Analysis of Turbulent Flow by Location Characteristics of Side Weir inlet in Meandering Channels (사행수로 구간의 횡월류위어 유입구 위치특성에 따른 흐름해석)

  • 횡월류위어(side weir)는 하천의 수위가 한계수위 이상으로 상승할 경우 본류로부터 저류지나 분수로(distributary channel)로 흐름을 전환하기 위하여 사용하는 수공구조물로 강변저류지나 off-line저류지의 유입부에 흐름방향과 평행하게 설치되어 유량관리 및 전환, 홍수통제, 에너지 소산, 수위조절, 일정 유량의 취수 및 분배, 초과 홍수량의 전환 등의 목적으로 이용되는 구조물이다. 횡월류 위어의 월류 흐름은 일반위어와 같이 위어마루부 직각방향으로 흐르지 않고 본류 흐름특성에 따라 비스듬하게 유입된다. 이러한 흐름특성으로 횡월류위어 월류량은 본류의 하폭, 흐름특성, 위어길이 및 설치위치 등에 따라 각기 다르게 산정된다. 현재 국내에서 진행된 횡월류위어 흐름특성에 관련된 연구는 대부분 직선수로에 집중되어 있으며 사행하천의 흐름특성에 따른 연구는 부족한 실정이다. 금회 연구에서는 3차원 상용프로그램인 FLOW-3D를 이용하여 사행하천구간 유입부 설치위치 특성에 따른 횡월류 위어 유입흐름 특성을 분석하였다. 사행하천 구간 횡월류위어 설치위치에 따른 3차원 흐름해석을 위해 AUTO CAD 프로그램을 이용하여 수로길이 30m, 수로폭 2m의 구형 사행수로를 구성하였고, 횡월류위어 유입부 위치를 20°~120°로 변화시키며 수치모형실험을 수행하였다. 해석결과 수로흐름은 유입부 설치각이 작을수록 상·하류 수위차가 작아지며 유속이 감소하며 설치위치각이 클수록 수로내 평균유속은 증가하는 것으로 확인되었다. 유입부 설치각이 작을수록 방류량이 증가하여 수로내 흐름분리현상 증가하였고 이로인한 지체현상이 발생하는 것으로 확인되었다. 본 연구로 사행하천구간에 횡월류위어가 설치된 경우, 월류량과 수리학적 흐름특성을 해석할 때 3차원 수치모형실험이 유용한 해석도구로 이용될 수 있음이 확인되었다. 이후 수치모형실험이 수공구조물 설계 및 해석 시 참고자료로 이용가능할 것으로 사료된다.

Characterization of peptide:N-glycanase from tomato (Solanum lycopersicum) fruits (토마토 (Solanum lycopersicum) 과육의 숙성정도에 따른 peptide:N-glycanase 발현 분석)

  • In eukaryotes, proteins that are secreted into ER are post-translationally modified by N-glycosylation, the patterns of which are significantly different between plant and animal cells. Biotechnology industry has already produced a number of therapeutic glycoproteins in plant cells. However, the aberrant glycosylation of therapeutic recombinant proteins in plant systems can cause immune problems in humans. Therefore, it is important to develop strategies for producing non-glycosylated forms to preserve biological activity and native conformation by a peptide: N-glycanase (PNGase). In this study, we try to isolate PNGase T gene from tomato, which can use as a platform plant for biotechnology industry. We isolated a cDNA (GenBank Accession number KM401550) from tomato leaves with 1,767 bp, which encoded a polypeptide of 588 amino acids with a predicted molecular mass of 65.8 kDa. We also investigated the expression patterns of PNGase T during fruit ripening of tomato. The transcripts of PNGase T, which were constitutively induced in tomato fruit from green stage, were significantly increased and reached a peak at orange stage. After which, those transcripts were continuously reduced. The expression pattern of PNGase T was coincided well with transcripts profiles of metacaspase gene, LeMCA, and senescence-related gene members of ACC synthase, LeACS2, LeACS4, and LeACS6, for ethylene biosynthesis during fruit ripening. These results suggest that PNGase T is involved in a de-glycosylation process associated with senescence and fruit ripening.

  • Self-injurious behavior is defined as deliberated harm to one's own body without suicidal intent. It usually occurs as head banging or hitting, body hitting, skin cutting, or finger biting and includes ocular, genital, and self-inflicted oral mutilation. Self-injurious behavior can occur with mental retardation, coma, psychotic problem, poisoning, or character disorders. In pediatric patients, self-injurious behavior usually is reported to lip, cheek and tongue biting, and many kinds of dental management methods have been introduced to prevent self-injurious behavior patients from self biting. This report presents two self-inflicted oral mutilation patients who were all treated successfully with appliances such as modified activator without wire for retention, modified tongue-rake appliance and mouth guard.

Fabrication of Tailor-Made 3D PCL Scaffold Using a Bio-Plotting Process (바이오-플로팅시스템을 통한 Tailor-Made 3D PCL Scaffold 제작)

  • Biomedical scaffold for tissue regeneration was fabricated by one of rapid prototyping processes, bioplotting system, with a biodegradable and biocompatible poly($\varepsilon$-carprolactone)(PCL). Through dynamic mechanical test, it was observed that the PCL scaffold manufactured by the bioplotting process has the superior mechanical properties compared to the conventional scaffold fabricated by a salt-leaching process, and the plotted scaffold could be employed as a potential scaffold to regenerating hard and soft tissue. The plotted scaffold was consisted of porous structures. which were interconnected with each pore to help cells be easily adhered and proliferated in the wall of pore tunnels, and metabolic nutrients can be transported within the matrix. By using the plotting system, we could adjust the pore size, porosity, strand pitch, and, strand diameter of PCL scaffolds, which were important parameters to control mechanical properties of the scaffolds, and consequently we could determine that the mechanically controlled scaffolds could be used as a matching scaffold for any required mechanical properties of the target organ. The fabricated 3D PCL scaffold showed enough possibility as a 3D biomedical scaffold, which was cell-cultured with chondrocytes.

SELF-INJURIOUS BEHAVIOR IN A PATIENT WITH AUTISM : A CASE REPORT (자폐 환자의 자해로 인한 구강 내 손상 : 증례 보고)

  • Self-injurious behavior (SIB) has been defined as the deliberate destruction or alteration of body tissue without conscious suicidal intent. It occurs in conjunction with a variety of psychiatric disorders as well as various developmental disabilities and some syndromes. The behavior is destructive and causes concern and distress to all involved in the care and treatment of the affected individual. A 13-year-old girl with autism, mental retardation and delayed development was reffered from her pediatrician because of severe and painful lower lip biting. An intraoral examination revealed a diffuse swelling of lower lip. It was covered with necrotic slough and the ulcer and scarring of the lower lip was observed. We chose to use an oral removable prosthesis for Conservative treatment. It was decided to use a soft silicone mouthguard in the maxillary arch. Initially, she could not tolerate the appliance inside her mouth but soon adapted with the appliance. After one month, she lost the mouth guard and started lip biting. So we made mouth guard again. There are no standard methods for preventing self-injurious behavior in a patient who is developmentally disabled. Appropriate preventive methods must be developed for each individual patient based on close observation and clinical findings. Behavior modification techniques, pharmacological treatment, extraction of teeth, orthognathic surgery and intra/extra oral appliances can be performed for adjust self-injurious behavior. A suitable oral guard could be tried initially before employing more invasive approaches.

Fabrication and characterization of porous hydroxyapatite scaffolds with PMMA addition using tertiary-butyl alcohol based freeze casting method (삼차부틸알코올 기반 동결주조 공정을 이용한 PMMA 첨가 다공질 수산화아파타이트 지지체의 제조 및 특성 평가)

  • In order to prepare porous scaffolds capable of pore control, PMMA powder serving as a pore-forming agent was added to HA powder to synthesize a slurry containing TBA as a solvent. And then, porous HA scaffolds where pillarshaped pore channels interconnected with each other were fabricated by freeze-casting and sintering. The crystal structure of the HA scaffolds according to the addition amount of PMMA powder was measured by XRD and the surface and inner cross section of the scaffolds were analyzed through SEM. It was found that removal of PMMA during sintering affects the internal structure of the scaffolds and the crystallinity of the HA powder. Furthermore, through evaluating the physical and mechanical properties of the scaffolds, it was confirmed that the porosity, pore size and compressive strength can be controlled by controlling the addition amount of the pore-forming agent. It was also found that the HA scaffolds produced in this study were similar in structure and properties to the natural cancellous bone. This suggests that porous HA scaffolds with PMMA can be used as an alternative to autogenous bone for tissue engineering as an artificial bone scaffold.

  • Cerebral palsy, a range of non-progressive syndromes of posture and motor impairment, is a common cause of disability in childhood. Self-injurious behavior(SIB) is deliberate harm to the body without suicidal intend, often involving repetitive actions that cause tissue damage. One of the most common orofacial self-injurious behavior is chewing tongue, lip or oral mucosa. This kind of SIB in children is not common in normal children. High occurrence rates are observered in cases of syndromatic, mentally retarded children, and children with congenital disease. Various methods such as behavior modification, behavior control by drugs, body restraints, application of dental appliance, surgery and extraction of teeth have been suggested to control those self-injurious behavior. Using mouthguard as one of dental applainaces is the most conservative and appropriate method in terms of reducing oral self-injurious habits and protection of tissue. This case report describes a child with cerebral palsy who presented with self-injurious ulceration of lip and buccal mucosa. A modified mouthguard was effective in preventing self-injurious oral trauma in a child with cerebral palsy.

Characteristics for Co-digestion of Food Waste and Night Soil using BMP Test (BMP실험을 이용한 음식물폐기물 및 분뇨의 병합소화 특성)

  • BMP test was carried out to evaluate the characteristics for co-digestion of night soil and food waste. 6 types of sludge were tested in 30 days which were raw, excess, digested, night soil/septic tank (1:1), food waste (food : dilution water = 1:1), and mixed sludge. Bio gas was produced actively after 2 days, and continued in 2 weeks. Gas generation amount was decreased rapidly after considerable space of time. Especially maximum productivity of gas was shown in 7~8 days. The ultimate methane yields of raw, excess, digested, night soil/septic tank, food waste, and mixed sludge were 64.63, 67.49, 66.45, 72.44, 107.85, and 46.71 mL $CH_4/g$ VS respectively from Modified Gompertz model. The lag growth phase time and maximum specific methane production rate of mixed sludge were 1.88 day and 80.4 mL/day respectively. The methane potential of mixed sludge was higher than individual sludge. So high methane potential was expected by controlling mixing ratio of food waste. Besides stable operation of digestion tank and the solution of oligotrophic problem were possible.

Direction of the study was concerntrated on three areas, with results and information collected, would like to suggest design change of X type collapsible chair in areas of size of SEAT(air pocket type), HANDRIM(anti-slip hump), ARMHEST(transfer movement, rubber padding. accent), and HANDLE(adjustable, round type). Preparation and BDNF Release Profile of BDNF-loaded PLGA Scaffolds for Tissue Engineered Nerve Regeneration (신경재생을 위한 BDNF를 함유한 PLGA 지지체의 제조 및 방출)

  • We manufactured poly (L-lactide-co-glycolide) (PLGA) scaffolds impregnated demineralized bone particle (DBP) and hyaluronic acid (HA) by ice-particle leaching method and tested their ability of sustained release of brain derived neurotrophic factor (BDNF). BDNF (50 and 200 ng) mixed with PLGA, DBP/PLGA, HA/PLGA and DBP/HA/PLGA scaffold. The release profiles of BDNF from BDNF loaded scaffolds were assayed using ELISA. Morphological changes of scaffolds by BDNF release were also observed by SEM. BDNF stably and sustainedly released from DBP/HNPLGA than from PLGA and DBP/PLGA scaffolds. DBP/HA/PLGA scaffolds showed the great structural changes, which demonstrated BDNF release amount from DBP/HA/PLGA scaffolds were highest in all groups. We suggest that BDNF loaded DBP/HNPLGA scaffold would be very useful for nerve regeneration.
