cAMP-CRP complex에 의해 전사조절을 받는 관련유전자의 구조, 특징, 조절양상, 기작과 CRP의 생화학적 및 분자생물학적이해 등을 살폈다. 그러나 대장균의 전체유전자의 구조해석이 연구사업으로 실행되어지고 있는 현실에서 볼 때 CRP의 조절하에 있는 유전자군이 보다 더 복잡 다양한 system으로 생각되어져서 CRP조절계의 전체적인 양상으로 해명하기 위한 연구도 이루어질 것으로 생각되어진다. 즉, cAMP-CRP의 조절하에 있는 조절영역의 전체를 파악함은 operon의 발현조절의 해명에 중요한 기틀이 될 뿐만 아니라 개별유전자의 이해를 위한 것보다 복잡한 세포 전체의 제어계를 분자적수준에서 해결하는데 중요한 과제라고 사료되어진다. 몇 가지 연구 결과에서는 cAMP-CRP가 결합은 하면서도 발현조절에는 관여하지 않는다든지, cAMP가 결합하지 않으면서도 전사조절을 받을 수 있는 유전자들이 밝혀지고 있다.

유전자 발현은 다양한 전사 인자들의 상호 작용에 의해서 조절되어진다 이러한 전사 인자들에 존재하는 모티프는 직접적으로 조절 작용을 위한 기능을 수행한다. 또한 대부분의 경우에서 여러 모티프가 함께 유전자 발현 기작을 위하여 조절 작용을 한다. 따라서 이러한 모티프들이 어떤 조합으로 함께 전사 과정에 관여하는지 여부를 밝히는 작업은 중요한 일이다. 본 논문에서 진화 연산을 응용하여, 다양한 조건 하에 전사 과정에 중요하게 작용하는 모티프들의 조합을 알아보았고, 그 결과를 기본적인 k-Means 알고리즘 등과 비교하여 제안한 방법이 유전자들의 상관관계에 있어서 보다 우수한 결과를 보임을 알 수 있었다.
Abstract Secondary cell wall is the most abundant biomass produced by plants. Plant secondary cell wall is composed of a complex mixture of cellulose, hemicellulose, and lignin. Lignin, a phenolic polymer that hinders the degradation of cell wall polysaccharides to simple sugars destined for fermentation to bio-ethanol. Cell wall biosynthesis pathway-specific biomass engineering offers an attractive 'genetic pretreatment' strategy to improve bioenergy feedstock. Recently, we found a transcription factor, MYB7, which is a transcriptional switch that may turns off the genes necessary for lignin biosynthesis. To gain insights into MYB7 mediated transcriptional regulation, we first established a dominant suppression system in Arabidopsis by expressing MYB7-SRDX. Then we used a transient transcriptional activation assay to confirm that MYB7 suppress the transcription of the lignin biosynthetic gene. Taken together, we conclude that MYB7 function as a repressor of the genes involved in the lignin biosynthesis.

락토페린은 주로 유즙에 많이 포함되어 있으며 인간 분비물 등에서도 발견되는 당단백질로써, 미생물 감염에 대한 방어작용이 있는 것으로 알려져 있다. 락토페린의 미생물에 대한 방어작용은 미생물 성장에 필요한 철이온이 락토페린에 결합하여 성장을 저해하기 때문인 것으로 알려져 있다. 락토페린은 이외에도 염증반응의 조절, 임파세포의 성장촉진 등 면역반응에도 관여하는데 이러한 활성은 철에 결합하는 성질과는 무관하게 일어나며 락토페린이 DNA에 결합하는 성질과 관련이 있는 것으로 추측되어진다. 락토페린은 DNA에 결합하여 유전자의 전사에 관여할 것으로 여겨지는데 그 동안 어떤 유전자의 발현에 관여하는지에 대해서 알려진 바가 없었다. 최근 본 연구팀은 락토페린이 포유세포의 세포유전자의 전사에 관여하는지를 분석한 결과 락토페린 결합부위를 가지고 있는 유전자중의 하나인 인간 인터루킨-1$\beta$ 유전자의 전사를 활성화시킨다는 연구 결과를 보여 주었다. 인간 myelogenous leukaemia 세포주인 K562 세포를 락토페린과 phorbolmyristate acetate(PMA)로 함께 처리하면 K562 세포의 인터루킨-1$\beta$ mRNA의 양은 PMA 단독으로 처리하였을 때 보다 상승적으로 더 많이 유도됨을 보여주었다. 또한 IL-1$\beta$/Luciferase 융합 유전자를 K562 배양세포에 넣어 전사 활성을 비교함으로써 락토페린에 의한 인터루킨-l$\beta$의 전사활성을 확인하였다. 락토페린을 전체, N-말단, 혹은 C- 말단 부위를 COS-1 세포에 발현시켜 전사 활성을 측정한 결과 C-말단 쪽은 전사활성이 없었으나 N-말단 90개 아미노산 부위(NIa라 명명)가 전사활성을 가지고 있음을 규명하였다. 본 연구결과는 락토페린이 인터루킨-l$\beta$의 유전자의 전사에 역할을 하고 있음을 보여 주고 있으며 또한 인터루킨-1$\beta$의 유전자 외에도 락토페린 결합 부위를 유전자의 조절부위에 포함하고 있는 세포 유전자의 전사도 관여할 수 있음을 제시하고 있다.

Background : Thyroid Transcription Factor-1(TTF-1) acts as a tissue specific transcription factor in the regulation of lung specific gene expression and as morphogenic protein during lung organogenesis. Currently, there is very little information on the cis-acting sequences and transcription factors that direct the TTF-1 gene expression. DNAse 1 hypersensitive (DH) sites represent a marker for active or potentially active chromatin and are likely to be especially important in gene regulation, being associated with many DNA sequences that regulate gene expression. It is clear that DH regions correlate with genetic regulatory loci and binding for sequence-specific DNA-binding proteins. Methods : We have used DH site assays to identify putative distal regulatory elements in H441 lung adenocarcinoma cells, which express the TTF-1 gene and HeLa cells. Results : There are four DH sites 5' of the TTF-1 gene. These sites are located at base pair approximately +150, -450, -800, and -1500 from the start of transcription. Conclusion : These data suggest that there may be at least one intragenic site and regulatory region 5' prime to the promotor region.
Long-chain fatty acids (LCFAs) are vital in cellular compartments, primarily regulating lipid metabolism. Fatty Acid-Binding Proteins (FABPs) facilitate LCFA transport, lipid synthesis, storage, and act as signaling molecules influencing various pathways, including inflammation. FABP4, in particular, is linked to vascular and cardio-related diseases, and it plays a role in macrophage-mediated inflammatory responses. Previous studies have identified FABP4 as not only a representative biomarker for lipogenesis but also as having correlations with immune responses. This study aims to investigate the regulation of the chicken FABP4 (chFABP4) gene by toll-like receptor 3 (TLR3) activation and determine the signaling pathways that are involved in chFABP4 transcriptional regulation. We analyzed the transcriptional regulation of chFABP4 in TLR3-stimulated DF-1 cells. The results showed that chFABP4 was up-regulated upon stimulation with polyinosinic-polycytidylic acid (PIC), a TLR3 ligand. Notably, chFABP4 transcription was independently regulated in the NF-κB signaling pathway. It was up-regulated in p38 inhibition, demonstrating that the p38 signaling pathway might suppress the transcription of chFABP4 within TLR3-activated DF-1 cells. In contrast, chFABP4 expression was down-regulated in JNK signaling pathway inhibition, suggesting the positive regulation of JNK signaling pathway for chFABP4 transcription in DF-1 cells in response to TLR3 activation, consistent with findings in macrophages. MEK pathway inhibition resulted in a similar regulation to NF-κB signaling. These results suggest that each MAPK contributes differentially to the transcriptional regulation of chFABP4 by in DF-1 cells in response to TLR3 activation.
The present study was aimed at investigating the metabolic regulation of insulin-like growth factor-I(IGF-I) expression in fasting animals. The expression of IGF-I gene was determined by a solution hybridization/RNase protection assay using total RNA from control, 4d-fasting, and 2d-fasting-refed rats. The levels of IGF-I transcripts were reduced in 4d-fasting than in control by decreasing its transcriptional rate, which was measured through nuclear nun-on assay. DNase I footprinting, which was performed using nuclear extracts from fasting rat, demonstrated protein binding to a sequence that extended from +179 to +210 (termed region B). These data suggest that the expression of IGF-I is transcriptionally regulated through DNA-liver enriched protein binding in a sequence which is located downstream from major transcription initiation site of IGF-I gene.
최근 Okamoto등(18)이 처음으로 VB receptor 유전자인 vbr A의 cloning 및 vbr A의 E. coli 유래의 필수 유전자인 Nus G와의 높은 상동성(36%)으로부터 전사, 번역기구의 필수 유전자로서 가능성의 보고와, Miyake등(19)은 A-factor receptor의 repressor로서의 기능을 보고함에 따라 이들 자기조절인자의 항생물질등 2차 대사산물의 생합성에 있어서 signal전달에 따른 전사조절의 switch on-off 기구 연구에 박차를 가하에 되었으며 본고에서는 VB를 중심으로한 유도인자의 기능면 및 응용면을 소개하고자 한다.

생물체에서 유전외적 변형의 하나인 DNA 메틸화는 cis-acting factor의 조성변화를 통하여 세포특이 유전자의 발현과 virus latency, genomic imprinting, mutagenesis등과 같은 생물학적 효과를 나타내는 것으로 알려져 있다.(reviewed by Olle Heby, 1995). 5-azaCR, 5-azaCdR 그리고 6-azaCR의 처리결과는 배아자체의 DNA 메틸화의 유지가 정상발생에 필수적임을 알 수 있으며, 메틸화에 의한 배아발생 조절기작이 존재함을 암시하고 있다. 이러한 과정에서 5-azaCR과 5-azaCdR은 서로다른 경로를 통하여 배아발생에 관여함을 보여주었다. 즉, 5-azaCdR은 주로 DNA에 incorporation되어 작용하는 것으로 여겨지며, 5-azaCR은 DNA 보다는 RNA에 incorporation되어 작용하는 것으로 나타났다. 그리고, 비록 소수의 유전자만이 조사되었지만, 5-azaCdR의 incorporation에 의한 cis-acting factor의 변화는 전사인자인 c-myc proto-oncogene과 fluid 수송에 관여하는 $Na^{+}$, $K^{+}$-ATPase 유전자의 전사를 억제하지 않았다. 반면, 5-azaCR의 RNA로의 incorporation은 전사인자인 c-myc proto-oncogene의 전사를 억제하였으며, 연이어 fluid 수송에 관련되어있는 $Na^{+}$, $K^{+}$-ATPase 유전자의 전사를 억제하였다. 이것은 아마도 RNA로 incorporation된 5-azaCR은 RNA의 post-transcriptional processing에 영향을 주어 trans-acting factor의 조성을 변화, 전사적 repression을 유발한 것으로 사료된다. 생쥐 착상전 초기배아에서 DNA 메틸화는 short-term하게는 cis-acting factor로써 전사적 수준에서 유전자발현 조절하며, 그리고 유전자발현을 통하여 long-term하게는 배아발생에 관여 할 것이라고 사료된다.
