Effects of Storage Condition, Storage Period, and Priming on Seed Germination of Corylopsis coreana (저장방법 및 priming 처리가 히어리 종자 발아에 미치는 영향)

  • Corylopsis coreana is an endangered Korean native plants. This is one of the genus that have high ornamental value for flowering plants available for garden shrub, bonsai, and pot plants. In this study, the methods to encourage seed germination rate were investigated for its ornamental uses. The germination rate of Corylopsis coreana seeds stored under dry-cold condition was very low, 12%, 12%, 8%, and 10%after 40, 70, 85, and 100 days storage respectively. But the germination rate of Corylopsis coreana seeds stored under wet-cold condition was higher than these, 20%, 54%, 78%, and 96% after 40, 70, 85, and 100 days storage respectively. Dry seeds sowed directly without $GA_3$ treatment showed no germination regardless of storage type(cold or room temp.) or storage periods. On the other hand, the soaking treatment with $GA_3$ 50~500 ppm for 24 hours was very effective to increase the germination rate. The most effective $GA_3$ levels was different by storage type(cold or room temp.) and storage periods. But the effect of $GA_3$ was decreased by prolonging of the storage period. Soaking treatment with $Ca(NO_3)_2$ 5, 10, 20 mM, $KNO_3$ 5, 10, 20 mM for 24 hours showed no effect.

Effects of Dehydration Methods and Storage Conditions on Germinability of Pelleted Carrot Seeds (당근 펠렛종자의 건조방법 및 저장조건이 발아성에 미치는 영향)

  • This study was conducted to identify the optimum condition for dehydration and storage for maintaining the seed vigor in pelleted carrot seeds. The water content of solid materials of un-pelleted seeds was 4.9% (F.W. basis), and that of pelleted seeds was 24%. When we dehydrated pelleted seeds for 3 hr at $25^{\circ}C$, $35^{\circ}C$, and $45^{\circ}C$, all seeds were dehydrated to 5% - the same level of water content as that before pelleting. Pelleted seeds did not show any significant difference in germination from un-pelleted seeds, and neither was there any significant time or germination difference between pelleted and un-pelleted seeds at the tested dehydration temperatures. However, the $T_{50}$ value of pelleted seeds increased about 3 days larger than that of un-pelleted seeds. Germination speed of pelleted seeds in which the dehydration period was prolonged at $45^{\circ}C$ was delayed compared to those prolonged at $25^{\circ}C$ or $35^{\circ}C$ for the same period. The optimum dehydration condition, which could be applied for large scale in the industry, was dehydration at $35^{\circ}C$ for 3 hr, and the optimum storage temperature which could maintain the seed performance was $5^{\circ}C$.

Effect of Storage Duration and $GA_3$ of Seed Germination of Celosia argentea (개맨드라미 종자 수확 후 저장 기간 및 $GA_3$처리가 발아율에 미치는 영향)

  • This experiment was conducted to improve germination rate of the seed treated duration of storage and $GA_3$ in Celosia argentea. With the seed directly after harvest, as increasing concentration of $GA_3$, ranged from 50ppm to 200ppm, and dipping hour, ranged from 6 to 24 hour, the rate of germination was also increased. The germination of seed dipping in 200ppm $GA_3$ for 24 hours was the highest with $52\%$ compared to the control with $12\%$. As increasing storage duration in $22{\pm}2^{\circ}C$, ranged from 2 weeks to 12 weeks, the rate of germination was also increased showing $84\%$ at 12 week. However, the rate of germination was not increased with the storage of $5^{\circ}C,\;0^{\circ}C,\;and\;-70^{\circ}C$. With the seed storage 16 weeks in $22{\pm}2^{\circ}C$ after harvest, the rate of germination was $84\%$. The rate of germination was increased $94\%\;to\;99\%$ by the $GA_3$ concentration, ranged from 50ppm to 400ppm, and dipping hour, ranged from 6 to 24 hour. The germination was finished 4 days after sowing.

Seed Structure and Effects of Storage on Germination of Astragalus membranaceus (황기 종자구조 및 저장이 휴면에 미치는 영향)

  • This experiment was conducted to examine the seed structure and to investigate the changes of seed vigor and germinability according to storage condition of Astragalus membranaceus. In case of the hard seed, there observed a gap between a testa and an embryo with abnormal shapes of an hilum and its micropyle. Seeds stored at room temperature maintained vitality for 4 months but continued to lose its vitality, while increase the hand seeds ratio as storing time longer. The content of Ca increased in proportion to the length of storing time.

  • 종자은행은 종자를 보존하는 가장 경제적이고 효율적인 보전방법이지만, 수종에 따라 저장특성의 차이가 커서 수종별 저장방법의 확립이 필요하다. 초저온저장 방법은 세포 분열, 대사 작용을 정지 상태로 유지하므로 현재까지 장기간 보존을 위한 비용이 효율적이며 안정적인 방법이다. 공시재료는 멸종위기 야생식물 등 22종을 대상으로 하였다. 액체질소에 7일간 종자를 침지한 후 $37^{\circ}C$ 항온수조에서 10분간 해동하였다. 휴면이 있는 수종은 파종 전 종자를 저온습윤 하였고, 저온습윤 처리 후에도 휴면타파 되지 않는 수종은 농황산, 지베렐린을 개별 또는 혼용으로 처리하였다. 발아 및 유묘 평가는 $25^{\circ}C/20^{\circ}C$(광12h/암12h)조건에서 진행하였으며, 매일 28일차까지 발아조사를 수행한 후 평가하였다. 진달래속의 꼬리진달래와 철쭉이 초저온 민감성을 보였으며, 오리나무속의 물오리나무, 사방오리는 부분 민감성을 보였다. 민산초나무는 지하부건물중이 유의하게 감소한 결과를 보였으며, 이외의 수종은 초저온 민감성을 보이지 않았으므로 장기보존을 위한 초저온 동결보존이 가능한 것으로 보였다.

Varietal Differences and Drying Storage Effects for Some Treatment Conditions of EMS and MMS on Rice Seeds (수도종자에 EMS와 MMS의 처리조건에 따른 건조저장효과와 품종간차이)

  • In order to study the biolgiocal effects of dry back following EMS (Ethyl methane sulfonate) treatment on seeds of 'Dunghan Shali' that belongs to indica-type rice with various presoaking(24, 36, and 40 hrs respectivily) and treatment time(l, 2 and 3 hrs respectivily), this experiment was done. Seedling heights of wet seeds presoaked for 36 hrs was only greatly decreased with EMS treatment compared with other periods. In germination rate of wet seeds, all of these damage was not changed. For the drying back seeds, otherwise, seedling height and germination rate were greatly reduced depend upon the time of treatment and presoaking as well as storage effects after drying back. Reduction patterns, both seedling height and germination rate, between 1 week and 8 weeks after dry back were quite similar. The other experiment was carried out in order to study for the influences on the seedling growth as biological damage with EMS and MMS(Methyl methane sulfonate) treatments among different varieties. The indica-type rice varieties; Taichung Native No.1, Dunghan Shali and IR-8, were used as the experimental materials. From this trial some results were obtained as follows; 1. Both seedling growth and germination rate on all varieties, used, MMS, treatment showed more toxic effects than those of EMS treatment. 2. Seeding growth injury of rice seeds dried back was increased gradually, and then was approached the maximum at 6-8 days after storing. In IR-s variety, otherwise, growth damage was appeared a little. Germination rate of these were quite smilar, even though chemical used is different and storage period was prolonged. 3. It was showed clearly that varietal differences of chemical mutagene treatments were recognized.

  • Rice is one of the most important food crops since it is consumed by approximately 60% of the world's population. The most abundant component of rice grain is starch that is an important source of energy. The second abundant component is protein, which is an important protein source for people in many developing countries that rarely take animal protein. However, the rice protein lacks the essential amino acid lysine. Therefore, nutritional improvement in the essential amino acid composition of rice proteins is required. On the other side, rice grain has attracted attention as a diet and health food in developed countries, because its proteins have superior physiological and food processing properties. Thus, nutritional improvements in rice seed proteins by changing amino acid composition or introducing an useful protein or peptide have been studied. This review aims at assessing the current research status of biosynthesis, accumulation, genetic improvement of seed storage proteins by mutation or genetic engineering in rice.

Effect of Storage Duration, Temperature and Priming Treatment on Seed Germination of Polygonatum odoratum var. pluriflorum (둥굴레의 종자발아에 관여하는 저장기간, 온도 및 프라이밍의 영향)

  • Present experiments are conducted to study the seed viability and optimum germination temperature of Polygonatum odoratum var. pluriflorum that is known to have low germination ability and long germination duration. To enhance germination rate, various growth regulators and inorganic salts were employed. Low germination rate was obtained with 4 year old seeds, but not with $1{\sim}2$ year old seeds. The seeds germinated very well under $22{\sim}25^{\circ}C$ and germinated speed was rapid. Especially, $1{\sim}2$ year old seeds germinated at $25^{\circ}C$ showed $70{\sim}71.2%$ germination rate. Priming treatments using $GA_3$, IAA, NAA, kinetin, $KNO_3$, $KH_2PO_4$, $Ca(NO_3)_2$ were effective compared to control except BA. 1 year old seeds treated with $GA_3$ 0.5 mM and IAA 1 mM showed 96% and 93% germination rate, respectively.

Change of Germination Rate for Chili Pepper and Chinese Cabbage Seed in Relation to Packaging Materials and Storage Conditions over 10 Years (보관용기 및 저장조건에 따른 고추 및 배추종자의 10년간 발아율 추이)

  • Seed viability is affected by storage conditions and rapid loss of viability in storage is the major cause for low germination. This study was carried out to examine the effect of packaging materials and storage temperature on seed germination rate over 10 years in two species (Capsicum annuum L. and Brassica rapa L. ssp. pekinensis) and determine effective storage conditions for maintaining seed viability. Seeds were packaged in aluminum poly pouches under vacuum, polyethylene bottles, and paper bags containing silica gel and stored under one of two controlled conditions ($15^{\circ}C$, RH 40% or $5^{\circ}C$, RH 30%) or at ambient condition. Seed germination was recorded at 6-month intervals for 10 years. The seeds of both species showed no decline in viability until 6.5 years at 15 or $5^{\circ}C$, irrespective of packaging materials. However, under ambient conditions, the seeds of chili pepper and Chinese cabbage in paper bags lost viability after 4 and 5 years, respectively. By contrast, seeds of both species in vacuum-aluminum poly pouches exhibited a 99% germination rate after 6 years under ambient conditions. Pepper seeds in the vacuum-aluminum poly pouches maintained a 93% germination rate after 10 years in ambient conditions. These results indicated that a special seed storage facility for maintaining viability of chili pepper and Chinese cabbage seed might not be essential and seed testing would not be necessary for 10 years, if chili pepper and Chinese cabbage seeds were packed in ambient/vacuum-aluminum poly pouches or $5^{\circ}C$/vacuum-aluminum poly pouches.

Effects of Storage Duration, Medium and Viscin on in vitro seed Germination in Endangered Species, Loranthus tanakae (저장기간, 배지종류 및 viscin이 멸종위기종 꼬리겨우살이의 기내 종자발아에 미치는 영향)

  • This study was conducted to establish the in vitro seed germination of Loranthus tanakae. A factorial experiment evaluated the effects of seed storage duration (0, 8, 16 weeks), media (MS, SH, White, WPM), presence of viscin, GA3, gelling agent and concentrations. Seed germinated after one week culture in in vitro condition and produced radicles. In vitro seed germination was optimal when the seeds removed viscin placed on SH medium (69%) and the addition of 0.35% gelrite (75%) in the same medium. As seed storage duration was expanded to 8 or 16 weeks, in vitro seed germination rate was reduced rapidly. Holdfasts were also produced at the side of radicles. The important factors to produce holdfast and haustorium was kind of media as an optimal condition in White medium without any supplements to be shown 98% and 8% respectively. Process of in vitro germination of Loranthus tanakae was followed to radicle elongation, holdfast development and then haustorium formation sequencially.
