Production of Fermented Feed from Food Wastes by Using Inoculation (종균 첨가에 의한 음식물 찌꺼기의 발효 사료화)

  • The fermentative conversion of food wastes into feed was investigated by seeding of mixed inoculum YM (Youngjin Environmental co.), and thermotolerant yeast Kl. marxianus. For 6 days' fermentaion, the fermentation method of 2 days' aerobic followed by 4 days' anaerobic was better for the production of organic acids and increasing total microbial population than 6 days' continuous aerobic or anaerobic fermentation. By seeding YM, the total microbial count increased about 100 times of the control group. In addition, Kl. marxianus seeding together with YM increased total viable cell count, but did not increase yeast count significantly.

Prevention and Control of composting Odors Using Microbial Inocula, KMT-199 (미생물 종균제(KMT-199)를 이용한 퇴비제조 공정의 악취제거)

  • Generation of gaseous ammonia has been a major problem in composting facilities. Microbial inocula. KMT-199(brand name: CompoBac$^{TM}$). was developed in INBI0NET CORPORATION and tested in the field for its ammonia reducing capability. When KMT-199 was applied. a ten-fold increase of mesophilic and thermophilic microorganisms was observed during the early stage of composting process. Also. the temperature and pH of early stage compost increased at a higher rate when compared to control. KMT-199 treated compost reached highest temperature of $75^{\circ}C$at day 9, indicating treatment could shift the maximum composting temperature to 3 days earlier The highest temperature also reached $3^{\circ}C$ higher than the control. The pH of compost gradually increased during composting. KMT-199 treated compost reached a plateau of pH 9.32 at day 15 after treatment, and then slowly decreased thereafter. On the other hand. pH of the control steadily increased until day 38 of composting. 29% reduction of gaseous ammonia generation during composting was observed compared to that of the control. KMT-199 amended compost resulted in a higher germination rate of radish seeds than the control. These results indicate that application of microbial inocula facilitates degradation of organic materials, including ammonia during the composting process.

  • The purpose of this study was to evaluate the potentials of food wastes as an animal feed. The food wastes from the Inha University and Hanyang University Hospitals were used for this fermentation study. As the additive, approximately 40~50% of dry bean curd dregs and sawdust was mixed for moisture adjustment, certain amount(l/300 of the total content) of inoculum was added into the 140L volume fermenter and fermented for 40~48 hours with the temperature of $46{\sim}52^{\circ}C$ maintained. Fermentation product with the dry bean curd dregs had the lower content of crude protein, crude ash, calcium and phosphorus than the commercial pig feed. Fermentation product with sawdust had the lower content in most components than the commercial pig feed except crude fiber content. Based upon these findings, it was considered that only the fermentation product with dry bean curd dregs as the additive could be used for the animal feed aid.

  • 1. 버섯종균 개선 버섯의 품질과 수량을 좌우하는 주요 요인은 종균의 활력이다. 그러나 라오스에서는 종균의 활력이 낮아 조그마한 100cc 병에 배양완성하는데 14일이 소요되는 등 볍씨종균의 균사생장과 균사밀도가 매우 낮음을 확인하였다. 따라서 100cc 병을 1000cc 비닐봉지로 개선하여 입봉하는데 소요되는 시간을 10배로 단축 개선하였다. 또한 곡립종균을 액체종균으로 대처한 결과 균사생장속도가 2배가 빨랐으며 작업속도가 4배가 빨라졌다. 2. 버섯배지제조 방법 개선 미세하고 건조한 톱밥에 수분을 첨가하는데 50%이하의 수분함량을 유지함으로서 균사가 표면에만 자라고 있는 것을 확인하고 버섯 배지제조 시 물리성을 개선하기 위하여 볏짚을 잘게 잘라 10%~100%까지 혼합하는 실험을 실시하였다. 그 결과 톱밥 50%에 볏짚 50% 혼합하는 것이 균사생장 속도와 밀도가 좋았다. 또한 왕겨도 30%정도 혼합한 배지처리에서 가장 좋았다. 또한 수분함량도 65%로 향상하여 혼합하도록 지도하였다. 3. 버섯배지살균 방법 개선 라오스에서는 왕겨를 태워 살균을 실시하고 있다. 화력이 약하여 살균한 배지에서 볍씨의 새싹이 돋아나오는 경우를 발견하였다. 이는 살균이 제대로 되지 않았음을 의미한다. 그래서 이동식 직화식 살균을 권하였으나 시설자재값이 감당이 안되어 엄두를 내지 못하였다. 따라서 가스버너를 도입하여 단시간에 화력을 높여 살균을 실시하므로서 세균의 증식을 막아 균사생장속도를 빠르게 하고 균사량의 축적을 높이는데 교육을 하고 컨설팅을 실시하여 개선하였다. 4. 병뚜껑과 형성틀을 대체한 링을 이용한 입봉작업시간 단축과 비용절감 버섯봉지에 배지를 담는 입봉작업을 할 때 병뚜껑과 그의 형성틀을 끼우고 솜을 조금 뜯어 톱밥배지 위에 놓는 작업을 한다. 이는 아주 잘못된 방법이다. 왜 그렇게 하는가하는 것은 이해가 간다. 그렇게 하는 이유는 접종할 때 실내에서 그냥 접종하므로 뚜껑을 열때 보호막 역할을 할 것으로 생각하고 그렇게 하는데 천만의 말이다. 어떻든 뚜껑을 열면 잡균이 들어가는 것은 마찬가지이다. 그래서 이 솜을 배지표면에 놓으면 마른 솜이 그렇지 않아도 수분이 적은데 이 솜이 수분을 또 빼앗아 가버린다. 그래서 균사생장이 늘려 15일이면 다 자라야할 균사배양기간이 한달씩 걸려도 표면만 살짝 잘린 결과가 빗어진다. 이렇게 표면만 균사가 사탕 발림식으로 자라면 품질이 저하되고 수량이 적고 병해충에 저항력이 약해지게 된다. 따라서 뚜껑과 형성틀, 솜을 모두 없애고 봉지 상단부위를 U자형으로 꺽어서 링을 끼우는 방법을 실제로 실험으로 보여준 결과 작업능률이 5배로 빠르고 작업공정이 빨라짐으로서 세균번식밀도가 적어 균사생장 속도가 2배로 빨라졌고 수량이 배가됨을 증명하고 보급하여 많은 호평을 받았다.

Antifungal Activity of EDTA and Combinatory Synergism of EDTA with Polygodial against Saccharomyces cerevisiae (Saccharomyces cerevisiae에 대한 EDTA의 항진균 활성 및 EDTA와 Polygodial 간의 병용 효과)

  • The antifungal activity of ethylenediaminetetraacetic acid (EDTA), an antioxidative food preservative, was investigated against Saccharomyces cerevisiae. At the general condition for antifugal test, EDTA exhibited the minimum inhibitory concentration (MIC) of 200 ${\mu}g/ml$ and the minimum fungicidal concentration (MFC) of 6,400 ${\mu}g/mL$, As seen with the antibacterial activity, the antifungal activity of EDTA was greatly decreased by high inoculum size, acidic medium, and cation ($Ca^{++},\;or\;Mg^{++}$) added to medium. On the other hand, when EDTA was combined with polygodial isolated from natural food spice, they showed strong synergism on the antifungal activity. Further, the diminishing antifungal activity of EDTA by high inoculum size, acidic medium, and cation ($Ca^{++}\;or\;Mg^{++}$) added to medium was considerably improved by the combination with polygodial.

Mycelial Growth of Phellinus linteus with Various Sawdusts (수종 톱밥에서 목질진흙버섯균 Phellinus linteus의 균사생장)

  • This study was carried out to obtain the basic data on artificial culture of Phellinus linteus. The suitable sorts of sawdusts artifical culture for Phellinus linteus were an oak sawdust, a mulberry sawdust, a peach sawdust. Rice bran, wheat bran and corn seed peel were found to be effective additives, and optimun mixing ratio of an oak sawdust and additives was from $70{\sim}80$ to $20{\sim}30$ percent. The suitable moisture content of sawdust media was obtained in the range of $65{\sim}70%$. The mixture of percent oak sawdust with 20 percent rice bran in polypropylene (850 cc) bottle was suitable for starter production, and it took 55 days to produce the starters at $25^{\circ}C$.

Effects of different drying methods on fermentation characteristics and viability of cold-adaptive yeast (저온 적응성 효모의 보존방법에 따른 균체의 생존율 및 발효특성)

  • The quality characteristics of Yakju and survival rate of yeast were investigated by modifying the drying method for the cold adapted yeast strain Saccharomyces cerevisiae Y297 (SCY297). Viability and fermentation characteristics of the freeze-dried, air blast-dried, and liquid SCY297 cultures were compared after storing them at $25^{\circ}C$. In addition, 5% skimmed milk, ${\alpha}$-lactose, or trehalose was added as a protective agent for examining the effects of drying methods. During the 15-week storage period, the liquid and freeze-dried SCY297 cultures containing a protective agent showed a survival rate of 80%. However, the air blast-dried SCY297 culture showed 80% survival rate only in the skimmed milk supplemented group. Compared to the untreated cells, the acidity and amino acidity of Yakju prepared using freeze-dried or air blast-dried cultures of SCY297 increased by 2 fold and 5.7 fold respectively, while the alcohol content decreased by 5.07%. Compared to the untreated cells, the pH and amino acidity of Yakju prepared using the liquid culture of SCY297 increased by 1.5 fold and 2.5 fold respectively. Although the alcohol content decreased by 2.9%, decrease rate was lower than that observed for the freeze-dried and air blast-dried yeast cultures. Therefore, the results of this study showed that using a liquid starter culture was more advantageous than using the conventional solid culture.

Effects of Concentration of Inhibitor on the Production of $\alpha$-amylase and Growth of Bacillus amyloliquefaciens (Bacillus amyloliquefaciens 성장과 $\alpha$-amylase 생합성에 미치는 저해제의 영향)

  • In this study, Bucillus amyloliquefaciens was adopted as bacterial source to investigate the concentration of carbon source by adding inhibitors in the batch culture. By adding acetic acid at $10g/\ell$ of initial glucose concentration, maximum dry cell density was obtained with the highest value of /$\3.9gell$ at $1.0g/\ell$ of initial acetic acid concentration. By adding acetic acid al 10g/$\ell$ of initial glucose concentration, maximum ${\alpha}$-amylase production was obtained with 331.55unit/m1 at $2.0g/\ell$ of initial acetic acid concentration. ${\alpha}$-Amylase production was decreased with the increase of initial acetic acrid concentration. By adding acetic acid to the medium, cell growth and ${\alpha}$-amylase production was higher in glucose than in maltose. By adding lactic acid to the medium, cell growth was decreased.

  • The production of doxorubicin by a mutant of Streptomyces peucetius subsp. caesius was studied. The optimal culture conditions, such as inoculum size and medium composition were established to improve the productivity of doxorubicin. The optimal medium composition was found to be 4% maltose, 0.5% HEPES, 0.02% $K_2HPO_4$, 0.01% $MgSO_4$. As an antiform agent, 0.001% KG(10% Adekanol+10% Silicone) was suitable one among various agents. Culture was carried out in 2.5 L jar-fermenter with different aeration rates of 1.5, 1.0, and 1.5 v/v min. The maximum production of doxorubicin(29 mg/l) was obtained at 1.5 v/v min of aeration rate, and even at 1.0 v/v min, the production of doxorubicin was increased up to 15% compared with that of shake-flask culture.

Studies on the Artificial Cultivation of Agrocybe aegerita (Brig) Sing using Pine Sawdust Substrate (소나무톱밥을 이용(利用)한 버들송이 인공재배(人工栽培)에 관한 연구(硏究))

  • Some factors effecting on mycelial growth and cultural substrates using pine sawdust were investigated to develop an artificial cultivation technique. The optimal temperature and media pH for the mycelial growth were at $25^{\circ}C$ and 4.0, respectively. The mycelial growth was not different among three isolates tested. Among them ASI 19003 isolate showed the highest mycelial density and fruitbody yield. Among supplements added into pine sawdust, wheat bran was the best and its appropriate percentage was 20 for the mycelial growth. The mycelial densities were uprisen according to the amounts of the supplement. The highest yield, 107g/bottle of the sporophores of the mushroom was obtained from the substrates supplemented by 30% of wheat bran, but no fruitbody was formed on non supplemented one. Optimal moisture contents of the substrates was different for the mycelial growth, pinheading and fruitbody yields. Sixty five, 60 and 75% of moisture contents of the substrates were optimal for the mycelial growth, pinheading and fruitbody yields, respectively.
