Microbial Removal Effects of Electrolyzed Acid Water on Lettuce by Washing Methods and Quality Changes during Storage (전해산화수에 의한 상치의 세척방법별 제균효과와 저장중 품질변화)

  • Effects of washing methods using electrolyzed acid water on lettuce(Lactuca sativa) and quality changes during storage were investigated. The multi-stage immersion treated 3 times in 2 min showed more effective than others to remove microorganisms. Total count of lettuce after immersion in electrolyzed acid water was decreased to 1/100-1/300 of $5.8\;{\times}\;l0^5CFU/g$ of non-immersed lettuce and $2.3\;{\times}\;l0^5CFU/g$ of tap water immersed lettuce. Also coliforms was significantly decreased to 1/3,000 of $3.1\;{\times}\;10^3CFU/g$ after electrolyzed acid water washing. However, microbial levels of electrolyzed acid water treated one became to be similar to those of non-treated lettuce after 3 days of storage at $10^{\circ}C$. The color values of L and b of lettuce treated with electrolyzed acid water were somewhat higher than those of others. Though chlorophyll content of lettuce just after immersion in electrolyzed acid water was 9% lower than those of non-treated one, the content was decreased to the same level of other treatments during storage. Decaying ratio showed the lowest value in lettuce immersed in electrolyzed acid water until 6 days of storage. The sensory tests for overall acceptability and appearance of lettuce immersed in electrolyzed acid water showed higher than those of others until $3{\sim}6$ days of storage.

Microbiological Cleaning Effect of Electrolyzed Acid Water by Containing Polysorbates (Polysorbates를 첨가한 전해산화수의 미생물학적 세정효과)

  • To enhance the cleaning and sterilization effect of cooled electrolyzed acid water on lettuce, several kinds of polysorbates were used at various concentrations in immersion washing process. In case of the treatment containing polysorbate 20, coliform count of lettuce was reduced to about 1/40 level of that in non-treated lettuce. The treatment containing polysorbate 60 did not show a significant sterilization effect. Otherwise, the total and coliform counts of lettuce during immersion in electrolyzed acid water containing 1 ppm of polysorbate 80 was reduced to about 1/300 and 1/1,700 level of those in non-treated one. And, the changes of ORP(oxidation-reduction potential), pH and color value of lettuce in electrolyzed acid water containing 1 ppm of polysorbate 80 were not quite different during 60 min. immersion, but HCIO content decreased from 10.28 ppm to 8.51 ppm after 20 min. immersion. Also, total and coliform count of lettuce immersed in electrolyzed acid water containing 1 ppm of polysorbate 80 was lower to about 1/1,800 after 20 min. and 1/5,550 after 30 min, compared with non-treated lettuce.

  • In this study, we investigated the effect of sterilization and the neutralization of treated ballast water using seawater electrolysis. The electrolysis apparatus has a cation-selective membrane for passing the cation and a titanium electrode in each cell. We examined the sterilization effect after an incubation period of 24 hr. The oxidation reaction during electrolysis caused, the solution to become strongly acidic due to the generation of a hydroxyl group, and the oxidation reduction potentials(ORP) was increased to 800 - 1200mV. After the reduction reaction, the solution became alkaline(pH 9 - 12), and ORP was decreased to - 900 - - 750 mV. It might be possible to control the pH of ballast water through electrolysis. In addition, we demonstrated the effects of sterilization of ballast water containing generated hypochlorous acid using electrolysis under high ORP condition.

Bactericidal Effects of Food-borne Bacteria using Chlorine Dioxide and Electrolyzed Water (이산화염소수와 전해수를 이용한 식중독균의 살균효과)

  • The present study investigated the bactericidal effects of chlorine dioxide (CD) and electrolyzed water (EW) on pathogenic bacteria, such as Bacillus cereus, Staphylococcus aureus, Salmonella Typhimurium, and Escherichia coli O157:H7, by treatment them with CD and EW, respectively, for 0, 2, 4, 6, 8, and 10 min. Additionally, the sensitivities of Gram-positive (B. cereus and S. aureus) and Gram-negative (S. Typhimurium and E. coli O157:H7) to CD and EW were compared, respectively. In CD, the D-values for B. cereus, S. aureus, S. Typhimurium, and E. coli O157:H7 were 1.85±0.64, 2.06±0.85, 2.26±0.89, and 2.59±0.40 min, respectively. In EW, the D-values for B. cereus, S. aureus, S. Typhimurium, and E. coli O157:H7 were 2.13±0.32, 1.64±0.64, 1.71±0.32, and 1.86±0.36 min, respectively. All strains decreased consistently for 10 min in both CD and EW. However, the D-values of each bacterial species did not differ significantly between CD and EW (P>0.05). When comparing the bactericidal effect of CD and EW, no difference in D-value was observed, even though the pH and available chlorine concentration of CD were significantly lower than those of EW. These data could be used for the application of CD and EW in the food industry, considering characteristics such as the selection of optimal disinfectants, determination of optimal concentrations, and sensitivity to disinfection targets.

Study on Reducing Methods of Natural Food-borne Pathogenic Microorganisms Originated from Saengshik (생식 중 자연환경유래 위해미생물 저감화 방법에 관한 연구)

  • In previous paper, contaminations of food-borne pathogenic bacteria of Saengshik was found to occur during processing, because detection rates of food-borne pathogenic bacteria in final products were higher than those of raw materials. In this study, methods to reduce food-borne pathogenic bacteria and improved manufacturing process were developed for microbial safety of Saengshik. Food-borne pathogenic bacteria in raw materials were reduced to about 0.5-2.0 log cfu/g when seven kinds of raw materials were washed with electrolyzed water and ozonated water, but food-borne pathogenic bacteria could not be removed completely. After improvement of manufacturing process, numbers of food-borne pathogenic bacteria were same or decreased to levels of raw materials. Gaseous ozone and Biocon could control air-borne bacteria under $1{\times}10^1$ cfu/1000 L of air in pulverization and mixing rooms.

Sterilization Effectiveness by Nominal and Absolute Filter in Pre-treatments of Honey (벌꿀의 전처리 공정 중 Nominal 및 Absolute Filter 적용을 통한 제균 효과에 관한 연구)

  • The coliform group, Salmonella typhimurium and Staphylococcus were analyzed for eliminating of microorganism spore which could be embeded in honey and eradication of studied for heat resistance of thermoacidophilic bacteria was studied for marketing of honey after producing honey drink. The method for analyzing of heat resistance thermoacidophilic bacteria was membrane-seperated cell culture with $0.45\;{\mu}m$ micro-filter and vacuum aspirator. The results of bacteria, coliform group, Salmonella typhimurium and Staphylococcus was negative, but normal method such as sterilization with electrolyzed water, normal micro-filter, high-temperature heating and microwave did not have effect on heat resistance thermoacidophilic bacteria. Also, absolute type micro-filter of $0.45\;{\mu}m$ and $0.8\;{\mu}m$ microfilteration showed higher effect on heat resistance thermoacidophilic bacteria than micro-filter of normal type, showing negative results at all treatments.

Effect of Surface Sterilization on Quality of Vacuum Packaged Fresh-cut Deodeok (Codonopsis lanceolata) during Storage (표면살균수 처리 후 진공포장된 신선편이 더덕의 저장 중 품질특성 변화)

  • This study investigated the changes in the quality of minimally processed sliced Deodeok (Codonopsis lanceolata) during storage at $7^{\circ}C$ in relation to vacuum packing using PE film after the various surface washing treatments. The surface washing treatments resulted in approximately 1~2 log CFU/g reduction of microbial load in the early storage day. After 20 days, the weight loss rate, deterioration rate, hardness, color, total microorganism levels and the coliform count of deodeok washed by ultrasonic wave water was deteriorated rapidly like the control. When measured by the fresh-cut deodeok surface using the video microscope system, washing with chlorine water and electrolyzed water didn't seem to make perceivable quality deterioration during the 10 days at $7^{\circ}C$. The hardness and color maintenance of the product vacuum packaged using PE film after washing with electrolyzed water, was better than that of other treatments.

Fungicidal Effect of Slightly Acidic Hypochlorous Water against Phytopathogenic Fungi (미산성 차아염소산수의 식물병원균류에 대한 살균 효과)

  • Slightly acidic hypochlorous water (SAHW) is well known for having a powerful and broad spectrum antimicrobial activity, and is harmless to the environment and humans. SAHW (pH 5~6.5, 20~30 ppm available chlorine concentration) was generated by electrolysis of dilute solution of HCl (4%) in a chamber of a non-membrane electrolytic cell. Our objective was to determine SAHW has a potential fungicidal activity on some phytopathogenic fungi. Spores of Botrytis cinerea, Colletotrichum acutatum and Phytophthora capsici were not culturable on agar media at approximately 10 seconds after treatment by SAHW. However, inactivation of Penicillium hirsutum was required over 3 min. Dilution of SAHW with sterilized distilled water (SDW) at the ratio of 1:1 (SAHW:SDW) against C. acutatum showed 100% inactivation but, the efficacy in 1:2 decreased until 63.2%. Control value of SAHW was 70.4% against C. acutatum on pepper fruits when applied upto 24 h postinoculation. SAHW has a powerful and wide spectrum antifungal activity and could be applied as a potential alternative to fungicidal agent for control of plant disease.

Quality Characteristics during Storage of Ginseng Washed by Different Methods (세척방법에 따른 인삼의 저장 중 품질특성)

  • We sought to improve the methods for washing fresh raw ginseng. The quality of ginseng surface-washed by different methods was evaluated during storage at 10C and $20^{\circ}C$. The raw ginseng surface-washing method was a full-cone spray-type procedure using water and air. The water for decontamination had an electrolysis value of 80 ppm, also known as electrolysis water $2^{\circ}C$ water and water containing 5 ppm chlorine dioxide, were also used for decontamination. The Hunter color (${\Delta}E$) of ginseng washed with water withan electrolysis value of 80 ppm, or water with 5 ppm chlorine dioxide, was greater than that seen after other washing methods were used. The weight loss after washing with 5 ppm chlorine dioxide water was similar to that seen after washing with $2^{\circ}C$ water or 80 ppm electrolysis water. Reductions in total microorganism levels, and counts of yeasts and molds, assayed 10 days after washing with 5 ppm chlorine dioxide water were greater than seen after use of other sterilization methods. Quality maintenance on storage, at both 10C and 20C, after washing with 80 ppm electrolysis water, was better than that noted after other sterilization methods. The moisture content of washed ginseng was similar under all storage conditions tested.
