Effect of External Factors on Heavy Metal Accumulation in the Cell of Heavy Metal-Tolerant Microorganisms (중금속내성균의 중금속 축적에 미치는 외부요인의 영향)

  • This study was performed to develop the biological treatment technology of wastewater polluted with heavy metals. Heavy metal-tolerant microorganisms, such as Pseudomonas putida, P. aeruginosa, P. chlororaphis and P. stutzeri possessing the ability to accumulate cadmium, lead, zinc and copper, respectively, were isolated from industrial wastewater and mine wastewater polluted with various heavy metals. The effect of several external factors, such as temperature, pH and heavy metal compounds on heavy metal accumulation in the cells was investigated. The amount of heavy metal accumulation into cells according to the kind of heavy metal compound was slightly increased in the case of the heavy metal compound with -nitrate group, but generally, there is little change according to the kind of compound in the amount of heavy metal accumulation. The amount of heavy metal accumulation according to the precultured time was increased in the case of the cell precultured for 24 hours, but generally the precultured time did not affect to the amount of heavy metal accumulation. Heavy metal accumulation into cells was affected by several external factors, such as temperature and pH. The optimum temperature and optimum pH of the accumulation of heavy metal into cells were $20{\sim}37^{\circ}C$ and pH $6{\sim}8$, respectively. By increasing the concentration of each heavy metal-tolerant microorganism in the solution, the total amount of heavy metal accumulated was increased, whereas the amount of heavy metal accumulated per cell(mg, heavy metal/g, dry cells) was decreased. These results indicated that the amount of heavy metal accumulated was not proportional to the concentration of microorganisms.

Molecular Characterization and Antimicrobial Susceptibility of Biofilm-forming Acinetobacter baumannii Clinical Isolates from Daejeon, Korea (대전지역에서 분리된 생물막 형성 Acinetobacter baumannii 임상분리주의 분자유전학적 특성과 항균제 감수성양상)

  • The emergence and dissemination of multidrug-resistant (MDR) Acinetobacter baumannii isolates have been reported worldwide, with most of these possessing the ability to form biofilms. Biofilm formation is an important virulence factor associated with the resistance to disinfection and desiccation. This study examined the genetic basis of antimicrobial resistance mechanisms of biofilm-forming A. baumannii clinical isolates. Imaging and quantification of biofilms were performed by a crystal violet assay and 46 biofilm-forming A. baumannii isolates were selected. Subsequently, 16 isolates belonging to different clones were identified using REP-PCR, and detection of the antimicrobial determinants in the isolates was carried out. The 16 isolates included 9 non-MDR and 7 MDR isolates. The mean biomass $OD_{560}$ values of the non-MDR (0.96) and MDR (1.05) isolates differed but this difference was not significant. In this study, most biofilm-forming MDR A. baumannii isolates contained various antimicrobial resistance determinants ($bla_{OXA-23}$, armA, and mutations of gyrA and parC). On the other hand, most biofilm-forming non-MDR A. baumannii isolates did not contain antimicrobial resistance determinants. These results suggest that there is little correlation between the biofilm-forming ability and antimicrobial susceptibility in A. baumannii isolates. In addition, the emergence of MDR A. baumannii clinical isolates is generally caused by mutations of the genes associated with antimicrobial resistance and/or the acquisition of various antimicrobial resistance determinants.

  • The emergence of carbapenem resistance among Pseudomonas aeruginosa has become an increasing problem worldwide. In particular, $metallo-{\beta}-lactamases$ (MBLs) are responsible for the high-level resistance to carbapenem. Sequence type 235 (ST235) has been found internationally in a multidrug-resistant clone and is involved in the dissemination of genes encoding IMP-6 and VIM-2. This study examined the prevalence of MBLs and the epidemiological relationship in carbapenem-resistant P. aeruginosa (CRPA) isolates obtained from a tertiary hospital in Daejeon, Korea, between March 2008 and June 2014. The antimicrobial susceptibilities were determined using the disk-diffusion method and PCR and DNA sequencing were used to identify the MBL genes. In addition, an epidemiological relationship was investigated by multilocus sequence typing (MLST). Among the 110 CRPA isolates, 32 isolates (29.1%) were MBL-producers; the major type was IMP-6 (29 isolates, 90.6%). VIM-2 was identified in 3 isolates (9.4%) of ST357. IMP-6-producing isolates were multidrug-resistant (MDR) and belonged to ST235. ST235 (55 isolates, 50.0%) was the clone most frequently detected and has gradually emerged during a seven-year period. To prevent the spread of MDR ST235 P. aeruginosa isolates, the current widespread use of carbapenems needs to be curtailed, and novel continuous monitoring strategies should be developed as soon as possible.

Transfer of Genes for Antimicrobial Resistance and Toxin of Hemolytic Escherichia coli Isolated from Feces of Pig Suffering Diarrhea to Human Isolates (설사 증상의 돼지 분변에서 분리된 용혈성 대장균의 항생제 내성과 독소의 인체로부터 분리된 균주로의 전이)

  • Between 1997 and 1998 in Korea, 56 isolates of Escherichia coli were obtained from pig suffering diarrhea. Among those, 38 isolates that showed the hemolytic activity, antimicrobial resistance, and toxin production were studied. Among 38 isolates, thirty-six isolates $(94.7\%)$ were resistant to tetracycline, 27 isolates $(71.0\%)$ were resistant to ampicillin, 26 isolates $(68.4\%)$ were resistant to chloramphenicol, and 21 isolates $(55.2\%)$ were resistant to trimethoprim, while none was resistant to aztreonam, amikacin, and norfloxacin. Among these iso­lates, 21 isolates $(55.3\%)$ were multiple drug resistant to at least four different class antimicrobial agents. Extended spectrum $\beta-lactamase$ producing isolates were not detected in the double disk synergy test. In these hemolytic Escherichia coli, heat-stable enterotoxin $(89.5\%)$ was the most prevalent toxin, followed by vero­toxins $(47.4\%),$ and then heat-labile enterotoxin $(31.6\%).$ Except 8 isolates $(21.0\%)$ which produced ST only, 12 isolates $(31.6\%)$ produced ST and LT, 13 isolates $(34.2\%)$ produced ST, VT, and VTe, and 5 isolates $(13.2\%)$ produced VT and VTe. However, none produced all 4 types of toxin, simultaneously. The predominant serotype could not be determined by the agglutination method. Sixteen isolates $(42.1\%)$ were strongly adhered to T-24 bladder cell and 17 isolates $(44.7\%)$ were to Caco-2 intestinal cell. Especially, 11 strains $(28.9\%)$ were evaluated as strongly adhesive to both T-24 cells and Caco-2 cells. Genes for toxin and the antimicrobial resistance were transferred to clinical isolates of Escherichia coli from human urine by the filter mating method. Results suggest the possibility that antimicrobial resistance and toxin can be transferred from animals to humans by direct con­tact of resistant bacteria as well as gene transfer, although there was no correlation between toxin production, adherent activity, and antimicrobial resistance among hemolytic E. coli isolated from pig suffering diarrhea.

Influence of Fish Compositions and Trophic/Tolerance Guilds on the Fishkills in Geum-River Watershed (Backje Weir) (금강수계(백제보)에서 발생된 어류폐사에 대한 종 조성 및 트로픽/내성도 길드 영향 분석)

  • The objectives of this study were to analyze structures of fish community and the ecological health using a multi-metric fish model, the Index of Biological Integrity (IBI) in the Backje Weir of Geum River during two periods namely before-the-fishkill ($B_f$) and after-the-fishkill ($A_f$). The total number of fish species observed were 32 and among them 10 species (35%) were Korean endemic species. The exotic species observed were 3 which decreased by 0.4% after-the-fishkill ($A_f$). The dominant species were Opsariichthys uncirostris amurensis (13%) at the Bf period and Squalidus japonicus coreanus (17%) at the Af period in the Backje Weir. At after the fishkill ($A_{f-I}$) total biomass was about 10 times lower than the biomass before-the-fishkill ($B_f$). The biomass of Carassius auratus decreased 98% after-the-fishkill and as time passed by the biomass recovered to nearly 100%. Similar decrease in the biomass occurred in the population of Opsariichthys uncirostris amurensis, while Rhinogobius brunneus population increased. According to the structure analysis of fish community, species richness index, evenness index and species diversity index were high but after-the fishkill, the values of indices decreased. Tolerant species (64%) dominated the fish community, and the sensitive species (2%) were rare, indicating the degradation of the ecosystem. According to analysis of the multi-metric model (IBI), the mean model value of the fish community in Backje Weir was estimated as 17.5 indicating a "fair condition".

Monitoring of Antimicrobial Resistant Bacteria from Animal Farm Environments in Korea (국내 축산 환경 중의 항생제 내성균 모니터링에 관한 연구)

  • The kinds and quantity of antimicrobial agents used for cattle (animal industry) may be considerable, suggesting the possibility that pathogenic bacteria which cannot be extirpated by the existing antimicrobial agents could appear. Ten cattle, pig and chicken farms, respectively, were randomly selected from 5 provinces in Korea and the samples were collected from excrement, manure, underground water, farmers' hands and the neishboring environment. h total of 299 samples were examined and 197 of Escherichia coli, 13 of Campylobacter jejun/coli, 223 of Enterococcus faecium/faecalis and 42 of Staphylococcus aureus isolates were collected. All isolates were screened for antimicrobial resistance: 69.4% of E. coli (137/197 strains), 78.6% of S. aureus (33/42 strains), and 82.1% of E. faecium/faecalis (183/223 strains) were resistant to one antimicrobial agent and all of C. jejuni/coli Isolates showed the resistance to one antimicrobial agent. Meanwhile, the multiple resistance ratio for more than 4 lines of antimicrobial agent was 19.2% of E. coli (38/197 strains), 11.9% of S. aureus (5/42 strains), 15.4% of C. jejuni/coli (2/13 strains) and 6.2% of E. faecium/faecalis (14/223 strains). The antimicrobial resistance ratio of bacteria isolated from the cattle farm showed lower than that of bacteria isolated from the pig or chicken farm, which might be related to the quantify of antimicrobial agents consumed. And one strain of vancomycin resistant E..faecium (VREF) were isolated from the excrement of chicken and stream, respectively. Generally, the ratio of VREF collected in animal farm environments is lower than that of VREF collected in medical environment.

Non-typhoidal Salmonella Gastroenteritis in Childhood: Clinical Features and Antibiotics Resistance (소아에서 비장티푸스성 살모넬라 위장관염의 임상양상과 항생제 내성률에 대한 연구)

  • Purpose: As the incidence of non-typhoidal salmonella strains resistant to antibiotics has been increased, we attempted to investigate clinical aspects of non-typhoidal salmonella gastroenteritis and antibiotics resistance. Methods: From January 2000 to June 2002, 99 children with positive stool culture of non-typhoidal salmonella were studied about clinical features, the incidence of antibiotics and multi-drug resistance and the difference of incidence of antibiotics resistance according to immune status. Results: There were 66 males and 33 females. The majority of them were under 5 years of age (71%). 25 children were immunocompromised due to chemotherapy, steroid or immunosuppressive treatment. Serogroup D was the most common isolates (65%) followed by B (16%), C (8%) and E (8%). Resistance rate of 30% to ampicillin, 12% to chloramphenicol, 20% to trimethoprim-sulfamethoxazole (TMP-SMX), 11% to cefotaxime and 8% to cefixime were obtained. All isolates were susceptible to ciprofloxacine. Resistance rate to cefotaxime and cefixime in immunocompromised patients was 24% and 14.3% respectively, which were significantly higher compared to that in immunocompetent patients (6.8%, 5.6%, p<0.05). 11 isolates were resistant to three or more antibiotics. The incidence of multi-drug resistant isolates was significantly higher in immunocompromised patients (24%) than that of immunocompetent patients (6.8%). Conclusion: Because of the high prevalence of non-typhoidal salmonella strains resistant to ampicillin, chloramphenicol and TMP-SMX, third-generation cephalosporin might be the treatment of choice in non-typhoidal salmonella gastroenteritis. In particular, antibiotics should be carefully selected in immunocompromised patients because non-typhoidal salmonellas from them showed the higher incidence of antibiotic resistance and multi-drug resistance.

  • 동서양을 망라하여 전 세계적으로 비알코올성 지방간(Nonalcoholic fatty liver disease[NAFLD])의 발생률은 증가추세에 있다. NAFLD는 간에 중성지방(triacylglyceride)이 비정상적으로 축적되어 발생하지만, 사람에 따라서는 간조직에 염증반응이 초래되고, 심지어 간경화, 간암으로까지 진행되기도 한다. NAFLD의 원인이 비만, 인슐린내성과 같은 대사증후군과 연관되기는 하지만, 정확한 병리기전은 아직 규명되지 않았다. 따라서, 치료방법도 충분히 개발되지 못하고 있다. 그럼에도 불구하고, NAFLD에 있어 분명한 점은, 예방과 악화방지가 가능하다는 사실이다.
