Production of a Fermented Korean Pear Puree using a New Strain Leuconostoc mesenteroides KACC 91495P Isolated from Kimchi (김치에서 신규 Leuconostoc mesenteroides KACC 91495P 균주의 분리 및 이를 이용한 배 발효물의 제조)

  • A lactic acid bacterial strain showing fast growth and high acid production in Korean pear puree was isolated from Kimchi. This strain was analyzed by API 50 CHL kit and 16S rRNA sequencing analysis and identified as Leuconostoc (Ln.) mesenteroides KACC 91495P. Korean pear puree was fermented using Ln. mesenteroides KACC 91495P strain at $30^{\circ}C$ for 18 h. The changes of pH, titratable acidity and viable cell number during fermentation were investigated. The pH and titratable acidity were reached to pH 3.86 and 1.09% after 18 h fermentation, respectively. The viable cell population of Ln. mesenteroides KACC 91495P was rapidly increased to $2.0{\times}10^9\;CFU/g$ during the 9 h of cultivation. The contents of lactic acid, acetic acid and malic acid were determined to be 0.213, 0.259, and 0.217% after 18 h fermentation, respectively. The content of polyphenolic compounds, known as antioxidants, in pear puree were enhanced by Ln. mesenteroides KACC 91495P cultivation. The level of total polyphenolic compounds was increased to around 140% of initial concentration. When the fermented pear puree was kept at $4^{\circ}C$, pH, titratable acidity and number of viable cells population were nearly maintained for 13 days.

Changes in Physicochemical Components of Cocoa Mass during Microwave Roasting (마이크로파 Roasting 중 Cocoa Mass의 이화학적 성분 변화)

  • This study was done to determine the changes in physicochemical components focused on the general compositions, pH, titratable acidity, organic acids, color and sugars during the microwave roasting of cocoa mass and to compare the changes with those during a conventional roasting. The cocoa mass was microwave-roasted at 110 to $150^{circ}C$ and for 10 to 40 min or conventionally roasted at $140^{circ}C$ for 30 min. General compositions of microwave-roasted cocoa mass were not different from those of raw cocoa mass or conventionally-roasted one. The pH changes did not affect the changes in titratable acidity or organic acids. Titratable acidity was different among the three samples. Microwave-roasted cocoa mass showed less sufficient effects on removing organic acids and less changes in color. Fructose, glucose and maltose decreased during a roasting but sucrose showed little changes.

Repeated Fed-Batch Fermentation of Wheat Flour Solution by Mixed Lactic Acid Bacteria (혼합 젖산균을 이용한 밀가루 용액의 반복 유가식 발효)

  • Effect of culture conditions on the fermentation of wheat flour solution by mixed lactic acid bacteria of Lactobacillus brevis, L. fermentum and L. plantarum was investigated. The optimum temperature for the fermentation of wheat flour solution was $35^{\circ}C$ because pH decreased the lowest value and TTA (total titrable acidity) increased the highest value at this temperature. In aerobic condition, fermentor was purged with air at 1.0 vvm and was purged with nitrogen gas at 1.0 vvm in anaerobic condition. The decrease of pH and the increase of TTA in aerobic condition were higher than those in anaerobic condition. In aerobic condition, the optimum condition of oxygen supply was found to be oxygen transfer rate coefficient of $60\;hr^{-1}$ which corresponded to agitation speed of 250 rpm in a 5 L fermentor. Repeated fed-batch cultures were performed using pH-stat in order to increase the productivity of fermented wheat flour. With increasing the repeated fraction of culture volume, mean cycle time increased but maximum operation time decreased. However, the volume of produced broth per culture volume per time and total volume of produced broth per culture volume were maximum at the repeated fraction of culture volume of 20%. In a repeated fed-batch fermentation of wheat flour solution using mixed lactic acid bacteria, the culture condition was optimum at temerature of $35^{\circ}C$, aeration rate of 1.0 vvm, oxygen transfer rate coefficient of $60\;hr^{-1}$, and repeated fraction of culture volume of 20%.

Quality characteristics of Mul-kimchi added with chlorella water extract (클로렐라 추출물을 첨가한 물김치의 품질특성)

  • The quality and microbiological characteristics of various Mul-kimchi prepared with addition of 0.02, 0.05, and 0.1% (w/v) chlorella water extract (CWE) were investigated during fermentation for 7 days at $10^{\circ}C$. The addition of CWE powder stimulated the growth of lactic acid bacteria and considerably improved the acid production. After 6 days fermentation, titratable acidity of CWE Mul-kimchi was 0.16-0.19% and was higher than that (0.14%) of Mul-kimchi made without CWE. The lactic acid bacterial counts were increased rapidly in CWE Mul-kimchi during fermentation for 4 days. The sensory quality of Mul-kimchi added with 0.05-0.1% CWE powder lightly enhanced in taste and overall acceptability among the tested Mul-kimchis preparations. When CWE Mul-kimchi preparations fermented for 6 days were incubated at $4^{\circ}C$ for 15 days, their titratable acidities were continuously increased up to 0.21-0.26% but number of viable lactic acid bacteria were well maintained above $10^7CFU/g$ through storage period.

Correlation between Sensory Quality and Instrumental Quality Attributes in 'Campbell Early' Grape (포도의 관능적 품질과 객관적 품질인자 상호간의 상관성)

  • In order to find correlations between the sensory evaluation and instrumental analysis in 'Campbell Early' grape, grapes were divided into three parts. One-half parts were used for the sensory evaluation and the other half were used as samples for instrumental analysis. Relationship between sensory evaluation and instrumental analysis were studied through correlation study using 48-paired data set obtained during storage. Soluble solid content analysis showed that instrumental quality attributes increased along the passage of storage time with increasing temperature. pH and titratable acidity decreased slightly at lower storage temperatures. Correlations between instrumental quality attributes in grapes were found to be very low. Positive correlation was found between pH and soluble solid content, and negative correlation was found between pH and titratable acidity. The relation of sourness and sweetness to sourness was higher than other sensory evaluation factors, and the shape of the grapes was found to have little relation to their sweetness. Texture ($r^2$=0.890) was found to be the most important factor in preference, followed next by sweetness with sourness ($r^2$=0.860). The correlations between the sensory evaluation and instrumental quality attributes were found to be very low, with overall acceptability given to a relatively high content of soluble solid content. Correlations between pH and titratable acidity in terms of overall acceptability were found to be very low.

Changes in Chemical Components of Cocoa Beans during Microwave Roasting (마이크로웨이브 Roasting 중 Cocoa Bean의 이화학적 성분 변화)

  • The physicochemical properties of microwave-roasted cocoa beans such as color, degree of browning, pH, titrable acidity, organic acid, and sugar content were determined and compared with those of conventionally-roasted beans. Microwave roasting temperatures were 110, 120, 130, 140 and $150^{\circ}C$. Roasting times were 0, 5, 10, 20, and 30 min. Conventional roasting was done at $120^{\circ}C$ and 15 min as a reference. The pH had no relations with roasting temperature or roasting time. As roasting time and roasting temperature were increased, the titrable acidity values of roasted cocoa beans became higher than those of raw bean. But there were no differences among each condition. Color, degree of browning, and sugar content changed as roasting temperature and roasting time increased. The organic acid content of roasted beans increased but the degree of increase was different from organic acid to organic acid. Roasting by microwaves resulted in higher inner temperature of samples than the surface temperature. To increase roasting efficiency, the other heat sources should be used together with microwave.

  • The effects of various nutrients added to soy protein concentrate (SPC)-yogurt on the growth of Lactobacillus acidophilus and the flavor of SPC-yogurt were investigated. Soy milk was prepared from SPC (4.2% as protein) and various nutrients. One hundred ml of the prepared soy milk was given proper heat treatment, inoculated with 2.5ml of 24 hr-old culture, and incubated for 24 hr at $37^{\circ}C$. The growth of L. acidophilus was estimated from viable cell counts, titratable acidity and pH. The growth curve obtained from the experiment suggested that the log phase ended after 6 hr and the stationary phase ended after 30 hr. Glucose and fructose greatly enhanced the acid production by L. acidophilus. The optimum concentration of these two sugars in the media was approximately 3% each. Yeast extract greatly stimulated the acid production by L. acidophilus, and the optimum concentration of this additive was approximately 0.5%. Higher concentration of SPC resulted in slightly higher acidity in soy yogurt.

Quality characteristics of home-made doenjang, a traditional Korean soybean paste (가정에서 제조된 전통된장의 품질특성)

  • To assess the quality characteristics of home-made Doenjang prepared by traditional methods, general components, organic acids, amino acids and fatty acid composition of Doenjang were investigated. The contents of moisture, crude protein, crude lipid, salt and the titratable acidity(as milliliter of 0.1 N NaOH consumed) of Doenjang were 57.3%, 11.6%, 8.3%, 13.9%, and 11.8, respectively. There were wide variation in the contents of crude protein and salt among all samples. The average content of amino type nitrogen was 308.4mg%(w/w), however, it ranged from 202.3 to 416.3 mg% in which the contents some samples were 1.3∼2.1 folds greater than others. Hunter color values of L(lightness), a(redness) and b(yellowness) were 37.3, +19.6, and +17.5, respectively. Browning index value(expressed as optical density at 425 nm) of water-soluble compound(2.23) was more than 5.7 times higher than that of water-insoluble compound(0.39). The contents of free and total amino acids were 2908.9 and 9019.3 mg%, respectively, and the range of free amino acids(1.8∼6.2 times) were much wider than the range of total amino acids(1.2∼2.0 times). Glutamic acid was most abundant in free and total amino acids as 576 and 1,126 mg%, respectively. The main organic acid was lactic acid as 326.8 mg% ranging from 59.4 to 613.4 mg%. Linoleic acid(49.24%) showed the highest content in total fatty acids. Unsaturated fatty acid comprised 81.97% of total fatty acids and polyunsaturated fatty acid ranged from 51.79 to 62.97%.

Development of the Formula for Natural Bread-making Starter (천연제빵 발효 Starter의 개발)

  • A starter formulation was developed to substitute a baker's yeast with natural starter when manufacturing bread products. To develop an active starlet, starter was formulated varying with types of wheat flours, level of water contents and various nutrients. Activities of starter were investigated in terms of viable counts of microbes and change of pH and total titratible acidity Domestic wheat flours contain 100 times more number of lactic acid bacteria than yeast regardless of types of wheat flours. The more protein contents in wheat flours, the more stable microbes in starter. This was considered to be the result of buffering effect of wheat proteins. The optimum level of protein content to ensure the activity of starter was more than 12.0%. Optimum level of water content in active starter was 110% based on strong flour. The more water or the less water had the tendency of decreasing viable counts of microbes. Addition of salt and sucrose had increased the activity of starters. However oligosaccharides did not affect the activity of starter. The optimum concentrations of salt and sucrose were 1.0% and 5.0% respectively. Bread with the starter was higher scored than breads with yeast in terms of all the quality and sensory characteristics except their volumes. In conclusion, a starter formulated with strong flour 100%, water 110%, salt 1% and sucrose 5% was considered to have high potential as a substitute of yeast in making natural bread.

On Chemical Characteristics of Sour Doenjang (Fermented Soybean Paste) (저장 유통중 시어진 된장의 화학적 성분 연구)

  • Doenjang (fermented soybean paste) is one of the korean traditional fermented soybean product which is consumed with cooked rice as a soup or paste. During the fermentation, soybean protein hydrolyzed into amino acids and various peptide, and various organic acids by mirobes related and enzymes produced by meju fermentation. Some commercial products locationally samples give more sour taste than normal due to abnormal fermentation which the reasons are not clear. Three samples that gave sour taste organoleptically were collected and analyzed their characteristics such as pH, moisture content, acidity and microbial counts. The pH of the sour sample were lower than the normal with higher acidity as pH 5.39 (normal) to pH 4.36 (S2) and 15.80 ml of(0.lN NaOH consumed) to 21.80 ml (S1) respectively. Salt and moisture contents were different with sour and normal Doenjang as 16.38% (normal) to 8.92% (S3) in salt and 55.94% (normal) to 49.34% (S1) in moisture content. Total viable counts were $4.1{\times}10^8$ (normal) to $8.0{\times}10^5$ (S2), and $3.4{\times}10^8$ (normal) to $8.0{\times}10^5$ (S2) in acid producing microbes at BCP plate. Yeast and mold were not detected. The composition of acids as mainly lactic acid and acetic acid of sour Doenjang. Total free amino acids content were lower the sour Doenjang than the normal.
