Immobilization of Trichoderma harzianum ATCC52445 into Natural Matrix (천연 담체를 이용한 Trichoderma harzianum ATCC52445의 고정화)

  • Spores of Trichoderma harzianum TCC 52445 for control of Rhlzocronia stem canker on potato were immobilized in various natural matrix, and germination rate and some rheological properties of the spore-matrix complex were Investigated. Germination rate of gelatin gel-spore complex and potato starch gel-spore complex were 2.8% and 2.9%, respectively, but hardness and cohesiveness of the gelstin gel-spoke complex were better than those of potato starch-spore complex. The hardness and cohesiveness were increased when the spores were immobilized in hybrid gelatin gel made by mixing several different types of natural matrix, but decreased their germination rate. Addition of corn steep liquor (1%) as spore nutrient in gelatin gel-spore complex was helpful for increasing the germination rate.

Industrial Application for Aquaculture of Fermented Soybean with Bacillus polyfermenticus SCD (양식업에서 바실러스 폴리퍼멘티쿠스 콩발효물의 상업적 응용)

  • Bacillus polyfermenticus SCD (B. polyfermenticus SCD) has been appropriately used for the therapy of long-term intestinal disorders, because live strains in the form of active endospores can successfully reach the target intestine in humans. B. polyfermenticus SCD produces the most antibiotic-related materials. In the present study, B. polyfermenticus SCD was fermented with soybeans (BPFS) and its probiotic properties were investigated. B. polyfermenticus SCD and BPFS showed a broad spectrum of antimicrobial activity against pathogenic Gram-positive (Streptococcus parauberis, Streptococcus iniae, Lactococcus garviae) and Gram-negative (Flexibacter tractuosus, Vibrio harveyi, Vibrio vulnificus, Vibrio ordalii) bacteria and moulds (Aspergillus niger, Aspergillus oryzae). Sebastes schlegeli were used to examine survival rate and cleanup action by BPFS. Bacterial infection resulted in a mortality of up to 99% in the commercial fodder fed groups. BPFS both enhanced the growth rate of fry by improving their appetite and had cleaned by decreased eutrophication. Therefore, it seems appropriate that BPFS should be developed as an antibiotic replacement, favorable fodder additive, and antifungal material in fish farming systems.

  • This study focuses on investigating roles of microorganisms in decontamination of reed rhizosphere in Sunchon Bay, Korea, which is considered one of the marsh and mud environment severely affected by human activities such as agriculture and fisheries. In general, the bay is known to play the role of the buffering zone to reduce the sudden impact or change by environmental stresses. In our initial efforts to elucidate the microbial functions in decontamination process in reed rhizosphere, pure bacteria capable of degrading aromatic hydrocarbons were isolated from reed (Phragmites communis) rhizosphere of Sunchon bay by enrichment culture using either benzene, toluene, ethylbenzene, or xylene (BTEX) as a sole source of carbon and energy. Measurement of the rates of BTEX degradation and cell growth during the incubation in BTEX media under several temperature conditions demonstrated maximized degradation of BTEX at $37^{\circ}C$ in both strains. Both strains were also resistant to all the heavy metals and antibiotics tested in this study, as well as they grew well at $42^{\circ}C$. Identification of the isolates based on 16S rRNA gene sequences, and a variety of phenotypic and morphologic properties revealed that the two strains capable of BTEX catabolism were among Microbacterium sp., and Rhodococcus sp. with over $95{\%}$ confidence, designated Microbacterium sp. EMB-1 and Rhodococcus sp. EMB-2, respectively This result suggested that in the rhizosphere of reed, one of major salt marsh plants they might play an important roles in decontamination process of reed rhizosphere contaminated with petroleum such as BTEX.

Production of air purification verification system using moss (이끼를 활용한 공기정화 검증 시스템 제작)

  • Fine dust aerated in the atomsphere penetrates our lungs and blood lines through respiratory. Recent fine dust problems in Korea leads to development of various air purifiers. The researchers used this to study systems that could replace chemical filters. In order to compare the effect of the reduction of moss and conventional chemical filter(Hepa), a 1 cubic meter cube was prepared and the amount of the concentration of fine dust reduction was compared under controlled environment. Under the high concentration of fine dust, a test was done to figure out the reduction rate of the fine dust concentration by using air purification system with moss, hepa, and no filter. The air purification system(moss, hepa, and no filter) were operated 90 times in total, 30 times each. The test explains that the reduction of the fine dust amount and the rate of fine dust concentration. The results illustrate that the reduction of the amount fine dust was 138.93 after using air purification system with moss filter. In contrast, the usage of air purification system with hepa filter reduced the amount of fine dust to 76.57. And the air purification with no filter shows that the slight reduction of fine dust amount at 0.10. In the rate of fine dust concentration, moss filter was significantly higher than that of hepa, no filter (0.2379, 0.1298 and 0.0063 each). The results have confirmed that moss is effective in reducing fine dust concentration, and it is expected that with further improvement it can be used as a means to replace or supplement existing chemical filters in air purifier.

Catalytic Oxidation of Volatile Organic Compounds Over Spent Three-Way Catalysts (배기가스 정화용 폐 자동차 촉매를 이용한 휘발성 유기화합물의 제거)

  • The optimum regeneration conditions for the regeneration of three way spent catalysts (TWCs), which were taken from automobiles with different driving conditions, were investigated to evaluate the suitability as alternative catalysts for removing VOCs. The spent catalysts were washed with five different acids ($HNO_3$, $H_2SO_4$, $C_2H_2O_4$, $C_6H_8O_7$, and $H_3PO_4$) to remove contaminants and examine the optimum conditions for recovering the catalytic activity. The physicochemical properties of spent and its regenerated TWCs were evaluated by using nitrogen adsorption-desorption isotherms, XRD, and ICP. The relative atomic ratios of contaminants and platinum group metals (PGMs) of the spent TWCs were greatly dependent on the placed positions. The main contaminants formed were lubricant oil additives and metallic components. Also, the regeneration treatment increased the PGMs ratio, BET surface area, and average pore diameter of TWCs. The catalytic activity results indicated that the spent TWCs have the possibility for removing VOCs. Moreover, the employed acid treatments greatly enhanced the catalytic activity of the spent TWCs. Especially, nitric and oxalic acids provided the most improvement in the catalytic behavior. The catalytic activities of the regenerated TWCs were significantly influenced by the containing platinum ratios rather than the removal ratios of contaminants and the changes in the structural properties offered by the acid treatments.

The Effects of Cadmium on Seed Germination and Growth of Sunflower and Rape (카드뮴이 해바라기와 유채 발아 및 성장에 미치는 영향)

  • Sunflower (Sunking4505) and Rape (Sunmang) are oil-seeds containing high oleic acid, and these are used for the production materials of bio-diesel and applying for phytoremediation. The effect of cadmium on germination rate and the growth of the plants is evaluated. Object seeds were placed in Cd (0, 1.5, 4, 12, 30, 60, 100, 150, 300, 500 mg/L) solutions for seven days, and germination rate, root length, shoot length, seedling length, and dry weight were observed. $IC_{50}$, seedling vigor index, and tolerance indices were computed, and data were statistically analyzed by Analysis of Variance (ANOVA). Germination rate as well as root, shoot, and seedling length decreased as the cadmium concentration increased except dry weight. The $IC_{50}$ of sunflower and rape are 112 and 10 mg-Cd/L, respectively. Only one of the sunflower seeds is germinated at 500 mg-Cd/L whereas rape seeds are not germinated more than 150 mg-Cd/L solution. Root has higher cadmium sensitivity than shoot, and sunflower has higher germination rate, growth, and seedling vigor index than rape. In case of tolerance indices, sunflower has lower value than rape at relatively low concentration, but has higher value at high concentration.

Particulate Matter Removal of Indoor Plants, Dieffenbachia amoena 'Marianne' and Spathiphyllum spp. according to Light Intensity (광량에 따른 실내식물 디펜바키아와 스파티필럼의 미세먼지 제거능)

  • This study investigated the effect of light intensity on the removal of particulate matter by Dieffenbachia amoena 'Marianne' and Spathiphyllum spp.. An acrylic chamber ($600{\times}800{\times}1200mm$, $L{\times}W{\times}H$) modeled as an indoor space and a green bio-filter ($495{\times}495{\times}1000mm$, $L{\times}W{\times}H$) as an air purification device were made of acrylic. The removal of particulate matter PM10 and PM1, the photosynthetic rate, stomatal conductance, and number of stomata of Dieffenbachia amoena 'Marianne' and Spathiphyllum spp. were measured according to three different levels of light intensity (0, 30 and $60{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}PPFD$). Regarding the length of time taken for PM10 to reach $1{\mu}g$, the Dieffenbachia amoena 'Marianne' showed a significant difference according to the presence or absence of light, and there was no significant difference shown between light intensity of 30 and $60{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}PPFD$. As for the Spathiphyllum spp., there was no significant difference between 0 and $30{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}PPFD$, while a significant difference was shown at $60{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}PPFD$. After 90 minutes, the PM1, PM10, and $CO_2$ residuals of the Spathiphyllum spp. were lowest at $60{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}PPFD$. The remaining amount of PM1 and PM10 was lower with the Spathiphyllum spp. than with the Dieffenbachia amoena 'Marianne', even at $0{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}PPFD$. With both plants, the higher the light intensity, the higher the photosynthetic rate, while the stomatal conductance did not show any significant difference. Spathiphyllum spp. showed a higher photosynthetic rate and stomatal conductance and a greater number of stomata than Dieffenbachia amoena 'Marianne', and stomata were observed in both the front and back sides of the leaves. The air purification effect of Spathiphyllum spp. is considered to be better than Dieffenbachia amoena 'Marianne' at the same light intensity due to such plant characteristics. Therefore, in order to select effective indoor plants for the removal of particulate contamination in an indoor space, the characteristics of plants such as the photosynthetic rate and the number and arrangement of stomata according to indoor light intensity should be considered.

Phytoremediation of diesel-contaminated soils using alfalfa (Alfalfa를 이용한 디젤오염토양의 phytoremediation)

  • In the past several years phytoremediation, defined as the use of plants for removing contaminants from media such as soils or water, has attracted a great deal of interest as a potentially useful remediation technology We attempted to assess the effectiveness of phytoremediation of diesel-contaminated soils in a green house. Screening test for selecting an appropriate plant was performed by observing the harmful effects of diesel dosage on the growth of 4 plants. Alfalfa was selected as a potentially useful plant among corn and barnyard grasses due to its high tolerance to the toxicity of diesel in growth. Bioremediation of the artificial diesel-contaminated soil packed in the PVC columns(0.3m in diameter $\times$ 1m in length) with air supplied, alfalfa planted, and alfalfa and air supplied was investigated for 100 days. The results of the column test showed plant effects on enhancing the biodegradation of diesel in the contaminated soils compared to the control column which had no plant. Injecting air to the columns during phytoremediation also showed additional effects on the removal rate of diesel. Comparison of microbial activity in each test column showed a beneficial effect of plants in the soil remediation processes. This results can be explained microbial activity in rhizosphere is a crucial factor for removing diesel.

Sand Elimination and Microbial Depuration in Surf clam, Mactra veneriformis, Harvested from Western Coast of Korea (서해산 동죽 (Mactra veneriformis)의 토사 배출 및 미생물 정화 조건)

  • Surf clam, Mactra veneriformis, is a important shellfish produced in south-western coast of Korea. But it's ready to be contaminated and have sand in flesh because it mainly inhabit in silt at tHe inside of coastal area. We determined optimum conditions for the sand elimination and microbial depuration in the shellfish harvested from western coast of Korea. It was found that the most effective conditions of process water for the sand elimination were $23^{\circ}C$, $32.9-35.0\%_{\circ}$ salinity and pH $7.9\~9.0.\;A$ surf clam contained about 210 mg of sand whose $94\%$ was eliminated after 24 hours in natural sea water ($32.9\%_{\circ}$ , pH 7.9) controlled at $23^{\circ}C$. To eliminate both sand and microorganisms contaminated in surf clam, the process water should be run during at least 36 hours for the former and 24 hours for the latter at 150 L/minute/$m^3$of shellfish, when its volume was above 4,000 L/$m^3$ of shellfish in 2 tons of tank.

The Effect of Salinity, Temperature and Diet on Larval Growth and Survival of Metapenaeopsis dalei (Rathbun) (Decapoda: Penaeidae) (산모양깔깔새우 (Metapenaeopsis dalei) 유생의 염분, 수온 및 먹이조건에 따른 성장 및 생존율)

  • Larvae of Metapenaeopsis dalei (Rathbun) were successfully spawned and reared in the laboratory under controlled conditions. The experiments were designed to examine effects of salinity (20, 25, 30 and 35), temperature (21, 24 and $27^{\circ}C$ ) and food (five items) on larval development, survival and growth, Salinity affected larval survival, from $\%$ at 20, to $60\%$ at 35 (protozoea 1 to first postlarva), The highest survival rate was obtained at $35\%_{\circ}$. Temperature affected larval survival, from $46\%$ at $27^{\circ}C$ to $54\%$ at $21^{\circ}C$(protozoea 1 to first postlarva). The highest survival rate was obtained at $24^{\circ}C$. Mortality was the highest from protozoea 1 to protozoea 3, but decreased considerably for all temperatures once the mysis stage was reached. Larval growth was affected by different of food items. Phytoplankton (Isochrysis galbana) was the optimum food for the larval survival and the growth from protozoea 3 to first postlarva.
