Isolation and Characterization of Cathepsin B inhilbitor Produced by Streptomyces luteogriseus KT-10 (Streptomyces luteogriseus KT-10 이 생산하는 Cathepsin B 저해물질의 분리 및 특성)

  • Isolation and Characterization of Cathepsin B inhibitor Produced by Streptomyces luteogriseus KT-IO. Han, Kil~Hwan and Sang~Dal Kim*. Department of Applied Microbiology, Yeungnam Universit}/t Kyongsan 712749, Korea - The cathepsin B inhibitor produced by Streptomyces luteogriseus KT-IO was very stable in heat, acidic and alkaline conditions. The cathepsin B inhibitor was isolated from the extracted fraction of culture broth with butanol, methanol and chloroform subsequently, the inhibitor was purified with following several column chromatography sLlch as DEAE-Sephadex A-25, Sephadex G-15, silica gel 60, Sephadex LH-20, and preparative HPLC. The cathepsin B inhibitor showed positively to detective reaction of ninhydrine, 5% H2S04, iodine, but negatively to the reaction of Ehrlich's reagent, DNS, aniline. The molecular formular of cathepsin B inhibitor was elucidated by JR, lH and 13C-NMR, FAB mass and elemental analyzer. Consequently, it was identified as C4HlI04N6. The cathepsin B inhibitor had the mode of competitive inhibition with the reaction of cathepsin B.

Allelopathic Inhibition by Extracts and Volatiles from Leaf and Seed of Sicklepod (Cassia tora L.) (결명자(決明子)(Cassia tora L.) 엽(葉)과 종자(種子)의 추출물(抽出物)과 휘발성분(揮發成分)의 타감성(他感性) 생육저해작용(生育沮害作用))

  • The growth of weeds and some other plants has been considered to be inhibited by sicklepod(Cassia tora L.) sharing the habitat. The study was conducted, for the first time, to propose that this phenomenon is not only due to competition for physical and nutritional conditions but also due to allelopathy. In addition, autotoxicity of sicklepod was examined. The results are summarized as follows. 1. Rice sheath length reduced progressively from 18 % to 36 % with increasing the concentration of treating aqueous extracts of sickle pod seeds, but rice germination was not affected. In contrast, radish hypocotyl length was not reduced by the aqueous extract treatment but radish germination was significantly reduced by 66 % at 1 : 10 and 1 : 5 treatment. 2. Total chlorophyll contents in rice seedling decreased from 50 % to 65 % by treatment of seed aqueous extracts diluted from 1 : 50 to 1 : 5 ratio. 3. Aqueous extracts of sicklepod leaves significantly reduced hypocotyl length and fresh weight in radish and germination in rice, but mung bean was slightly affected by aqueous extracts only in fresh weight. 4. Volatiles from fresh, immature seeds with husk reduced the radish germination and seedling growth and radish root growth appeared to be more sensitive to the exposure to volatiles from fresh immature seeds than both germination and hypocotyl growth. 5. Volatiles from sickepod leaves inhibited germination and growth of radish, rice and mung bean, and seedling growth was more sensitive to volatiles from leaves than germination. 6. Volatiles from sicklepod leaves reduced germination and radicle length of sicklepod itself. 7. Collectively, it is concluded that there are water-soluble and volatile substances responsible for allelopathy in sicklepod.

Hyaluronidase Inhibitory Activity of Extracts from Doenjang, Chungkookjang and Miso (된장, 청국장 및 미소추출물의 Hyaluronidase 저해활성)

  • The inhibitory effects of water and methanol extracts of Doenjang, Chungkookjang and Miso on bovine hyaluronidase were examined. The extracts were vacuum-dried and the resulting pellets were dissolved with 0.1 M acetate buffer with 1$\%$ DMSO to a final concentration of 50 mg/mL. The water extracts of Doenjang, Chungkookjang and Miso inhibited 62$\%$, 70$\% $, 56$\%$ of hyaluronidase activity, respectively, while the extract of crushed soybean inhibited 24$\%$ of the activity. Under the same condition, disodiumcromoglycate known as an anti-allergic drug inhibited 46$\%$ of hyaluronidase activity at the concentration of 0.35 mg/mL as a positive control. Also the methanol extracts of Doenjang, Chungkookjang and Miso inhibited 48$\%$, 52$\%$, 70$\%$ of hvaluronidase activity, respectively, while the extract of crushed soybean inhibited 46$\%$ of the activity. After heat treatment of the water extracts of Doenjang, Chungkookjang and Miso at 100"C for 10 min, hyaluronidase inhibitory effects of them were reduced approximately to half. However, hyaluronidase inhibitory effects of methanol extracts were maintained well even after heat treatment.

  • 현재 고해상도의 원격탐사 영상을 이용하기 위해서는 고가의 비용을 부담해야 하고 데이터의 용량도 매우 커서 실제로 사용에는 대부분의 사람들이 매우 소극적이다. 이미 수집된 저해상도의 활발한 활용을 위해서는 값이 저렴하면서도 해상도가 좋은 분광력이나 지상해상도를 높여주어야 한다. 따라서 본 연구에서는 해상도가 각기 다른 영상을 관련 자료들을 근거로 20년 전에 저해상도인 30m의 위성영상과 5m의 고해상도 위성사진과의 합성을 통하여 저해상도에서 판독할 수 없었던 여러 종류의 지형지물을 파악할 수 있는 고해상도의 퓨젼 영상을 생성시킨 것이다.

  • Casein을 인공 소화관 내의 조건으로 처리한 가수분해물의 ACE 저해활성 측정 및 이를 유제품에 적용하기 위한 기초자료 수집을 위해 관능검사를 실시하였다. Casein을 인공위액과 인공장액으로 각각 단독으로 처리했을 때의 ACE 저해 활성보다는 인공 위액과 인공 장액의 연속처리를 받은 가수분해 분해물의 ACE저해 활성이 더 높게 나타났다. 또한 가수분해물의 관능검사를 실시한 결과에서는 백색 시유에는 0.3% 정도의 첨가가 적당하였으며, 초코우유나 커피우유에 첨가할 경우에는 0.5% 정도까지 첨가하여도 맛에 크게 변화를 주지 않았다.

Production of Anti-dementia Acetylcholinesterase Inhibitor from Pleurotus ostreatus (Heuktari) and Inhibitory Effect on PC12 Neuron Apoptosis (흑타리버섯으로부터 항치매성 Acetylcholinesterase 저해물질의 생산 및 PC12 신경세포사 저해 효과)

  • To develop a new antidementia acetycholinesterase (AChE) inhibitor from edible mushrooms, the inhibitory effects on AChE of water and ethanol extracts from various edible mushrooms were measured. Among the tested compounds, 70% ethanol extracts from Tremella fuciformis showed the highest AChE inhibitory activity, at 25.3% (IC50: 9.9 mg). Water extracts from the fruiting body of Pleurotus ostreatus (Heuktari) showed AChE inhibitory activity of 20.2% (IC50: 12.4 mg). However, the yield (40.8%) from Pleurotus ostreatus (Heuktari) was higher than that from Tremella fuciformis (5.0%). Therefore, we selected Pleurotus ostreatus (Heuktari) as the most promising candidate for a mushroom containing anti-dementia AChE inhibitors. The AChE inhibitor from Pleurotus ostreatus (Heuktari) was optimally extracted when its fruiting body was treated with water for 6 h at 30℃. The anti-dementia effects of the partially purified AChE inhibitor from Pleurotus ostreatus (Heuktari) were observed in PC12 nerve cells.

Purification and Characteristics of Tyrosinase Inhibitor Produced by Actinomycetes F-97 (방선균 F-97이 생산하는 Tyrosinase 저해제의 정제 및 특징)

  • An actinomycetes F-97 producing tyrosinase inhibitor was isolated from soil samples. Isolation and purification of tyrosinase inhibitor produced by F-97 was performed as follows: IRC-120 ($NH_4^+$ type) column chromatography, silica gel column chromatography, $C_{18}$ column chromatography and Sephadex LH-20 column chromatography were used successively after the centrifuged supernatant was adjusted to pH 4.0. To identify the purity of the inhibitor, octadecylsilyl(ODS) HPLC was carried out with 5% methanol as a mobile phase. Finally, the purification yield of a tyrosinase inhibitor was 5.24%. The inhibitor was very soluble in water, methanol and ethanol but insoluble in acetone, butanol, ethylacetate and chloroform. The ${\lambda}_{max}$ value of this inhibitor in water was 194nm under UV light. The biochemical test of the inhibitor was positive in Molish, Benedict, cone. $H_2SO_4$, and $KMnO_4$ tests but negative in iodine, ninhydrin, Million, Sakaguchi, xanthoproteic and Emerson tests. The tyrosinase inhibitor was stable against heat treatment of $100^{\circ}C$ for 50 minutes and pH $4{\sim}9$. The $IC_{50}$ value of this inhibitor was $19.2{\mu}g/ml$ for mushroom tyrosinase. In $1,000{\mu}g/ml$ inhibitor concentration, inhibition zone was 27 mm for Streptomyces bikiniensis NRRL B-1049. The inhibition of F-97 against mushroom tyrosinase was competitive with tyrosine.

Inhibition Characteristics of Chlorsulfuron and Imazaquin on Acetolactate Synthase Activity of Corn Plants (Chlorsulfuron 및 Imazaquin에 의한 옥수수 Acetolactate Synthase 활성의 저해특성)

  • The inhibition characteristics of chlorsulfuron [CHL, 2-chloro-N-[{ (4-methoxy-6-methyl-1,3,5-triazin-2-yl)amino}carbonyl]benzenesulfonamide] and imazaquin [IMA, 2-{4,5-dihydro-4-methyl-4-(1-methy-lethyl)-5-oxo-1H-imidazol-2-yl}-3-quinolinecarboxylic acid] on acetolactate synthase(ALS) activity of corn plants were investigated. CHL and IMA rapidly inhibited ALS activity of corn plants in vitro. Their $I_{50}$ values for ALS activity were 100nM and $5{\mu}M$, respectively, indicating that CHL had 50 times more inhibitory effect on ALS activity than IMA. The first applied herbicide had a dominant inhibitory effect on ALS activity when the two herbicides were applied sequentially. Branched-chain amino acids, valine(Val), leucine(Leu), and isoleucine(Ile) showed a feedback inhibition on ALS activity ; Val or Leu had a more inhibitory effect on ALS activity than Ile. Branchedchain amino acids and CHL or IMA exhibited an additive effect on inhibiting ALS activity. This suggests that branched-chain amino acids inhibit ALS activity by a different mechanisms) from that of CHL or IMA. Apparent ALS activity, which was measured on the basis of the conversion of pyruvate to acetolactate, was decreased by the addition of 2-ketobutyrate into the ALS reaction mixture in a concentration-dependent manner. In addition, kinetic studies revealed that CHL acts as a noncompetitive inhibitor, while IMA acts as an uncompetitive inhibitor to ALS with respect to pyruvate.

Inhibitory Effects of Copper on the Anaerobic Degradation of Propionate (프로피온산의 혐기성 분해시 구리의 저해 효과)

  • The effects of copper on the anaerobic degradation of propionate were studied using anaerobic batch reactors. The apparent inhibitory effects of copper on the anaerobic degradation of propionate could be observed from behaviors of intermediates, ultimate methane yield(UMY) and specific methanogenic activity(SMA) There was little inhibition at the concentration of $2.5mg\;Cu^{2+}/L$. Beyond this concentration, the inhibitory effects increased with increasing dose of coppers. The 50% inhibition of UMY and SMA occurred at copper dosage of 33.8 and $24.1mg\;Cu^{2+}/gVSS$, respectively. The inhibitory effect based on the UMY was gradually reduced with the operation time dueprobably to the acclimation of microorganisms and/or binding of the added copper by ligands(and possibly ion exchange sites)contained on the cell membrane and extracellular polymer matrix whereas it based on the SMA might exclude the this phenomena. Therefore, the methodology for interpretation of inhibition data based on the SMA was more accurated than the UMY. There was no inhibitory effect in batch reactors supplemented with sulfate due to an antagonistic action of the sulfate reducing bacteria. Propionate degradation was initially retarded for copper inhibited samples but it gradually degraded afterward. Based on the mass removal considering take into account the propionate to acetate conversion, propionate degradation may appeal more affected than acetate. This result revealed that the hydrogenotrophic methanogens were the most affected by copper.

Screening of Tyrosinase Inhibitor from Plants (Tyrosinase 활성을 저해하는 식물체의 탐색)

  • In order to screen natural inhibitor of tyrosinase which catalyzes an enzymatic browning of some foods and in vivo synthesis of melanin, inhibitory effect of 129 edible plants and 15 chemical compounds on the in vivo melanin synthesis by mushroom tyrosinase was analyzed. Among leafy vegetables tested, radish bud, red chicory, Shepherd's purse and small green onion were found to have more than 50% tyrosinase inhibition effect in the descending order. Chinese radish and garlic in root vegetables, and nameko, shiitake and oyster mushroom in mushrooms, and teas showed also more than 50% inhibition effect. Among fruit vegetables tested, red pepper, Chinese quince and avocado were found to have more than 50% tyrosinase inhibition effect, while fruits generally showed low inhibitory effect. Medicinal plants which inhibit tyrosinase more than 50% were mume fructus>cinamomi ramulus>rubi fructus>mori cortex>biotae orientalis folium>puerariae radix, and herbs with more than 50% inhibitory effect were allspice>clove>mustard. In some chemical compounds tested, 4-hexylresorcinol, L-cysteine, glutathione, sodium bisulfite and kojic acid showed powerful inhibition effect on mushroom tyrosinase.
