Analysis of Genetic Relationships of Korean Native Polygonatum spp. (우리 나라 자생 둥굴레속 식물의 유연관계 분석)

  • Germplasms having valuable characters are increasjngly important for modern breeding programs. This study was conducted to obtain the basic data for effective use of genetic resources of Polygonatum. The relationship of seven Polygonatum species collected widely in Korea was analyzed by RAPD markers. Total number of alleles amplified by nineteen random primers were 114 to 157, and variation in number of alleles was also diverse among seven species examined. The seven species were divided into two groups; one was of Polygonatum stenophyllum and Polygonatum humile. the other was of Polygonatum inflatum, Polygonatum lasianthum, Polygonatum odoratum var. pluriflorum (1), (2), and for variegatum.

Genetic Relationships of Internal Transcribed Spacer (ITS) Regions on Entomopathogenic Fungi by RFLP (Entomopathogenic Fungi의 ITS 영역에 대한 RFLP 분석)

  • A similarity coefficient were analyzed by RFLP of fourteen species of entomopathogenic fungi, isolated from inhabiting pupa and adult insect at forest. Each rDNA ITS I and ITS II with primers of ITS 1 and ITS 4 was amplified by PCR. The amplified products were conserved to 500 bp were not demarcated between genus and species. Four Paeciliomyces tenuipes, two Beauveria bassiana and six Cordyceps militaris were treated by seven restriction enzymes and confirmed in species except JB3 by electrophoresis band. However, the band of C. scarabaeicola showed the identity with B. bassiana. The result of this experiment indicated that the teleomorph of C. scarabaeicola was the same as that of B. bassiana. CfoI and HpaII of seven restricted enzymes were easily discriminating in the genus between Paecilomyees and Cordyceps. Especially, CfoI was more effective to classify the genera of Paecilomyees, Cordyceps and Beauveria than other restriction enzymes. The band patterns of RFLP of P. tenuipes, C. militaris, C. scarabaeicola and B. bassiana were also analyzed by UPGMA program of NTSYS-pc and showed 100% significance. Thus, the similarity coefficient tended to be lower between genera by RFLP analysis, but was higher between species.

Genetic Variations and Phylogenetic Relationship of and Pueraia lobata Ohwi (Fabaceae) and Related Taxa by RAPD Makers (RAPD분자마커를 이용한 칡(콩과) 및 근연분류군의 유전적 변이 및 유연관계)

  • RAPD analyses were performed to investigate genetic relationships and useful molecular maker for 3 species and their 17 regional populations of the Pueraria lobata and related taxa. The length of amplified DNA fragments ranged from 200 to, 2,800 bp. Two hundred and eight scorable polymorphic makers and three scorable monomorphic makers were found from the PCR reactions with 15 random oligo primers, and those were analyzed by Nei's genetic distance coefficient. Based on the UPGMA phenogram from RAPD analyses, two major groups (9 populations from Korea; 3 populations from foreign countries) were recognized. And it showed distinct genetic differences from related taxa. The RAPD results was very useful to define the samples by geographical distribution and to discuss the relationships among the populations and their related taxa of the Pueraria lobata.

Isozyme Variations of the Genus Semisulcospira ( Pleuroceridae : Gastropoda ) in Korea (한국산 Semisulcospira속 ( 다슬기과 : 복족강 ) 패류 5종의 동위효소 변이)

  • 한국산 다슬기류의 종별 유전적 변이를 알아보고 계통분류학적 근거를 제공하기 위하여 다슬기속(Simisulcospira)에 속하는 다슬기(S. libertina),주름다슬기,(S. forticosta), 곳체다슬기(S. gottschei), 좀주름다슬기(S. tegulata)및 참다슬기(S. coreana)등 5종을 전국 각지에서 채집하여 전기영동(starch gelelectrophoresis)를 이용해 분자유전학적 분석을 실시한 결과를 용약하면 아래와 같다. 유전적 다형형의 빈도는 58%에서 83%로 대단히 높았고 이형접합자의 평균 빈도도 50%내외였다. Rogers' D를 근거로 한 각 종간 유연관계는 주름다슬기와 곳체다슬기가 근연관계를 이루고 있었으며 이들 두 군과 다슬기는 유전적으로 거리를 두고 있었다. 이러한 결과는 패각의 형채와 비교할 때 유사한 경향을 보였다.

Analysis of Genetic Relationship Among Native Taraxacum and Naturalized Taraxacum species using RAPD (RAPD를 이용한 자생 민들레 종과 귀화 민들레 종간의 연관계 분석)

  • The genetic relationships between 4 Korean native Taraxacum and 2 naturalized Taraxacum species were analyzed using the random amplified polymorphic DNA (RAPD) method. Because 141 polymorphic bands were generated from 30 random primers selected through the primer screening, it was possible to analyze the genetic relationship among 6 Taraxacum species. In RAED with the primer OPC12, OPD16, OPK16, OPK17, OPK20, OPS1 or OPS8, many specific polymorphic bands have been appeared in each species. Especially RAPD with the primer OPS8, a specific polymorphic band at 564bp was appeared only in the naturalized Taraxacum officinale. Based on RAPD analysis, Korean native Taraxacum and naturalized Taraxacum species are divided into two groups. T. officinale and T. laevigatum are classified into group I which is a naturalized Taraxacum species group, and T. mongolicum, T. hallasanensis, T. ohwianum and T. coreanum are classified into group II which is a Korean native Taraxacum species group. The result from the RAPD method was very similar to the result from the Bootstrap method. From the examination of the physical characteristics of 6 Taraxacum species populated in Korea, flowering period of Taraxacum species in group I are longer than Taraxacum species in group ll, and the direction of involucral bract of Taruxacum species in the group I was also different comparing to the group ll. Because the flowering color, leaf direction, and the specificity of seed germination of T. coreanum were different compared to the other species in the group II, T. coreanum would be genetically divergent and showed the highest dissimilarity index score.

Epicranial Bony System of Six Species in Family Cynoglossidae (Pisces, Pleuronectiformes) from Korea (한국산 참서대과 Cynoglossidae 어류 6종의 상두개골계(epicranical bony system))

  • The epicranial bony system of the six species of the family Cynoglossidae were examined based on the specimens collected from the coast of Korea from 1992 to 1994. They were divided into four groups by the morphological features of epicranial bony system as follow: 1. Paraplagusia japonica, 2. Cynoglossus joyneri and C. robustus, 3. C. semilaeuis and C. abbreuiatus, and 4. C. interruptus. Among them, P japonica showed the most derived character in the erisma and frontal elements of the epicranial bony system. And it was remarked that the number of proximal radials supporting erisma show much differences among the species. It is discussed that the morphological differences in the epicranial bony system are significant character for interspecific relationships in family Cynoglossidae.

A Morphological Study of Symplocaceae in Korea (한국산 노린재나무과의 형태학적 연구)

  • The morphological characters for 4 taxa of Korean Symplocaceae were examined. Based on morphological examination, numerical analysis was performed to clarify the taxonomic relationships among the taxa. It was found that epidermal characters of leaves, cellular deposition of petals, cellular boundary of petioles, growth of stigmas and pollen grains and their surfaces were useful diagonostic characters. From the results of PCA analysis, four taxa were grouped as species clusters including each populations. Four species of Symplocos were clustered as species groups with clear delimitation. Morphological, numerical, and palynological analyses supported the previous morphological studies on this family.

Variation of Peroxidase Isoenzymes in Rhododendron plants (Rhododendron에 있어서 Peroxidase 동위효소(同位酵素)의 변이성(變異性))

  • Peroxidase banding patterns were obtained by disc electrophoresis of leaf extractions from flowering plants, R. mucronulatum, R. yedoense var. poukhanese, and R. schlippenbachii. A total of 21 peroxidase isoenzymes were resolved from these species, R. mucronulatum, P. yedoense var. poukhanese, and R. schlippenbachii. Five identical peroxidase isoenzymes were resolved in R. yedoense var. poukhanese and R. schlippenbachii. It means that there is a close relationships between two species. The peroxidase banding patterns of all three Rhododendron species were not correlated with each other on percentage total density of each band. The correlation coefficients for peroxidase band densities compared with leaf areas and leaf width/length ratios of Rhododendron plants were not significant at the 5% level. Wide variation was observed in R. yedoense var. poukhanese and R. schlippenbachii compared with R. mucronulatum.

  • choChorion(egg-shell) morphology of the F1 hybrid between Bombyx mori and Bombyx mandarina has been observed by scanning electrom microscope and chorion protein was analyzed by electrophoresis. The chorion surface structure of F1 hybrids in the lateral(flat) region was similar to that of maternal line. The F1 hybrids chorion was found to have basically a three layer structure. The middle and inner layer were very much like those of the Bombyx mandarina and Bombyx mori. There were many conic pillar structures in the outer layer of the F1 hybrid, which was similar to Bombyx mandarina. This conic pillar structure had a thin cover layer was more clear in the dorsal and ventral side of the F1 hybrid chorion. The conic pillar structure of Bombyx mandarina was found to be dominant in F1 hybrid chorion irrespective of their maternal line. Major components of chorion protein were analyzed by two-dimensional electrophoresis and found to have isoelectric points in the range of pH 4.0-6.5 and molecular weight 10 to 50 kd. F1 hybrid chorion protein components related directly to those of the maternal line. The conic pillar structure was dominat characteristic and it was present in all F1 hybrid.

Phylogenetic study of the Genus Suaeda(Chenopodiaceae) based on chloroplast and nuclear DNA sequences from Korea (엽록체 DNA 및 핵 DNA 염기서열에 근거한 한국산 나문재속(명아주과)의 분류학적 연구)

  • The purpose of this study was to identify the phylogenetic relationships of the plants in the Korean genus Suaeda and to find out the molecular markers that could confirm the interspecies relationships in the family tree through molecular phylogenetic studies. We used the nuclear ribosomal DNA ITS and the chloroplast DNA matK, psbA-trnH, and trnL-trnF as the molecular markers. We could not distinguish between S. japonica and S. maritima and between S. maritima and S. australis in the ITS region and could not distinguish between S. japonica and S. australis with the base sequence in the psbA-trnH and trnL-trnF region. However, we analyzed the combinations of four molecular marker regions and confirmed that each of five plant species of the genus Suaeda formed the independent line. Therefore, it is considered that combinations of molecular markers would be useful for the analysis of phylogenetic relationships in the genus Suaeda. Further investigations of the ecological and morphological characteristics would be needed to understand the phylogenetic relationship and lineage diversification in the genus Suaeda.
