The search for tumor-avid agents for use in nuclear medicine imaging or therapy is a field of ongoing importance. Metallothionein (MT) is an intracellular protein that binds many metals with isotopes having imaging or radiotherapeutic potential. The purpose of the study was to determine whether uptake of radioisotopes that bind to MT is increased in tumor. We measured the uptake of Cd-109 and Ga-67 in tumor and normal tissues of sarcoma-bearing mice. Tumors were grown subcutaneously in female Balb/C mice from cultured Balb/3T3 cells transformed by the Moloney murine sarcoma virus (MMSV). When the tumors reached about 1 cm in diameter, mice were injected subcutaneously with Cd-109 and Ga-67. Eighteen and seventy-two hours later, the mice were sacrified. Organs and tissues were removed, weighed, and activity per mg tissue determined by gamma well-counting. Uptake of Cd-109 by MMSV tumors exceeded that by normal tissues examined, with the exception of liver and kidney (the organs known to be richest in MT). The tumor-to-tissue ratios of uptake for Cd-109 were far greater than those for Ga-67 for many normal tissues of great importance in terms of background activity (bone, intestine, fat, muscle, and blood). We concluded that metals that bind to MT may be useful for oncologic imaging or rediotherapy of cancer.

We prepared 2 different crude polysaccharides from persimmon leaves, by hot water (PLW-0) and pectinase digestion (PLE-0), respectively. PLW-0 and PLE-0 showed similar sugar compositions and contained 11 different sugars, including rarely observed sugars such as 2-methyl-fucose, 2-methyl-xylose, apiose, and aceric acid. However, the uronic acid content of PLE-0 was lower than that of PLW-0, because of pectinase treatment. In normal mice, administration of PLW-0 and PLE-0 increased the spleen index and splenocyte proliferation. The effect of PLE-0 on the spleen index and splenocyte proliferation was greater than that of PLW-0. We subsequently assessed the immunomodulatory activities of PLW-0 and PLE-0 on cyclophosphamide (CY)-induced immunosuppressed mice. We revealed that mice treated with PLW- 0 or PLE-0 showed increased splenocyte proliferation, NK cell activity, and white blood cell numbers. Taken together, our results indicate that PLW-0 and PLE-0 can enhance immune function in normal mice and modulate CY-induced immune suppression.
Benzo(a)pyrene(B(a)P), an extensively studied polycyclic aromatic hydrocarbon(PAH), is a common contaminant produced through the burning of fossil fuels, particularly coal, and from the exhaust products of internal combustion engines. It produces a wide range of toxicities, including carcinogenicity in experimental animals. B(a)P has been shown to suppress systemic immunity in experimental animals, which may contribute to the growth of the chemical-induced tumors. Using colorimetric MTT assay natural killer(NK) cell-mediated growth inhibition of tomor cell was measured in normal and B(a)P-exposed C57BL/6 mice. Non-adherent splenocytes of normal or B(a)P-exposed mice were cultured with Yac-1 cells at four different effector/target(E/T) cell ratios ranging from 200/1, 100/1, 50/1, and 25/1 in an assay volume of 0.1 ml. After the optical density of culture wells containing MTT solution was measured at a wavelength of 540 nm, the percentage of dead cells relative to the control target cell number was calculated. The NK activity of B(a)P-exposed mice was markedly lower than that of non-exposed mice group at all E/T ratios. These results indicated that suppression of NK cell activity may play a role in allowing for the growth of tumors.

The aim of this study was to evaluate the inhibitory effect of nude pack containing black tea water extract (NPBT) on skin wrinkle formation in hairless mice. Skin wrinkles were induced by UVB irradiation to the backs of hairless mice for 5 weeks. And at the same time, NPBT was applied topically. Wrinkle formation, histological changes, expression of matrix metalloproteinase-3 (MMP-3) and protein activities of MMP-2 and MMP-9 were observed or analyzed. Wrinkles for the control group were formed as a pattern of deep furrows and thick crests. Whereas wrinkles for the NPBT treated group were formed as a pattern of shallow furrows and thin crests, and their wrinkle areas were significantly (p<0.001) lower than the control group. Collagen fibers were arranged irregularly and sparse in density and some elastic fibers were degenerated in the control group, while they were almost intact in the NPBT treated group. MMP-3 mRNA expression in the control group was significantly (p<0.001) higher than the normal group, and that of NPBT treated group was significantly (p<0.001) lower than the control group. The NPBT treated group showed remarkably lower protein activities of MMP-2 and MMP-9 than the control group. NPBT could have a considerable inhibitory effect on skin wrinkle formation in hairless mice.
To investigate the effect of the squalene against the lead toxicity. A healthy male of ICR mice were used for experiment. The SOD was observed after the intraperitoneal injection in mice. The ultrastructural changes of the liver were observed after the intraperitoneal injection in mice. The experimental groups were divided into two groups. Group A was control group that squalene was not treated after intraperitoneal injection of lead acetate. Group B was squalene treatment group that squalene solution was injected after intraperitoneal injection of lead acetate. All groups were used to 10 mice. The results were as follow: SOD activity in the liver, Group A was lower than in normal. But, Group B was higher than in Group A (P<0.05). In the histological observation, Group A were showed that the inner cavity of mitochondria swellen and development of cristae weakened. Swelling of lamellae of rough endoplasmic reticulum (rER) was showed. It was concluded that the SQ will be effective for the recovery of hepatic cell at lead intoxication.
In this study, we investigated the effect of water, extract of Terminalia Chebula (TC) on physiological activity in mice. TC water extract showed hemagglutination against several different types of red blood cells. $LD_{50}$ of TC extract was 390 mg/kg (po). Treatment of TC water extract orally administered 200, 300 mg/kg daily for one week. Hepatic cytosolic enzymes, xanthine oxidase and aldehyde oxidase activities were significantly increased comparison with normal group. Treatment of TC water extract increased hepatic malondialdehyde (MDA) formation, and reduced glutathione content. We also found that the decreased activities of glutathione S-transferase and glutathione reductase but was not affected activities of $\gamma$-glutamylcysteine synthetase after treatment of TC water extract. These results suggested that increase of the hepatic lipid peroxide is caused by glutathione reduction.
Although the adenosine triphosphate (ATP) efflux pathway is thought to play a major regulatory role in RVD in some cells, including cholangiocarcinoma cells, the role of ATP in regulatory volume decrease (RVD) of normal cholangiocytes is not well defined. Thus, this study was conducted to investigate the role of extra cellular ATP and ATP pathways of BDCCs isolated from normal mice. Changes in cell volume of BDCCs were indirectly assessed by measurement of the cross-sectional area (CSA) by quantitative videomicroscopy. The relative CSA of BDCCs from normal mice increased with hypotonic maneuver to $1.20{\pm}0.02$ (n=20) within 10 min, but decreased to $1.06{\pm}0.03$ at 40 min. Administration of ATP, ATP hydrolase apyrase or the P2 receptor blocker suramin during RVD had no significant effects compared with untreated controls. In addition, treatment with the PKC inhibitors, Bisindolamide I and Ro 31-8220, during RVD had no significant effects when compared with untreated controls. These results indicate that unlike the results from cholangiocarcinoma cells, ATP plays no significant role in the RVD of normal mouse cholangiocytes.

This study was conducted to investigate effects of fructans (chicory inulin, fructooligosaccharide and chicory inulin oligosaccharide) on blood glucose, activities of disaccharidases in small bowel and kidneys, and splenocyte proliferation in streptozotocin-induced diabetic mice. Sixty ICR male mice were divided into one normal group and four diabetic groups. Diabetes was induced by injecting streptozotocin after 2 weeks of experimental diets feeding. Experimental diets based on AIN93G diet were control diet, 6$ \%$ fructooligosaccharide (FOS) diet, 6$\%$ chicory inulin oligosaccharide (CIOS) diet, 6$\%$ chicory inulin (Cl) diet, and given for 25 days after streptozotocin injection. Plasma glucose was lower in Diabetic-Cl group as compared to Diabetic-control group. Plasma insulin level was not different among diabetic groups. Specific activities of jejunal maltase and sucrase in diabetic groups were about double as that of Normal group. Jejunal maltase activity and plasma glucose were positively correlated (r=0.643). However, specific activity of renal maltase in diabetic groups was not significantly different as compared to Normal group. Stimulation index of splenocyte proliferation by lipopolysaccharide (LPS) was significantly increased in Diabetic-CIOS as compared to Diabetic-control. Stimulation index of splenocyte proliferation by Concanavalin A (ConA) tended to be higher in Diabetic-CIOS group. Concentrations of interleukin-2 and interferon- $\gamma$ secreted from splenocytes induced by ConA were not significantly different among all groups. In conclusion, fructans may be effective for lowering plasma glucose, possibly by lowering disaccharidase activity and for increasing immune responses in diabetic con-ditions, where their effects can be different depending on degree of polymerization.
In addition to tumors, normal tissues, such as the brain and myocardium can intake $^{18}F$-FDG, and the amount of $^{18}F$-FDG intake by normal tissues can be altered by the surrounding environment. Therefore, a process is necessary during which the contrasts of the tumor and normal tissues can be enhanced. Thus, this study examines the effects of glucose levels on FDG PET images of brain tissues, which features high glucose activity at all times, in small animals. Micro PET scan was performed on fourteen mice after injecting $^{18}F$-FDG. The images were compared in relation to fasting. The findings showed that the mean SUV value w as 0.84 higher in fasted mice than in non-fasted mice. During observation, the images from non-fasted mice showed high accumulation in organs other than the brain with increased surrounding noise. In addition, compared to the non-fasted mice, the fasted mice showed higher early intake and curve increase. The findings of this study suggest that fasting is important in assessing brain functions in brain PET using $^{18}F$-FDG. Additional studies to investigate whether caffeine levels and other preprocessing items have an impact on the acquired images would contribute to reducing radiation exposure in patients.
Effects of water extracts of antler on blood sugar levels were studied in normal rats and KK-mice. a spontaneously diabetic strain of murine. Blood sugar levels were determined at 30, 60, and 120 minutes after intraperitoneal administration of 1 ml/kg or 2ml/kg AE into normal rats. A moderate but significant decrease in blood sugar level was noticed only in 30 minutes following 2 ml/kg AE administration. On the other hand no significant changes in blood sugar level was observed in KK-mice to which 1ml/kg/day AE had been given for 10 weeks. The results of present study strongly indicate that antler extract exerts no significant effect on blood sugar levels in experimental animals.
