Drug resistance is one of the most significant impediments to successful chemotherapy of cancer. Multidrug-resistance Is characterized by decreased cellular sensitivity to anticancer agents due to the overexpression of P-glycoprotein. By using adriamycin-resistance CL02 cancer cells, we undertook the screening fur agents which were effective to multidrug-resistant cancer cells from strains of the species Sorangium cellulosum isolated in our laboratory. Sorangium cellulose, cellulose-degrading myxobacteria have recently proved to be a rich source of novel anticancer agents. One of the significant examples is the promising anticancer agent epothilone. JW 1006 is the first strain of Sorangium cellulosum which was selected by us for the isolation of a metabolite by a biological screening because of a high cytotoxic activity against the CL02 cancer cells. Cytotoxicity-guided chromatographic fractionation of the culture broth led to the Isolation of two active principles, disorazole $A_1$ and $A_2$. They showed potent cytotoxicity against CL02 cancer cells with $IC_{50}$ values in the picomolar range, and were as active against drug-resistant cancer cells CL02 and CP70 as against the corresponding sensitive cells.
We prepared culture extracts of 174 Corallococcus and 207 Myxococcus strains isolated in Korea, and compared their antimicrobial activity against Candida albicans, Pseudomonas aeruginosa, and Staphylococcus aureus. The percentage of strains showing antifungal activity was lower in Corallococcus (7.5% [13 of the 174 strains]) than in Myxococcus (51.7% [107 of the 207 strains]). However, the percentage of strains exhibiting antibacterial activity was higher in Corallococcus (12.1% [21 strains]) than in Myxococcus (1% [2 strains]). The culture extracts of 6 Corallococcus strains inhibited both P. aeruginosa and S. aureus and displayed similar high-performance liquid chromatography chromatograms, although the shapes of their fruiting bodies were dissimilar. The rate of production of antibacterial substances was the highest when the strains were cultured in CYS medium for more than 6 days.
Phylogenetic analysis of two cellulase genes, xynB1 and bglA2, and the groEL1 gene from 34 Sorangium cellulosum strains isolated in Korea suggested that there are at least five subgroups in S. cellulosum, which is the most proficient producer of secondary metabolites among myxobacteria. This analysis also revealed diversity among the isolated S. cellulosum. It appeared that at least 30 out of 34 strains are different each other.
Fifty isolates of myxobacteria were isolated from soils from several areas in Korea during 1996-1997 and bioactivity against plant pathogenic fungi of these isolates was examined. A myxobacterial isolate KR025 showed good antifungal activities against Pyricularia oryzae, Cryphonectria parasitica, Colletotrichum lagenarium, and C. gloeosporioides but did not against Rhizoctonia solani, Fusarium oxysporum and Pythium ultimum. The bacterium was identified as Myxococcus fulvus based on morphological and physiological characteristics. Antifungal substances were extracted from culture broth and bacterial cell of Myxococcus fulvus KR025 by ethyl acetate. Antifungal substance of Myxothiazole (100 ${\mu}{\textrm}{m}$/ml) produced by Myxococcus fulvus KR 025 controlled 97.0% rice blast, tomato late blight, wheat leaf rust, and barley powdery mildew and showed 45.0 and 82.6% disease control of rice sheath blight and cucumber gray model, respectively.
Glycerol-spore formation of wild myxobacteria isolated in Korea has been tested. One third of the wild myxobacterial isolates belonged to the genus Myxococcus and Corallococcus produced a large number of glycerol-spores in the presence of glycerol or dimethyl sulfoxide. However, two third of the other wild myxobacterial isolates belonged to the genus Myxococcus and Corallococcus produced a negligible number of spores under the same condition. In addition, the myxobacteral isolates belonged to the other genus, such as Stigmatella, Archangium, and Cystobacter, failed to produce a noticeable number of glycerol-spores.
The relationship between soluble salts and microbial growth on stone surface was investigated. Samples from deteriorated surface layers of several stone monuments were collected, and the soluble salts in samples were chemically analysed. According to the results of chemical analyses, $Na_2SO_4$, $CaSO_4$$2H_2O$, and NaCl and so on were added to isolation media. As a result of preliminary experiments, 15 bacteria were selected. They are all Gram positives and fourteen of them were rods. Every bacteria could grow not only in media without salts, but also in media with $CaCO_3$, $CaSO_4$$2H_2O$, and $Ca(OH)_2$. Three bacteria, Bl6, B56, and B6l, secreted viscous substances in these $Ca^{2+}$ added media. $CaCl_2$$2H_2O$ was the most hygroscopic among 11 soluble salts, but it seemed to interfere with bacterial growth. Ammonium ion was the inhibitor, too.
Vibrio parahaemolyticus and V. vulnificus are known to be infected to human via fisheries products. Therefore, food safety of fisheries products is important for public health and fish industry. This paper was conducted to know how well these human isolates can survive in olive flounder (Paralichthys olivaceus). The growth of V. parahaemolyticus and V. vulnificus showed about 50~60% reduced rates at 25℃ than at 37℃ and did not show any differences according to NaCl concentration of media except the increasing in the growth of V. vulnificus in medium containing 3% NaCl. Artificial infection of 1×106 CFU/fish was carried out to confirm the sensitivity of olive flounder against V. parahaemolyticus and V. vulnificus. After 1 week from injection, no fish was dead. To evaluate nonspecific defense of olive flounder against V. parahaemolyticus and V. vulnificus, the antibacterial potency of serum and epidermal mucus were tested. The number of the vibrios exposed to serum obtained from olive flounder significantly decreased after 3 hours, and epidermal mucus showed decrease of the bacteria over than 90% until 12 hours from exposure. Phagocytosis of head kidney leucocytes of healthy olive flounder against V. parahaemolyticus and V. vulnificus showed in over 70% of leucocytes at the 2 hours. Therefore, cultured olive flounder only as vehicle for human pathogen in environmental water is well developed its antibacterial potency against human pathogens, so the viability of V. parahaemolyticus and V. vulnificus in cultured olive flounder was considered very low.
Bacillus licheniformis 9945a releases a natural ${\gamma}-poly(glutamic\;acid)({\gamma}-PGA)$ into fermentation broth and shows a mucoid phenotype on the solid agar medium. Transformation of mucoid cells of Bacillus species has not been simple and straightforward. The transpositional activity of Tn10 in B. licheniformis also has not been own either. Thus, a spontaneous non-mucoid derivative of the B. licheniformis was obtained first. Shuttle vector pHV1248 containing mini-Tn10 was introduced into the non-mucoid derivative by the method of protoplast transformation. The resulting transformant was reverted to the wild mucoid phenotype, and then mutated randomly with the mini-transposon by heat induction. Auxotrophs requiring arginine, lysine, or tryptophan were isolated by replica plating method. Southern blotting and DNA-DNA hybridzation analysis showed that these auxotrophs were generated by mini-Tn10 insertion into the chromosomal DNA. This method of transformation and mutation using pHV1248 would be useful for the generation of diverse mutants of B. licheniformis 9945a.(Received January 24,1997; accepted March 10, 1997)
