Analysis of Gene Mutation and Expression Level of Follicle Stimulating Hormone Receptor in Premature Ovarian Failure(POE) Patients (조기 난소 부전증(Premature Ovarian Failure, POF) 환자에서 난포 자극 호르몬 수용체 유전자 변이 및 발현 양상에 대한 분석)

  • This study was investigated to analyze the inactivating point mutation and expression level of follicle-stimulating hormone(FSH) receptor mRNA. In first experiment, we analyzed the point mutation. Peripheral blood was collected from each patient. To screen individuals for the C566T mutation, PCR was performed for exon 7 of the FSH receptor gene in 10 patients. No inactivating point mutation of FSH receptor gene was identified in women with premature ovarian failure. To analyze the expression level of FSH receptor, mRNA expressions were examined by RT-PCR method using specific primers for the FSH receptor. The amount of FSH receptor mRNA expressed in POF patients was lower than that in the control group. But it was not significantly different. These finding suggests that lower expression of FSH receptor in premature ovarian failure patients might be the cause of the low response to the gonadotropin during the hyperstimulation in IVF-ET cycles.

  • Gonadotropin receptors are members of the seven-transmembrane (TM) receptor family, Point mutations in the lutropin/choriogonadotropin receptor (LH/CGR) have been shown to cause constitutive activation which results in precocious puberty in affected males, We introduced one of the mutation, D556Y, into the LH/CG receptor and the same high affinity binding mutant (D556Y) receptor clone cell for wild type LH/CGR (LH/CGR-wt) was chosen for further analysis, In contrast to cells expressing LH/CGR-wt, it was demonstrated that the mutant receptor exhibited markedly increased basal cAMP production in the absence of agonist, suggesting that autonomous Leydig cell activity in familial male-precocious puberty (FMPP) is caused by a constitutively activating LH/CGR.

Effects of Electrical Stimulation on Physicochemical and Rheological Properties of Plaice, Paralichthys olivaceus Muscle at Early Period after Death (전기자극이 넙치육의 사후조기의 물리화학적 및 물성적 변화에 미치는 영향)

  • This study was undertaken to clarify the effects of electrical stimulation on physicochemical and rheological properties of the plaice (Paralichthys olivaceus) muscle at early period after death. The plaices were electrically stimulated in seawater bath (110V/60Hz) for 15sec., 35sec., and 60sec. and killed instantly with spiking at the head. Killed samples were stored at $5^{\circ}C$, and the changes in rigor index, ATP breakdown, lactate accumulation, and breaking strength of muscle through storage were investigated. Electrical stimulation effectively accelerated rigor-mortis, lactate accumulation , and ATP breakdown. As the time of electrical stimulation was lengthened, the onset of rigor-mortis of all samples were accelerated Just after killing, and the amount of lactate was rapidly increased, But, significant differences were not observed in variance of rigor-mortis and lactate concentration. Electrically stimulated plaices showed decreasing in ATP to $4.58{\mu}mole/g$ for 15sec., $4.13{\mu}mole/g$ for 35sec., and $2.39{\mu}mole/g$ for 60sec. samples as compared with $5.5{\mu}mole/g$ of unstimulated samples. As the time of electrical stimulation was lengthened, ATP in samples were decomposed more rapidly. The rate constant of ATP breakdown were $0.244hr^{-1}$ for 15sec., $0.358hr^{-1}$ for 35sec., and $0.479hr^{-1}$ for 60sec.. The level of breaking strength in muscle of the plaice was $1050.30\pm50.23g$ immediately after killing. Values of breaking strength in samples electrically stimulated for 35sec. increased rapidly just after killing among all samples. However, the breaking strength was not increased through the whole storage time in samples stimulated for 60sec.. The value and time roaching to the maximum breaking strength for each samples stimulated electrically for 15, 35 and 60 second were $1264.43\pm35.76g$ and 2hr, $1357.68\pm22.50g$ and Ohr, and $1012.18\pm57.36g$ and Ohr. Breaking strength in all samples electrically stimulated decreased significantly (P<0.05) after reaching the maximum values.

  • The purpose of this study was to examine the effect of neuromuscular electrical stimulation(LFES) during hindlimb suspension on weight and function of rat hindlimb muscles. Sprague-Dawley rats(body weight 300-350g) were randomly assigned to five groups: a HLS(n=5) that were hindlimb suspended for 14 days, a WB(n=5) that kept as control, a ES14 that were hind limb suspended for 14days with pre-application of LFES for 14 days, ES11 that were hindlimb suspended for 14 days with pre-application of LFES for 11 days, a LFES 7 that were hindlimb suspended for 14 days with pre-application of LFES for 7 days. Gastrocnemius muscle weight, stride length were significantly decreased and toe out angle were significantly increased in HLS and ES7 groups, whereas muscle weight, stride length, toe out angle were maintained in ES14, ES11. this indicated that LFES for 14 days, LFES for 11 days could prevent muscle atrophy and retain function.

An Experimental Study on the Early Compressive Strength Improvement of Cement Mortar Mixed with Blast Furnace Slag using Powdered Stimulants (분말형 자극제를 이용한 고로슬래그미분말 혼입 시멘트 모르타르의 조기 압축강도 향상에 관한 실험적 연구)

  • Based on previous research and existing literature, this study examines the development of admixture, which increases the early concrete strength (1 and 3 day) by mixing blast furnace slag cement and concrete stimulant. The research on early strength development of concrete is necessary in dealing with the drawbacks of slow early strength concrete on site and to shorten the construction time. The study confirmed that when a high alkaline mortar mixture is mixed with blast furnace slag, the early strength of admixture exceeds that of ordinary portland cement (OPC). The use of calcium chloride ($CaCl_2$) promotes hydration of cement at low temperature and show similar strength as the blast furnace slag admixture. Although calcium chloride seems economically advantageous, it causes steel corrosion and its use in concrete should be further studied in-depth.

Effect of GnRH analogue on the bone mineral density of precocious or early pubertal girls (성조숙증 혹은 조기 사춘기가 있는 여아에서 단기간의 성선자극호르몬 방출호르몬 효능약제 사용이 골밀도에 미치는 영향)

  • Purpose:Treatment of precocity with gonadotropin releasing hormone analogue (GnRHa) might theoretically exert a detrimental effect on the bone mass during pubertal development. We investigated the short-term changes in bone mineral density (BMD) during GnRHa treatment and the enhancement in the changes with the co-administration of GnRHa and human growth hormone (hGH). Methods:Forty girls with precocious or early puberty who were using GnRHa for more than 1 year were enrolled. Of them, 14 concurrently received hGH. Lumbar bone mineral density was measured before and after the treatment, and bone mineral density-standard deviation scores (BMD-SDSs) were compared according to chronologic age (CA) and bone age (BA), as well as according to the administration of GnRHa alone (Group I) or the co-administration of hGH and GnRHa (Group II). Results:BMDs before and after treatment were in the normal range according to CA but were significantly lower according to BA (P<0.05). During treatment, BMD-SDSs did not change according to CA but significantly increased according to BA (P<0.05). BMD-SDSs in group I did not change during treatment according to CA or BA, while those in group II increased significantly according to BA (P<0.05), but not according to CA. Conclusion:Lumbar BMD was adequate according to CA at initial manifestation of precocity but was lower if compared to BA, that is, BMD did not increase with BA. Because co-treatment with hGH significantly increased BMD-SDSs according to BA, hGH co-treatment could be considered during GnRHa therapy.

An Experimental Study on Early Strength and Drying Shrinkage of High Strength Concrete Using High Volumes of Ground Granulated Blast-furnace Slag(GGBS) (고로슬래그 미분말을 대량 사용한 고강도 콘크리트의 조기강도 및 길이변화 특성에 관한 실험적 연구)

  • For high strength concrete of 40~60 MPa, the effects on the early strength and concrete dry shrinkage properties replacing 60~80% of Ordinary Portland Cement with Blast Furnace Slag Powder and using the Alkali Activator (Modified Alkali Sulfate type) are considered in this study. 1% Alkali Activator to the binder, cumulative heat of hydration for 72 hours was increased approximately 45%, indicating that heat of hydration contributes to the early strength of concrete, and the slump flow of concrete decreased slightly by 3.7~6.6%, and the 3- and 7- strength was increased by 8~12%, which that the Alkali Activator (Modified Alkali Sulfate type) is effective for ensuring the early strength when manufacturing High Strength Concrete (60%) of Blast Furnace Slag Powder. Furthermore, the dry shrinkage test, both 40 MPa and 60 MPa specimens had level of length changes in order of BS40 > BS60 > BS60A > BS80A, and the use of the Alkali Activator somewhat improved resistance to dry shrinkage.
