Total Polyphenol and Flavonoid of Fruit Extract of Opuntia humifusa and Its Inhibitory Effect on the Growth of MCF-7 Human Breast Cancer Cells (천년초선인장 열매추출물의 폴리페놀, 플라보노이드 함량과 유방암 세포(MCF-7)에 대한 성장 억제효과)

  • Opuntia humifusa, widely distributed in the southern regions of the Korean peninsula, is known to have bioactive functions and medicinal benefits for treating various diseases such as arteriosclerosis, diabetes mellitus, gastritis, and hyperglycemia. In this study total polyphenol and flavonoid contents of fruit and its anticarcinogenic effects on human breast cancer were investigated. As expected, O. humifusa showed high concentrations of total polyphenol as well as flavonoid as compared to other kinds of cactus. Effects of the water extracts of O. humifusa on the proliferation, G1 arrest and apoptosis of the MCF-7 human breast cancer cells were also examined using the MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assays, and G1 cycle arrest and apoptotic effect of O. humifusa were analyzed by flow cytometry. When MCF-7 cells were treated with different concentrations of hexane, ethyl acetate and water extracts of O. humifusa, water extracts of the fruit significantly decreased viable cell numbers in a concentration dependent manner. A G1 arrest in MCF-7 cells was induced as well. The overall results indicate that water extracts of fruit of O. humifusa would inhibit MCF-7 human breast cancer cell proliferation and induce G1 arrest.

Protective Effect of Rhus Semialata M. extract on Epidermal Stem Cells against UV Irradiation (자외선 조사된 상피 줄기세포에 대한 붉나무 추출물의 보호 효과)

  • Human epidermal stem cells(ESCs) residing in the basement membrane of the skin have an important role in maintenance of skin homeostasis of epidermal layer. Although, ESCs provide new cells to repair damaged tissue in response to tissue injury, subsets of stem cells remain in the quiescent state protected from differentiation and senescence for prolonged survivals. In this perspective, the stem cell niche, which is specific microenvironment composed of niche cells and an extracellular matrix(ECM), supplies the relevant signal to save stem cells from microenvironmental damages. The expression of stemness marker on the surface of ESCs contributes to the attachment on their ECM of the basement membrane, which lead to growth potential and apoptotic resistance against environmental stimuli. In this study, we observed that UV irradiation, a major factor of environmental stimuli, reduced the expression of α2, β1 and α6 integrin in ESCs. Rhus Semialata M extract(RSE) showed inhibitory effect on the UVB-induced reduction of integrin expression. Furthermore, RSE could upregulate the expression of Col-IV and Laminin, which contribute to the attachment of ESCs. These results indicated that RSE could be a potent ingredient for the protection of ESCs from UV irradiation by increasing the expression of integrin and substrate ECM components at their niche.

Development of Organ Culture Medium for Long Term Culture of Human Hair Follicle (인체 두피 모낭의 장기간 배양을 위한 기관 배양 배지의 개발)

  • We successfully isolated human anagen hair follicles from human scalp skin by microdissection and tried to culture them under various conditions. First we confirmed negative effect of serum on human hair follicle organ culture. As a next step serum-free medium compositions, Philpott medium, IMDM, and DHGM (Dongguk hair growth medium) were tried. Philpott medium is a general medium for hair organ culture based on Williams' E medium and DHGM is a special self-developed medium containing high amino acids and vitamins (B group) composition. As results, hair follicle in Philpott medium and IMDM showed anagen phase morphological structure, but rapid loss of hair elongation, low alkaline phosphatase expression, and very low expression of CK19. It is thought these hair follicles rapidly regressed from apoptosis. However, hair follicles in DHGM showed long term anagen phase morphological structure, continuous hair elongation, high alkaline phosphatase, and CK19 expression. These results demonstrate that high amino acids and vitamins (B group) composition are essential to in vitro long term human hair follicle organ culture and this culture medium will be useful in basic study of hair biology or application study to the development of alopecia treatment drugs.

Evaluation of Bactericidal Capacity of Silver Ion against Bovine Mastitis Pathogens (소 유방염 원인균에 대한 은이온의 향균력 평가)

  • Bovine mastitis, defined as an inflammation of the mammary gland, is usually associated with bacterial infection. Thus, treatment and control of mastitis relies primarily on antimicrobial therapy. This study investigated the bactericidal actions of silver ion against causing various bovine mastitis pathogens. Minimal inhibitory concentrations (MICs), minimum bactericidal concentrations (MBCs) and bactericidal activity times and concentrations of silver ion against pathogens were determined. The effect of silver ion on bacterial morphology was studied by scanning electron microscopy (SEM). The MICs and MBCs of silver ion for various bacteria strains ranged from 1.9-15.6 ${\mu}g$/ml. SEM images indicated formation of a pit, distortion and disruption of cell walls in silver treated Staphylococcus aureus and Escherichia coli. The results demonstrate that silver ion has a bactericidal capacity against causing various pathogens of bovine mastitis and suggest that silver ions may be exploitable as a therapeutic/preventative tool of bovine mastitis.

Characteristics of Adhesive Disks in Parthenocissus tricuspidata during Attachment (착생에 따른 담쟁이덩굴 흡착근의 부착 특성)

  • Parthenocissus tricuspidata is an epiphyte that lacks a main axial stem, but develops adhesive disks along the stem for climbing support. In this study, scanning electron microscopy (SEM) and energy dispersive X-ray spectroscopy (EDS) were utilized to examine the brick wall surface and the adhesive disks of P. tricuspidata that attached to the surface successfully. The study was mainly focused the outermost layers of both structures before and after adhesion to find out whether there has been some structural and/or physical interactions between the two. The adhesive disks adhered firmly to the brick wall by secreting adhesive materials that help them for a tight attachment to the surface. The rough wall surface appeared facilitating better attachment of the adhesive disks by infiltrating the materials into those spaces leading to some degree of interactions at the interface. EDS analysis on the outermost layers of the adhesive disks that were separated from the substrates was also consistent with the SEM data on the interaction between the adhesive disks and the substrate surface. EDS analysis of the brick wall surface and the adhesive disks demonstrated similar elements of O, Si, Fe, Al, K, Mg, and Na in their components.

Screening of Potential Anticancer Compounds from Marketed Drugs: Aripiprazole, Haloperidol, Miconazole, and Terfenadine Inhibit Cytochrome P450 2J2 (시판 약물의 시토크롬 2J2 약물대사효소 저해능 탐색)

  • Cytochrome P450 2J2 (CYP2J2) plays important roles in the metabolism of endogenous metabolites such as arachidonic acid as well as therapeutic drugs. CYP2J2 is overexpressed in human cancer tissues and cancer cell lines, as well as in epoxyeicosatrienoic acids (EETs) and CYP2J2-mediated metabolites, and prevent apoptosis of cancer cells. This study aimed to screen marketed drugs for inhibitory potential on CYP2J2 isoforms using human liver microsomes. The initial screen isolated 4 compounds, from 120 marketed drugs, that inhibited the CYP2J2-mediated astemizole O-demethylation more than 50% in the following the order: haloperidol (75%) > terfenadine (56%) > aripiprazole (55%) > miconazole (52%). Miconazole strongly inhibited CYP2J2-mediated ebastine hydroxylation ($IC_{50}$=11.2 ${\mu}M$) and terfenadine hydroxylation ($IC_{50}$=2.2 ${\mu}M$), and terfenadine also inhibited CYP2J2-mediated ebastine hydroxylation ($IC_{50}$=13.6 ${\mu}M$) in a dose dependent manner. The present data suggest that these drugs are potential candidates for further evaluation for their anti-cancer activities.

Preparation and Characterization of Double-Layered Coated Capsule Containing Low Molecular Marine Collagen and γ-Aminobutyric Acid Producing Lactobacillus brevis CFM20 (저분자 해양성 콜라겐과 γ-Aminobutyric Acid 생성 Lactobacillus brevis CFM20을 함유하는 이중코팅캡슐의 제조 및 특성)

  • This study was performed to encapsulate low molecular weight marine collagen and ${\gamma}$-aminobutyric acid (GABA)-producing lactic acid bacteria to inhibit degradation and improve survival rate during exposure to adverse conditions of the gastro-intestinal tract. Calcium-alginate method was used for the manufacture of a double-layered coated capsule. The inner core material was composed of collagen and lactic acid bacteria, and the coating materials were alginate and chitosan. The sizes and shapes of the double-coated capsule were affected mainly by centrifuge speed and pH. Manufactured capsules were observed with a scanning electron microscope and by confocal laser scanning microscopy to confirm the micromorphological changes of capsules and bacterial cells. As a result, double-layered coated capsules were not degraded at pH 1.2, whereas degradation occurred at pH 7.4. In addition, GABA and collagen were maintained in stable state at pH 1.2. Therefore, double-layered coated capsules developed in this study would not be degraded in the stomach and could be stably delivered to the small intestine to benefit intestinal and dermatic health.

Functional Analysis of Fibroblastic Reticular Cells Derived from Mouse Lymph Node via Bidirectional Crosstalk with T Cells (T세포와 양방향 작용을 통한 마우스 림프절로부터 분리된 fibroblastic reticular cell의 기능적 분석)

  • Fibroblastic reticular cells (FRCs) form the structural backbone of the T zone provide a guidance path for immigrating T cells in the lymph node (LN). FRCs may contribute directly to developing T-cell biology in the LN and allow analyses of fundamental aspects of FRC biology related to T cells. FRCs inhibited T-cell apoptosis, and FRC culture supernatants strongly induced the expression of Bcl-xL in T cells against doxorubicin. Coculture of FRC and T cells resulted in rearrangements of the actin cytoskeleton, as well as global changes in the morphology of the FRCs. In addition, when cocultured, the T cells adhered to the FRC monolayer, and the membrane intercellular adhesion molecule (ICAM)-1 was slightly increased by day-dependent manner. In contrast, the expression of soluble ICAM-1 was dramatically increased in a day-dependent manner. Several chemokines, such as CCL5, CXCL1, CXCL5, CXCL16, CCL8, CXCL13, and ICAM-1, and MMPs were expressed in FRCs sensed by tumor necrosis factor (TNF) families. Nuclear factor kappa B ($NF{\kappa}B$)-RelA of the $NF{\kappa}B$ canonical pathway was translocated into FRC nuclear by $TNF{\alpha}$. In contrast, p52 proteolyzed from p100, a counterpart of RelB of the noncanonical $NF{\kappa}B$ pathway, accumulated in the peripheral FRC nucleus by agonistic anti-$LT{\beta}R$ antibody. In summary, we propose a model in which FRCs engage in bidirectional crosstalk to increase the efficiency of T-cell biology. This cooperative feedback loop may help to maintain tissue integrity and function during immune responses.

Effects of High-fat Diet on Type-I Muscle Loss in Rats (고지방식이가 쥐의 Type-I 근육손실에 미치는 영향)

  • The term lipotoxicity has been used to describe how excess lipid accumulation leads to cellular dysfunction and death in non-adipose tissues, including skeletal muscle. While lipotoxicity has been found in cultured skeletal muscle cells with high-fat feeding, the consequences of lipotoxicity in vivo are still unknown, particularly in Type-I muscle, which is metabolically affected by lipotoxicity. The aim of this study was to investigate the effects of a high-fat diet on changes in the morphology and apoptotic protein expression of Type-I muscle loss in rats. The rats were fed either a high-fat diet or a normal diet for six weeks, and then lipid accumulation, inflammation response, and nucleus infiltration were measured, and PARP protein expression was cleaved by Oil Red O staining, H & E staining, and Western blot, respectively. Lipid accumulation, inflammation response, nucleus infiltration, and cleaved PARP protein expression were significantly (p<0.05) higher in the high-fat diet group than they were in the normal diet group. The weight of Type-I muscle tended to be lower in the high-fat diet group compared to the normal diet group, but the difference was not statistically significant. These results indicate that a high-fat diet triggers cell death in Type-I muscle via lipotoxicity, which suggests that a high-fat diet may be associated with sarcopenia.

Comparison of Immuno-Modulatory Regulatory Activities of Rubus coreanus Miquel by Ultra High Pressure Extracts Process (초고압 공정에 의한 복분자의 면역조절효능 비교)

  • This study was performed to investigate the enhancement of anticancer activities and immuno modulatary activities from R. coreanus. by ultra high pressure extracts process. The cytotoxicity on human kidney cell (HEK293) was showed below 19.5% in adding 1.0 $mg/m{\ell}$ concentration. The anticancer activity was increased over 10% by high pressure processing in AGS and A549 cells. The immune cell growth using human immune B and T cells was improved by the high pressure extracts of Rubus coreanus in adding 1.0 $mg/m{\ell}$ concentration. The secretion of two kinds of cytokine, the IL-6 and $TNF-{\alpha}$ from human immune B and T cells were also enhanced in adding extracts by high pressure process of R. coreanus. The ultra high pressure extraction technique showed high efficiency in extracting of bioactive compound. The ultra high pressure technique could be used combined with other technique to improve the extracting rate and extracting efficiency.
