. In addition, the uptake of $P_2O_5$ and CaO in the paddy rice negatively correlated with the content of butyric acid in the soil. 5. The content of total volatile fatty acids positively correlated with the content of $Fe^{++}$ and $Mn^{++}$ in the soil at 25 days after transplantation. A significantly positive correlation was observed between $Fe^{++}$ and acetic acid contents in the soil.

  • The root of lxeris dentata forma albiflora was extracted with 80% aqueous MeOH and solvent fractionated with EtOAc, n-BuOH and water, successively. From the EtOAc and n-BuOH fractions, four sesquiterpene compounds were isolated through the repeated silica gel and ODS column chromatographies. The chemical structures were determined as zaluzanin C (1), $9{\alpha}-hydroxyguaian-4(l5),10(14),11(13)-triene-6,12-olide$ (2), $3{\beta}-O-{\beta}-D-glucopyranosyl-8{\alpha}-hydroxyguaian-4(15),10(14 )-diene-6,12-olide$ (3), and $3{\beta}-O-{\beta}- D-glucopyranosyl-8{\beta}hydroxyguaian-10(14)-ene-6,12-olide$ (4) through the interpretation of several spectral data including 2D-NMR. Some showed the inhibitory effects on DGAT (Diacylglycerol acyltransferase), ($IC_{50}$ values of 1, 2: 0.13, 0.10 mM), the catalyzing enzymes of the intracellular esterification of diacylglycerol and FPTase (Famesyl-protein transferase), ($IC_{50}$ values of 1, 2: 0.15, 0.18 mM), the farnesylation enzyme for Ras protein charge of cancer promotion.

Lipids and Free Sugar Composition in Ginseng Classified by Years (한국(韓國) 인삼(人蔘)의 연근별(年根別) 지질(脂質) 및 유리당조성(遊離糖組成))

  • Lipid contents, fatty acid compositions and free sugar contents of dried ginsengs grown for different years were studied. The lipid and free sugar were separated and quantified by silicic acid column chromatography, thin layer chromatography, gas liquid chromatography and high performance liquid chromatography. Ginseng contained $1.07{\sim}67%$ crude lipid and the hightest lipid content was shown in 3 year old root. The lipid fractions obtained by silicic acid column chromatography were mainly composed of neutral lipid$(51.35{\sim}72.30%)$. The contents of phospholipid and glycolipid in total lipids were $15.03{\sim}34.59%$ and $11.83{\sim}20.72%$, respectively. As the ginseng grew, neutral lipid content increased gradually but glycolipid decreased to the half of the one year old root. Seven components of neutral lipid separated by thin layer chromatography were identified but the other two components were not able to be identified. Triglyceride content was $14.42{\sim}23.91%$, sterol ester and unidentified material(II) were $13.58{\sim}21.26%$ and $7.13{\sim}15.83%$ respectively. The major fatty acids were linoleic acid, palmitic acid, oleic acid and linolenic acid. Linoleic acid content of total and neutral lipid was $61{\sim}65%$ palmitic acid content in phospholipid was $26{\sim}39%$ and one year old root showed the hightest contents. The fatty acid composition of neutral lipid was similar to the pattern of total lipid. Linolenic acid content of glycolipid decreased gradually. Free sugars were composed of rhamnose, fructose, glucose, sucrose, maltose and an unidentified material. The content of sucrose was $92{\sim}94%$ of total free sugars. Two year old root showed the highest sugar content and it decreased since then.

The Taste Compounds of Damchi-jeotguk -Concentrated Sea Mussel Extract- (담치젓국의 정미성분(呈味成分))

  • We have examined to evaluate the taste compounds of damchi-jeotguk(concentrated sea mussel extract) which is a kind of traditional processed sea food in Korea. The contents of such compounds as free amino acids, nucleotides their and related compounds, non-volatile organic acid and fatty acid composition were analyzed. The content of total free amino acids was 10520.5mg/100g on dry basis and the major ones were glycine, arginine, aspartic acid and glutamic acid. These amino acids were resulted as 55.7% of total free amino acids in damchi-jeotguk. Hypoxanthine was the highest content($8.77\;{\mu}mole/g$, dry basis) of nucleotides and their related compounds. Free amino acid-N was the most abundant, resulting 53.3% of extractive nitrogen which was 3490.2mg/100g on dry basis and next ammonia-N, nucleotide-N and TMAO-N in order. The major non-volatile organic in damchi-jeotguk were succinic acid(125.5mg/100g, dry basis) and lactic acid 91.9mg/100g, dry basis). In this fatty acid composition of total lipid, polyenoic acid was abundant holding 45.5%. The major fatty acids were 16 : 0, 18 : 2, 22 : 6, 18 : 1 and 20 : 5. It was concluded from the omission test and chemical analysis that the major taste compounds of damchi-jeotguk were free amino acids and non-volatile organic acids.

Studies on Freezing Tolerance of Mulberry, Morus species -Relation between Freezing Tolerance and Some Substances in Mulberry Branches- (뽕나무 내한성에 관한 연구 -특히 지조함유물질과 내한성과의 관계에 대하여-)

  • Some substances and freezing tolerance in the mulberry (Morus species) branch have been studied on the basis of varietal differences and harvesting times along with harvesting methods in autumn. The results obtained are summarized as follows; 1. The highest freezing tolerance was shown in the varieties of Yongcheon-chou, Jasan, Kang-weon No. 3 and Ichihei, the medium in Roso. Kairyonezumigaeshi, Yanagida and Kokuso No. 28, and the lowest in Ichinose, Mokuso, Kokuso No. 21 and Suweousang No. 3. 2. There was a signifiant negative correlation (r= -0.59*) between death atop percentage in the field and the temperature required to kill 50% of the mulberry buds (T$_{50}$) with the harvesting times and methods in autumn. Cold hardening occurred in the early through the end of September with the peak at the mid-september. During this period, leaf harvest decreased freezing tolerance with remarkable decrease due to picking all the leaves and leaving several leaves at the base of branch. Greater cold hardening was induced by leaving several leaves after topping. 3. Negative correlations were observed between freezing tolerance and the contents of soluble (r =-0.70*) and crude (r= -0.70*) protein. However, positive correlations were shown between freezing tolerance and total carbohydrate contents per crude (r=0.31*) and per soluble (r=0.71*) protein . There were also positive correlations between freezing tolerance and total sugar (r=0.67*) and RNA content (r=0.99**). No relationships of dry matter. fat. total carbohydrate and DNA contents were observed to the freezing tolerance. 4. Such sugars as raffinose. lactose, sucrose, glucose, fructose. arabinose. xylose. ribose (assumed) and rhamnose were detected in winter mulberry branch. Major sugars such as sucrose, glucose, and fructose were supposed to have higher relationship to the freezing tolerance than the other sugars. 5. Late harvesting increased RNA content except in the case of total leaf picking at mid-September. Leaf picking decreased RNA content. Some amount of RNA was, however, maintained by leaving several leaves after topping Leaving upper-middle leaves of a branch showed high RNA content. Leaving young leaves at the top and the overmatured leaves at the base showed low content. A positive correlation (r=0.51*) was noted between RNA content and freezing tolerance in the different harvesting methods.s.

Biological Treatment of Piggery Liquid Manure by Malodor Reducing Bacteria (악취 저감용 세균에 의한 돈분뇨의 생물학적 처리)

  • Sulfur-oxidizing, ammonium-oxidizing, and nitrogen-oxidizing media were used to isolate bacteria to degrade malodor gas effectively in piggery manure or soil. Twelve different strains were isolated: Paenibacillus amylolyticus, Rhodococcus jostii, Rhodococcus qingshengii, Rhodococcus opacus, Alcaligenes faecalis, Alcaligenes faecalis, Kastia adipate, Kastia adipata, Microbacterium oxydans, Halomonas campisalis, Acinetobacter oleivorans, and Micrococcus luteus. By inoculating each strain in the piggery liquid manure by 1%, the pH in most strain treatments was maintained at 8.0. Total bacterial counts were maintained at 7.3~7.9 log CFU/ml until 15 days, and then they dropped dramatically down to 5.1~5.5 log CFU/ml. On the 30th day, the treatment group inoculated with Rhodococcus opacus SK2659 showed a relatively high level of ammonium nitrogen removal, which was 39% of that of the control group. When Rhodococcus opacus SK2659 was inoculated, H2S concentration after 100 days was 3.23% compared with the control (no inoculation), suggesting that Rhodococcus opacus SK2659 is an excellent strain for removing malodor gas. The gas production of the treatments was lower than that of the control. The total accumulated amount of gas production in most strain treatments was a quarter of the gas production compared to the control throughout the experimental periods. Acinetobacter oleivorans SK2675 showed the lowest level at 12.39% compared to the control in gas production. In conclusion, the use of mixture strains, such as Rhodococcus opacus SK2659 and Acinetobacter oleivorans SK2675 isolated in this study could increase the efficacy of malodor gas reduction in the biological treatment of piggery manure.

Yearly Occurrence of Thrips Infesting Hot Pepper in Greenhouses and Differential Damages of Dominant Thrips (시설 고추재배지 총채벌레 연중 발생 및 주요 총채벌레의 차등 해충성)

  • Andong is a place to culture the great amount of hot pepper (Capsicum annuum L.) in Korea. This study reports a yearly occurrence (March 31~October 25, 2021) of thrips infesting the hot pepper in Andong. Thrips caught to yellow sticky traps were diagnosed by morphological characters and showed two dominant species: Frankliniella occidentalis and F. intonsa. During this period, a total of 107,874 thrips were caught and included F. occidentalis at about 82%, F. intonsa at about 17%, and the other thrips at about 0.3%. There were two main peaks at May~June and at September~October, respectively, in which the total number of thrips was higher in the second peak and most were F. occidentalis. Interestinly, a low level of thrips occurred during July~August was observed and explained by their susceptibility to high temperatures. A laboratory experiment by exposing thrips to high temperatures showed that thrips were susceptible to temperatures higher than 35℃ and not tolerant to 45℃ for 1 h. Indeed, high temperatures higher than 45℃ were recorded in the greenhouses in Andong during July~August. F. occidentalis was more tolerant to the high temperatures than F. intonsa. On the other hand, the thrips showed the highest occurrence peak at July~August in hot pepper-culturing greenhouse in Kangwon, where the average temperatures were mostly lower than those of Andong and no high temperatures higher than 45℃ were recorded during July~August. A viral disease caused by tomato spotted wilt virus (TSWV) was observed in the hot peppers cultured in Andong greenhouses. Multiplex PCR was used to detect the virus along with identification of thrips. With a high record of about 30%, the virus-infected thrips were detected during all the monitoring period. The virulent thrips were identified to be only F. intonsa. These results suggest that F. occidentalis gives a direct damage especially during harvesting period with their high populations while F. intonsa gives indirect damage by transmitting TSWV.

Change in the Sensitivity to Propiconazole of Fusarium graminearum Species Complex Causing Head Blight of Barley and Wheat in Jeolla Province (전남북 지역 맥류 붉은곰팡이병균의 Propiconazole 약제에 대한 감수성 변화)

  • Fusarium head blight is an important disease of small grains. It is mainly caused by members of the Fusarium graminearum species complex (FGSC). Barley and wheat growers spray fungicides, especially demethylation-inhibitor fungicides, to suppress the disease. The objective of this study was to examine the changes in the sensitivity of the FGSC population to the triazole fungicide, propiconazole. A total of 124 and 350 isolates of FGSC were obtained from barley and wheat in Jeolla Province during 2010-2016 and 2020-2021, respectively. The species identity and trichothecene chemotypes of the FGSC isolates were determined based on polymerase chain reaction assays targeting translation elongation factor 1-alpha and TRI12 genes, respectively. Sensitivity to propiconazole was determined based on the effective concentration that reduced 50% of the mycelial growth (EC50) using the agar dilution method. Of all isolates, F. asiaticum with the nivalenol chemotype was the most common (83.9% in 2010-2016 and 96.0% in 2020-2021), followed by F. asiaticum with the 3-acetyl deoxynivalenol chemotype (12.1% in 2010-2016 and 2.9% in 2020-2021). The EC50 values of the isolates collected in 2010-2016 and 2020-2021 ranged from 0.0180 to 11.0166 ㎍/mL and 1.3104 to 17.9587 ㎍/mL, respectively. The mean EC50 value of the isolates increased from 3.8648 ㎍/mL in 2010-2016 to 5.9635 ㎍/mL in 2020-2021. The baseline resistance to propiconazole was determined to be 7 ㎍/mL, based on the EC50 value of isolates collected in 2010-2016, and the ratio of resistant isolates increased from 9.7% in 2010-2016 to 28.6% in 2020-2021.

Microbiological and Enzymological Studies on the Flavor Components of Sea Food Pickles (젓갈등속(等屬)의 정미성분(呈味成分)에 관(關)한 미생물학적(微生物學的) 및 효소학적(酵素學的) 연구(硏究))

  • More than thirty kinds of sea food pickles have been eaten in Korea. Out of these salted yellow tail pickle, salted clam pickle, salted oyster pickle, and salted cuttlefish pickle were employed for the analysis of their components, identification of main fermenting microbes, and determination of enzyme characteristics concerned. Also studied was the effect of enzymic action of microbes, which are concerned with the fermenting of pickles, on the production of flavorous 5'-mononucleotides and amino acids. The results are summarized as follows: 1. Microflora observed in the pickles are: (a) Total count of viable cells after 1-2 months of pickling was found to be $10^7$ and that after 6 months decreased to $10^4$. (b) Microbial occurence in the early stage of pickling was observed to be 10-20% Micrococcus spp., 10-20% Brevibacterium spp., 0-30% Sarcina spp., 20-30% Leuconostoc spp., ca 30% Bacillus spp., 0-10% Pseudomonas spp., 0-10% Flavobacterium spp., and 0-20% yeast. (c) Following the early stage of pickling, mainly halophilic bacteria such as Bacillus subtilis, Leuconostoc mesenteroides, Pediococcus halophilus and Sarcina litoralis, were found to exhibit an effect on the fermentation of pickle and their enzyme activities were in direct concern in fermentation of pickles. (d) Among the bacteria participating in the fermentation, Sarcina litoralis 8-14 and 8-16 strains were in need of high nutritional requirement and the former was grown only in the presence of purine, pyrimidine and cystine and the latter purine, pyrimidine and glutamic acid. 2. Enzyme characteristics studied in relation to the raw materials and the concerned microbes isolated are as follows: (a) A small amount of protease was found in the raw materials and 30-60% decrease in protease activity was demonstrated at 7% salt concentration. (b) Protease activity of halophilic bacteria, Bacillus subtilis 7-6, 11-1, 3-6 and 9-4 strains, in the complete media decreased by 10-30% at the 7% salt concentration and that of Sarcina litoralis 8-14 and 8-16 strains decreased by 10-20%. (c) Proteins in the raw materials were found to be hydrolyzed to yield free amino acids by protease in the fermenting microbes. (d) No accumulation of flavorous 5'-mononucleotides was demonstrated because RNA-depolymerase in the raw materials and the pickles tended to decompose RNA into nucleoside and phosphoric acid. (e) The enzyme produced in Bacillus subtilis 3-6 strain isolated from the salted clam pickles, was ascertained to be 5'-phosphodiesterase because of its ability to decompose RNA and thus accumulating 5'-mononucleotide. (f) It was demonstrated that the activity of phosphodiesterase in Bacillus subtilis 3-6 strain was enhanced by some components in the corn steep liquor and salted clam pickle. The enzyme activity was found to decrease by 10-30% and 40-60% at the salt concentration of 10% and 20%, respectively. 3. Quantitative data for free amino acids in the pickles are as follows: (a) Amounts of acidic amino acids such as glutamic and aspartic acids in salted clam pickle, were observed to be 2-10 times other pickles and it is considered that the abundance in these amino acids may contribute significantly to the specific flavor of this food. (b) Large amounts of basic amino acids such as arginine and histidine were found to occur in salted yellow tail pickle. (c) It is much interesting that in the salted cuttlefish pickle the contents of sulfur-containing amino acids were exceedingly high compared with those of others: cystine was found to be 17-130 times and methionine, 7-19 times. (d) In the salted oyster pickle a high content of some essential amino acids such as lysine, threonine, isoleucine and leucine, was demonstrated and a specific flavor of the pickle was ascribed to the sweet amino acids. Contents of alanine and glycine in the salted oyster pickle were 4 and 3-14 times as much as those of the others respectively. 4. Analytical data for 5'-mononucleotides in the pickles are as follows: (a) 5'-Adenylic acid and 3'-adenylic acid were found in large amounts in the salted yellow tail pickle and 5'-inosinic acid in lesser amount. (b) 5'-Adenylic acid, especially 3'-adenylic acid predominated in amount in the salted oyster pickle over that in the other pickles. (c) The salted cuttlefish pickle was found to contain only 5'-adenylic acid and 3'-adenylic acid. It has become evident from the above fact that clam and the invertebrate lack of adenylic deaminase and contain high content of adenylic acid. Thus, they were demonstrated to be the AMP-type. (d) 5'-Inosinic acid was contained in the salted yellow tail pickle in a significant concentration, and it might be considered to be IMP-type. 5. Comparative data for flavor with regard to the flavorous amino acids and the contents of 5'-mononucleotides are: (a) A specific flavor of salted yellow tail pickle was ascribed to the abundance in glutamic acid and aspartic acid, and to the existence of a small amount of flavorous 5'-inosinic acid. The combined effect of these components was belived to exhibit a synergistic action in producing a specific fiavor to the pickle. (b) A specific flavor of salted clam pickle has been demonstrated to be attributable to the richness in glutamic acid and aspartic acid rather than to that of 5'-mononucleotides.
