Identification of Flavor Components in Tomato Fruit (토마토 향기성분의 동정)

  • The volatiles of whole and homogenated tomato fruits collected by the headspace trapping method using Tenax GC and the simultaneous steam distillation method were identified by GC and GC-MS. Among over 100 GC peaks, 10 alcohols, 6 aldehydes, 4 ketones, 3 esters, 1 phenol and 1 acid were identified from whole tomato fruits, whereas 12 alcohols, 6 aldehydes, 5 ketones, 5 esters, 2 phenols, 1 hydrocarbon and 1 acid were identified from homogenated tomato fruits. By simultaneous steam distillation-extraction, 19 alcohols, 13 hydrocarbons, 9 esters, 9 ketones, 8 aldehydes, 2 phenols, 2 lactones,2 furans, 1 acids and 2 others were identified among over 300 peaks.

PVA 분해용 균주 분리${\cdot}$동정 및 특성 연구

  • Through the PVA degrading test, 8 species of microbes were finally isolated, and two species among them were identified. To search PVA degrading rate by using 8 species of microbes, single species of microbes and combination of each species were tested. As a result, single species of microbes showed 96% of PVA degrading rate, and the similar result was obtained by using combination of two species. And 78% of PVA degrading rate was obtained by using enzyme which was secreted from good PVA degrading microbes. As a result of identification, this good PVA degrading microbes were Paenibacillus sp. and Microbacterium barkeri.

  • Two kinds of heat treatment method for the selective isolation of soil fungi to eliminate the commonest fungi and also to examine the vertical and seasonal distributions of the fungal population were applied to soil samples from two plots around Seoul area. The incubation method at $42^{\circ}C$ and heat treatment at $70^{\circ}C$ were used in this experiment. In the incubation method, the almost all the fungi isolated from two plots were mesophile, while the thermotolerant fungi was Aspergillus fumigatus and thermophilic fungi were Sporotrichum thermophile and Malbranchea pulchella var. sulfrea. The most dominant species isolated by this method was A. fumigatus. Nine genera and fourteen species were isolated from the two plots, and S. thermophile, Talaromyces ucrainicus,Malbranchea pulchella var. sulfrea were new to Korea. From the selection method by heat treatment at $70^{\circ}C$, ten genera and twenty species were isolated. Among these, the most fungi were also mesophile and thermotolerant fungus was A. fumigatus. The most dominant species isolated by this method was T. stipitatus, Talaromyces helicus var. major, Emericella nidulans var. nidulans, Chaetomium subspirale and Neosartorya fisheri var. fisheri were new to Korea. From the two isolation methods, it was found that the total number of soil fungi and frequency of species appeared including dominant ones were the highest at the soils of upper layer while the lowest at the soils of lower layer in its vertical distribution.

  • Allelopathic activity of the volatiles from leaves of Perilla frutescens an Artemisia asiatica was determined on the basis of bioassay, which tested germination and seedling growth of radish, rice, mung bean and lettuce. Seedling growth was more inhibited by phytotoxic volatiles than germination. Volatile components collected by headspace cold trapping-Tenax GC adsorption were analyzed by GC-MS. Fifteen volatile components in P. frutescens and 15 components in A. asiatica were identified. By steam distillation-extraction, 4 flavor components in P. frutescens and 10 components in A. asiatica were identified. The inhibitory activity of the fractions, obtained by steam distillation-extraction, was determined by virtue of bioassay on radish. Volatile allelochemicals of the most active fraction, neutral fraction, isolated from P. frutescens contained 9 components. In A. asiatica, 24 volatile allelochemicals were identified.

Isolation and Diversity of Yeasts from Wild Flowers in Ulleungdo and Yokjido, Korea (경북 울릉도와 경남 욕지도 야생화들로부터 효모의 분리 및 종 분포 특성)

  • Various yeasts from wild flowers of Ulleungdo in Gyeongsangbuk-do and Yokjido in Gyeongsangnam-do, Korea were isolated and identified by comparison of nucleotide sequences for PCR-amplified D1/D2 region of 26S rDNA using BLAST. Forty eight yeast strains of twenty two species and sixty yeast strains of twenty five species were isolated from wild flowers of Ulleungdo and Yokjido, respectively. Only seven species were overlapped from the two different islands areas: Cryptococcus albidus, Cryptococcus laurentii, Metschnikowia reukafii, Pichia scolyti, Rhodotorula glutinis, Rhodotorula graminis and Rhodotorula mucilaginosa. Among forty species from two different islands, other thirty three species were restricted to specific collection site suggesting that each area has distinctive yeast flora.

  • The studies were carried out to optimize a new starter for bread fermentation. Two strains of lactic acid bacteria and yeast were isolated from Kimchi. These strains showed good condition for quality bread fermented. The strains identified as Leuconostoc mesenteroides, Lactobacillus brevis, Saccharomyces fermentati and Saccharomyces cerevisiae. The mixed culture of four strains was due to the synergistic effect by interaction of these strains.

Microfungal flora of Tricholoma matsutake producing and nonproducing sites in the forest of Pinus densiflora (적송 (Pinus densiflora) 림내 송이(Tricholoma matsutake) 발생지와 미발생지의 토양 균류의 수직 분포)

  • The vertical distribution of the fungal population for the soil samples from two sites of producing and nonproducing of Tricholoma matsutake, song-yi mushroom, were examined at Yang­yang and Myung-joo, Gangweon province. By the dilution plate method, a total number of propagu­les of fungi per gram of soil was observed to be low at the song-yi producing sites but high at the song-yi nonproducing sites under the communities of Pinus densiflora. The tendency of the number of fungal propagules were decreased with the increasing vertical depth. In the incuhation method at $42^{\circ}C$, six genera and nineteen species of the fungi were isolated from two sites; Aspergillus fumigatus, Acremonium sp., Talaromyces stipitatus, Penicillium lilacinum, P. oxalicum and Westerdykella multispora. The most dominant species by this method was A. fumigatus. From heat treatment method at $70^{\circ}C$, seven genera and nineteen species were isolated; Aspergillus fumigatus, Alternaria alternata, Neurospora sitophila and Mucor sp.. In the ethanol treatment method, one genera and one species was isolated Mortierella sp.. From the three isolation methods, it was found that the total number of the soil fungi and the frequency of species appeared were the highest at the soil of upper layer whereas the lowest at the soils of lower layer in its vertical distribution.

Antifungal activity against cheese fungi by lactic acid bacteria isolated from kimchi (김치 분리 유산균의 치즈 곰팡이 항진균 활성)

  • The antifungal activity against cheese fungi by lactic acid bacteria isolated from kimchi was investigated. Eight fungi were isolated from cheese in the cheese ripening room. Two of them were identified as Penicillium and Cladosporium via ITS-5.8S rDNA analysis. Twenty-two species of lactic acid bacteria with antifungal activity were isolated from kimchi. Two of them were identified as Lactobacillus and Pediococcus via 16S rRNA sequence analysis. Of the 22 lactic acid bacteria species, six were selected (L. sakei subsp. ALJ011, L. sakei subsp. ALI033, L. sakei subsp. ALGy039, P. pentosaceus ALJ015, P. pentosaceus ALJ024 and P. pentosaceus ALJ026) due to their higher activity against the eight fungi isolated from cheese in the cheese ripening room; and among the six species, the P. pentosaceus ALJ015 and P. pentosaceus ALJ024 isolates from the Jeonju area kimchi and the L. sakei subsp. ALI033 isolate from the Iimsil area kimchi had higher antifungal activity than the other lactic acid bacteria. The minimum inhibitory concentration (MIC) of L. sakei subsp. ALI033 against the eight fungi isolated from cheese in the cheese ripening room was $62.5{\mu}g/mL$.

Diversity of Endophytic Fungi Isolated from Roots of Cypripedium japonicum and C. macranthum in Korea (광릉요강꽃과 복주머니란의 뿌리에서 분리한 내생균의 다양성)

  • This study was conducted to investigate diversity of orchid endophytic fungi from roots of two terrestrial orchids, Cypripedium japonicum and Cypripedium macranthum in Korea. The endophytic fungi were identified using morphological and DNA sequences isolated from the roots. Totally, 11 taxa of the endophytic fungi were identified from the roots of C. japonicum and 15 taxa from C. macranthum. Three species of the fungi were common in both host species; Leptodontidium orchidicola, Humicola fuscoatra var. fuscoatra, Umbelopsis dimorpha. Six species of the fungal isolates were the first report in Korea; Oidiodendron echinulatum, Pseudogymnoascus roseus, Geomyces vinaceus, Cryptosporiopsis ericae, U. dimorpha, Chaetomium cupreum.

Entomophathogenic Fungi, Beauveria and Matarhizium in the Southern Korea (우리나라 남부지방에 분포하는 곤충병원성곰팡이 Beauveria spp. 및 Metarhizium spp.)

  • Korean entomopathogenic fungi were serveryed from soil samples throughout the five provinces and within three city limits using larvae of great wax moth, Galleria mellonella as baits. The fungi were also isolated from the eight species of dead insects. Of the total 200 soil samples, 40(20.0%) were positive for entomopathogenic fungi with 21(10.5%) containing Beauveria and 19(9.5%) containing Metarhizium. Positive sample sites in each habitat included 25 of the 110(22.7%) from forests, 2 of the 20(11.0%) from agricultural fields, 6 of the 11(54.5%) from agricultural fallow areas, and 7 of the 19(36.8%) from riparian areas. In addition, Metarhizium was isolated from Japanese walking stick (Phraortes elongatus) and Beauveria from the other 7 species of dead insects.
