Evaluation on Growth Characteristics of Red Pepper (Capsicum annuum L.) and Soil Chemical Properties by Continuous Application of Food Waste Compost with Manure (음식물류폐기물 혼합 가축분 퇴비 연용에 따른 고추(Capsicum annuum L.) 생육 및 토양 화학적 특성 평가)

  • Food waste compost with high salt content produced by aerobic digestion, but concerns about application of cropland. To address this issue, food waste is being composted by mixing it with livestock manure, which has a lower salt content. Therefore, this study aimed to evaluate the growth characteristics of red pepper and soil chemical properties for continuous application with different amounts of food waste compost with manure (FWC). Treatments were consisted of no fertilizer (NF), inorganic fertilizer (N-N-P2O5-K2O, 19.0-11.2-14.9 kg 10a-1), and inorganic fertilizer + food waste compost with manure (NPKFWC). FWC treatment was applied at three treatment rates based on soil organic matter content: 900 kg 10a-1, 1,800 kg 10a-1, and 2,700 kg 10a-1 ( referred to a s FWC 1, 2, 3 , respectively). As a r esult of the red pepper yield was about 1.8 times higher in NPKFWC 1 and NPKFWC 2 than that in the NF, but decreased in the NPKFWC 3, 300% of the recommended FWC application rate. Yield decreased in all FWC treatments with continuous application for three year and also decreased about 40% from 3,265 kg 10a-1 in the first year to 1,948 kg 10a-1 in the third year. For the soil chemical properties, the content of soil organic matter, available P2O5, and exchangeable cations increased in the FWC treatments, and were higher than the NF. Exchangeable sodium in all treatments was increased slightly compared to the soil before used, and no significant salinization was observed in the FWC. This study confirmed that excessive use of FWC not only reduced nutrient use efficiency, but also decreased the red pepper yield. Therefore, it is concluded that optimum usage of FWC is effective for agroecological impacts.

  • The human leukocyte antigen (HLA) system, which is part of the major histocompatibility complex (MHC) plays a vital role in immune responses by differentiating between itself and foreign cells and antigens. The significant diversity of alleles affects disease susceptibility and immune responses within different populations. Specifically, the HLA-A and HLA-DRB1 alleles are associated with various immune-related diseases, and understanding the frequency and haplotype associations of these alleles is vital for genetic and immunological research. To investigate the distribution of these characteristics in Koreans, we isolated peripheral blood mononuclear cells (PBMCs) from blood samples donated by volunteers at the Seoul Central Blood Bank and performed HLA typing on 790 samples. Our study found that the HLA-A and HLA-DRB1 alleles are widely distributed within the Korean population, with HLA-A*24:02 (21.7%) and HLA-DRB1*09:01 (9.9%) being the most frequent. Significant haplotype associations between specific HLA-A and HLA-DRB1 alleles were identified using the Chi-square test, suggesting that certain genetic combinations may influence disease onset. This insight could contribute to the development of predictive and preventative strategies for various diseases. The unique genetic characteristics of the Korean population highlight the importance of studying the HLA allele and the haplotype distributions in this group as key indicators for understanding disease susceptibility.

Optimization of Conditions for Conidial Production in Bipolaris oryzae Isolated from Rice (벼 깨씨무늬병 Bipolaris oryzae의 포자 형성 방법 개선)

  • Conidial production is a critical factor in testing pathogenicity and studying the physiology and ecology of fungal pathogens. Therefore, selecting an appropriate condition and medium for consistent conidia production is essential. In this study, we investigated light conditions and suitable medium conditions using the slide culture method to establish optimal conditions for continuous spore acquisition of Bipolaris oryzae. Primarily, we observed conidial production using two B. oryzae isolates, CM23-042 and 23CM10, under two different light conditions: (1) consistent near-ultraviolet (NUV) with fluorescent light, and (2) a 12-hr shift of the NUV-dark cycle. Secondly, we examined conidial formation under seven different media on potato dextrose agar (PDA), V8-Juice agar, minimal medium (MM), sucrose-proline agar (SPA), rabbit food agar (RFA), rice bran agar (RBA), and rice leaf agar (RLA). Under consistent NUV light with fluorescent conditions, conidia were induced in both isolates, whereas conidia were not produced under other conditions after 7 days post-inoculation (dpi). Moreover, B. oryzae isolate CM23-042 produced the highest number of conidia in MM, while isolate 23CM10 yielded the highest number of conidia in PDA after 7 dpi. In summary, our data demonstrated that the consistent NUV light with fluorescent conditions were most conducive for conidia induction in B. oryzae. The selection of a medium for conidiation may vary depending on the B. oryzae isolates, but using MM and PDA or SPA and RFA medium could be effective for spore induction. These findings will contribute to improving conidiation according to the characteristics of collected isolates of B. oryzae.

A Study on the Stainability and DNA Conservation of Tissue Slides according to Fixation Time and Temperature (고정시간과 온도에 따른 조직 슬라이드의 염색성 및 DNA 보존성 연구)

  • In this paper, the factors affecting tissues during fixation on slides were determined by changing the tissue type, fixation time, and temperature. Also, stainability and DNA conservation were evaluated. The tissues selected were parenchymal and intestinal tissue. The stainability was evaluated using hematoxylin and eosin staining and a special stain suitable for the tissue. DNA conservation was evaluated using the DNA integrity number (DIN) for stability evaluation after purity measurement. The results showed that, at all temperatures, as time increased, there was no histomorphological difference and the stainability tended to intensify. The intestinal tissue tended to have less mucosal change and proper muscle layer degeneration. There was little difference in the purity. However, the longer the fixation time, the lower the DIN value for DNA. Significant differences were identified at 25℃ and 50℃. This means that fixation at 4℃ is the more safe for DNA. This experiment confirmed that, between the time and temperature conditions associated with fixation, time had a greater effect on both staining and DNA conservation. The results of this study are expected to provide basic data for future research on the setting appropriate conditions for fixation for histopathological examinations or diagnoses.

Seismic Wave Velocity Characteristics of the Mudeungsan Tuff under the Influence of Freeze-Thaw (동결-융해에 따른 무등산 응회암의 탄성파 속도 특성)

  • We analyzed the changes in the properties of the Mudeungsan tuff by conducting an artificial weathering experiment based on the climatic conditions of Mudeungsan National Park, to evaluate the long-term stability of the columnar jointing in the tuff. The climate of Mudeungsan National Park over 20 years suggests the temperature conditions for freeze-thaw are -20 to 30℃. The change in tuff properties due to weathering were estimated by measuring the elastic wave velocity, which was measured after every 40 freeze-thaw cycles. Based on the origin of the Mudeungsan tuff and fracture distribution in the tuff, the elastic wave velocity in samples from 24 locations was measured at regular intervals in the axial and radial directions. The axial elastic wave velocity of the Mudeungsan tuff is 5,187~5,367 m/s, and the radial elastic wave velocity is 4,001~5,290 m/s. As a result of 200 freeze-thaw cycles, the axial elastic wave velocity decreased by 5.53% for sample MT-1, 4.89% for MT-2, and 5.36% for MT-3. The radial elastic wave velocity decreased by 20.00% for MT-1, 17.02% for MT-2, and 19.84% for MT-3. The decrease in elastic wave velocity due to the freeze-thaw cycles is greater for low values of elastic wave velocity. For the axial elastic wave velocity, the weathering is accelerated after 120 cycles and, for the radial elastic wave velocity, weathering actively progresses from the start of the freeze-thaw cycles. In summary, for a low elastic wave velocity, experimental weathering results in a large decrease in elastic wave velocity. In addition, the Mudeungsan tuff and its columnar joints have a distinct anisotropy.

Fucoidan Reduces Cellular and Mitochondrial Injury and Improves Impaired Osteogenic Activity in MC3T3-E1 Cells Treated with Advanced Glycation End-products (MC3T3-E1 세포에서 최종당산화물에 의한 세포와 미토콘드리아 손상, 조골세포 분화능, 조골 및 파골 활동성 변화에 미치는 후코이단의 효과)

  • Fucoidan is a polysaccharide found in brown algae, which is known for its various bioactive effects, including immune enhancement, anti-cancer, and anti-inflammatory properties. In this study, the effects of fucoidan on cellular and mitochondrial damage, as well as changes in osteogenic and osteoclastic activities induced by advanced glycation end-products (AGEs) in MC3T3-E1 osteoblast-like cells, were investigated. Treatment with AGEs resulted in a time- and dose-dependent decrease in MTT reduction capacity, activation of caspases (-3, -8, and -9), and an increase in apoptosis. Pre-treatment with fucoidan significantly alleviated these cellular damage markers caused by AGEs. In addition, fucoidan protected against AGEs-induced mitochondrial dysfunction by significantly mitigating the loss of mitochondrial membrane potential, reduction in intracellular ATP levels, and occurrence of mitochondrial permeability transition in AGEs-treated cells. Fucoidan also markedly suppressed the production of reactive oxygen species and, lipid and protein peroxidation induced by AGEs. In cells exposed to AGEs, gene expression related to osteogenic differentiation and markers of osteogenic activity increased, while markers of osteoclastic activity decreased. Fucoidan significantly moderated these changes. In conclusion, AGEs induce mitochondrial dysfunction and apoptosis in MC3T3-E1 cells, while decreasing osteogenic differentiation and activity, and increasing osteoclastic activity. Fucoidan appears to reduce cellular and mitochondrial damage and improve osteogenic activity impaired by AGEs.

A study on the case of education to train an archivist - Focus on archival training courses and the tradition of archival science in Italiy - (기록관리전문가의 양성교육에 관한 사례연구 -이탈리아의 기록관리학 전통과 교육과정을 중심으로-)

  • Conserving the recored cultural inheritance is actually the duty of all of us. Above all, the management and conservation of archives and documents is up to archivists who have technical knowledge about archival science. Archivists have to not only conserve archives and documents but also carry out classifying and appraising them in order to define them as current historic ones. The fundamental education about archival science is made up of history and law. Because Archive is the organisation which manage archives and documents produced by legal and administrative actions. Although there are still arguments about technical knowledge and degree archivists have to acquire, most of them prefer the studies related with history and emphasize legal studies to be the general boundary of archivits' ideology and trust. The training course about conservation of archives is conducted in about 9 National Archives of Torino, Milano, Venezia, Genova, Bologna, Parma, Roma, Napoli, Palermo. The training course in 19th was mostly based on the lectures of Phaleography, Diplomatics. There were not the education about archival science yet. Toward the end of 19th and 20th, people stressed the most basic subject in the training course of National Archive was not Phaleography and Diplomatics but archival science. The goal of archival science is to study the institution and organisation transferring archives and documents to Archive. And also it help archivists not wander about with ignorance of organisational and original procedures and divisions but know exactly theirs works. Like this, the studies on institution and organisation have got in the saddle as a branch of archival science since a few ten years. While archival science didn't evoke sympathy among people and experienced the tedious and difficult path in italy and other countries, Archive was managed by experts of other branches. As a result, there were a lot of faults in Archival Science. Specializing training course for Italian archivists came into being under the backdrop of Social Science Institute of Roma National University in 1925. The archival course of universities accomplished by the studies of history, law and economy. And such as Eugenio Casanova and Giorgio Cencetti were devoted archival science was abled to settle down in national archive. The training course for experts of 'archival science, 'Phaleography and Diplomatics' in National Archive of Bologna(Archivio di Stato di Bologna) is one of courses conducted in 17 National Archives in italy. This course is gratuitous and made up of 8 subjects(Archivistica, Paleografia, Diplomatica, Storia dell' Archivio, Notariato e documenti privati, istituzione medievale, istituzione moderna, istituzione contemporanea) students have to complete for two years. Students can receive the degree through passing twice written exam and once oral test. After department of Culture and education finally puts the marks of students, the chief Nationa Archive of Bologna confer the degree of 'archival science Phaleography and Diplomatics' on students passing the exams. This degree authenticates trainees' qualification which enables him to work at the archive in province, district and administrative capital city and archive of comunity and so on. Italian training course naturally leads archivists to keep in contact with valuable cultural inheritance through training in Archive. And it shows the intention to strengthen the affinity with each documents in the spot of archival management before training archivists. Also this is appraised as one of positive policies to conserve the local cultual inheritante in connection with the original qualitity of national archive with testify the history of each region. Traning course for archivist in Italy shows us the way how we have to prepare and proceed it. First, from producing documents to conserving than forever there has introduced 'original order that is to say a general rule to respect the first order given at the time producing documents'. Management of administrative documents is related consistently with one of historical documents. Second, the traning course for archivist is managing around 17 national archives. because italian national archive lay stress not or rducation of theory bus on train for archivest working in the first time of archival science. Third, diplomatics and phaleography for studies about historical document support archives. Forth, the studies on history id proceeding by cooperation between archivist and historian around archive. How our duties is non continuinf disputer who has to conserve and manage document and archives, but traing experts who having ability, vision and flexible thought, responsibility about archivals.

Studies on the Internal Changes and Germinability during the Period of Seed Maturation of Pinus koraiensis Sieb. et Zucc. (잣나무 종자(種字) 성숙과정(成熟過程)에 있어서의 내적변화(內的變化)와 발아력(發芽力)에 대(對)한 연구(硏究))

  • The author intended to investigate external and internal changes in the cone structure, changes in water content, sugar, fat and protein during the period of seed maturation which bears a proper germinability. The experimental results can be summarized as in the following. 1. Male flowers 1) Pollen-mother cells occur as a mass from late in April to early in May, and form pollen tetrads through meiosis early and middle of May. Pollen with simple nucleus reach maturity late in May. 2) Stamen number of a male flower is almost same as the scale number of cone and is 69-102 stamens. One stamen includes 5800-7300 pollen. 3) The shape is round and elliptical, both of a pollen has air-sac with $80-91{\mu}$ in length, and has cuticlar exine and cellulose intine. 4) Pollen germinate in 68 hours at $25^{\circ}C$ with distilled water of pH 6.0, 2% sugar and 0.8% agar. 2. Female flowers 1) Ovuliferous scales grow rapidly in late April, and differentiation of ovules begins early in May. Embryo-sac-mother cells produce pollen tetrads through meiosis in the middle of May, and flower in late May. 2) The pollinated female flowers show repeated divisions of embryo-sac nucleus, and a great number of free nuclei form a mass for overwintering. Morphogenesis of isolation in the mass structure takes place from the middle of March, and that forms albuminous bodies of aivealus in early May. 3. Formation of pollinators and embryos. 1) Archegonia produce archegonial initial cells in the middle and late April, and pollinators are produced in the late April and late in early May. 2) After pollination, Oespore nuclei are seen to divide in the late May forming a layer of suspensor from the diaphragm in early June and in the middle of June. Thus this happens to show 4 pro-embryos. The organ of embryos begins to differentiate 1 pro-embryo and reachs perfect maturation in late August. 4. The growth of cones 1) In the year of flowering, strobiles grow during the period from the middle of June to the middle of July, and do not grow after the middle of August. Strobiles grow 1.6 times more in length 3.3 times short in diameter and about 22 times more weight than those of female flower in the year of flowering. 2) The cones at the adult stage grow 7 times longer in diameter, 12-15 times shorter diameter than those of strobiles after flowering. 3) Cone has 96-133 scales with the ratio of scale to be 69-80% and the length of cone is 11-13cm. Diameter is 5-8cm with 160-190g weight, and the seed number of it is 90-150 having empty seed ratio of 8-15%. 5. Formation of seed-coats 1) The layers of outer seed-coat become most for the width of $703{\mu}$ in the middle of July. At the adult stage of seed, it becomes $550-580{\mu}$ in size by decreasing moisture content. Then a horny and the cortical tissue of outer coats become differentiated. 2) The outer seed-coat of mature seeds forms epidermal cells of 3-4 layers and the stone cells of 16-21 layers. The interior part of it becomes parenchyma layer of 1 or 2 rows. 3) Inner seed-coat is formed 2 months earlier than the outer seed-coat in the middle of May, having the most width of inner seed-coat $667{\mu}$. At the adult stage it loses to $80-90{\mu}$. 6. Change in moisture content After pollination moisture content becomes gradually increased at the top in the early June and becomes markedly decreased in the middle of August. At the adult stage it shows 43~48% in cone, 23~25% in the outer seed-coat, 32~37% in the inner seed-coat, 23~26% in the inner seed-coat and endosperm and embryo, 21~24% in the embryo and endosperm, 36~40% in the embryos. 7. The content compositions of seed 1) Fat contents become gradually increased after the early May, at the adult stage it occupies 65~85% more fat than walnut and palm. Embryo includes 78.8% fat, and 57.0% fat in endosperm. 2) Sugar content after pollination becomes greatly increased as in the case of reducing sugar, while non-reducing sugar becomes increased in the early June. 3) Crude protein content becomes gradually increased after the early May, and at the adult stage it becomes 48.8%. Endosperm is made up with more protein than embryo. 8. The test of germination The collected optimum period of Pinus koraiensis seeds at an adequate maturity was collected in the early September, and used for the germination test of reduction-method and embryo culture. Seeds were taken at the interval of 7 days from the middle of July to the middle of September for the germination test at germination apparatus.

Effects of Gestation Housing and Parity on the Farrowing Performance and Behaviour of Sows during the Pregnancy, Farrowing and Lactation (임신돈의 수용형태와 산차가 임신, 분만 및 비유기간 중 모돈의 행동과 분만성적에 미치는 영향)

  • This study was designed to determine the effect of gestation housing and parity on the farrowing performance and behaviour of sows during pregnancy, farrowing and lactation periods. Total 18 Duroc sows were randomly assigned after 4 weeks of gestation to either an individual gestation stall or a groups of three with pen gestation system for three parities(1st, 2nd${\sim}$3rd, and 4th${\sim}$5th). Approximately 7 days before predicted farrowing date, sows were transferred to farrowing crates where they remained until 21 days post-partum. Behaviour was recorded on day 60 and 90 of pregnancy for 24 hours, on day of farrowing for farrowing duration and on day 10 of lactation for 2.5 hours in the farrowing crate. The results obtained from this study were summarized as follows: On 60 and 90 day of pregnancy, all sows in both groups made attempts for ventral lying more than for other postures. Sows in the group gestation pen spent more time on walking than those in individual gestation stall(p<0.01); however, the group housed sows were spent less time on drinking than the individually housed sows(p<0.01). There was no significant difference between gestation housings in the occurrence of stereotypy of pregnant sows. On day 60 of pregnancy, sows in the individual gestation stall showed the highest frequency of vacuum chewing and head weaving. However, floor licking and bar licking behaviours were highest in day 90 pregnant sows individually housed. The sows individually housed during the gestation period spent more time on sitting and eating on the day of farrowing in the farrowing crate than the sows group housed. On day 10 of lactation in the farrowing crate, the group housed sows during the gestation period made significantly more attempts for ventral lying than sows individually housed. In conclusion, the occurrence of stereotypy of sows during the gestation and lactation did not differ between gestation housings and farrowing performance of sows during the gestation period was not affected by gestation housing. Thus, the group housing for pregnant sows is a feasible means to improve welfare of sows and to increase the normal behaviour of sows.

Effect of Garlic Stalk Silage on Performance and Carcass Characteristics of Hanwoo Steers (마늘대 사일리지 급여가 한우거세우의 성장 및 도체특성에 미치는 영향)

  • This study was conducted to investigate the effect of feeding of garlic stalk silage on performance and carcass characteristics of Hanwoo steers. Feeding trial was conducted with 27 heads of Hanwoo steers and these animals were divided into two groups of control(rice straw) and garlic stalk silage. Animals consuming rice straw were fed concentrates and rice straw for 22 months from the 5 months of age and animals consuming garlic stalk silage were fed concentrates, rice straw and garlic stalk silage for 22 months from the 5 months of age. Animals fed rice straw and garlic stalk silage were fed 1.34 and 1.47, 1.69 and 1.74, 1.65 and 1.66% concentrates to body weight for the growing period, fattening period and finishing period, respectively. Animals fed rice straw and garlic stalk silage as roughage sources were fed 1.37 and 1.38, 0.65 and 0.63, 0.43 and 0.43% roughages to body weight for the growing period, fattening period and finishing period, respectively. Also, animals fed garlic stalk silage were offered 0.22 and 0.33% garlic stalk silage to body weight for the fattening and finishing period, respectively. Animals fed rice straw and garlic stalk silage as a roughage sources did not differ in average daily gain during the whole feeding periods. Animals of control group consumed less concentrates and roughage than those fed garlic stalk silage during the whole feeding periods. However, feed efficiency was not significantly different between both treatments. Beef yield including backfat thickness, eye muscle area and carcass weight was slightly lower in the animals fed garlic stalk silage than in the animals fed rice straw even though there were no differences between both treatments. However, beef quality including beef color, fat color, texture, maturity and marbling score was slightly higher in the garlic stalk silage-fed animals than in the animals fed rice straw although there were not statistically different between both treatments. Animals consuming garlic stalk silage was significantly(p<0.05) lower in shear value than those fed rice straw. Amino acid composition including essential amino acid and non-essential amino acid was not different between animals fed rice straw and garlic stalk silage. Eye muscle area of animals fed garlic stalk silage contained slightly higher oleic acid, less linoleic acid and arachidonic acid and more linolenic acid than that of animals fed rice straw only as a roughage sources. So eye muscle area of animals fed garlic stalk silage contained more mono-unsaturated fatty acid than that of animals fed rice straw and $\omega$6/$\omega$3 ratio was narrower in the animals fed garlic stalk silage than in the animals fed rice straw. Economic income was higher by 20% in the animals fed garlic stalk silage than in the animals fed rice straw. Therefore, It may be concluded that feeding of garlic stalk silage as a roughage sources to steers during the fattening period seems to improve meat quality, fatty acid composition and economic income.
