Pure Dyestuff Extract from Polygonum tinctoria (천연 쪽의 순수 염료 개발)

  • In order to make pure dyestuff extract from polygonum tinctoria, we tried to traditionally-using cockle shell which the extracted water should be filtered in indigo for 8 hours and at the $40^{\circ}C$, and 4.0 pH. The 1.631g of powder dye could be produced when the 10.0 ml of aqueous ammonia was added into the solution, which had been filtered for 8 hours with the 300 g of indigo plant and the 2.51 of water. The main components of two maded -dye which has traditionally made of this and has purely made of that compared by TLC, HPLC, LC/MC techniques. Finally, the antibacterial activities and deodorization ratio of silk fabrics with natural indigo were carried out, too.

Study on Preparation of Environmental-Friendly Specialty Paper Using Functional Antibiotic Nano-Particle (II) (기능성 항균 나노입자를 이용한 친환경성 특수지 제조에 관한 연구(II))

  • For the purpose of antibacterial and photocatalystic deodorization functions, the papermaking inorganic fillers and pigments were surface-modified with Ag nano-colloidal solution and $TiO_2$ by using the hybridization technique. The functional specialty sheets and coating papers were produced with the surface-modified fillers and pigments, and evaluated by halo test and inhibition growth test in their antibacterial and photocatalystic characteristics. For the application of specially produced antibacterial handsheet to the wallpaper usages, the photocatalyst efficiency test of benzene in volatile organic compound dissolution experiment of antibacterial and deodorization function wallpaper showed that the efficiency was 45~50% for 80 min of reaction time and 90% of attained resolution was reached at approximately 30 min of response time.

Effects of the Acute Exposure Oxytetracycline on the Behavior and Endocrine Response in Adult Zebrafish (Oxytetracycline의 단기 노출이 제브라피쉬의 행동 및 내분비 반응에 미치는 영향)

  • Zebrafish (Danio rerio) has been more widely used to study pharmacology. Oxytectracycline (OTC) is a broad-spectrum antibiotic and works by interfering with the ability to produce essential proteins of bacteria. The aim of this study was to identify the effects of exposure to OTC on behavioral changes or endocrine response in zebrafish. The behavioral effects of exposure to OTC (50, 100 or 200 mg/l) were characterized in several novelty-based paradigms such as the novel tank or open field test in zebrafish. Moreover, to investigate effects of exposure to OTC on endocrine response, we measured whole-body cortisol level using cortisol ELISA kit. As results of novel tank test, duration in top and immobile duration were significantly increased by the exposure to OTC in a concentration-dependent manner (p<0.05). In addition, moving distance, highly mobile, velocity and zone transition were significantly decreased by the exposure to OTC in a concentration-dependent manner (p<0.05). As results of open field test, the exposure to OTC increased immobile duration significantly (p<0.05). However, moving distance, mobile duration and velocity were significantly decreased by the exposure to OTC in a concentration-dependent manner (p<0.05). Besides, the exposure to OTC elevated whole-body cortisol levels in zebrafish. These results suggest that the exposure to OTC may induce chemical stress in zebrafish.

  • In children and adolescents, oral and maxillofacial trauma is one of the most common causes of dental and periodontal damage, which often induces crown fracture of the permanent anterior teeth. Frequently, these traumatized teeth lose their vitality, and require routine endodontic treatment if their root apices are closed. However if their apices are not fully closed, further root formation should be promoted by apexification or apexogenesis. Calcium hydroxide is a biocompatible & bacteriostatic material, and is widely used for apexification. However it has several disadvantages which include the need for multiple visits and patient cooperation, low strength and technical sensitivity in a broad apex. In one-visit apexification using IRM or SuperEBA, patient's visits can be minimized. However, their biocompatibility is questionable. Mineral trioxide aggregate(MTA) is a relatively new material. It is considered biocompatible with periapical hard tissue and has good marginal sealing ability. MTA is also known to help facillitate the growth of the cementum around it. In this case report, apexification with MTA was attempted on traumatized maxillary central incisors with immature root apices, and favorable clinical results were achieved.

  • This research was performed to estimate the properties of particleboard based on the press time and temperature which was made of chip of larch that grows in Korea. The results in this study were as follows: 1) Even though the chips, 1:1-35 ratio between length and thickness, are relatively bad condition, the surface smoothness that can easily spread the adhesive evenly and thoroughly and bonding ability of chips can give proper physical properties. 2) It shows more mechanical properties at the press time of 10 min. in MOR (Modulus of Rupture), MOE (Modulus of Elasticity) and SHA (Screw Holding Ability). 3) It is not significant according to the press time 20 min. in MOR, IBS (Internal Bonding Strength) and SHA, for the reciprocal actions between the accelerating aging effect of chip and the softening effect of adhesion are occured. 4) IBS is rising according to the increasing temp at the press time of 10 min. Because it needs to transfer the plate heat to make the proper hardening temp. In the layer. 5) The heat treatment effects have greatly influenced the stahility of dimension by falling the absorption, anisotropy and inhomegenity. As a result of these the values of thickness and linear expansion ratio were respectively dropped by the increase of press temp and the time and so did absorption.

Combination Effects of Potassium Sorbate and Sodium Benzoate with sodium Chloride on the Growth Inhibition of Escherichia coli and Salmonella typhimurium (Escherichia coli 와 Salmonella typhimurium 의 생육억제에 미치는 식염과 Potassium Sorbate, Sodium Benzoate의 병용효과)

  • An experiment was performed to investigate the combined effect of preservatives and the synergistic effect of sodium chloride to them on the inhibition of bacterial growth. Escherichia coli and Salmonella typhimurium were cultured with or without shaking in liquid media (pH 6) of tryptone-glucose-yeast extract or tryptic soy broth which contained 0.1% potassium sorbate and/or 0.03% sodium benzoate, equivalent to half of the maximum permissible levels, respectively. The growth of E. coli was more inhibited with one or both of the two preservatives by shaking culture than by non-shaking culture. For S. typhimurium the single treatment of the preservatives did not show inhibitory effect whereas the combined treatment of them showed bacteriostatic effect in shaking culture and a prolongation of lag phase in non-shaking culture. Addition of 2% sodium chloride to either potassium sorbate or potassium sorbate plus sodium benzoate remarkably increased the growth inhibition of E. coli for non-shaking cultivation but no effect observed for shaking cultivation. S. typhimurium was more sensitive to the addition of sodium chloride than E. coli in both shaking and non-shaking culture to show lower viable cell counts than initial numbers.

Effects of a Peroxidase-System-Containing Toothpaste on Whole Saliva in Vivo (Peroxida System을 함유한 세치제가 생체에서 전타액에 미치는 영향에 대한 연구)

  • Peroxidase system은 구강내 고유의 항균체계로서 항세균물질인 $HOSCN/OSCN^-$을 생산하여 다양한 그램 양성 그리고 그램 음성 세균에 대해 살균 및 정균작용을 나타낸다. 본 연구는 peroxidase system을 함유한 치약을 사용한 후 건강한 대상자의 자극성 전타액내 생성되는 $HOSCN/OSCN^-$의 양을 측정하여 최대 생성시간을 결정하였다. 그 후 건강한 대상자에게 임의로 실험치약과 대조치약을 할당하여 2주간 사용하도록 한 후 $HOSCN/OSCN^-$ 최대 생성 시간에 타액을 채취하여 치약 사용전과 2주후의 타액분비량, pH, 점조도 그리고 S. mutans와 lactobacilli에 대한 항균효과를 비교하였다. 구강건조증 환자를 대상으로 한 두 번째 연구에서도 peroxidase system을 함유한 세치제를 사용한 후 생성되는 $HOSCN/OSCN^-$의 최대생성시간을 측정하였으며, peroxidase system을 함유한 세치제가 S. mutans와 lactobacilli에 미치는 영향을 양치전과 양치후 $HOSCN/OSCN^-$의 최대생성시간에 타액을 채취하여 비교하였다. 이와같은 실험을 통해 다음과 같은 결론을 얻었다. 1. 건강한 대상자의 자극성 전타액내의 $HOSCN/OSCN^-$의 농도는 양치후 30분 후 대조군에 비해 peroxidase systems을 함유한 세치제를 사용한 군에서 유의성있게 높게 나타났다 (p < 0.05). 2. 건강한 대상자의 자극성 전타액의 타액분비량, pH, 타액점조도는 peroxidase system을 함유한 세치제를 2주간 사용한 후에도 유의한 변화를 나타내지 않았다. 3. 2주간 실험치약을 사용한 후 건강한 대상자의 자극성 전타액내 S. mutans와 lactobacilli의 양은 peroxidase system을 함유한 세치제를 사용한 군에서 상대적으로 낮게 나타났으나 통계학적 유의성은 없었다(p = 0.206 : lactobacilli, p = 0.944 : S. mutans). 4. 구강건조증 환자의 자극성 전타액내의 $HOSCN/OSCN^-$의 농도는 양치후 30분(p < 0.01)과 60분(p < 0.05)에서 대조군에 비해 peroxidase system을 함유한 세치제를 사용한 군에서 유의성있게 높았다. 5. 구강건조증 환자의 양치 후 30분에 채취한 자극성 전타액내의 S. mutans와 lactobacilli의 양은 peroxidase system을 함유하거나(p < 0.01) 함유하지 않은(p < 0.05) 치약을 사용한 군 모두에서 양치전에 비해 유의성있게 감소하였다. 6. 양치후 30분에 채취한 구강건조증 환자의 자극성 전타액내 S. mutans 양은 peroxida system을 함유한 세치제를 사용한 군에서 대조군에 비해 유의성있게 낮았다(p < 0.05). 7. 양치후 30분에 채취한 구강건조증 환자의 자극성 전타액내 lactobacilli양은 peroxidase system을 함유한 세치제를 사용한 군에서 대조군에 비해 상대적으로 낮게 나타났으나(p = 0.067) 통계학적 유의성은 없었다.

Influence of Phenobarbital on the Circadian Rhythm of Opiate Receptor in Rat Brain (백서의 뇌내 Opiate 수용체의 일중 변동에 미치는 Phenobarbital의 영향)

  • To investigate the influence of phenobarbital sodium on the action of morphine and on the diurnal rhythms of both opiate receptor binding and ${\beta}-endorphin$ contents, the amount of specifically bound $(^3H)$-morphine and immunoreactive ${\beta}-endorphin$ were measured in the midbrain of phenobarbital-treated rats at 4h intervals in a day. Rats were housed and adapted to a controlled cycle of either 12 h light-12 h dark or 24 h constant dark. After 3 weeks of adaptation, 0.5 ml of physiological saline or phenobarbital sodium (20mg/kg/day, i.p.) were administered twice a day for 2 weeks. Highly significant diurnal rhythms of opiate receptor binding and ${\beta}-endorphin$ were present in rat midbrain. In control group, the peak of maximum $(^3H)$-morphine binding was observed at 22:00 h, whereas the peak of ${\beta}-endorphin$ content was found at 06:00 h. Even in the absence of time cues these diurnal rhythms persisted, but they were highly modified with respect to the wave form as well as differences in the timing of peak and nadir. In the phenobarbital-treated group, these diurnal rhythms were also modified in shape, phase and amplitude, as well as in timing of peak and nadir. In this group, 24 h mean of opiate receptor binding was significantly decreased, while the 24 h mean level of ${\beta}-endorphin$ content was highly increased. However, Kd values in all experimental groups did not change. This indicates that differences in binding were not due to changes in the affinity, but in the number of binding sites. Statistical analysis of regression line indicates that changes of receptor binding were closely correlated with the changes of ${\beta}-endorphin$ content. These results suggest that phenobarbital may influence the action of morphine by changing the number of opiate receptors and that the modification of diurnal rhythm of opiate receptor by the agent is possibly due to changes of ${\beta}-endorphin$ content.

Suppressive Mechanism of Soil-borne Disease Development and its Practical Application -Isolation and Identification of Species of Trichoderma Antagonistic to Soil diseases and its activities in the Rhizosphere- (토양병의 발병억제 기작과 그 실용성 -길항성 Trichoderma spp.의 분리, 동정 및 근권내 활동-)

  • Trichoderma spp. are an effective control agent for damping-off or other plant diseases. The interaction between. T. hamatum and Rhizoctonia solani on the rhizosphere or surface soil were examined to assess the possible roles of antibiosis or competition in the mechanisms of biological control agents as a basic research. In a proportional comparison, total bacteria, fungi, actinomycetes and Trichoderma spp were 65%, 8.8%, 25.9% and 0.28% respectively in their distribution in the soil. Among Trichoderma spp isolated, the 5 species of Trichoderma spp were indentified as T. koninggii, T. pseudokoninggii, T. aureoviridi, T. hamatum and T. viride respectively. In a mycoparasitic test, one isolate of T. hamatum strain Tr-5 showed an enzymatic ability to break fungal hyphae into piecies and infected on the R. solani hyphae showing a parasitism. Spore germination of the all isolates of Trichoderma spp showed a 1.7-7.3% of germination in natural soil conditions, but the percentage was high in sterile soil indicating all the natural soil were fungistatic on conidia of Trichoderma spp. In rhizosphere competent assay in pea plant, the antagonistic T. hamatum, T. viride, T. koninggii, T. pseudokoninggii showed a colonizing upper soil depth in rhizosphere around 1-3 cm in root zone, but the colonizing ability was much reduced along the deeper the soil depth. Propagule density was decreased in deeper the soil layer. Disease development rate treated alone with plant pathogens, Fusarium solani, Rhizoctonia solani, Cylindrocarpon destructans increased, but disease incidence rate reduced in treatment with combinations with antagonistic T. hamatum strain Tr-5.

Studies on the Ecology of Occurrence and Identification of Typhula Snow Mold of Graminous Plants -II. Several Factors Affecting Growth of Typhula incarnata- (화본과식물에 발생하는 설부소입균핵병균(雪腐小粒菌核病菌)의 동정 및 발생상태에 관한 연구 -II. Typhula incarnata의 생육에 미치는 몇 가지 요인-)

  • Typhula incarnata grew over a temperature range of -5 to $20^{\circ}C$ with maximum growth at 10 to $15^{\circ}C$. Sclerotial production for T. incarnata was greatest at the higher temperature. Maximum mycelial growth of this pathogen occurred from pH 5.4 to 6.2. When carbon sources were added to a basal salt medium (Czapek's dox agar) at 5 g carbon sources/l, inulin, soluble starch, galactose, glucose, mannose, manitol, sucrose, maltose, cellobirose, trehalose, raffinose, and dextrin supported growth better than other carbon sources did. Of the twenty-three nitrogen sources tested, glycine, serine, ammonium sulfate, asparagine, asparatic acid, and ${\beta}-alanine$ were the most favorable for mycelial growth of T. incarnata. Cystine and cysteine were poor nitrogen sources. Ammonium salt of nitrogen sources supported growth better than nitrate salt of nitrogen sources. Potato dextrose agar, oat meal agar, and V-8 juice agar were the most favorable for mycelial growth and sclerotial formation. Appropriate addition of pepton to PDA decreased mycelial dry weight, but sucrose supported good growth of T. incarnata. Percent viable sclerotia of T. incarnate buried in bentgrass soil decreased from 2 months after treatment remarkably. Trichoderma riride and bacteria were isolated from non-germinated sclerotia. Live orchard grass leaf pieces within the soil were colonized by T. incarnata better than sterile and unsterile dead leaf pieces at $0^{\circ}C$. Saprophytic ability of T. incarnate on sterile leaf sheath occurred better at $0^{\circ}C$ than at $10^{\circ}C$. Saprophytic microflora consisting of Cladosporium sp., Fusarium sp., Mucor sp., Pythium sp., and unidentified fungi were the competitors for the sterilized and unsterilized substrate, but their colonization was not find on live leaf sheath buried in the soil at $0^{\circ}C$. In the effect of fungicides to Typhula snow mold disease of creeping bentgrass, mixture of polyoxin and thiram was the most effective, followed by iprodione, mixture of iprodione and oxine copper, thiophanate-methyl, myclobutanil, and tolclofos-methyl.
