Phylogenetic analysis of the medicinal mushroom and taxonomical positions of their commercial products (약용버섯의 계통분류 및 국내유통 Inonotus속내 종간 구별을 위한 신속동정법 개발)

  • The Aphyllophorales is a large order containing about 2,000 known species. Many of these are the bracket and coral fungi. The vast majority of these fungi are saprophytic on the plant debris. Many species are significant in decomposing plant remains, as they are able to digest cellulose or lignin that occurs in plant cell walls. Many of these fungi have been involved in everyday human affairs. A few were used medicinally by the Greeks and Romans as a remedy for many complaints, including colic, fractured limbs and bruises. Other bracket fungi have been used as curry combs for horses, as snuff, as razor strops and as a source of dye for clothing. The texture of the basidiocarp may be similar to that of cork, wood, leather, paper, or cartilage. Unlike the basidiocarps of the Order Agaricales, the basidiocarps of the Aphyllophorales are not fleshly and moist. Division of the members of the Aphyllophorales into genera was originally made on the basis of gross morphology of the basidiocarp and hymenium and Donk(1964) recognizes 22 families in this order. The species and genus whose typical in Aphylloporales were listed in Table. with related information. The ITS region sequence of some genus were found by BLAST search. Sequences retrieved from GenBank were visually aligned by the program CLUSTAL G. As a result, the medicinal mushroom was separated in four groups. In this multiple alignment, the sequence analysis among Fomes group, Inonotus group and Phellinus group showed high genetic similarity except Hericium group and Sparassis group.

Interspecific Relationship of Polygonatum Species Collected from Gyeongnam Area Using Cluster Analysis (경남지역 둥굴레속의 Cluster 분석에 의한 종간 유연관계)

  • Polygonatum species are a useful medical crop in Korea but basic study in the species was not well conducted. The study was carried out to analyse genetic diversity and intraspecific relationship of 47 Polygonatum species collected from Gyeongnam province. Their analysis was done through principle component analysis and average linkage cluster analysis with their twelve morphological traits. The result of principle component analysis showed the Prin 1, Prin 2 and Prin 3 represented 79% of total variation. By the 0.7 average distance of the cluster analysis and the calculated Euclidian distance, the 47 collected species were grouped into five groups. Group I included 22 collected species representing P. ordoratum var. pluriflorum, group II did 5 ones representing P. involucratum, group III was divided into two subclasses, 2 species including P. inflatum and 7 species including P. thunbergii, group IV also consisted of 2 subclasses, a species similar to P. thunbergii and P.involucratum, respectively, and finally group V included 8 species representing P.lasianthum var. coreanum. meaning that the useful germplasms can be collected from relatively small area.

Phylogeny of Korean Rhus spp. Based on ITS and rbcL Sequences (ITS 및 rbcL 염기서열에 근거한 한국 자생 옻나무속의 계통분류)

  • This study was carried out to confirm the phylogenetic relationships in Korean Rhus species. Sequences from internal transcribed spacers (ITS) of nuclear ribosomal DNA and rbcL gene of chloroplast DNA were determined. Cotinus coggygria was selected as outgroup because it is closest allied with Rhus in Anacardiaceae. Also, ingroup was limited as six Korean Rhus species. ITS 1 sequences in six species of Rhus and one species of Cotinus ranged from 246 to 253 bp and ITS 2 sequences from 234 to 244 bp. Concerning the G+C content of the studied taxa, ITS 1 sequences ranged from 58.0 to 68.13% and ITS 2 from 59.75 to 68.46%. On the other hand, rbcL sequences were same size in the all species examined by 1,428 bp. G+C contents of rbcL sequences were ranged from 43.56 to 43.77% which means there are nearly no different from interspecies each other. Phylogenetic tree strongly supports the colse relationships between R. succedanea and R. sylvestris. Rhus javanica and Cotinus coggygria were also closely allied with each other in ITS and rbcL trees. Therefore, R. javanica was regarded as most primitive species among the Korean Rhus species. ITS 1 region of nuclear ribosomal DNA was suggested as very useful taxonomical marker for genus Rhus.

Genetic Relationship of Pleurotus ferulae Strains (아위버섯(Pleurotus ferulae) 균주의 유전적 유연관계)

  • This study was carried out to investigate the genetic relationship of Pleurotus ferulae, an edible mushroom found on a medicinal plant, Ferula assa-foetida, in central China. The genetic relationships of 15 Pleurotus species strains, including five P. ferulae strains were analyzed. The strains were divided into seven groups at 80% genetic similarity level according to random amplified polymorphic DNA (RAPD) analysis. Four out of the seven groups consisted of two to four strains, while the other three groups consisted of three strains. In each of the three groups, the three strains were from each of three different Pleurotus species (P. cornucopiae, P. florida and P. sajorcaju). Other strains grouped together for genetic similarity were P. eryngii 26060 and P. fuscus var. ferulae 26065, three strains of P. ostreatus, and four P. ferulae strains (Bakdal, Awi, Cheonsan 1, and Yesan). However, Japanese Seolyi which belongs to P. ferulae and Heukpyung which belongs to P. ostreatus were together in a separate group.

Numerical Taxonomy of Korean Orostachys (Crassulaceae) (한국산 바위솔속(돌나물과) 식물의 수리분류학적 연구)

  • Numerical analyses using 28 morphological characters from 14 populations of four Korean Orostachys species were conducted to investigate infra-specific variation and to test species relationships. The resulting phenogram recognized three distinct clusters, and the populations of O. malacophyllus, O. iwarenge and O. minutus were closely related each other. The populations of O. japonicus from Gaeksanri (I) and Gadeuk-do were isolated from the remaining populations of the species, and it suggests that the ranges of morphological variation within the species are significant, and related to the previous cytological variation. The Jungdongjin population of O. malacophyllus was not grouped together with the same species, but was clustered with the populations of the O. japonicus, and suggested that it seems to be an unrelated population in O. malacophyllus. The results of the PCA analysis recognized two distinct groups: the populations of O. japonicus and remaining species populations. The populations of O. malacophyllus, O. minutus and O. iwarenge placed within the closely related species.

Diversity and Geographical Relationships by SSR Marker in Subgenus Soja Originated from Korea (SSR 마커에 의한 한국 원산 Soja 아속의 다양성과 지리적 유연관계)

  • This study was carried out to investigate polymorphism, gene diversity, and geographical relationships of 81 Korean wild (Glycine soja) and 130 cultivated soybeans (G. max) using seven simple sequence repeat (SSR) markers. A total of 144 alleles were observed in 211 accessions with an average of 20.6. Each SSR loci showed 13 (Satt532) to 41 (Sat_074) multialleles. The range of alleles within the loci was wider in wild soybean than the cultivated soybeans. The average genetic diversity values were 0.88 and 0.69 in wild and cultivated soybeans, respectively. In a scatter diagram of wild and cultivated soybeans based on canonical discriminant analysis, CAN1 accounted for 84.2% while CAN2 did 8.5%. Two species were grouped into three: group I (G. max), group II (G. soja), and group III (complex of G. max and G. soja). The geographical relationships of wild soybean were distinguished into two groups: Gyeonggi for Group I, and Gyeongsang, Jeolla, Gangwon, and Chungcheong for Group II. Those of cultivated soybeans were distinguished into Gyeonggi, Gangwon, and Gyeongsang for Group I, and Jeolla and Chungcheong for Group II. Therefore, the geographical relationships of wild soybeans were well typified based on the ecosystems of the Korean peninsula.

Phylogenetic Relationship of Araliaceae in Korea by Seed Morphological Characteristics (종자 외부형태학적 특성에 의한 한국산 두릅나무과(Araliaceae) 식물의 종간 유연관계)

  • The purpose of this study is to describe and compare the morphological characteristics of seeds of 12 taxa of Araliaceae, and to evaluate their possible use for taxonomic considerations. For light microscope observations and measurements, the seeds were observed using Image Analyzing System. The observations were made on twenty randomly selected seeds of each species. Obtained data were statistically processed using analysis of variance. Principal Components Analysis indicated four groups of characters, the genera Aralia, Eleutherococcus, Panax and the others, which explained 65.47% of the total variation. As a results of Cluster Analysis using the eleven variables, 12 species of Araliaceae were also discriminated into four groups. Eleutherococcus senticosus and E. gracilistylus were closely related, which is well supported by the results from recent molecular studies. Also, the genera Dendropanax and Eleutherococcus were closely related in terms of seed characters.

Mitochondrial DNA Variations among Three Species of Korean Planorbid Snails : Gyraulus convexiusculus, Hippeutis cantori and Segmentina hemisphaerula (한국산 또아리물탈행이과 3종 내의 미토콘드리아 DNA 변이)

  • 한국산 담수 또아리물달팽이과(Planorbidae)에 속하는 또아리물달팽이(Gyraulus convexiusculus),수정또아리물달팽이(Hippeutis cantori) 및 배꼽또아리물달팽이(Segmentina hemisphaerula)3종에 대한 종간 유전적 변이와 이들 상호간의 분류학적 유연 관계를 생화학적 측면에서 밝히고자 하였다. 즉, 모계유전으로 자손에 유전되고 있는 미토콘드리아 DNA(mitochondrial DNA; mt DNA)의 변이를 보기위하여 제한효소(restriction enzyme)를 처리하고 잘라진 mtDNA절편들을 상호 비교하는 restriction fragmint length polymorphism(RFLP)기법을 응용하였다. 본 실험에서 10개의 제한효소 중 CIa I, Dra I, Eco RI, Hin dIII, Kpn I및 pst I의 6개 제한효소에서 좋은 결과를 얻어 종간의 공통절편(shated fragmints)을 비교하였고, 염기분화율(nucleotide divergince rate)을 각각 측정하였다. 미토콘드리아 DNA 크기(genome size)는 또아리물달팽이가 12.08 kb, 수정또아리물달팽이가 14.4 kb, 그리고 배꼽또아리물달팽이가 12.93 kb로 관찰되었다. 염기분화율(p)는 또아리물달팽이/수정또아리물달팽이 군에서 p=12.7%, 배꼽또아리물달팽이와 상기 2종군 사이의 염기분화율은 P=56.6%여서 배꼽 또아리물달팽이류는 타 2종보다 그 분화율이 매우 높음을 알 수 있었다. 이상의 결과로 보아 분류군(taxa)의 mtDNA 변이에 의한 RELP기법이 앞으로 한국산 담수 패류 연구에 널리 응용될 수 있음이 확인되었다.

Identification and Phylogenetic Relationships of Inonotus obliquus Strains by PCR-RFLP of ITS sequences and STS markers (ITS 부위의PCR-RFLP 및 STS 마커를 이용한 차가버섯의 종 및 계통간 유연관계 분석)

  • Because the import of Inonotus obliquus have been rapidly increased in Korea, we developed the Inonotus species-specific marker by using various sequences including ITS sequences, PCR-RFLP and STS primers and used this marker to determine both genetic relatedness and strains discrimination of Inonotus spp. Total 17 different Inonotus spp. were examined by using ITS sequences and classified into 2 different groups. One strain, ASI74008 isolated from Kamchaka island of Siberia, showed the high sequence identity (98%) at the nucleotide level to the other I. obliquus DSM strain, indicating the ASI74008 belong to I. obliquus species. Comparison of banding patterns after restriction enzyme digestions with PCR amplicons of ITS region revealed some variations depending on the species and strains. However, PCR products amplified with STS primer showed species specific patterns.Therefore, use of both STS primers and PCR-RFLP could help for better strain identification.

Analysis of Genetic Relationship and Cultural Characteristics of Penicillium species Isolated from Postharvest Decay of Pear by Random Amplified Polymorphic DNA (저장 배에서 분리한 Penicillium속의 배양적 특성과 RAPD에 의한 유연관계 분석)

  • Fifteen isolates from pear, and sixteen isolates from KCTC, KCCM, and Chungnam Univ. of Penicillium spp. were investigated for the analysis of their relationships of cultural characteristics and RAPD genetic variation by RAPD. The cultural characteristics of Penicillium spp. were shown different growth rate, morphology, and color. In addition, the cultural characteristics and RAPD analysis were conducted for the pear rot pathogens and related isolates. RAPD patterns were applied to compare the taxonomic and genetic diversity of the Penicillium species between 15 groups isolated from pear fruits and 16 standard species. The genomic DNA were amplified from $0.1{\sim}2.0kb$ by five URP primer and 744 bands were detected. The cluster analysis showed four genomic DNA RAPD groups and its similarity was 47.7%. Intraspecific relationships were 87.4, 97.5 and 95.2%, in P. expansum, P. solitum, and P. crustosum, respectively. These results appeared to be that there were high similarities between isolates, and consistent with the results of cultural morphological characteristics analysis.
