Analysis of Inter-simple sequence repeat (ISSR) markers in cultivars and collected strains of button mushroom (Agaricus bisporus) (양송이 품종과 수집 균주간의 Inter-simple sequence repeat (ISSR) 마커 분석)

  • A. bisporus is the fifth most cultivated mushroom in Korea, and approximately 10,757 tons were cultivated in 2015. The genetic diversity of collected strains in Korea and commercial cultivars was analyzed using inter-simple sequence repeat (ISSR) markers. ISSR markers known to be comparable among A. bisporus spp. were selected from various markers. Totally, 16 markers, namely the ISSR markers 807, 808, 810, 811, 834, 835, 836, 841, 842, P3, P8, P17, P22, P30, P38, and P39, were evaluated to discriminate between ASI 1110, 1114, 1115, 1238, 1246, 1365, 1366, and 1369 for selecting suitable markers in 16 markers. The ISSR markers P31, P38 and P39 exhibited various fingerprints that could help classify the strains in species. Using the three markers, genetic relationships among 39 strains, including commercial cultivars, such as SaeA and SaeYeon, were analyzed using the UPGMA method. The results of the analysis of the genetic relationships between commercial cultivars and collected strains in Korea confirmed that the commercial cultivars were different from the collected strains in Korea. These results suggested that the ISSR markers P31, P38, and P30 could be used for selecting the commercial cultivars of A. bisporus.

Genetic variation in five species of Korean Orostachys (Crassulaceae) (한국산 바위솔속(돌나물과) 5종에 대한 유전적 변이)

  • Starch gel electrophoretic studies using 24 populations of five Korean Orostachys species were conducted to investigate allozyme variation and to test hypotheses of systematic relationships among species. The resulting phenogram showed that the populations of five Korean Orostachys species were divided into two major groups. And they were concordant with molecular and morphological data in suggesting that Orostachys was divided into two groups corresponding to the subsect. Appendiculatae and subsect. Orostachys. The low genetic identities among Korean Orostachys species indicated that the species of Orostachys have diverged gradually through the model of geographical species. Comparing the previous genetic data from the species with similar life history and mode of reproduction, most of Korean Orostachys species revealed a significant low genetic variation, while the widespread O. japonicus showed a relatively high genetic variation among the Korean species. This kind of genetic variation pattern might be the results of the isolated habitats, limited numbers of individuals within the populations, destruction of habitats, inbreeding and asexual reproduction in Korean Orostachys populations. The Jungdongjin population (POP 21) of O. malocophyllus was genetically unrelated with remaining populations of the same species, and this interpretation was consistent to the results from the previous palynological and morphological studies. Our allozyme data supported the taxonomic treatment of recently proposed taxa, O. iwarenge (Makino) Hara for. magnus and O. margaritifolius.

Taxonomical Classification and Species-specific Detection of Genus Some Phellinus using Phylotype (Phylotype에 의한 수종의 Phellinus속의 분류체계 확립 및 종간구별을 위한 신속동정법 개발)

  • This study was carried out to identify the phylogenetic relationship of Phellinus species and to know its distribution by comparing the DNA sequences of internal transcribed spacer regions(ITS1 and IST2) and 5.8S ribosomal DNA (rDNA) repeat unit. The Phellinus species had their specific sequences in IST1 and 2 regions depending on suedes. The comparison of the ITS sequences of standard strains indicated that the sequences of ITS1 were more variable than those of ITS2. Nine strains of the commercial products of Phellinus species used in this study were identified as P. lintues, P. baumii, P. igniarius, and P. pini. Most of commercial species were P. pini and P. baumii, and P. gilvus was not found. Also, P. linteus was only found in form of mycelial culture rather than fruiting body. Moreover, the species-specific primers were designed based on ITS sequence data. Each species-specific primers were bound in P. lintues(ITSF-PL2R), P. baumii(PB1F-ITS4R), P. igniarius(IF1-IR3), P. pini(PF1-PR3), and P. gilvus(GF2-GR4), respectively. These primer sets would be useful fer the detection of specific-species among unidentified Phellinus species rapidly.

Classification of the Genus Lilium Native to Korea by Pollen Size (우리나라 자생나리의 종별 화분형태 분석)

  • This study was carried out to estimate the size and exine pattern of pollen grains in seven species of the genus Lilium by using scanning electron microscope. Exine of pollen grains, which were all monad, took the prolate forms with a pollen aperture. However, there were various L/W ratio between length (L) and width (W) of pollen grains among selected Lilium species. The pollen of L. distichum was oblong shape which had the highest L/W ratio of 2.59. On the other hand, that of L. concolor var. partheneion was broadly elliptical shape close to circle which had the lowest L/W ratio of 1.82. The surface patterns of exine were reticulate in all Lilium species that have showed diverse forms in their lumina. Pollen length and width in L. lancifolium had the greatest value, 115.90 and $46.54{\mu}m$, respectively, in comparison with those of L. tsingtauense, 70.33 and $38.72{\mu}m$, respectively. Except L. distichum and L. leichtlinii var. maximowiczii which had a similar pollen length within standard errors, the others in Lilium species showed different sizes, respectively. Therefore, these results indicated that pollen length could be a better standard for the classification of Lilium species than exine surface patterns or pollen dissection area.

Karyotypes, DNA Values and Nuclear Sizes of Several Scups (Teleostomi : Perciformes) (돔류(類) 몇종(種)의 염색체(染色體), DNA함량(含量)과 핵(核)의 크기에 관한 연구(硏究))

  • An examination of the karyotypes, DNA values and nuclear sizes of three scups was undertaken as part of the study of cytogenetical evolution of order Perciformes. The chromosome number 2n=48 was the same in all three species but the numbers of chromosome arm were not identical. The distribution of genome size and nuclear volumes among species was continuous ranging from 1.287 pg and $20.78\;{\mu}m^3$ for Pagrus major down to 1.237 pg and $20.56\;{\mu}m^3$ for Acanthopagrus schlegeli. Above results indicate the possible role of pericentric inversions in the karyotypic evolution of these species.

Genetic Relationship among Garlic Cultivars Based on RAPD Analysis (RAPD에 의한 마늘의 유연관계 분석)

  • RAPD analysis using random primers were tried to evaluate the genetic variation and diversity of the nine garlic cultivars including two foreign varieties. Thirty-two primers out of 70 primers screened were used to amplify genomic DNA of garlic cultivars using polymerase chain reaction(PCR). Among a total of 151 bands amplified by 32 primers, 125 polymorphic bands were subjected to analysis for genetic relationship of garlic cultivars. The estimated size of amplified PCR products were in the range of 932 to 4,060 base pairs. Nine garlic cultivars were classified into two groups, such as group I corresponded to Changnyung and Hungary cultivars, and group II, Namdo, Sandong from China, Yecheon, Euiseong, Youngweol, Danyang, Jeongsun cultivars, with the genetic distance value of 0.271. The major ecological types of garlics, so called southern and northern types, was grouped in the genetic distance value of 0.200. The results presented in this study suggest that RAPD analysis are likely to be useful for identification of cultivars and evaluation of genetic origin in garlics.

Fruit Wall Anatomy of Ocotea (Lauraceae)

  • The fruit wall anatomy of Ocotea was investigated on the basis of 14 species within the genus to contribute to a better understanding of specific relationships and homogeneity of genus. The species have a similar mature fruit wall structure, but diUerences among the species are found with respect to whether or not sdc.nchyma cells are present in the mesocarp. if present, whether or not they are present in particular positions and forms. Comparisons with species studied suggested that at least a few groups of species can be distinguished in Ocotea. They arc divided into five groups on the basis of anatomical structures. i.e., group 1) O. atrriensis, O. cujumari, O. helicterifolia, O. rubra and O. schomburgkiana; group 2) O. aeiphylla, O. javitensis, and O. sp. [Werff et ai. 12676]; group 3) O. tonduzii: group 4) O. foetens, O. quixos, and O. veraguensis; and group 5) O. floribunda and O. nitida. These various variations in Ocntea were also discussed to invite its respective systematic revisions. By the comparisons with species, on the other hand, it suggested that the specialized species are evolved from non-specialized species.

Phylogeny and Speciation of Entomobryoidea (Collembola, Insecta) from Korea (한국산 털보톡토기상과 (톡토기목, 곤충강)의 계통과 종분화)

  • In order to understand affinities and phylogeny of three families of the Superfamily Entomobryoidea allozyme analysis was performed with three species presumably representing each family, in addition to four species belonging to the neighboring Superfamily Poduroidea together. Electrophoresis for this purpose produced data for calculating allele frequency and enabled to obtain genetic distance and to depict dendrograms as well. The species of Isotomidae, Entomobryidae and Tomoceridae were clustered together whereas those of Hypogastruridae, Onychiuridae and Neanuridae were grouped as monophylies, respectively. Even though Tomoceridae and Entomobryidae were brought together they are considered complete families by showing high genetic distance value between them, thus supporting the hypothesis on their affinity among others. Gulgastrura reticulosa has been revealed to be separated from Hypogastruridae by being clustered rather with Onychiuridae. However, its high genetic distance value suggests the status as an independent family. The present result agreed with the phylogeny of Suborder Arthropleona divided into two Superfamilies mentioned above as in conventional systematics and also when compared with analysis of data of their morphological characters as well as 18S rDNA performed and published elsewhere by the present writers.

Genetic Diversity of Penicillium isolates Isolated from Pears with Postharvest Decay in Storage (수확 후 저장 배에서 분리한 Penicillium속 균의 유전적 다양성)

  • This study was carried out to identify the genetic diversity of Penicillium isolates that were isolated from pears with postharvest decay in storage. URP-PCR was used to detect DNA diversity of 84 Penicillium isolates. Based on URP-PCR profiles, 18 Penicillium isolates were selected and their PCR polymorphic bands were produced by additional primers URP1F, URP2R, URP2F, and URP4R. UPGMA cluster analysis using the polymorphic bands showed four clustered groups and futhermore cultural and morphological features characterized the 18 Penicillium isolates. Group 1 was dominant, which occupies 70% in the four clustered groups and identified as P. expansum based on ITS sequence and morphological features.

Comparative Studies on the Developmental Capacity by Interspecific Nuclear Transplantation (이종간의 핵치환에 의한 발생수행능력의 비교 연구)

  • In order to investigate the role of nuclei and cytoplasm in early embryogenesis, interspecific nuclear transplantation was carried out Nuclei of one species of amphibian was transplanted into enucleated egg of another species. In interspecific hybrids between Rana species, development was arrested before or right after the dorsal lip formation. In intergenetic hybrids between Rana, Xenopus and axolod, developmental arrest took place at late blastula stage. Comparing the developmental capacity of each nudeocytoplasmic hybrid, the more distantly related the species, the earlier does development arrest. The general rule is that nuclear transfers between any amphibian species will form a regulary cleaved late blastula. Two plausible factors relate to limitation of tite developmental capacity of nudeocytaplasmic hybrids. One is an irreversible change in grafted nuclei and another is tunctional incompatibility between the recipient cytoplasm and transplanted nucleus. The later is postulated a more possible cause of the arrestrnent.
