Effects of Extrusion and Enzyme Treatment on Characteristics of Whole Grains (압출성형 및 효소처리가 전곡립의 특성에 미치는 효과)

  • Extrusion with enzyme treatment improved processing characteristics and physiological activity of whole grains, strongly suggesting industrial potential of whole grains for production of cereal-based functional foods containing activated nutraceuticals. Physicochemical properties of whole grains prepared by extrusion were characterized in terms of vitamin, mineral, and resistant starch contents, gelatinization, and digestibility in comparison with those of raw and roasted grains. Extrusion increased grain gelatinization, with degree of gelatinization in extruded barley being seven times higher than that of raw barley. Extrusion improved digestibility of whole grains in comparison with those of raw and roasted grains. Dispersibility of starch was improved through extrusion, while content of resistant starch decreased. Release of ${\beta}-sitosterol$ present in whole grains increased 13-fold when both extrusion and enzyme treatment were used.

Digestive, Physical and Sensory Properties of Cookies Made of Dry-Heated OSA-High Amylose Rice Starch (변성 고아미 쌀전분을 이용한 쿠키의 소화율과 물리적 및 관능적 특성)

  • Cookies containing wheat flour mixed with 10, 30 or 50% esterified with octenylsuccinic anhydride (OSA, 3%) and dry-heated ($130^{\circ}C$, 2 hr) high amylose rice (Goami 2) starch (DH-OSAR) were prepared and then their physical and digestive properties were evaluated. When the amount of added DH-OSAR increased, the hardness and brittleness of the cookies decreased, and L (brightness) value increased. For the digestive properties, the cookies containing 50% DH-OSAR significantly increased the amount of slowly digestible starch (SDS), and decreased the amount of rapidly digestible starch (RDS), resulting in the lowest expected Glycemic Index (eGI) among tested cookies. Although the cookies containing DHOSAR were inferior to the control, the addition of xanthan gum (0.5% based on total powder amount) significantly improved their textural and sensory properties. Specially, the cookies containing 50% DH-OSAR and the addition of 0.5% xanthan gum showed the lowest eGI value, maintaining the improved textural and sensory properties.

Formation of Enzyme Resistant Starch by Extrusion Cooking of High Amylose Corn Starch (고아밀로즈 옥수수전분의 압출성형에 의한 난소화성화)

  • Extrusion cooking treatment was compared with autoclaving/cooling treatment for formation of enzyme resistant starch of high amylose corn starch (HACS). Effects of barrel temperature $(100^{\circ}C,\;120^{\circ}C,\;140^{\circ}C)$ and feed moisture content (25%, 35%, 45%) on extrusion processing in a co-rotating twin-screw extruder under fixed screw speed (100 rpm) were investigated by measuring enzyme resistant starch (RS) yield. RS yield were estimated by in-vitro pancreatin digestion method and enzymatic-gravimetric method using termamyl. Barrel temperature and yield of RS were negatively correlated and feed moisture content and yield of RS was positively correlated as determined by in-vitro pancreatin method. The highest yield (38.4%) of RS was obtained from HACS extrudate processed at the barrel temperature of $100^{\circ}C$ and the feed moisture content of 45%, while the yield of RS by 5 times of autoclaving/cooling was 25%. The yield of RS by in vitro pancreatin digestion method was 20.7% with high amylose corn starch and 8.2% with ordinary corn starch (CS), respectively, under the same extrusion condition (barrel temperature $120^{\circ}C$, feed moisture content 35%). At the same condition, the yields of RS by enzyme-gravimetric method were 14.6% with HACS and 6.8% with CS, respectively. The yield of RS increased during the storage at $4^{\circ}C$ for 4 weeks and the highest yield (60%) was obtained by the storage of HACS extrudates extruded at $100^{\circ}C$ and 45% feed moisture content.

  • An in vitro digestibility methods have been developed to mimic an in vivo system in the past decades. Because the in vivo techniques cause high cost, intensive labor, longer research periods and ethnical problems. In this review, the digestive systems from stomach to large intestine of pig as a monogastric animal were addressed to understand an in vivo digestion. The innovative in vitro technique using the Daisy II incubator was performed and the in vitro ileal and fecal dry matter (DM) digestibility of corn, rice, wheat and barley was determined.

A study on Reducing Power, Degree of Gelatinization and Retrogradation Rate of Soybean Jeolpyon (콩절편의 소화율, 호화도 및 노화속도)

  • Various kinds of jeolpyon (cubed rice cake) differing in roasted soybean flour. content (0, 5, 10, 15, 20%) were prepared to study digestibility, degree of gelatinization and retrogradation rate. Digestibility slightly in creased as soybean content increased. Degree of gelatinization also increased as soybean content increased. Retrogradation rate obtained from the changes in hardness of soybean jeolpyon with the control was 0.528 day and time constant (1/k) was 1.89 days.

  • Cultivation condition of yeast on the utilization of fermentable substrate from the cellulosic wastes such as rice hull, rice straw and corn starch cake was investigated. The results obtained were summarized as follows;1. Corn starch cake was respectively added to rice hull and rice straw in order to increase sugar concentration in the hydrolyzate, and then hydrolyzed. As the result, concentration of sugar in hydrolyzed solution of rice hull was 9.12%, in that of rice straw was 7.98%. 2. It was found that calcium carbonate as a neutralizer was the most effective to prepare the culture broth of yeast. 3. An optimal growth of Hansenula subpelliculosa GFY-2 was observed in the medium prepared by adding 0.3% of ammonium sulfate, 0.4% of potassium phosphate dibasic, 0.02% of magnesium sulfate, sodium chloride and calcium chloride to hydrolyaed sugar solution, respectively. 4. Hansenula subpellicuiosa GFY-2 cultured in the substrate solution which of rice hull and rice straw added to corn starch cake was assimilated more than 90% of sugar in the hydrolyzate within 48 hours. The yeast cells yielded in rice hull was 46.5%, and that of rice straw 45.4% to utilized sugars.

Production of Single-Cell Protein from Starchy Material by the Fusant (전분이용성 세포융합 효모를 이용한 단세포단백질 생산)

  • The production of single cell protein using the amylolytic fusant obtained from cell fusion between Hansenula anomala and Saccharomyces cerevisiae was studied. The fusant12 strain was selected for single cell protein production from starchy materials among five fusants. Optimum nitrogen source and its concentration for the growth of fusant12 were ammonium sulfate and 0.1%, respectively. Optimum concentration of soluble starch and optimum pH of the basal medium were lord and pH 5.6, respectively. Autolysis of fusant12 was effectively carried out by addition of 5% (v/v) ethyl acetate to the cell suspension and liquidization for 30 min before incubation for 24 hr at 3$0^{\circ}C$. Coculture of fusant12 and non-amylolytic yeast, Torulopsis candida YA-l5, resulted in the increase of the mass as compared to the monoculture of fusant12. The cell mass on tapioca medium was increased about 2.5 times as on soluble starch medium. The content of crude protein and nucleic acid of the dry cell were 39% and 5.8%, respectively.

  • This study was conducted to evaluate the effect of different processing of rice on rumen fermentation in in vitro and in situ experiments. Different processing treatments (extruding, roasting, and steaming) were used in this study and all treatments were ground through a cyclone mill (Foss, Hillerød, Denmark) fitted with a 1 mm screen. Non-treated rice was considered to a control substrate. Then, all treatments were used in in vitro and in situ experiments. Total gas production and dry matter digestibility in control were lower than any other treatment at all incubation times (P<0.01). The lowest ammonia nitrogen ($NH_3-N$) concentration was observed in control among treatments at 6, 12, and 24 h incubation (P<0.01). Extruding had a highest total volatile fatty acids (VFA) concentration at 6, 12 h incubation (P<0.01) and Steaming exhibited a highest total VFA at 24 h (P<0.01). The lowest total VFA concentration was observed in control at 6, 12, and 24 h (P<0.01). In an in situ, The highest value of soluble fraction, degradation rates, effective degradability was observed in extruding (P<0.01). It was considered that feed processing increased dry matter digestibility, total VFA concentration, and decreased pH as well as $NH_3-N$ concentration indicating that processing may increase nutrient degradation of rice in the rumen.

Portulaca oleracea L. Extract Lowers Postprandial Hyperglycemia by Inhibiting Carbohydrate-digesting Enzymes (쇠비름(Portulaca oleracea L.) 추출물의 탄수화물 소화 효소 저해와 식후 고혈당 완화 효과)

  • Postprandial hyperglycemia plays an important role in the development of Type 2 Diabetes and diabetic complications. Controlling postprandial hyperglycemia is the most important factor for reducing the risks of diabetic complications in Type 2 diabetic patients. This study was designed to determine whether Portulaca oleracea L. extract suppresses the activation of carbohydrate-digesting enzymes, and lowers postprandial hyperglycemia in diabetic mice through streptozotocin. P. oleracea was extracted with either 80% ethanol (PEE) or water (PWE), and the extract solutions were concentrated. The ${\alpha}$-glucosidase and ${\alpha}$-amylase inhibition assays were performed using the chromogenic method. Normal mice and STZ-induced diabetic mice were orally treated with PEE, PWE (300 mg/kg of body weight) or acarbose (100 mg/kg of body weight), with soluble starch (2 g/kg of body weight). The ${\alpha}$-glucosidase and ${\alpha}$-amylase inhibitory effectiveness by PEE were markedly more effective than PWE, and both extracts indicated a higher effectiveness than the acarbose (positive control). The rise in postprandial blood glucose due to starch loading was markedly inhibited in the PEE group when compared to the control group in diabetic and normal mice. Furthermore, the area under the concentration-time curve values were markedly declined by the PEE injection in the diabetic group when compared to that exerted for the control group. These results demonstrate that P. oleracea extracts lower postprandial hyperglycemia by inhibiting carbohydrate-digesting enzymes, and that the ethanol extract is more efficacious than the water extract.

  • To study the effect of preventing the browning reaction between casein and glucose by protecting the reactants using corn starch with coating material, rats were fed for 30 days nonbrowning diet, nonprotected browning diet, browning diet protected only casein, browning diet protected both casein and glucose and browning diet supplement sucrose simultaneously with protecting both casein and glucose. The amounts of fructosyl-lysine excreted through urine were greater than those through feces regardless of diets and the both side of the excreted amounts of fructosly-lysine through urine or feces were greater for rats fed browning diets regardless protecting compared to rats fed the nonbrowning diet. Through urine, the excreted amounts of fructosyl-lysine were decreased for rats fed the browning diet supplemented sucrose simultaneously with protecting both casein and glucose than those for rats fed the nonprotected browning diet and through feces, were decreased for rats fed protected browning diets regardless of protecting method than the nonprotected browning diet. The excreted amounts of all individual essential free amino acids through feces were increased for rats fed browning diets irrespective of protecting compared to the nonbrowning diet, but through urine, were increased or similar level for rate fed the nonbrowning diet compared to browning diets except histidine. The excreted amount of free lysine through feces were decreased for rats fed protected browning diets than nonprotected browning diet. Fructosyl-lysine contents of serum, liver and small intestinal digesta were increased for rats fed browning diets regardless of protecting by starch compared to nonbrowning diet but, of serum and small intestinal digesta were decreased for rats fed protected browning diets than the nonprotected browning diet.
