다슬기로부터 칼슘락테이트의 제조와 품질특성

  • 다슬기의 분말(PSB)과 그 회화분(ASB)으로부터 체내 흡수력이 높은 젖산칼슘의 제조조건과 색상, 용해도 및 관능적 품질을 조사하였다. PSB로부터 제조한 젖산칼슘(PSB-CL)의 PSB 및 젖산 100 mL에 대한 수율은 젖산농도가 10%일 때는 300% 및 15 g, 20%일 때는 260% 및 20 g이었으며, ASB로 제조한 젖산칼슘(ASB-CL)의 ASB 및 젖산 100 mL에 대한 수율은 10% 젖산에서는 400% 및 60 g, 20% 젖산에서는 329% 및 66 g으로 원료량을 기준으로 하였을 때는 다같이 젖산농도의 증가에 따라 감소하였으나 젖산의 부피를 기준으로 하였을 때는 젖산농도의 증가에 따라 증가하였다. Dehydated PSB-CL 및 ASB-CL 제조의 적정온도와 시간은 10$0^{\circ}C$에서는 각각 4 및 5시간, 12$0^{\circ}C$에서는 3 및 4시간, 15$0^{\circ}C$에서는 1 및 2시간으로 ASB-LA의 경우가 짧았다. IR 및 H-NMR spectrum의 분석결과 PBS-LA와 ASB-LA의 구조는 Ca($CH_3$CHOH$CO_2$)$_2$임이 확인되었다. 무수 PSB-CL 및 ASB-CL의 칼슘함량은 각각 15.4%(w/w)와 17.3%(w/w)로 이론 값의 각각 84.2%와 94.5%를 나타내었으며, 미량의 Fe, Na, Mn Zn을 함유하였다. PBS-CL와 ASB-CL의 색상은 각각 연한 황색과 연녹색을 지닌 백색을 띠었다. pH 3~8로 조정한 증류수에서 PSB-CL과 ASB-CL의 평균 용해도는 각각 5.43 g/100 mL 및 6.11 g/100 mL로 standard CL의 4.74 g/mL에 비하여 높았다. 국간장을 제외한 대부분의 액체식품(3% 소금물, 소주, 진간장, 국간장, 포도주스 및 오렌지주스)에서의 용해도는 PSB-CL(3.14~5.03 g/100 mL)과 ASB-CL(4.69~6.05 g/100 mL)이 standard CL(2.94~5.84 g/100 mL)에 비하여 높았다. 관능검사 결과, ASB-CL은 신맛이 낮아 사용범위가 높은 것으로 평가되었으며 PSB-CL는 쓴맛은 높으나 떫은맛이 낮고 구수한 맛이 강하여 식품에의 응용이 기대된다.

Partitioning of Lactobacillus helveticus Cells and Lactic Acid in Aqueous PEI/HEC Two-Phase Systems. (수용성 이상계에서의 젖산과 Lactobacillus helveticus세포의 분배특성)

  • For an ideal extractive bioconversion in aqueous two-phase systems, the product has to be preferentially partitioned into the phase opposite to the one in which the biocatalyst is located. Partitioning behaviors of Lactobacillus helveticus IAM 11090 and lactic acid in aqueous two-phase systems composed a polycation, poly(ethylenimine) (PEI), and an uncharged polymer (hydroxyethyl)cellulose (HEC) were investigated. L. helveticus cells were preferentially partitioned to the HEC-rich top phase while about 85% of lactic acid was partitioned to the PEI-rich bottom phase. These results indicate that extraction of charged, low molecular weight products in an aqueous two-phase systems can be promoted by using an oppositely charged polymer as one of the phase-forming polymer. By the ideal partitioning of the cells and lactic acid, an aqueous PEI/HEC two-phase system can be used as a potential system for the extractive lactic acid fermentation of cheese whey.

Effects of Sports Drink Including the Extract from Prunus mume on the Changes of Respiratory Variables, Heart Rate, and Blood Lactate Concentration in Submaximal Exercise (매실함유 음료섭취가 장시간 운동시 심박수, 호흡가스 변인 및 혈중 젖산농도 변화에 미치는 영향)

  • This study was designed to investigate the effects of sports drink including the extract from Prunus mume on the changes of oxygen uptake, minute ventilaton, heart rate, and blood lactate concentration during 1 hour treadmill running exercise corresponding to 75%VO$_2$max. Subjects were 16 male athletes(long distance runners and tennis players). The changing patterns of oxygen uptake and minute ventilation showed no significnat difference among all types of sports drink, but the minute vetilation of the placebo group showed a higher value than type I group during submaximal exercise. Although the changing patterns of heart rate and blood lactate concentration showed no significnat difference among all types of sports drink, but type E group showed a lower heart rate compared to Placebo group. And blood lactate concentration of placebo group showed a higher value compared to the value corresponding to lactate threshold during submaimal exercise. but the other types of sports drink showed a lower value of blood lactate concentration. Especially blood lactate threshold of type D and E groups showed a lower value (range from 1.44 to 2.00mM) during submaximal exercise. In these results, the sports drink including the extract from Prunus mume showed no significant effects to the changes of oxygen uptake, minute ventilaton, heart rate, and blood lactate concentration during 1 hour treadmill runnuing exercise corresponding to 75%VO$_2$max. But it could be suggested the positive effects of the intake of sports drink including the extract from Prunus mume on the inhibition of exercise-induced fatigue during submaximal exercise, because of the lower changing Patterns of heart rate, blood lactate concentration, and ventilation efficiency in this results.

Quality Characteristics of Rice Wine according to the Rice Wine Seed Mash with Lactic Acid Concentration (젖산농도별 주모에 따른 막걸리의 품질 특성)

  • The quality characteristics of rice wine seed mash and rice wine made with different lactic acid concentrations were investigated. The pH decreased along with the lactic acid concentration. The total titratable acid content of the rice wine seed mash was lowest when 0.5% lactic acid was added, and the ethanol contents of the mash samples were not significant. The results of the measurement of the microorganism number, C (cell numbers of the total bacteria and the lactic acid bacteria), decreased along with the lactic acid concentrations of the mash samples. The yeast cell numbers of the rice wine seed mash samples according to the lactic acid concentrations were high (0.5, 0.3, 1.0, control, and 0.7%, respectively). The pH and total titratable acid levels of rice wine according to the lactic acid concentration were stable during fermentation, according to the increase in the amount of lactic acid. The organic acids in the rice wines were highest in the lactic acid. The rice wines to which lactic acid had been added had lower acetic acid contents than the control. Also, the acetic acid contents decreased along with the lactic acid concentrations, except in the rice wine treated with 1.0% lactic acid. The ethanol contents of the tested rice wines were not significant. All in all, in the sensory evaluation, the rice wines treated with 0.5 and 0.7% lactic acid scored higher than the other treatments.

  • Accumulation of toxic waste byproducts in hybridoma culture can limit cell growth and monoclonal antibody (MAb) productivity, and one of the major toxic metabolites is lactate produced via glycolytic pathway of glucose metabolism. The factors affecting the glucose to lactate conversion rate were investigated. The conditions of high initial glucose concentration and high growth rate stimulated glucose to lactate conversion rate. The glucose-controlled fed-batch culture was investigated, and 19% reduction in lactate formation and 41% enhancement of MAb titer could be achieved by fed-batch culture.

Isolation of an acid-tolerant Leuconostoc mesenteroides LM3 from Kimchi (김치에서 산내성을 가진 Leuconostoc mesenteroides LM3의 분리)

  • In order to understand stress response of Leuconostoc mesenteroides against lactic acid, a new Leuconostoc sp. which has acid tolerance was isolated from various Kimchi samples. And it identified as Leuconostoc mesenteroides LM3 by comparing its fatty acid composition with reference strain. Its growth pattern was investigated by adding a given concentration of lactic acid at the lag phase to the stationary phase. In the DeMan, Rogosa, Sharpe (MRS) media containing over 0.4% (final v/v) lactic acid, this strain slowed slowly After exposure of the stationary phase cells to 4% of lactic acid for 60 min, this strain could survive, whereas a reference strain, Leuconostoc mesenteroides KCTC3505, showed no survival. And changes of trehalose concentration, the activity of trehalase and ATPase in the growth of Leuconostoc mesenteroides LM3 after addition of 0.6% (final v/v) lactic acid were investigated : After exposure to lartic acid, trehalose concentration in this strain was increased in comparison with no treatment, but its trehalase activity was not changed. And its ATPase activity was constant, and intracellular pH was almost constant. This result meant Leuconostoc mesenteroides LM3 should have a tolerance against lactic acid. It remains to further study the mechanism of this acid tolerance.

The Effect of Kinesio Taping and Icing on Blood Lactate Density and Heart Rate Recover Pace After Shuttle Run Test (키네시오 테이핑과 아이싱 처치가 셔틀런 테스트 후 혈중 젖산 농도 및 심박수의 회복속도에 미치는 영향)

  • This study will be targeting 21-24 years old college students majoring in physical education. This study covers the effect of kinesiology taping and icing treatment on the recovery rate of blood lactate concentration, and cardiovascular function so that it can provide the basic data for improving physical performance. As a result of this study, Kinesio taping and icing aid the recovery rate of the maximum load of lactic acid recovery rate and 30 minutes after exercise heart rate after exercise, but increased, after 15 minutes, immediately after exercise heart rate, exercise lactate and heart rate, and 30 minutes after exercise lactate showed no difference. In conclusion, Kinesio taping and icing treatment, reduce pain and fatigue but it is not satisfactory. Further study, applying a more accurate and technical form of exercise programs needs to continuously research the effects of Kinesio taping and icing treatment

Enhanced Resistance to Lactic Acid by Laboratory Adaptive Evolution of Saccharomycopsis fibuligera (실험실 적응진화를 이용한 Saccharomycopsis fibuligera의 젖산에 대한 내성 증대)

  • Saccharomycopsis fibuligera is an amylolytic yeast that exhibits raw starch-degrading activity. In this study, adaptive laboratory evolution was performed to improve the tolerance of S. fibuligera to lactic acid by prolonged repeated batch fermentation in which the lactic acid concentration was gradually increased. The evolved S. fibuligera strain exhibited a significantly enhanced tolerance to lactic acid at concentrations up to 2.5% (w/v), as assessed by determining its specific growth rate using a plate assay. Scanning electron microscopy revealed an elongated and perforated morphology of the parent strain under lactic acid stress, indicating that its membrane might be more prone to damage caused by lactic acid than that of the evolved strain.

A Comparison of Cardiopulmonary Function, RPE, and Blood Lactate following in Wheelchair Treadmill and Arm Ergometer GXT Test through Convergence (융복합을 활용한 휠체어 트레드밀과 암에르고미터 점증부하운동검사 시 심폐기능, 운동자각도 및 젖산농도 비교)

  • The purpose of this study was to compare cardiopulmonary function, RPE(Rating of perceived exertion), and blood lactate when examining the GXT(Graded exercise testing) of wheelchair treadmill and arm ergometer. Participants were 11 wheelchair basketball players with non disability. While examining the GXT of wheelchair treadmill and arm ergometer, cardiorespiratory functional capacity was measured by using $Quarkb^2$ and Polar and RPE was measured through Borg Scale. The lactate analyser, YSI-2000 was used to measure blood lactate level when resting, right after exercise, two minutes, four minutes, six minutes, and ten minutes of recovery. Data was analyzed by paired t-test using SPSS 18.0 program and significance for all statistical analysis was fixed at .05 confidence level(p<0.05). The conclusion of this study is below. First, maximal oxygen uptake which is a factor of cardiopulmonary function showed the highest with arm ergometer, the rate of respiratory exchange showed the highest with wheelchair treadmill, and maximal heart rate showed the highest with wheelchair treadmill. Second, subjective exercise intensity showed the highest with arm ergometer at the end point. Third, blood lactate level showed the highest with arm ergometer right after exercise.

The Review of Metabolic Acidosis During Exercise (운동 시 대사적 산성화에 관한 고찰)

  • The development of acidosis during intense exercise has traditionally been explained by the increased production of lactic acid which causes the release of a proton and the formation of the acid salt sodium lactate. Through this explanation, when the rate of lactate production is high enough to exceed cellular proton buffering capacity, cellular pH is decreased. This biochemical process has been termed lactic acidosis. This belief has been an interpretation that lactate production causes acidosis and fatigue during intense exercise. However, this review provides clear evidence that there is no biochemical support for lactate production causing acidosis and fatigue. Metabolic acidosis is caused by an increased reliance on nonmitochondrial ATP turnover. Lactate production is essential for muscle to produce cytosolic $NAD^+$ to support continued ATP regeneration from glycolysis. In addition, Lactate production consumes protons. Although lactate accumulation can be a good indirect indicator for decreased cellular and blood pH, that is not direct causing acidosis.
